1Microstructure,mechanical and corrosion properties of Mg-2Dy-xZn(x=0,0.1,0.5 and 1 at.%)alloys显示文摘Microstructure,mechanical and corrosion properties of as-cast Mg-2Dy-xZn(x=0,0.1,0.5,and 1)(at.%)alloys were investigated.The microstructures of the as-cast Mg-2Dy and Mg-2Dy-0.1Zn alloys mainly consisted ofα-Mg phase and Mg_(24)Dy_(5)eutectic phase.With 0.5 at.%Zn addition,Mg_(12)ZnDy phase with 18R-type long period stacking ordered(LPSO)structure and Mg_(2)Dy phase precipitated at the grain boundaries.When the content of Zn is 1 at.%,only the Mg_(3)Zn_(3)Dy_(2)phase formed in theα-Mg matrix.The electrochemical measurements and immersion testing results indicated that the Mg-2Dy-0.1Zn alloy exhibited the best corrosion resistance.It revealed that the morphology,scale,amount and distribution of the second phase have a great effect on the corrosion resistance of alloy.Additionally,the tensile testing results showed that the Mg-2Dy-0.5Zn alloy exhibited the higher tensile strength and good elongation,especially at 200℃.The improvement of mechanical properties was mainly due to the strengthening of LPSO phase and grain refinement of α-Mg.Guangli Bi Yuandong Li Shijun Zang Jianbin Zhang Ying Ma Yuan Hao 2014Journal of Magnesium and Alloys2014,2,1:13
2Association of KIR Genotypes and Haplotypes with Susceptibility to Chronic Hepatitis B Virus Infection in Chinese Han Population显示文摘Killer immunoglobulin-like receptor (KIR) genes can regulate the activation of NK and T cells upon interaction with HLA class I molecules. Hepatitis B virus (HBV) infection has been regarded as a multi-factorial disorder disease. Previous studies revealed that KIRs were involved in HCV and HIV infection or clearance. The aim of this study was to explore the possibility of the inheritance of KIR genotypes and haplotypes as a candidate for susceptibility to persistent HBV infection or HBV clearance. The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 150 chronic hepatitis B (CHB) patients, 251 spontaneously recovered (SR) controls, and 412 healthy controls. The frequencies of genotype G, M, FZ1 increased in CHB patients compared with healthy control subjects. The frequency of genotype AH was higher in SR controls than that in both CHB patients and healthy controls. The carriage frequencies of genotype G and AH were higher; while, the frequencies of AF and AJ were lower in SR controls than those in healthy control subjects. The frequency of A haplotype was lower, whereas, the frequency of B haplotype was higher in CHB patients and SR controls than those in healthy controls. In healthy controls, haplotype 4 was found lower compared with that in CHB patients and SR controls and the frequency of haplotype 5 was higher in SR controls than that in other two groups. Based on these findings, it seems that the genotypes M and FZ1 are HBV susceptive genotypes; AH, on the other hand, may be protective genotypes that facilitate the clearance of HBV. 3Jianpijiedu Fang improves survival of hepatocarcinoma mice by affecting phosphatase and tensin homolog, phosphoinositide 3-kinase, and focal adhesion kinase显示文摘OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. 4Surrounding rock control mechanism of deep coal roadways and its application显示文摘Aiming at the surrounding rock control problem of mining and preparation entries in Xingdong mine with large mining depth, and the comprehensive control countermeasures including high pre-stress cable truss system, this study put forward powerful anchor support system and anchor cable adaption technology to surrounding rock deformation. Furthermore, the control measures possess the supporting performance with ‘‘primary rigid-following flexible-new rigid, and primary resistance-following yield-new resistance'', which suits deep roadway surrounding rock control. The mechanical model of truss anchor supporting roof beams was established, and the inverted arch deflection produced by the cable pre-stress with stress increment effect and roof beam deflection were obtained. And then the system working mechanism was illustrated. Finally, the surrounding rock support parameters were determined by means of comprehensive methods, and put into practice. The results show that surrounding rock deformation realized secondary stability after three months. The roadway sides convergence value was less than 245mm, and roof subsidence was less than 124mm. 5Gene expression profiling of rat livers with Yin-deficiency-heat syndrome显示文摘OBJECTIVE: To explore the nature of 'Yin internal heat caused byYin-deficiency,' in terms of the theory of Traditional Chinese Medicine, by studying energy metabolism in rats with Yin-deficiency-heat syndrome and analyzing the gene expression profile of their livers. METHODS: A Yin-deficiency-heat syndrome model was induced in rats using three Chinese medicinal herbs. Glycogen and triglycerides in blood plasma, and the enzyme activity of ATP in livers were measured colorimetrically. Triiodothyronine (T3 ), thyroxine (T4 ), and thyroid stimulating hormone levels in blood plasma were also measured with enzyme linked immunosorbent assay. The gene expression profile of livers was detected with gene chip analysis. Differentially expressed genes were screened out and classified according to Gene Ontology. The accuracy of results were