2Genome Analysis of Dasineura jujubifolia Toursvirus 2,A Novel Ascovirus显示文摘So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic wasps.Here,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura jujubifolia.Initially,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of D.jujubifolia(Cecidomyiidae,Diptera)which is a major pest on Ziziphus jujuba.Then a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and apoptosis.Multi-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus genome.Phylogenetic analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of evolution.The results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.Jun Wang Minglu Yang Haibing Xiao Guo-Hua Huang Fei Deng Zhihong Hu 2020Virologica Sinica2020,35,2:4
3Detection and quantification of enteroviruses in coastal seawaters from Bohai Bay,Tianjin,China显示文摘An 8-month survey was conducted to detect and quantify enteroviruses in Tianjin coastal seawaters of Bohai Bay to assess coastal water quality. Ten water samples were collected from Bohai Bay for the detection and quantification of enteroviruses by conventional reverse transcription polymerase chain reaction (RT-PCR) and SYBR Green real-time quantitative RT-PCR (qRT-PCR). Total viral nucleic acid was extracted from 500 mL of seawater samples concentrated by Centricon plus-70 centrifugal filter devices. The viral recovery rate was 29.1% based on viral seeding study. The centrifugal ultrafiltration method applied is effective for viral recovery from small volume of polluted water, which may have broader applications to monitoring human virus in aquatic environment. Our results indicated that there was a severe viral contamination in seawater of Bohai Bay. Enteroviruses were detected at concentrations ranging from 1.7×106 to 6.3×107 copies/L by qRT-PCR. Sequencing analyses identified that all of the twenty clones as poliovirus type 2. This is the first quantitative report of human viruses in coastal waters of a metropolitan city in China. 8Efficient control of chronic LCMV infection by a CD4 T cell epitope-based heterologous prime-boost vaccination in a murine model显示文摘CD4^(+)T cells are essential for sustaining CD8^(+)T cell responses during a chronic infection.The adoptive transfer of virus-specific CD4^(+)T cells has been shown to efficiently rescue exhausted CD8^(+)T cells.However,the question of whether endogenous virus-specific CD4^(+)T cell responses can be enhanced by certain vaccination strategies and subsequently reinvigorate exhausted CD8^(+)T cells remains unexplored.In this study,we developed a CD4^(+)T cell epitope-based heterologous prime-boost immunization strategy and examined the efficacy of this strategy using a mouse model of chronic lymphocytic choriomeningitis virus(LCMV)infection.We primed chronically LCMV-infected mice with a Listeria monocytogenes vector that expressed the LCMV glycoprotein-specific I-Ab-restricted CD4^(+)T cell epitope GP61–80(LM-GP61)and subsequently boosted the primed mice with an influenza virus A(PR8 strain)vector that expressed the same CD4^(+)T cell epitope(IAV-GP61).This heterologous prime-boost vaccination strategy elicited strong anti-viral CD4^(+)T cell responses,which further improved both the quantity and quality of the virusspecific CD8^(+)T cells and led to better control of the viral loads.The combination of this strategy and the blockade of the programmed cell death-1(PD-1)inhibitory pathway further enhanced the anti-viral CD8^(+)T cell responses and viral clearance.Thus,a heterologous prime-boost immunization that selectively induces virus-specific CD4^(+)T cell responses in conjunction with blockade of the inhibitory pathway may represent a promising therapeutic approach to treating patients with chronic viral infections.Ran He Xinxin Yang Cheng Liu Xiangyu Chen Lin Wang Minglu Xiao Jianqiang Ye Yuzhang Wu Lilin Ye 2018Cellular & Molecular Immunology2018,15,9:1

10Super-assembled sandwich-like Au@MSN@Ag nanomatrices for high-throughput and efficient detection of small biomolecules显示文摘Small biomolecules(m/z<500)are the material basis of organisms and participate in life activities,but their comprehensive and accurate detection in complex samples remains a challenge.Matrix-assisted laser desorption/ionization mass spectrometry(MALDI-MS)is a powerful detection tool for molecular analysis with high throughput.The development of a new matrix is essential to improve the efficiency of the MALDI-MS for molecular compound detection.In this work,the sandwich-like gold nanoparticles@mesoporous silica nanocomposite@silver nanoparticles(Au@MSN@Ag)nanospheres were prepared by layer-by-layer super-assembly strategy,and can be used