1Coal and gas outburst prevention using new high water content cement slurry for injection into the coal seam显示文摘As coal and gas outburst is one of the most serious mine disasters, it is very important to at least control it if not prevent it from occurring. Injecting cement slurry or grouting into the coal seam can strengthen the seam, increase its rigidity coefficient(f), and reduce the volumetric expansion due to gas energy release.This paper reports the results of laboratory experiments on cement-based high water content slurry having different water-cement ratios(W/C) to be used for coal injection. The results show that as the W/C increases, the mobility of the slurry and its setting time increase. The compressive strength and rupture strength, however, are reduced. Furthermore, high W/C grout shows early strength after 7 days, which can be 80% of its 14-day compressive strength. To achieve rapid setting and early strength, the addition of Na_2SiO_3has proven to give the best result, when the concentration of the additive is 3%. The initial and final setting times are 13 and 21 min shorter than samples without Na_2SiO_3, while the compressive strength is more than double. As a retarder, the initial setting time can be extended to 83 min when tartaric acid of 0.4% concentration is added. Through the orthogonal experiment, the optimum recipe of the new high water content slurry has been determined to be: W/C = 2, tartaric acid = 0.2%, Na_2SiO_3= 3%, and12% bentonite. Reinforcement by injection simulation experiments show that the grouting radius of the new slurry mix is 250 mm when the applied grouting pressure is 60 k Pa, 7-day rupture strength and compressive strength are 5.2 and 6.4 MPa, respectively, and are 37% and 88% higher than ordinary cement grout. 2Effects of continuous and pulse irrigation with different nitrogen applications on soil moisture,nitrogen transport and accumulation in root systems显示文摘Pulse and continuous irrigation can affect water distribution in the soil,thereby affecting the growth and distribution of root systems.Different base and topdressing fertilizers can affect nitrogen transport in the soil and root distribution,thus affecting root water and nitrogen uptake.This study used pot experiments to set up four different nitrogen levels,including two different irrigation methods,to research the effects of soil moisture,nitrogen transport,nitrogen accumulation and root systems under the conditions of continuous and pulse irrigation,as well as different nitrogen applications.Result showed that continuous irrigation can increase the moisture content around the drip emitter position and reduce the soil moisture content in the horizontal spreading compared with pulse irrigation,especially reducing the horizontal spreading in the deep soil layers.One treatment in this study did not apply base fertilizer,and then supply topdressing fertilizer at a ratio of 2:4:4 in the seedling stage,jointing stage and tasseling stage,respectively.This treatment is beneficial for improving nitrogen utilization efficiency.Furthermore,this treatment can increase the root length density and root surface area in the deep soil layers,especially in the 20-35 cm soil depth,and to some extent increase the deep growth of the root system.Lingmiao Huang Peiling Yang Shumei Ren Haobo Cui 2018International Journal of Agricultural and Biological Engineering2018,11,5:2
3Immune‑Ageing Evaluation of Peripheral T and NK Lymphocyte Subsets in Chinese Healthy Adults显示文摘Ageing is often accompanied with a decline in immune system function,resulting in immune ageing.Numerous studies have focussed on the changes in different lymphocyte subsets in diseases and immunosenescence.The change in immune phenotype is a key indication of the diseased or healthy status.However,the changes in lymphocyte number and phenotype brought about by ageing have not been comprehensively analysed.Here,we analysed T and natural killer(NK)cell subsets,the phenotype and cell differentiation states in 43,096 healthy individuals,aged 20–88 years,without known diseases.Thirty-six immune parameters were analysed and the reference ranges of these subsets were established in different age groups divided into 5-year intervals.The data were subjected to random forest machine learning for immune-ageing modelling and confirmed using the neural network analysis.Our initial analysis and machine modelling prediction showed that na.ve T cells decreased with ageing,whereas central memory T cells(Tcm)and effector