examined with reversetranscription-polymerase chain reaction. RESULTS: Compared with the control group, body weight (P<0.05) and hepatic glycogen (P<0.05) were significantly lower in the Yin-deficiency-heat syndrome group. Moreover, toe temperature (P< 0.01) and triglyceride (P<0.05), Na+ -K+ -ATPase (P< 0.01), Mg2+ -ATPase (P<0.01), T3 (P<0.05), and T4 (P< 0.01) levels were significantly higher. There were 99 differentially expressed genes in livers from the Yin-deficiency-heat syndrome group. Genes were mainly related to sterol synthesis (Pc=0.0392), defense response (Pc=0.0448), and sterol metabolism (Pc=0.0533). CONCLUSION: Abnormal expression genes in rats with Yin-deficiency-heat syndrome prompted the synthesis and metabolism of cholesterol, increased energy consumption, and reduced defense response. 6Nonlinear dynamics of a flapping rotary wing:Modeling and optimal wing kinematic analysis显示文摘The analysis of the passive rotation feature of a micro Flapping Rotary Wing(FRW)applicable for Micro Air Vehicle(MAV) design is presented in this paper. The dynamics of the wing and its influence on aerodynamic performance of FRW is studied at low Reynolds number(~10~3).The FRW is modeled as a simplified system of three rigid bodies: a rotary base with two flapping wings. The multibody dynamic theory is employed to derive the motion equations for FRW. A quasi-steady aerodynamic model is utilized for the calculation of the aerodynamic forces and moments. The dynamic motion process and the effects of the kinematics of wings on the dynamic rotational equilibrium of FWR and the aerodynamic performances are studied. The results show that the passive rotation motion of the wings is a continuous dynamic process which converges into an equilibrium rotary velocity due to the interaction between aerodynamic thrust, drag force and wing inertia. This causes a unique dynamic time-lag phenomena of lift generation for FRW, unlike the normal flapping wing flight vehicle driven by its own motor to actively rotate its wings. The analysis also shows that in order to acquire a high positive lift generation with high power efficiency and small dynamic time-lag, a relative high mid-up stroke angle within 7–15° and low mid-down stroke angle within -40° to -35° are necessary. 7Rational cutting height for large cutting height fully mechanized top-coal caving显示文摘Large cutting height fully mechanized top-coal caving is a new mining method that improves recovery ratio and single-pass production. It also allows safe and efficient mining. A rational cutting height is one key parameter of this technique. Numerical simulation and a granular-media model experiment were used to analyze the effect of cutting height on the rock pressure of a fully mechanized top-coal caving work face. The recovery ratio was also studied. As the cutting height increases the top-coal thickness is reduced. Changing the ratio of cutting to drawing height intensifies the face pressure and the top-coal shattering. A maximum cutting height exists under a given set of conditions due to issues with surrounding rock-mass control. An increase in cutting height makes the top-coal cave better and the recovery ratio when drawing top-coal is then improved. A method of adjusting the face rock pressure is presented. Changing the cutting to drawing height ratio is the technique used to control face rock pressure. The recovery ratio when cutting coal exceeds that when caving top-coal so the face recovery ratio may be improved by over sizing the cutting height and increasing the top-coal drawing ratio. An optimum ratio of cutting to drawing height exists that maximizes the face recovery ratio. A rational cutting height is determined by comprehensively considering the surrounding rock-mass control and the recovery ratio. 8Liquid crystal integrated metalens with tunable chromatic aberration显示文摘Overcoming chromatic aberrations is a vital concern in imaging systems in order to facilitate fullcolor and hyperspectral imaging.By contrast,large dispersion holds opportunities for spectroscopy and tomography.Combining both functions into a single component will significantly enhance its versatility.A strategy is proposed to delicately integrate two lenses with a static resonant phase and a switchable geometric phase separately.The former is a metasurface lens with a linear phase dispersion.The latter is composed of liquid crystals(LCs)with space-variant orientations with a phase profile that is frequency independent.By this means,a broadband achromatic focusing from 0.9 to 1.4 THz is revealed.When a saturated bias is applied on LCs,the geometric phase modulation vanishes,leaving only the resonant phase of the metalens.Correspondingly,the device changes from achromatic to dispersive.Furthermore,a metadeflector with tunable dispersion is demonstrated to verify the universality of the proposed method.Our work may pave a way toward active metaoptics,promoting various imaging applications.Zhixiong Shen Shenghang Zhou Xinan Li Shijun Ge Peng Chen Wei Hu Yanqing Lu 2020Advanced Photonics2020,2,3:7
