1SHY2 as a node in the regulation of root meristem development by auxin, brassinosteroids, and cytokinin显示文摘In multicellular organisms,the balance be-tween cell division and differentiation determines organ size,and represents a central unknown in devel-opmental biology.In Arabidopsis roots,this balance is mediated between cytokinin and auxin through a reg-ulatory circuit converging on the IAA3/SHORT HYPO-COTYL 2(SHY2)gene.Here,we show that crosstalk between brassinosteroids(BRs)and auxin occurs in the vascular transition zone to promote root meristem de-velopment.We found that BR increases root meristem size by up-regulating expression of the PINFORMED 7(PIN7)gene and down-regulating expression of the SHY2 gene.In addition,BES1 could directly bind to the pro-moter regions of both PIN7 and SHY2,indicating that PIN7 and SHY2 mediate the BR-induced growth of the root meristem by serving as direct targets of BES1.Moreover,the PIN7 overexpression and loss-of-function SHY2 mutant were sensitive to the effects of BR and could partially suppress the short-root phenotypes associated with deficient BR signaling.Interestingly,BRs could inhibit the accumulation of SHY2 protein in response to cytokinin.Taken together,these findings suggest that a complex equilibrium model exists in which regulatory interactions among BRs,auxin,and cytokinin regulate optimal root growth.Taotao Li Xinke Kang Wei Lei Xiuhong Yao Lijuan Zou Dawei Zhang Honghui Lin 2020Journal of Integrative Plant Biology2020,62,10:5

4Sinoatrial node pacemaker cells share dominant biological properties with glutamatergic neurons显示文摘Activation of the heart normally begins in the sinoatrial node(SAN).Electrical impulses spontaneously released by SAN pacemaker cells(SANPCs)trigger the contraction of the heart.However,the cellular nature of SANPCs remains controversial.Here,we report that SANPCs exhibit glutamatergic neuron-like properties.By comparing the single-cell transcriptome of SANPCs with that of cells from primary visual cortex in mouse,we found that SANPCs co-clustered with cortical neurons.Tissue and cellular imaging confirmed that SANPCs contained key elements of glutamatergic neurotransmitter system,expressing genes encoding glutamate synthesis pathway(G/s),ionotropic and metabotropic glutamate receptors(Grina,Gria3,Grm1 and Grm5)t and glutamate transporters(Slc17a7).SANPCs highly expressed cell markers of glutamatergic neurons(Snap25 and S/-c17a7)t whereas Gad1,a marker of GABAergic neurons,was negative.Functional studies revealed that inhibition of glutamate receptors or transporters reduced spontaneous pacing frequency of isolated SAN tissues and spontaneous Ca2+transients frequency in single SANPC.Collectively,our work suggests that SANPCs share dominant biological properties with glutamatergic neurons,and the glutamatergic neurotransmitter system may act as an intrinsic regulation module of heart rhythm,which provides a potential intervention target for pacemaker cell-associated arrhythmias.Dandan Liang Zhigang Xue Jinfeng Xue Duanyang Xie Ke Xiong Huixing Zhou Fulei Zhang Xuling Su Guanghua Wang Qicheng Zou Yi Liu Jian Yang Honghui Ma Luying Peng Chunyu Zeng Gang Li Li Wang Yi-Han Chen 2021Protein & Cell2021,12,7:1

13Photosynthetic microorganisms coupled photodynamic therapy for enhanced antitumor immune effect显示文摘The ideal photodynamic therapy(PDT)should effectively remove the primary tumor,and produce a stronger immune memory effect to inhibit the tumor recurrence and tumor metastasis.However,limited by the hypoxic and immunosuppressive microenvironment,the PDT efficiency is apparently low.Here,Chlorella(Chl.)is exploited to enhance local effect by producing oxygen to reverse hypoxia,and release adjuvants to reverse immunosuppressive microenvironment to enhance abscopal effect afterwards.Results from different animal models indicated that Chl.could enhance local effect and PDT related immune response.Ultimately,Chl.coupled PDT elicited anti-tumor effects toward established primary tumors(inhibition rate:90%)and abscopal tumors(75%),controlled the challenged tumors(100%)and alleviated metastatic tumors(90%).This Chl.coupled PDT strategy can also produce a stronger anti-tumor immune memory effect.Overall,this Chl.coupled PDT strategy generates enhanced local tumor killing,boosts PDT-induced immune responses and promotes anti-tumor immune memory effect,which may be a great progress for realizing systemic effect of PDT.Haoran Wang Honghui Liu Yunfei Guo Wenjing Zai Xianghui Li Wei Xiong Xiaozhi Zhao Yingfang Yao Yiqiao Hu Zhigang Zou Jinhui Wu 2022Bioactive Materials2022,7,6:0
14Rare inherited missense variants of POGZ associate with autism risk and disrupt neuronal development显示文摘Excess de novo likely gene-disruptive and missense variants within dozens of genes have been identified in autism spectrum disorder(ASD)and other neurodevelopmental disorders.However,many rare inherited missense variants of these high-risk genes have not been thoroughly evaluated.In this study,we analyzed the rare missense variant burden of POGZ in a large cohort of ASD patients from the Autism Clinical and Genetic Resources in China(ACGC)and further dissected the functional effect of diseaseassociated missense variants on neuronal development.Our results showed a significant burden of rare missense variants in ASD patients compared to the control population(P=4.6×10-5,OR=3.96),and missense variants in ASD patients showed more severe predicted functional outcomes than those in controls.Furthermore,by leveraging published large-scale sequencing data of neurodevelopmental disorders(NDDs)and sporadic case reports,we identified 8 de novo missense variants of POGZ in NDD patients.Functional analysis revealed that two inherited,but not de novo,missense variants influenced the cellular localization of POGZ and failed to rescue the defects in neurite and dendritic spine development caused by Pogz knockdown in cultured mouse primary cortical neurons.Significantly,L1CAM,an autism candidate risk gene,is differentially expressed in POGZ deficient cell lines.Reduced expression of L1cam was able to partially rescue the neurite length defects caused by Pogz knockdown.Our study showed the important roles of rare inherited missense variants of POGZ in ASD risk and neuronal development and identified the potential downstream targets of POGZ,which are important for further molecular mechanism studies.Wenjing Zhao Jieqiong Tan Tengfei Zhu Jianjun Ou Ying Li Lu Shen Huidan Wu Lin Han Yanling Liu Xiangbin Jia Ting Bai Honghui Li Xiaoyan Ke Jingping Zhao Xiaobing Zou Zhengmao Hu Hui Guo Kun Xia 2019Journal of Genetics and Genomics2019,46,5:0
