1Seamless and early gap healing of osteochondral defects by autologous mosaicplasty combined with bioactive supramolecular nanofiber-enabled gelatin methacryloyl(BSN-GelMA)hydrogel显示文摘Autologous mosaicplasty is a common approach used to treat osteochondral defects in clinical practice.Gap integration between host and transplanted plugs requires bone tissue reservation and hyaline cartilage regeneration without uneven surface,graft necrosis and sclerosis.However,poor gap integration is a serious concern,which eventually leads to deterioration of joint function.To deal with such complications,this study has developed a strategy to effectively enhance integration of the gap region following mosaicplasty by applying injectable bioactive supramolecular nanofiber-enabled gelatin methacryloyl(GelMA)hydrogel(BSN-GelMA).A rabbit osteochondral defect model demonstrated that BSN-GelMA achieved seamless osteochondral healing in the gap region between plugs of osteochondral defects following mosaicplasty,as early as six weeks.Moreover,the International Cartilage Repair Society score,histology score,glycosaminoglycan content,subchondral bone volume,and collagen II expression were observed to be the highest in the gap region of BSN-GelMA treated group.This improved outcome was due to bio-interactive materials,which acted as tissue fillers to bridge the gap,prevent cartilage degeneration,and promote graft survival and migration of bone marrow mesenchymal stem cells by releasing bioactive supramolecular nanofibers from the GelMA hydrogel.This study provides a powerful and applicable approach to improve gap integration after autologous mosaicplasty.It is also a promising off-the-shelf bioactive material for cell-free in situ tissue regeneration.Hongwei Wu Yuna Shang Wei Sun Xinyi Ouyang Wenyan Zhou Jieji Lu Shuhui Yang Wei Wei Xudong Yao g Xiaozhao Wang Xianzhu Zhang Yishan Chen Qiulin He Zhimou Yang Hongwei Ouyang 2023Bioactive Materials2023,,1:1
2PROTACs:great opportunities for academia and industry (an update from 2020 to 2021)显示文摘PROteolysis TArgeting Chimeras(PROTACs)technology is a new protein-degradation strategy that has emerged in recent years.It uses bifunctional small molecules to induce the ubiquitination and degradation of target proteins through the ubiquitin–proteasome system.PROTACs can not only be used as potential clinical treatments for diseases such as cancer,immune disorders,viral infections,and neurodegenerative diseases,but also provide unique chemical knockdown tools for biological research in a catalytic,reversible,and rapid manner.In 2019,our group published a review article“PROTACs:great opportunities for academia and industry”in the journal,summarizing the representative compounds of PROTACs reported before the end of 2019.In the past 2 years,the entire field of protein degradation has experienced rapid development,including not only a large increase in the number of research papers on protein-degradation technology but also a rapid increase in the number of small-molecule degraders that have entered the clinical and will enter the clinical stage.In addition to PROTAC and molecular glue technology,other new degradation technologies are also developing rapidly.In this article,we mainly summarize and review the representative PROTACs of related targets published in 2020–2021 to present to researchers the exciting developments in the field of protein degradation.The problems that need to be solved in this field will also be briefly introduced.Ming He Chaoguo Cao Zhihao Ni Yongbo Liu Peilu Song Shuang Hao Yuna He Xiuyun Sun Yu Rao 2022Signal Transduction and Targeted Therapy2022,7,7:1

5Computed tomography-guided percutaneous radiofrequency thermocoagulation for primary trigeminal neuralgia in older and younger patients显示文摘We evaluated the immediate and long-term clinical efficacy of computed tomography (CT)-guided radiofrequency thermocoagulation for primary trigeminal neuralgia (RTPTN) in 852 patients includ-ing 502 patients aged ≥ 60 years and 350 patients aged < 60 years.After discharge,the incidence of complications was 1.0% and 0.9% in patients aged ≥ 60 years and patients aged < 60 years,respectively.Over 3-year follow-up after CT-guided RTPTN,96.8% of the patients aged ≥ 60 years and 98.6% of the patients aged < 60 years were completely pain-free,and there was no significant difference between these two age brackets.In addition,there were no significant differences in quality of life scores and numbness scores between these two age brackets.These findings suggest that CT-guided RTPTN is a safe and effective method and is recommended for older and poor-risk patients.Guanghui Lai Jiaxiang Ni Baishan Wu Mingwei He Liqiang Yang Jianning Yue Yuna Guo 2011Neural Regeneration Research2011,6,24:0