9Antibody drug conjugate:the“biological missile”for targeted cancer therapy显示文摘Antibody–drug conjugate(ADC)is typically composed of a monoclonal antibody(mAbs)covalently attached to a cytotoxic drug via a chemical linker.It combines both the advantages of highly specific targeting ability and highly potent killing effect to achieve accurate and efficient elimination of cancer cells,which has become one of the hotspots for the research and development of anticancer drugs.Since the first ADC,Mylotarg®(gemtuzumab ozogamicin),was approved in 2000 by the US Food and Drug Administration(FDA),there have been 14 ADCs received market approval so far worldwide.Moreover,over 100 ADC candidates have been investigated in clinical stages at present.This kind of new anti-cancer drugs,known as“biological missiles”,is leading a new era of targeted cancer therapy.Herein,we conducted a review of the history and general mechanism of action of ADCs,and then briefly discussed the molecular aspects of key components of ADCs and the mechanisms by which these key factors influence the activities of ADCs.Moreover,we also reviewed the approved ADCs and other promising candidates in phase-3 clinical trials and discuss the current challenges and future perspectives for the development of next generations,which provide insights for the research and development of novel cancer therapeutics using ADCs.Zhiwen Fu Shijun Li Sifei Han Chen Shi Yu Zhang 2022Signal Transduction and Targeted Therapy2022,7,4:5
10Contribution of Baroreflex Afferent Pathway to NPY-Mediated Regulation of Blood Pressure in Rats显示文摘Neuropeptide Y (NPY), a metabolism-related cardiovascular factor, plays a crucial role in blood pressure(BP) regulation via peripheral and central pathways. The expression of NPY receptors (Y1R/Y2R) specific to baroreflex afferents impacts on the sexually dimorphic neural control of circulation. This study was designed to investigate the expression profiles of NPY receptors in the nodose ganglion (NG) and nucleus tractus solitary (NTS) under hypertensive conditions. To this end, rats with hypertension induced by NG-nitro-L-arginine methylester (L-NAME) or high fructose drinking (HFD), and spontaneously hypertensive rats (SHRs) were used to explore the effects/mechanisms of NPY on BP using functional, molecular, and electrophysiological approaches. The data showed that BP was elevated along with baroreceptor sensitivity dysfunction in model rats;Y1R was up-or down-regulated in the NG or NTS of male and female HFD/L-NAME groups,while Y2R was only down-regulated in the HFD groups as well as in the NG of the male L-NAME group. In SHRs,Y1R and Y2R were both down-regulated in the NTS, and not in the NG. In addition to NPY-mediated energy homeostasis, leptin-melanocortin activation may be essential for metabolic disturbance-related hypertension. We found that leptin and a-melanocyte stimulating hormone (aMSH) receptors were aberrantly down-regulated in HFD rats. In addition, a-MSH concentrations were reduced and NPY concentrations were elevated in the serum and NTS at 60 and 90 min after acute leptin infusion. Electrophysiological recordings showed that the decay time-constant and area under the curve of excitatory post-synaptic currents were decreased by Y1R activation in A-types, whereas, both were increased by Y2R activation in Ah-or C-types. 11Deer Body Adaptive Threshold Segmentation Algorithm Based on Color Space显示文摘In large-scale deer farming image analysis,K-means or maximum between-class variance(Otsu)algorithms can be used to distinguish the deer from the background.However,in an actual breeding environment,the barbed wire or chain-link fencing has a certain isolating effect on the deer which greatly interferes with the identification of the individual deer.Also,when the target and background grey values are similar,the multiple background targets cannot be completely separated.To better identify the posture and behaviour of deer in a deer shed,we used digital image processing to separate the deer from the background.To address the problems mentioned above,this paper proposes an adaptive threshold segmentation algorithm based on color space.First,the original image is pre-processed and optimized.On this basis,the data are enhanced and contrasted.Next,color space is used to extract the several backgrounds through various color channels,then the adaptive space segmentation of the extracted part of the color space is performed.Based on the segmentation effect of the traditional Otsu algorithm,we designed a comparative experiment that divided the four postures of turning,getting up,lying,and standing,and successfully separated multiple target deer from the background.Experimental results show that compared with K-means,Otsu and hue saturation value(HSV)+K-means,this method is better in performance and accuracy for adaptive segmentation of deer in artificial breeding scenes and can be used to separate artificially cultivated deer from their backgrounds.Both the subjective and objective aspects achieved good segmentation results.This article lays a foundation for the effective identification of abnormal behaviour in sika deer.Yuheng Sun Ye Mu Qin Feng Tianli Hu He Gong Shijun Li Jing Zhou 2020Computers, Materials & Continua2020,,8:5
