1Effects of GBE50 on hippocampal CA1 synaptic plasticity,learning and memory in an experimental rat model of aging显示文摘The content of total flavonoids in an extract of Ginkgo biloba,called GBE50,is 44% by weight.This is significantly greater than that in a standard extract of Ginkgo biloba,designated EGB761.To date,the mechanisms by which GBE50 and EGB761 function remain poorly understood.In the present study,an experimental rat model of aging was induced by intraperitoneal injection of D-galactose,followed by intragastric perfusion of GBE50 (30,60mg/kg),or EGB761 (60mg/kg).The water maze scores and hippocampal CA1 synaptic plasticity were evaluated.In the place navigation test,the GBE50 group rats did better than EGB761,while similar scores were obtained in the spatial probe test,and in the platform-switched test.In addition,long-term potentiation was significantly enhanced following high-frequency stimulation in the GBE50 and EGB761 groups,compared with the model group.These results demonstrate that GBE50 and EGB761 improved the learning and memory of aging rats.In particular,GBE50 administered at the 60mg/kg dose exhibited superior effects over EGB761 at the same 60mg/kg dose.Furthermore,the enhancement of hippocampal synaptic plasticity may be an underlying mechanism.Lili Wu Xianwen Dong Gaiying He Zhixiong Zhang Ying Xu Xingyu Wang Yun Li 2011Neural Regeneration Research2011,6,12:9
2Design and experiment of variable rate orchard sprayer based on laser scanning sensor显示文摘During different growth periods,canopy size and density in orchards are variable,which need application conditions(flow rate and air flow)to be adjusted to match the canopy’s characteristics.In order to improve orchard sprayer’s automatic operating performance,an automatic variable-rate orchard sprayer(VARS)fixed with 40 electromagnetic valves and 8 brushless fans was developed based on the canopy’s spatial dimensions.Each solenoid valve and brushless motor can be individually adjusted in real-time through pulse width modulation(PWM)signals emitted by a control system to adjust each nozzle’s spout and fan rotation speed.A high-precision laser scanning sensor(light detecting and ranging,LIDAR)was adopted as the detector to measure the canopy volume using the variable rate algorithm principle.Field experiments were conducted in an apple orchard,and conventional air blast sprayer(CABS)and directed air-jet sprayer(DAJS)were tested as a comparison.Results showed that on average,46%less spraying solution was applied compared to conventional applications,while penetration rate was similar to DAJS.Normalized deposition in the canopy with variable application was higher than that of conventional applications,indicating that electronic sprayers are more efficient than conventional sprayers.It was also observed that VARS could significantly reduce off-target loss.The field experiment showed that the newly developed variable-rate sprayer can greatly reduce pesticide use and protect the environment for the orchard fruit production,and also provide a reference for design and performance optimization for plant protection machinery.Longlong Li Xiongkui He Jianli Song Yajia Liu Aijun Zeng Liu Yang Chaohui Liu Zhixiong Liu 2018International Journal of Agricultural and Biological Engineering2018,11,1:8
3Identification and characterization of rice blast resistance gene Pid4 by a combination of transcriptomic profiling and genome analysis显示文摘Map-based cloning of plant disease resistance (R) genes is time-consuming. Here, we reported the isolation of blast R gene Pid4 using comparative transcriptomic profiling and genome-wide sequence analysis. Pid4 encodes a coiled-coil nucleotide-binding site leucine-rich repeat(CC-NBS-LRR) protein and is constitutively expressed at diverse developmental stages in the rice variety Digu. The Pid4 protein is localized in both the nucleus and cytoplasm. Introduction of Pid4 into susceptible rice cultivars confers race-specific resistance to leaf and neck blast. Amino acid sequence comparison and blast resistance spectrum tests showed that Pid4 is a novel R gene, different from the previously reported R genes located in the same gene cluster. A Pid4 Indel marker was developed to facilitate the identification of Pid4 in different rice varieties. 