as a novel matrix for the quantitative detection and enrichment of small biomolecules by LDI-MS.The sandwich-like nanospheres form a unique plasma resonant cavity that effectively absorbs the laser energy,while the homogeneous mesoporous structure of MSN can lock the analyte,which is essential for efficient LDI of small molecules.Compared to traditional matrices,Au@MSN@Ag shows the advantages of low background,wide application range,high sensitivity,super high salt and protein tolerance,and good stability.For example,the detection limit of glucose was as low as 5 fmol,and showed a good linear relationship in the range of 1−750μg/mL.Au@MSN@Ag assisted LDI-MS allows the enrichment and detection of small molecules in traditional Chinese medicine(TCM)without derivatization and purification,classification of herbs using the accurate quantitative results oligosaccharides,and identification of gelatin by amino acid content.This research could help in designing more efficient nanostructure matrices and further explored the application of LDI-MS.Dantong Zhao Chunxia Ma Meng Gao Yong Li Bo Yang Hui Li Runhao Zhang Minglu Hao Jing Huang Kang Liang Pu Chen Lei Xie Rong Rong Biao Kong 2022Nano Research2022,15,3:0
11Abdominal Grey Color Variation along an Elevation Gradient in a Lizard显示文摘Color variation among populations is thought to be either due to genetic variation or phenotype plasticity.In this study,we found that an agamid lizard,Phrynocephalus theobaldi,inhabiting elevations greater than any other reptile,displayed a significantly darker spotted marking on the central ventral abdomen in higher populations than in lower populations.The grey spotted area only occurred in elevations>4200 m,and also increased with increased elevation.The ratio of the size of the dark colored area to the degree of skin luminance/reflectance was used to test whether the lizard has a larger and darker spotted area at higher elevations.This ratio was significantly positively associated with elevation rather than substrate color.The lizard did not display sexual difference in this phenotype.Temperature had a significant influence on the brightness of central abdomen,but temperature-related plasticity of this trait was much smaller than the magnitude of the difference between higher(>4200 m)and lower populations(<4200 m).Common garden experiments showed that genetic inheritance,rather than phenotype plasticity,played a dominant role in this variation.Our research addressed the darkness of a body region that is rarely analyzed because it is shielded from direct insolation.The idea that ventral color could be related to thermoregulation is shown to be worthy of further research.Zian WEI Jiasheng LI Tao WANG Minglu YANG Yuanting JIN 2023Asian Herpetological Research2023,14,2:0
12Relationship between the expression of TLR4 and TIRAP and sepsis in peripheral blood显示文摘Objective:To explore the relationship between the expression of TLR4 and TIRAP and sepsis severity.Methods:The study selected 30 healthy examinees as the control group and 53 patients with sepsis as the observation group.The patients in the observation group were assessed by Acute Physiology and Chronic Health Evaluation Ⅱ(APACHE Ⅱ).40 patients with APACHE Ⅱ score≤20,were classified into the low-score sepsis group;13 patients with APACHE Ⅱ score>20,were classified into the high-score sepsis group.The levels of TLR4 and TIRAP in venous serum were detected in all subjects by enzyme-linked immunosorbent assay(ELISA).Results:The levels of TLR4 and TIRAP in serum were 0.886±0.058 ng/ml and 5.216±0.410 ng/ml in the control group;2.253±0.379 ng/ml and 9.540±2.294 ng/ml in the low-score sepsis group;4.494±0.709 ng/ml and 19.206±1.755 ng/ml in the high-score sepsis group.The observation group(low-score and high-score sepsis groups)was significantly different from the control group(p=.000),and the low-score sepsis group was significantly different from the high-score sepsis group(p=.000).With APACHE II score of 20 as the cut-off point,the low-score sepsis group was consisted of low-risk patients and the high-score group was consisted of the high-risk,which can indicate the sepsis severity.According to Pearson’s correlation analysis,the level of TLR4 was positively correlated with sepsis severity(r=0.931,p<.05),the level of TIRAP was also positively correlated with the sepsis severity(r=0.972,p<.05);the level of TLR4 was positively correlated with the level of TIRAP(r=0.936,p<.05).Conclusions:The levels of TLR4 and TIRAP in septic patients can be used to predict and determine the severity of sepsis.Minglu Qi Jingping Yang Xue Yin Si Chen Xiyuan Xu 2016Discussion of Clinical Cases2016,3,4:0
