| 4 | Improving CLM4.5 Simulations of Land–Atmosphere Exchange during Freeze–Thaw Processes on the Tibetan Plateau显示文摘Soil is heterogeneous and has different thermal and hydraulic properties, causing varied behavior in heat and moisture transport. Therefore, soil has an important effect on land–atmosphere interactions. In this study, an improved soil parameterization scheme that considers gravel and organic matter in the soil was introduced into CLM4.5(Community Land Model). By using data from the Zoige and Madoi sites on the Tibetan Plateau, the ability of the model to simultaneously simulate the duration of freeze–thaw periods, soil temperature, soil moisture, and surface energy during freeze–thaw processes, was validated. The results indicated that:(1) the new parameterization performed better in simulating the duration of the frozen, thawing, unfrozen, and freezing periods;(2) with the new scheme, the soil thermal conductivity values were decreased;(3) the new parameterization improved soil temperature simulation and effectively decreased cold biases;(4) the new parameterization scheme effectively decreased the dry biases of soil liquid water content during the freezing, completely frozen, and thawing periods, but increased the wet biases during the completely thawed period; and(5) the net radiation, latent heat flux, and soil surface heat flux of the Zoige and Madoi sites were much improved by the new organic matter and thermal conductivity parameterization. | Siqiong LUO Xuewei FANG Shihua LYU Yu ZHANG Boli CHEN | 2017 | Journal of Meteorological Research2017,31,5: | 13 |

| 6 | Prognostic significance of lymphovascular infiltration in overall survival of gastric cancer patients after surgery with curative intent显示文摘Objective: Lymphovascular infiltration(LVI) is frequently detected in gastric cancer(GC) specimens. Studies have revealed that GC patients with LVI have a poorer prognosis than those without LVI.Methods: In total, 1,007 patients with curatively resected GC at Department of Gastric Cancer, Tianjin Medical University Cancer Institute and Hospital were retrospectively enrolled. The patients were categorized into two groups based on the LVI status: a positive group(PG;presence of LVI) and a negative group(NG;absence of LVI). The clinicopathological factors corrected with LVI and prognostic variables were analyzed. Additionally, a pathological lymphovascular-node(lvN) classification system was proposed to evaluate the superiority of its prognostic prediction of GC patients compared with that of the eighth edition of the N staging system.Results: Two hundred twenty-four patients(22.2%) had LVI. The depth of invasion and lymph node metastasis were independently associated with the presence of LVI. GC patients with LVI demonstrated a significantly lower overall survival(OS) rate than those without LVI(42.8% vs. 68.9%, respectively;P<0.001). In multivariate analysis,LVI was identified as an independent prognostic factor for GC patients(hazard ratio: 1.370;95% confidence interval: 1.094-1.717;P=0.006). Using strata analysis, significant prognostic differences between the groups were only observed in patients at stage I-IIIa or N0-2. The lvN classification was found to be more appropriate to predict the OS of GC patients after curative surgery than the pN staging system. The-2 log-likelihood of lvN classification(4,746.922) was smaller than the value of pN(4,765.196), and the difference was statistically significant(χ^2=18.434, P<0.001).Conclusions: The presence of LVI influences the OS of GC patients at stage Ⅰ-Ⅲ a or N0-2. | 7 | Identification and characterization of rice blast resistance gene Pid4 by a combination of transcriptomic profiling and genome analysis显示文摘Map-based cloning of plant disease resistance (R) genes is time-consuming. Here, we reported the isolation of blast R gene Pid4 using comparative transcriptomic profiling and genome-wide sequence analysis. Pid4 encodes a coiled-coil nucleotide-binding site leucine-rich repeat(CC-NBS-LRR) protein and is constitutively expressed at diverse developmental stages in the rice variety Digu. The Pid4 protein is localized in both the nucleus and cytoplasm. Introduction of Pid4 into susceptible rice cultivars confers race-specific