1Dietary nano-selenium alleviated intestinal damage of juvenile grass carp(Ctenopharyngodon idella)induced by high-fat diet:Insight from intestinal morphology,tight junction,inflammation,anti-oxidization and intestinal microbiota显示文摘In recent years,high-fat diet(HFD)has been widely applied in aquaculture,which reduces the intestinal health of cultured fish.The current study evaluated the protective effects of nano-selenium(nano-Se)on intestinal health of juvenile grass carp(Ctenopharyngodon idella)fed with HFD.A total of 135 experimental fish were fed with a regular diet(Con),a HFD(HFD)and a HFD containing nano-Se at 0.6 mg/kg(HSe)for 10 weeks.The results showed that dietary nano-Se significantly improved the survival rate and feed efficiency which were reduced by HFD in juvenile grass carp(P<0.05).Also,nano-Se(0.6 mg/kg)supplement alleviated intestinal damage caused by the HFD,thus maintaining the integrity of the intestine.Moreover,it significantly up-regulated the expression of genes related to tight junction(ZO-1,claudin-3 and occludin),anti-oxidization(GPx4a and GPx4b),and the protein of ZO-1 in the intestine of juvenile grass carp,which were depressed by the HFD(P<0.05).Furthermore,nano-Se supplementation significantly suppressed the expressions of genes related to the inflammation,including inflammatory cytokines(IL-8,IL-1β,IFN-γ,TNF-αand IL-6),signaling molecules(TLR4,p38 MAPK and NF-kB p65),and protein expression of NF-kB p65 and TNF-αin the intestine of juvenile grass carp which were induced by the HFD(P<0.05).Besides,dietary nano-Se normalized the intestinal microbiota imbalance of juvenile grass carp caused by the HFD through increasing the abundance of the beneficial bacteria,e.g.,Fusobacteria.Finally,dietary nano-Se increased the production of short chain fatty acids(SCFA)in the intestine,especially for butyric acid and caproic acid,which were negatively related to the increase of intestinal permeability and inflammation.In summary,supply of nano-Se(0.6 mg/kg)in HFD could effectively alleviate intestinal injury of juvenile grass carp by improving intestinal barrier function and reducing intestinal inflammation and oxidative stress.These positive effects may be due to the regulation of nano-Se on intestinal microbiota and the subsequently increased beneficial SCFA levels.Sha Liu Haibo Yu Pengju Li Chi Wang Guohao Liu Xiaotian Zhang Cheng Zhang Meng Qi Hong Ji 2022Animal Nutrition2022,,1:4

3Construction of nanofibrous scaffolds with interconnected perfusable microchannel networks for engineering of vascularized bone tissue显示文摘Vascularization and bone regeneration are two closely related processes during bone reconstruction.A three-dimensional(3D)scaffold with porous architecture provides a suitable microenvironment for vascular growth and bone formation.Here,we present a simple and general strategy to construct a nanofibrous poly(L-lactide)/poly(ε-caprolactone)(PLLA/PCL)scaffold with interconnected perfusable microchannel networks(IPMs)based on 3D printing technology by combining the phase separation and sacrificial template methods.The regular and customizable microchannel patterns within the scaffolds(spacings:0.4 mm,0.5 mm,and 0.6 mm;diameters:0.8 mm,1 mm,and 1.2 mm)were made to investigate the effect of microchannel structure on angiogenesis and osteogenesis.The results of subcutaneous embedding experiment showed that 0.5/0.8-IPMs(spacing/diameter=0.5/0.8)and 0.5/1-IPMs(spacing/diameter=0.5/1)scaffolds exhibited more vascular network formation as compared with other counterparts.After loading with vascular endothelial growth factor(VEGF),VEGF@IPMs-0.5/0.8 scaffold prompted better human umbilical vein endothelial cells(HUVECs)migration and neo-blood vessel formation,as determined by Transwell migration,scratch wound healing,and chorioallantoic membrane(CAM)assays.Furthermore,the microangiography and rat cranial bone defects experiments demonstrated that VEGF@IPMs-0.5/0.8 scaffold exhibited better performance in vascular network formation and new bone formation compared to VEGF@IPMs-0.5/1 scaffold.In summary,our results suggested that the microchannel structure within the scaffolds could be tailored by an adjustable caramel-based template strategy,and the combination of interconnected perfusion microchannel networks and angiogenic factors could significantly enhance vascularization and bone regeneration.Jiani Gu Qianqian Zhang Mengru Geng Weizhong Wang Jin Yang Atta ur Rehman Khan Haibo Du Zhou Sha Xiaojun Zhou Chuanglong He 2021Bioactive Materials2021,6,10:1

6Safety and feasibility of laparoscopic surgery for colorectal and gastric cancer under the Chinese multi-site practice policy:admittance standards of competence are needed显示文摘Background:The multi-site practice(MSP)policy has been practiced in China over 10 years.This study aimed to investigate the safety and feasibility of performing laparoscopic surgery for colorectal cancer(LSCRC)and gastric cancer(LSGC)under the Chinese MSP policy.Methods:We collected and analysed the data from 1,081 patients who underwent LSCRC or LSGC performed by one gastrointestinal surgeon in his original hospital(n=573)and his MSP institutions(n=508)between January 2017 and December 2020.Baseline demographics,intraoperative outcomes,post-operative recovery,and pathological results were compared between the original hospital and MSP institutions,as well as between MSP institutions with and without specific competence(surgical skill,operative instrument,perioperative multi-discipline team).Results:In our study,690 patients underwent LSCRC and 391 patients underwent LSGC.The prevalence of post-operative complications was comparable for LSCRC(11.5%vs 11.1%,P=0.89)or LSGC(15.2%vs 12.6%,P=0.46)between the original hospital and MSP institutions.However,patients in MSP institutions without qualified surgical assistant(s)and adequate instruments experienced longer operative time and greater intraoperative blood loss.The proportion of patients with inadequate lymph-node yield was significantly higher in MSP institutions than in the original hospital for both LSCRC(11.5%vs 21.2%,P<0.01)and LSGC(9.8%vs 20.5%,P<0.01).Conclusion:For an experienced gastrointestinal surgeon,performing LSCRC and LSGC outside his original hospital under the MSP policy is safe and feasible,but relies on the precondition that the MSP institutions are equipped with qualified surgical skills,adequate operative instruments,and complete perioperative management.Zhenghao Cai Haiqin Song Zhenfeng Huang Abraham Fingerhut Ximo Xu Hao Zhong Zhigang Li Yingjie Zhang Dachong Sha Dandan Bao Haibo Wang Binghua Cai Shangbo Hua Yanhui Zhang Jianguang Sun Ke Ye Jianwen Li Yong Lu Bo Feng 2022Gastroenterology Report2022,10,1:0