4Regulation of non-classical immune parameters in immune thrombocytopenic purpura mice by a spleen-invigorating,qi-replenishing and blood-containing formula显示文摘Objective:This study investigated the regulatory effect of non-classical immune parameters on immune thrombocytopenic purpura(ITP)mice by a spleen-invigorating,qi-replenishing and blood-containing formula(SQBF).Method:A total of 80 BALB/c mice were randomly divided into four equal groups(20 mice each):control group,model group,prednisone group and spleen-invigorating,qi-replenishing and blood-containing(SQBF)group.Mice in the model group,prednisone group,and SQBF group were administered anti-platelet serum to induce ITP.The dynamic variations of platelet counts in ITP mice were measured with an automatic blood analyzer before modeling and 48 h,and 8,12 and 15 days following APS injection.Levels of b-endorphin(b-EP),vasoactive intestinal peptide(VIP)and salivary IgA(SIgA)were detected by enzyme-linked immunosorbent assay(ELISA)on 15th day of experiment.Results:SQBF enhanced peripheral blood platelet counts in ITP mice similar to that of prednisone,and both groups showed a statistically significant response compared with the model group(P<.01).The SQBF significantly decreased b-EP levels compared with the model and prednisone intervention groups(P<.05),significantly increased the levels of VIP and SIgA in ITP mice compared with the model group(P<.05)and had significant protective effects on the thymus of ITP mice compared with the model group(P<.01).Conclusions:The SQBF had a similar effect to prednisone with regards to enhancing peripheral blood platelet counts in ITP mice.Furthermore,it decreased b-EP levels and increased VIP and SIgA,and protected the thymus.This shows that,on base of the brain-gut axis functions,some non-classical immune vascular active factors or neurotransmitters are also involved in immune responses,and also have relationship with the onset of ITP and bleeding and/or hemostasis.It needs further study to determine whether a change in these active factors is related to immediate hemostasis.Tiantian Li Hao He Li Hou Yue Xu Xiaoyong Wu Yanping Sun Lei Zheng Zhixiong Chen Xinyi Chen Bei Qin 2015Journal of Traditional Chinese Medical Sciences2015,2,2:6

6Rumen-protected glucose supplementation in transition dairy cows shifts fermentation patterns and enhances mucosal immunity显示文摘Manipulation of perinatal diets,such as supplementing feed with rumen-protected glucose(RPG),has been positively regarded as a strategy to improve milking performance.This study was conducted to assess the effects of RPG on the fermentation profiles,resident microbiota and mucosal immunity in the cecum.Ten Holstein dairy cows were randomly assigned to either a 25 g/kg RPG diet(DM basis)or a 11 g/kg coating fat diet(control,CON).Compared with the CON group,the acetate-to-propionate ratio was lower in the RPG group.Gene expression analysis indicated that RPG supplementation tended to upregulate the expression of Na^(+)/H^(+)hydrogen exchanger 3(NHE3)(P=0.076).RPG supplementation downregulated the expression of genes involved in self-rehabilitation such as matrix metalloproteinase 1(MMP1),MMP3,MMP9 and MMP13.Additionally,the mRNA expression of genes involved in immunity including Toll-like receptors(TLR4,TLR6 and TLR7)and proinflammatory cytokines(immune interferon gamma[IFNG]and interleukins interleukin 17 A[IL7 F],IL17 A,IL22),was downregulated by RPG supplementation.Nonetheless,no differences existed in the bacterial copy number and beta diversity between the 2 groups.Overall,supplementation with RPG would probably cause a shift towards propionate production in the cecal digesta,and promote the immune homeostasis of the cecal mucosa in transition dairy cows.Our results extended the basic understanding of RPG supplementation and utilization in transition dairy cows in terms of host microbe interplay in the cecum.Xiaoli Zhang Xiaopeng Li Jian Wu Jinzhen Jiao Zhixiong He Zhiliang Tan Xuefeng Han 2021Animal Nutrition2021,,4:4
7Profiling of hepatocellular carcinoma neoantigens reveals immune microenvironment and clonal evolution related patterns显示文摘Objective: Neoantigens derived from tumor-specific genomic alterations have demonstrated great potential for immunotherapeutic interventions in cancers. However, the comprehensive profile of hepatocellular carcinoma(HCC) neoantigens and their complex interplay with immune microenvironment and tumor evolution have not been fully addressed.Methods: Here we integrated whole exome sequencing data, transcriptome sequencing data and clinical information of 72 primary HCC patients to characterize the HCC neoantigen profile, and systematically explored its interactions with tumor clonal evolution, driver mutations and immune microenvironments.Results: We observed that higher somatic mutation/neoantigen load was associated with better clinical outcomes and HCC patients could be further divided into two subgroups with distinct prognosis based on their