resistance to leaf and neck blast. Amino acid sequence comparison and blast resistance spectrum tests showed that Pid4 is a novel R gene, different from the previously reported R genes located in the same gene cluster. A Pid4 Indel marker was developed to facilitate the identification of Pid4 in different rice varieties. | 9 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 11 | Modified hyaluronic acid hydrogels with chemical groups that facilitate adhesion to host tissues enhance cartilage regeneration显示文摘Stable integration of hydrogel implants with host tissues is of critical importance to cartilage tissue engineering.Designing and fabricating hydrogels with high adhesive strength,stability and regeneration potential are major challenges to be overcome.This study fabricated injectable adhesive hyaluronic acid(HA)hydrogel modified by aldehyde groups and methacrylate(AHAMA)on the polysaccharide backbone with multiple anchoring mechanisms(amide bond through the dynamic Schiff base reaction,hydrogen bond and physical interpenetration).AHAMA hydrogel exhibited significantly improved durability and stability within a humid environment(at least 7 days),together with higher adhesive strength(43 KPa to skin and 52 KPa to glass),as compared to commercial fibrin glue(nearly 10 KPa)and HAMA hydrogel(nearly 20 KPa).The results showed that AHAMA hydrogel was biocompatible and could be easily and rapidly prepared in situ.In vitro cell culture experiments showed that AHAMA hydrogel could enhance proliferation(1.2-folds after 3 days)and migration(1.5-folds after 12 h)of bone marrow stem cells(BMSCs),as compared to cells cultured in a culture dish.Furthermore,in a rat osteochondral defect model,implanted AHAMA hydrogel significantly promoted integration between neo-cartilage and host tissues,and significantly improved cartilage regeneration(modified O’Driscoll histological scores of 16.0±4.1 and 18.3±4.6 after 4 and 12-weeks of post-implantation in AHAMA groups respectively,12.0±2.7 and 12.2±2.8 respectively in HAMA groups,9.8±2.4 and 11.5±2.1 respectively in untreated groups).Hence,AHAMA hydrogel is a promising adhesive biomaterial for clinical cartilage regeneration and other biomedical applications. | Jiaqing Chen Jiabei Yang Li Wang Xuewei Zhang Boon Chin Heng Dong-An Wang Zigang Ge | 2021 | Bioactive Materials2021,6,6: | 6 |
| 12 | Differential expression analysis and regulatory network reconstruction for genes associated with muscle growth and adipose deposition in obese and lean pigs显示文摘During the growth and development of skeletal muscle cells and adipose cells, the regulatory mechanism of micro-effect polygenes determines porcine meat quality, carcass characteristics and other relative quantitative traits. Obese and lean type pig breeds show obvi- ous differences in muscle growth and adipose deposition; however, the molecular mechanism underlying this phenotypic variation remains unknown. We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with muscle growth and adipose deposition in longissimus dorsi muscle at six growth stages (birth, 1, 2, 3, 4 and 5 months) of Landrace (a leaner, Western breed) and Taihu pigs (a fatty, indigenous, Chinese breed). Variance analysis (ANOVA) revealed that differences in the expression of 18 genes in Landrace pigs and three genes in Taihu pigs were very significant (FDR-adjusted permutation, P < 0.01) and di?erences for 22 genes in Landrace pigs and seven genes in Taihu pigs were significant (FDR-adjusted permutation, P < 0.05) among six growth stages. Clustering analysis revealed a high level of signi?cance (FDR-adjusted, P < 0.01) for four gene expression patterns, in which genes that strongly up-regulated were mainly associated with the positive regulation of myofiber formation and fatty acid biogenesis and genes that strongly down-regulated were mainly associated with the inhibition of cell proliferation and positive regulation of fatty acid b-oxidation. Based on a dynamic Bayesian network (DBN) model, gene regulatory networks (GRNs) were reconstructed from time-series data for each pig breed. These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of muscle growth and adipose deposition between the two pig breeds; from these results, some potential key genes