memory T cells(Tem)increased cluster of differentiation(CD)28-associated T cells.This is the largest study to investigate the correlation between age and immune cell function in a Chinese population,and provides insightful differences,suggesting that healthy adults might be considerably influenced by age and sex.The age of a person's immune system might be different from their chronological age.Our immune-ageing modelling study is one of the largest studies to provide insights into‘immune-age’rather than‘biological-age’.Through machine learning,we identified immune factors influencing the most through ageing and built a model for immune-ageing prediction.Our research not only reveals the impact of age on immune parameter differences within the Chinese population,but also provides new insights for monitoring and preventing some diseases in clinical practice.Zhenghu Jia Zhiyao Ren Dongmei Ye Jiawei Li Yan Xu Hui Liu Ziyu Meng Chengmao Yang Xiaqi Chen Xinru Mao Xueli Luo Zhe Yang Lina Ma Anyi Deng Yafang Li Bingyu Han Junping Wei Chongcheng Huang Zheng Xiang Guobing Chen Peiling Li Juan Ouyang Peisong Chen Oscar Junhong Luo Yifang Gao Zhinan Yin 2023Phenomics2023,3,4:1
4The chemical reprogramming of unipotent adult germ cells towards authentic pluripotency and de novo establishment of imprinting显示文摘Although somatic cells can be reprogrammed to pluripotent stem cells(PsCs)with pure chemicals,authentic pluripotency of chemically induced pluripotent stem celis(CipsCs)has never been achieved through tetraploid complementation assay.Spontaneous reprogramming of spermatogonial stem cells(ssCs)was another non-transgenic way to obtain PsCs,but this process lacks mechanistic explanation.Here,we reconstructed the trajectory of mouse SsC reprogramming and developed a five-chemical combination,boosting the reprogramming effciency by nearly 80-to 100-folds.More importantly,chemical induced germline-derived PsCs(5C-gPSCs),but not gpsCs and chemical induced pluripotent stem cells,had authentic pluripotency,as determined by tetraploid complementation.Mechanistically,ssCs traversed through an inverted pathway of in vivo germ ceil development,exhibiting the expression signatures and DNA methylation dynamics from spermatogonia to primordial germ cells and further to epiblasts.Besides,ssC-specific imprinting control regions switched from biallelic methylated states to monoallelic methylated states by imprinting demethylation and then re-methylation on one of the two alleles in 5c-gPsCs,which was apparently distinct with the imprinting reprogramming in vivo as DNA methylation simultaneously occurred on both alleles.Our work sheds ight on the unique regulatory network underpinning SsC reprogramming,providing insights to understand generic mechanisms for cell-fate decision and epigenetic-relateddisorders in regenerative medicine.Yuhan Chen Jiansen Lu Yanwen Xu Yaping Huang Dazhuang Wang Peiling Liang Shaofang Ren Xuesong Hu Yewen Qin Wei Ke Ralf Jauch Andrew Paul Hutchins Mei Wang Fuchou Tang Xiao-Yang Zhao 2023Protein & Cell2023,14,7:0
5Determination of Median Nerve Somatosensory Evoked Potentials in 27 Patients with Hemiplegia显示文摘Summary: Twenty-seven in-patients with hemiplegia following brain injury were studied by using upper extremity median nerve somatosensory evoked patentials (SVEP), Brunnstrom assessment in hemiplegic hand and assessment of the patients’ activities of daily lioing (ADL) (Barthel index). The upper extremity median nerve SEP on the affected and normal sides was determined. By using Kovindha standard, upper extremity median nerve SEP was graded in accordance with N20. The correlation between the differences of SEP N20 amplitude and the latencies on the both sides and the Barthel index scores was analyzed. A Spearman correlation analysis was made between the median nerve SEP N20 grades and Brunnstrom stages in hand or ADL on the affected side. The results showed that upper extremity median nerve SEP grades were positively correlated with those of the Brunnstrom stages in hand (r 1=0.6925, P 1<0.01). The correlation coefficient between SEP N20 grades and patients’ ADL grades was r 2=0.5015, P 2<0 01. It was concluded that upper extremity median nerve SEP could be used as a sensitive electrophysiological predictor to clinically assess hemiplegic hand function. 