neoantigen expression patterns. HCC subgroup with neoantigen expression probability high(NEP-H) showed more aggressive pathologic features including increased incidence of tumor thrombus(P=0.038), higher recurrence rate(P=0.029),more inclined to lack tumor capsule(P=0.026) and with more microsatellite instability sites(P=0.006). In addition,NEP-H subgroup was also characterized by higher chance to be involved in tumor clonal evolution [odds ratio(OR)=46.7, P<0.001]. Gene set enrichment analysis revealed that upregulation of MYC and its targets could suppress immune responses, leading to elevated neoantigen expression proportion in tumor cells. Furthermore, we discovered an immune escape mechanism that tumors could become more inconspicuous by evolving subclones with less immunogenicity. We observed that smaller clonal mutation clusters with higher immunogenicity in tumor were more likely to involve in clonal evolution. 8Maternal intake restriction programs the energy metabolism, clock circadian regulator and m TOR signals in the skeletal muscles of goat offspring probably via the protein kinase A-cAMP-responsive element-binding proteins pathway显示文摘The biological mechanism by which maternal undernutrition increases the metabolic disorder risk of skeletal muscles in offspring is not fully understood.We hypothesize that maternal intake restriction influences metabolic signals in the skeletal muscles of offspring via a glucagon-mediated pathway.Twentyfour pregnant goats were assigned to the control group(100%of the nutrients requirement,n=12)and restricted group(60%of the control feed allowance from pregnant days 45 to 100,n=12).Blood and Longissimus thoracis muscle were sampled from dams(100 d of gestation),fetuses(100 d of gestation),and kids(90 d after birth)in each group.The data were analyzed using the linear MIXED model,with the multiple comparison method of SIDAK applied.Intake restriction reduced(P<0.05)the total blood protein of dams and fetuses.Maternal restriction decreased(P<0.05)the cAMP-responsive element-binding protein 1(CREB1),CREB-binding protein(CREBBP),protein kinase A(PKA),aryl hydrocarbon receptor nuclear translocator-like protein 1(BMAL1),protein kinase B(AKT1),mammalian target of rapamycin(mTOR),and regulatory-associated protein of mTOR(RPTOR)mRNA expression in the fetuses,and reduced(P<0.05)the CREBBP,nuclear receptor subfamily 1 group H member 3(NR1 H3),D-box binding PAR bZIP transcription factor(DBP)and PKA mRNA levels in the kids,but increased(P<0.05)the peroxisome proliferator-activated receptor gamma coactivator 1 alpha(PGC1 A)and tuberous sclerosis 2(TSC2)mRNA levels in the fetuses.The mRNA expression of clock circadian regulator(CLOCK)and TSC2 genes was increased(P<0.05)in the restricted kids.The protein expression of total PKA and phosphorylated PKA in the restricted fetuses and kids were downregulated(P<0.05),and the protein expression of total mTOR and phosphorylated mTOR were reduced(P<0.05)in the restricted fetuses and kids.Maternal intake restriction regulated fat oxidation,protein synthesis,and circadian clock expression in the muscles of the offspring probably via the glucagon-mediated PKA-CREB pathway,which reveals a noteworthy molecular pathway that maternal undernutrition leads to metabolic adaptation of skeletal muscle in offspring.Xiaoling Zhou Qiongxian Yan Hong Yang Ao Ren Zhixiong He Zhiliang Tan 2021Animal Nutrition2021,,4:2
9A mathematical model to describe the diurnal pattern of enteric methane emissions from non-lactating dairy cows post-feeding显示文摘Enteric methane emission is not only a source of energy loss in ruminants, but also a potent contributor to greenhouse gas production. To identify the nature and timing of interventions to reduce methane emissions requires knowledge of temporal kinetics of methane emissions during animal husbandry.Accordingly, a mathematical model was developed to investigate the pattern of enteric methane emissions after feeding in dairy cows. The model facilitated estimation of total enteric methane emissions(V,g) produced by the residual substrate(V1, g) and newly ingested feed(V2, g). The model was fitted to the10 h methane emission patterns after morning feeding of 16 non-lactating dairy cows with various body weights(BW), and the obtained parameters were used to predict the kinetics of 24 h methane emission for each animal. The rate of methane emission(g/h) reached a maximum within 1 to 2 h after feeding,followed by a gradual post-prandial decline to a basal value before the next feeding. The model satisfactorily fitted curves for each cow according to the criterion of goodness-of-fit, and provided biological descriptions for fluctuations in methane emissions based on basal V1 and feeding V2 in response to the changes in BW and dry matter intake(DMI) of different dairy cows. The basal V1 and feeding V2 are probably maintained by slow-and readily-degradable substrates, respectively. The former contributed at least 0.6 of methane production. 