could be identified. Quantitative real-time RT-PCR (QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both two pig breeds at different growth stages (r = 0.876 ± 0.095). The rice genome contains two SERK genes,OsSerk1 and OsSerk2. We previously demonstrated that OsSerk2 is required for rice Xa21-mediated resistance to Xanthomonas oryzae pv. oryzae(Xoo) and for normal development. Here we report the molecular characterization of OsSerk1. Overexpression of OsSerk1 results in a semi-dwarf phenotype whereas silencing of OsSerk1 results in a reduced angle of the lamina joint. OsSerk1 is not required for rice resistance to Xoo or Magnaporthe oryzae. Overexpression of Os Serk1 in OsSerk2-silenced lines complements phenotypes associated with brassinosteroid(BR) signaling defects, but not the disease resistance phenotype mediated by Xa21. In yeast, OsSERK1 interacts with itself forming homodimers, and also interacts with the kinase domains of OsSERK2 and BRI1, respectively.OsSERK1 is a functional protein kinase capable of auto-phosphorylation in vitro. | 18 | Fine mapping of the rice bacterial blight resistance gene Xa-4 and its co-segregation marker显示文摘An F2 population developed from the Xa-4 near isogenic lines, IR24 and IRBB4, was used for fine mapping of the rice bacterial blight resistance gene, Xa-4. Some restriction fragment length polymorphism (RFLP) markers on the high-density map constructed by Harushima et al. and the amplified DMA fragments homologous to the conserved domains of plant disease resistance (R) genes were used to construct the genetic linkage map around the gene Xa-4 by scoring susceptible individuals in the population. Xa-4 was mapped between the RFLP marker G181 and the polymerase chain reaction (PCR) marker M55. The R gene homologous fragment marker RS13 was found co-segregating with Xa-4 by analyzing all the plants in the population. | 19 | Alterations in cecal microbiota of Jinhua piglets fostered by a Yorkshire sow显示文摘Gut microbiota play important roles in host health and diseases.Several factors,in particular antibiotics,affect the gut microbiota of pigs.Cross-fostering has been applied as a regular practice to equalize litter size,reduce pre-weaning mortality and increase body weight.However,the effect of cross-fostering on cecal microbiota is unclear.In this study,we fostered three Jinhua pigs to a Yorkshire sow.The fostered Jinhua piglets grew significantly faster than their biological siblings.To explore whether the cecal microbiota of piglets will alter during fostering,we characterized the cecal microbiota of allpiglets by examining the V3 hypervariable region of 16S rDNA.We observed altered cecal microbiota in these piglets using the Illumina HiSeq 2000 platform,and this was accompanied with an increase in growth rate after fostering.The relative abundance of Bacteroidetes in the fostered Jinhua piglets was decreased compared with their biological siblings,although still higher in comparison with their new littermates.Beta-diversity analysis also showed that the cecal microbiota of the adopted Jinhua piglets differed from their biological siblings with a shift toward their step-siblings.Our data show that cecal microbiota of piglets were altered after cross-fostering while the growth rate increased. | Lingjin Xian Ying Li Zhi Jiang Jideng Ma Long Jin Lei Chen Chaowei Zhou Jie Zhang Yingkai Liu Li Zhu An-an Jiang Guoqing Tang Jian Xiao Jinyong Wang Bo Zeng Mingzhou Li Xuewei Li | 2014 | Chinese Science Bulletin2014,59,32: | 4 |
| 20 | Fabrication of superstable gold nanorod-carbon nanocapsule as a molecule loading material显示文摘In this work, we fabricated a monodisperse nanocomposite by coating gold nanorods(Au NRs) with a layer of biocompatible, stable carbon, obtaining Au NR@Carbon core–shell nanocapsules, which without any functionalization could be used as a molecule loading material due to its high surface areas. In this system, the Au NR core had a high-absorption cross section for conversion of near-infrared light to heat, which could be explored for local hyperthermia. The carbon shell, which was biocompatible and stable even under concentrated acidic and alkaline conditions, was able to adsorb molecules with p–p interactions or electrostatic interactions.