8A Reactive Oxygen Species-Tyrosinase Cascade-Activated Prodrug for Selectively Suppressing Melanoma显示文摘“Off-target effect”is one of the obstacles for targeted prodrugs in chemotherapy.To circumvent this issue,herein we propose a dual biomarker cascadeactivated prodrug strategy based on high levels of reactive oxygen species(ROS)and tyrosinase(TYR)in melanoma cells.A representative prodrug,Coumarin-Quinazolinone-phenylBoronic acid pinacol ester(CQB),was prepared.The prodrug contained Coumarin-Quinazolinone(CQ)as mitochondria targeting and monitoring moiety and phenylboronic acid pinacol ester as a cascade-activated prowarhead.After activation by the endogenous ROS and subsequent TYR in melanoma cells,CQB can be converted to the final active reagent Coumarin-Quinazolinone-o-Quinone(CQQ)that contains o-quinone group as the reactive species.CQQ may interact with cellular nucleophiles to exert its genotoxic effect and disrupt the cellular redox balance.This enables CQB to selectively suppress melanoma.CQB accumulates in the mitochondria and causes mitochondrial dysfunction via affecting mitochondrial DNA integrity.This cascade-activated prodrug may provide a precise strategy for melanoma theranostics.Guorui Li Yi Yang Yaya Zhang Peiling Huang Jiangyu Yan Zhibin Song Quan Yuan Jing Huang 2022CCS Chemistry2022,4,5:0
9Dual Diomarkers Triggered Prodrugs for Precise Treatment of Melanoma:Design,Synthesis and Activities显示文摘Targeted prodrug strategy,which utilizes the endogenous biomarkers in cancer cells as activators to release the active drug,has been well established either in the fundamental research or the clinical treatment.However,many prodrugs suffer from safety concern due to“off-target activation”.Dual or multiple biomarkers triggered prodrug may provide an effective strategy to overcoming the“off-target effect”.Melanoma cells have both high levels of reactive oxygen species(ROS)and tyrosinase(TYR),which makes them significantly different from other tumor cells and normal cells.Here we reported a series of quinazolinone-aryl boronic acid/ester-based prodrugs,which can be activated by the cascade of ROS and TYR and selectively kill melanoma cells.The structure-activity relationship(SAR)analysis revealed that mitochondria-targeting property was vital for their cytotoxicity and the dual activated effector played a significant role in their selectivity towards melanoma cells.Among these candidates,compound 4b showed the highest toxicity to B16,leading to an imbalance of the redox system in melanoma cells,causing mitochondrial DNA damage,and then promoting melanoma cells death.RUAN Yawen HUANG Peiling YAN Jiangyu LI Guorui HUANG Jing 2022Chemical Research in Chinese Universities2022,38,4:0
10Effects of topdressing ratio and frequency on the dry matter, yield, and quality of tomato and celery under a small amount of continuous subsurface drip irrigation显示文摘Compared with conventional full irrigation,the existing water-saving irrigation method achieves the purpose of water saving by actively controlling soil moisture.High-frequency irrigation can make the ideal conditions for soil water movement and root absorption of water and nutrients.This research used a plot experiment in a greenhouse and set up different fertilization amounts and frequencies during the growth stage to study the effects of small-amount and continuous subsurface drip irrigation on the dry matter,yield,and quality of tomato and celery.The results showed that the frequency of topdressing had a great influence on the dry matter accumulation of tomatoes,and the amount of topdressing had little effect on the fresh weight of the upper part of a tomato plant.The application of high-frequency fertilizer increased the dry matter accumulation in the underground part of the tomato.Under the premise given amount of total fertilization,the growth rate and yield of tomatoes were positively correlated with the amount of topdressing.The optimum fertilization frequency was 1 time during the first fruit stage,3 times for the second fruit stage,and 5 times for the third fruit stage,the yield during the fourth stage was increased with higher frequency,and the topdressing was started ahead of the fourth fruit stage when the diameter of fruit was 40 mm.The lower fertilization frequency during the early stage and higher fertilization frequency during the later stage can increase the yield of celery.The higher ratio of topdressing,the higher frequency during the early growth stage,and the lower frequency during the later stage can achieve the best quality of celery.Lingmiao Huang Peiling Yang Haobo Cui Zhanghao Sun Shumei Ren Zhaocheng Wang 2023International Journal of Agricultural and Biological Engineering2023,16,3:0