13Deletion of a Dna K protein gene causes seedling green-revertible albino by retarding chloroplast development in rice显示文摘Green-revertible albino mutants are important sources for studying chloroplast structure, chloroplast development, chlorophyll biosynthesis, and plant photosynthesis. In the present study, we characterized a greenrevertible albino mutant gra(k), which was obtained from the tissue-cultured rice Kitaake. The mutant gra(k) exhibited albino on its first three leaves. The leaf color started to turn green at the four-leaf stage. The chlorophyll contents were deeply reduced at the seedling stage, and the chloroplast development was delayed in gra(k). The greenrevertible albino(gra) phenotype of the mutant gra(k) was temperature dependent. The main agronomic traits,including plant height, tilling number per plant, seed setting rate, and thousand-grain weight, slightly decreased in gra(k) comparing to those in the wild-type Kitaake.Genetic analysis showed that the gra phenotype was controlled by a single recessive nucleic gene. By using 5,168 recessive F2individuals derived from the cross of gra(k) 9 Jodan, the locus of the gene Gra(k) was delimited in a DNA region of 200 kb between the makers B-31 and P11 on chromosome 5. Sequencing analysis indicated thatthe three functionally annotated genes, LOC_Os05g23700,LOC_Os05g23720, and LOC_Os05g23740, were all deleted in the 200 kb region in the mutant gra(k). Transgenic test revealed that the gra(k) plants over-expressing LOC_Os05g23740CDS were restored to normal green as the wild-type Kitaake. 15Lipid metabolism and m^(6)A RNA methylation are altered in lambs supplemented rumen-protected methionine and lysine in a low-protein diet显示文摘Background:Methionine or lysine has been reported to influence DNA methylation and fat metabolism,but their combined effects in N6-methyl-adenosine(m^(6)A)RNA methylation remain unclarified.The combined effects of rumen-protected methionine and lysine(RML)in a low-protein(LP)diet on lipid metabolism,m^(6)A RNA methylation,and fatty acid(FA)profiles in the liver and muscle of lambs were investigated.Sixty-three male lambs were divided into three treatment groups,three pens per group and seven lambs per pen.The lambs were fed a 14.5%crude protein(CP)diet(adequate protein[NP]),12.5%CP diet(LP),and a LP diet plus RML(LP+RML)for 60 d.Results:The results showed that the addition of RML in a LP diet tended to lower the concentrations of plasma leptin(P=0.07),triglyceride(P=0.05),and non-esterified FA(P=0.08).Feeding a LP diet increased the enzyme activity or m RNA expression of lipogenic enzymes and decreased lipolytic enzymes compared with the NP diet.This effect was reversed by supplementation of RML with a LP diet.The inclusion of RML in a LP diet affected the polyunsaturated fatty acids(PUFA),n-3 PUFA,and n-6 PUFA in the liver but not in the muscle,which might be linked with altered expression of FA desaturase-1(FADS1)and acetyl-Co A carboxylase(ACC).A LP diet supplemented with RML increased(P<0.05)total m^(6)A levels in the liver and muscle and were accompanied by decreased expression of fat mass and obesity-associated protein(FTO)and alk B homologue 5(ALKBH5).The m RNA expressions of methyltransferase-like 3(METTL3)and methyltransferase-like 14(METTL14)in the LP+RML diet group were lower than those in the other two groups.Supplementation of RML with a LP diet affected only liver YTH domain family(YTHDF2)proteins(P<0.05)and muscle YTHDF3(P=0.09),which can be explained by limited m^(6)Abinding proteins that were mediated in m RNA fate.Conclusions:Our findings showed that the inclusion of RML in a LP diet could alter fat deposition through modulations of lipogenesis and lipolysis in the liver and muscle.These changes in fat metabolism may be associated with the modification of m^(6)A RNA methylation.Kefyalew Gebeyew Chao Yang Hui Mi Yan Cheng Tianxi Zhang Fan Hu Qiongxian Yan Zhixiong He Shaoxun Tang Zhiliang Tan 2023Journal of Animal Science and Biotechnology2023,14,1:0