12Strengthening in Al-,Mo-or Ti-doped CoCrFeNi high entropy alloys:A parallel comparison显示文摘In the current work,a parallel comparison of the influence of Al,Mo and Ti,on the microstructure and strengthening of the CoCrFeNi alloy was conducted.To achieve this,inconsistencies on variables including the extent of alloying,thermomechanical processing and property-evaluation method were avoided.Microstructurally,following cold-rolling,annealing of the 4 at.%Al-doped alloys at 800-1000℃ did not result in phase separation;nevertheless,that of the 4 at.%Mo-and Ti-doped alloys led to the respective formation ofσandηphase and,consequently,caused extra strengthening through the Orowan dislocation bypassing mechanism.Our systematic qualitative analysis and DFT calculations showed that Al and Ti are more effective than Mo in reducing the stacking fault energy(SFE)of the CoCrFeNi alloy,because they can induce more considerable deformation of electronic density,making the gliding of atomic layers easier.Following identical thermomechnical processing,Al-,Mo-,and Ti-doping causes different extent of solid solution strengthening and grain boundary strengthening.Mo causes the most pronounced solid solution strengthening but does not benefit the grain boundary strengthening;in contrast,the effectiveness of grain boundary strengthening is boosted by the doping Al and Ti.Current analyses support that Labusch instead of Fleischer mechanism is applicable to explain the differences in solid solution strengthening,and the observed differences in grain boundary strengthening arise from the different tendency of Al,Mo and Ti to reduce the SFE of CoCrFeNi.In addition,we determined the value of the dimensionless parameter f in the Labusch model for CoCrFeNi-based alloys and observed a close relation between Hall-Petch slope and SFE.Although more in-depth studies are needed to provide full and mechanistic understandings,both these findings in fact presents significant values toward designing novel singlephase high-strength CoCrFeNi-based alloys through manipulating the solid solution and grain boundary strengthening by compositional tuning.Xi Li Zhongtao Li Zhenggang Wu Shijun Zhao Weidong Zhang Hongbin Bei Yanfei Gao 2021Journal of Materials Science & Technology2021,,35:4
13Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4

15Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4
16Synthesis and DNA Adsorption of Poly(2-Vinyl-4,6-Diamino-1,3,5-Triazine)Coated Polystyrene Microspheres显示文摘To take advantage of the polymer's special properties, we synthesized PVDAT coated polystyrene(PVDAT@PS) microspheres through semi-continuous precipitation polymerization, and demonstrated the possibility of controlling the size of microspheres by using different sized PS cores. Mechanism of the microsphere synthesis was investigated. 18Transforming growth factor β1 is a differentially expressed candidate protein of congestive heart failure with Qi-deficiency-blood-stasis syndrome显示文摘OBJECTIVE: To investigate the role of tongue coating fluid protein in regulation of congestive heart failure(CHF) in Qi-deficiency-blood-stasis syndrome.METHODS: We studied patients with CHF(3 patients with Qi-deficiency-blood-stasis syndrome and 3 without Qi-deficiency-blood-stasis syndrome) to investigate differentially expressed proteins. We also included a control group. A biotin label-based antibody array was used for testing tongue coating fluid samples from patients. Net-work analysis of these differentially expressed proteins was conducted using the STRING database,which can predict the relations between differentially expressed proteins and CHF with Qi-deficiency-blood-stasis syndrome.RESULTS: A total of seven differentially expressed proteins were identified, and among these, transforming growth factor β1(TGF-β1) gets a particular attention for us has drawn specific attention.Network analysis showed a homologous relationship of TGF-β1 with bone morphogenetic protein15, which is associated with myocardial fibrosis.CONCLUSION: Occurrence and development of CHF may result from certain DE-proteins and associated signaling pathways. 20Genetic variation in growth traits and stem–branch characteristics and their relationships to Eucalyptus clones显示文摘Eucalyptus has become an important genus in China because it displays adaptability to a wide range of site conditions and produces pulp wood and veneer on short rotations. The aim of this study was to estimate genetic parameters and relationships and consider implications for development of clonal forestry. We assessed growth traits, stem–branch characteristics, crown diameter and height of fresh branch for 20 Eucalyptus hybrid clones in China measured at the age of 44 months. Analysis of variance showed that there were significant differences in growth traits, height of fresh branch and stem straightness among clones. Significant differences in height, volume,crown diameter, height of fresh branch and branch size among replicates were also recorded. Coefficients of variation ranged from 9.84 to 28.54 % for growth traits, 12.03 to 17.25 % for stem–branch characteristics, 18.26 % for crown diameter and 11.73 % for height of fresh branch.Estimates of repeatabilities for height, diameter at breast height over bark, volume, crown diameter, height of fresh branch, stem straightness and branch size at clone mean level were 0.86, 0.80, 0.80, 0.54, 0.85, 0.77 and 0.44 respectively. Diameter at breast height over bark and height had strongly positive phenotypic and genotypic correlations with volume, ranging from 0.96 to 1.00.
