1Genome Analysis of Dasineura jujubifolia Toursvirus 2,A Novel Ascovirus显示文摘So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic wasps.Here,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura jujubifolia.Initially,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of D.jujubifolia(Cecidomyiidae,Diptera)which is a major pest on Ziziphus jujuba.Then a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and apoptosis.Multi-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus genome.Phylogenetic analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of evolution.The results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.Jun Wang Minglu Yang Haibing Xiao Guo-Hua Huang Fei Deng Zhihong Hu 2020Virologica Sinica2020,35,2:4

4Folic acid-modified Exosome-PH20 enhances the efficiency of therapy via modulation of the tumor microenvironment and directly inhibits tumor cell metastasis显示文摘High accumulation of hyaluronan(HA)in the tumor microenvironment leads to an increase in the interstitial pressure and reduction perfusion of drugs.Furthermore,high molecular-weight(HMW)-HA suppresses M1 macrophage polarization,enhances M2 polarization,and induces immunosuppression.Hyaluronidase treatment have attempted to decrease the quantity of HA in tumors.However,hyaluronidase-driven HA degradation driven accelerates tumor cell metastasis,which is a major cause of mortality in cancer patients.Thus,we designed a novel exosome-based drug delivery system(DDS),named Exos-PH20-FA,using genetic engineering to express human hyaluronidase(PH20)and self-assembly techniques to modify the exosomes with folic acid(FA).Our results show that Exos-PH20-FA degraded HMW-HA to low-molecular-weight(LMW)-HA.Moreover,LMW-HA polarized macrophages to the M1 phenotype and reduced the number of relevant immunosuppressive immunocytes which changed the immune microenvironment from an immunosuppressive to immunosupportive phenotype.Furthermore,we demonstrated Exos-PH20-FA directly reduced hyaluronidase-induced metastasis of tumor cells.This tumor treatment also allowed an enhanced delivery of chemotherapy by tumor-targeting effect with FA modification.Our findings indicate that Exos-PH20-FA improves tumor treatment efficiency and reduces the side effects of hyaluronidase treatment,namely tumor cell metastasis.This all-in-one exosome-based HA targeting DDS maybe a promising treatment that yields more efficient and safer results.Chunxiang Feng Zhiyong Xiong Cheng Wang Wen Xiao Haibing Xiao Kairu Xie Ke Chen Huageng Liang Xiaoping Zhang Hongmei Yang 2021Bioactive Materials2021,6,4:1

6Experimental Analysis of Radial Centrifugal Pump Shutdown显示文摘Centrifugal pumps are widely used in the metallurgy,coal,and building sectors.In order to study the hydraulic characteristics of a closed impeller centrifugal pump during its shutdown in the so-called power frequency and frequency conversion modes,experiments were carried to determine the characteristic evolution of parameters such as speed,inlet and outlet pressure,head,flow rate and shaft power.A quasi-steady-state method was also used to further investigate these transient behaviors.The results show that,compared to the power frequency input,the performance parameter curves for the frequency conversion input are less volatile and smoother.The characteristic time is longer and the response to shutdown is slower.The quasi-steady-state theoretical head-flow curves match the experimental head-flow curves more closely at low flow rates when the frequency conversion input is considered.Moreover,in this case,the similarity law predicts the hydraulic performance more accurately.Xiao Sun Jiangbo Tong Yuliang Zhang Haibing Cai Wen Zhou Xiaoqi Jia Litao Ou 2024Fluid Dynamics & Materials Processing2024,20,4:0
7Drying-rewetting cycles reduce bacterial diversity and carbon loss in soil on the Loess Plateau of China显示文摘With global climate change, soil drying-rewetting(DRW) events have intensified and occurred frequently on the Loess Plateau of China. However, the extent to which the DRW cycles with different wetting intensities and cycle numbers alter microbial community and respiration is barely understood. Here,indoor DRW one and four cycles treatments were implemented on soil samples obtained from the Loess Plateau, involving increase of soil moisture from10% water-holding capacity(WHC) to 60% and 90% WHC(i.e., 10%–60% and 10%–90% WHC, respectively). Constant soil moistures of 10%, 60%,and 90% WHC were used as the controls. The results showed that bacterial diversity and richness decreased and those of fungi remained unchanged under DRW treatments compared to the controls. Under all moisture levels, Actinobacteriota and Ascomycota were the most dominant bacterial and fungal phyla,respectively. The bacterial network was more complex than that of fungi, indicating that bacteria had a greater potential for interaction and niche sharing under DRW treatments. The pulse of respiration rate declined as the DRW cycle increased under 10%–60% WHC, but remained similar for different cycles under 10%–90% WHC. Moreover, the DRW treatments reduced the overall carbon loss, and the direct carbon release under 10%–60% WHC was larger than that under 10%–90% WHC. 8Hybridization of iron phthalocyanine and MoS_(2)for high-efficiency and durable oxygen reduction reaction显示文摘Hybrid catalysts based on iron phthalocyanine(FePc)have raised much attention due to their promising applications in electrocatalytic oxygen reduction reaction(ORR).Various hybridization strategies have been developed for improving their activity and durability.However,the influence of different hybridization strategies on their catalytic performance remains unclear.In this study,Fe Pc was effectively distributed on molybdenum disulfide(MoS_(2))forming Fe Pc-based hybrid catalysts,namely Fe Pc-MoS_(2),Fe Pc*-MoS_(2),and Fe Pc-Py-MoS_(2),respectively,to disclose the related influence.Through direct hybridization,the stacked and highly dispersed Fe Pc on MoS_(2)resulted in Fe Pc-MoS_(2),and Fe Pc*-MoS_(2),respectively,in which the substrate and Fe Pc are mainly bound through van der Waals interactions.Through covalent hybridization strategy using pyridyl(Py)as a linker,Fe Pc-Py-MoS_(2)hybrid catalyst was prepared.Experimental and theoretical results disclosed that the linker hybridization of Fe Pc and MoS_(2)facilitated the exposure of Fe-N4 sites,maintained the intrinsic activity of Fe Pc by forming a more dispersed phase and increased the durability via Fe-N bonding,rendering the Fe Pc-Py-MoS_(2)an excellent ORR hybrid catalyst.Compared with van der Waals hybridized Fe Pc-MoS_(2)and Fe Pc*-MoS_(2)in alkaline media,the linker hybridized Fe Pc-Py-MoS_(2)showed an obviously enhanced ORR activity with a half-wave potential(E_(1/2))of 0.88 V vs RHE and an ultralow Tafel slope of 26 m V dec-1.Besides,the Fe Pc-Py-MoS_(2)exhibited a negligible decay of E_(1/2) after 50,000 CV cycles for ORR,showing its superior durability.This work gives us more insight into the influence of different hybrid strategies on Fe Pc catalysts and provides further guidance for the development of highly efficient and durable ORR catalysts.Haibing Meng Xiaolong Liu Xiao Chen Ying Han Chenhui Zhou Qinyuan Jiang Ting Tan Rufan Zhang 2022Journal of Energy Chemistry2022,31,8:0