17Clinical and pathological features of intracranial meningiomas in children:A retrospective analysis显示文摘BACKGROUND: The attack of meningiomas has sex and age differences. Previous studies indicated that children and adult patients had different clinical and pathological features. Due to the limitation of conditions and technique of diagnosis and treatment, some clinical and pathological features of meningiomas in children should be further recognized. OBJECTIVE: To summarize the clinical and pathological features in 15 children with intracranial meningiomas. DESIGN: A retrospective case analysis. SETTING: The hospitals which the first author had been working in. PARTICIPANTS: Fifteen children with intracranial meningiomas were selected from the hospitals the first author had been working in from June 1996 to June 2006. There were 9 boys and 6 girls, aged 1.8-15 years, with an average of 9.8 years. The duration from attack to diagnosis was 1 month to 2.5 years, with an average of 13 months. Inclusive criteria: ① Clinically diagnosed, operated and confirmed pathologically; ② ≤ 15 years; ③ The children and their relatives were all informed and agree with the detection. METHODS: The clinical manifestations, imaging data, preoperative diagnosis and misdiagnosis, site and size of tumor, operative treatment and pathological data were retrospectively summarized. Thirteen of 15 children with intracranial meningiomas were followed up by means of reexamination for 1-8 years, and the postoperative recurrence were observed. MAIN OUTCOME MEASURES: Clinical manifestations, imaging data, preoperative diagnosis and misdiagnosis, site and size of tumor, operative treatment, pathological data, recurrence conditions of 1 to 8 years follow-up. RESULTS: Thirteen of the 15 children with intracranial meningiomas were involved in the analysis of results, and 2 lost to the follow-up. ① The main clinical manifestations at diagnosis were headache in 11 cases (73%, 11/15), vomiting in 8 cases (53%, 8/15), papilledema in 9 cases (60%, 9/15), hypopsia in 6 cases (40%, 6/15), epilepsy in 5 cases (33%, 5/15), hemiplegia in 4 cases (27%, 4/15), cranial nerve damage in 4 cases (27%, 4/15), instability of gait and incoordination in 2 cases, conscious disturbance in 3 cases, nystagmus in 2 cases and lump under scalp in 1 case. ② Imaging examination: Most of tumor CT plain scans were slightly high-density imaging, most enhanced scanning appeared well-distributed enhancement and clear boundary. MRI examination observed the tumor appeared equal or long T1 or longer T2 abnormal mixed singles with clear boundary. Two cases had cystic lesion of tumor and 8 cases had peritumor edema. Digital substraction angiography (DSA) observed that there obvious tumor staining and displacement of corresponding brain vessels due to compression in 2 cases. ③ Preoperative misdiagnosis: Six children (40%, 6/15) were misdiagnosed to have other diseases due to the untypical symptoms, including diseases of gastrointestinal tract in 3 cases, eye diseases in 2 cases, and encephalitis in 1 case. Most cases were diagnosed after CT or MRI scanning. ④ Sites and sizes of tumors: Tumor located at supratentorial parts in 13 cases (87%, 13/15), including cerebral convexity in 7 cases (46.7%, 7/15), intraventricle in 2 cases (13.3%, 2/15), cerebral parenchyma in 2 cases (13.3%, 2/15), parafalx and parasagittal sinus in 2 cases respectively. Tumor located at infratentorial part in 2 cases (13.3%, 2/15), which at cerebellar hemisphere and cerebellopontine angle respectively. The maximal diameter of tumor was 1.3-8.0 cm with an average of 4.6 cm. ⑤ Pathological types: The pathological examination reported that the meningioma was benign in 11 cases (73.3%, 11/15) and malignant in 4 cases (26.7%, 4/15). Among the benign meningiomas, it was syncytial in 5 cases, fibrous in 4 cases, and transitional in 2 cases. ⑥ Treatments: All the menigiomas of children were resected, and 14 cases (93.3%, 14/15) had the tumor resected totally (Simpson grades Ⅰ-Ⅲ), including Simpson grade Ⅰ resection in 7 cases, grade Ⅱ in 4 cases, and grade Ⅲ in 3 cases. One case only underwent subtotal resection (Simpson Ⅳ), this case had invasive medial sphenoid ridge-diaphragma sellae meningiomas, the tumor grew towards cavernous sinus and anterior skull base, and wrapped ipsilateral internal carotid artery and optic nerve. No one died postoperatively. ⑦ Follow up and recurrence: Of the 15 children with meningiomas, 13 cases were followed up, 4 cases (31%, 4/13) recurred, including 1 case recurred at 1 year, which was malignant meningioma, another 3 cases recurred at 3.5, 5.5 and 7 years after operation respectively, which were benign meningiomas. CONCLUSION: Meningiomas in children, most of which are benign, mostly locate at supratentorial part, have higher incidence rate in boys than in girls, higher recurrence rate, untypical symptoms, and easy to be misdiagnosed.
