Induced regulatory T cells suppress Tel cells through TGF-β signaling to ameliorate STZ-induced type 1 diabetes mellitus显示文摘Type 1 diabetes mellitus(T1D)is a chronic autoimmune condition in which the immune system destroys insulin-producing pancreatic β cells. In addition to well-established pathogenic effector T cells, regulatory T cells(Tregs)have also been shown to be defective in T1D.Thus,an increasing number of therapeutic approaches are being developed to target Tregs. However, the role and mechanisms of TGF-β-induced Tregs(iTregs)in T1D remain poorly understood. Here, using a streptozotocin(STZ)-induced preclinical T1D mouse model,we found that iTregs could ameliorate the development of T1D and preserve β cell function. The preventive effect was associated with the inhibition of type 1 cytotoxic T(Tel)cell function and rebalancing the Treg/Tc1 cell ratio in recipients.Furthermore,we showed that the underlying mechanisms were due to the TGF-β-mediated combinatorial actions of mTOR and TCF1.In addition to the preventive role, the therapeutic effects of iTregs on the established STZ-T1D and nonobese diabetic(NOD)mouse models were tested, which revealed improved β cell function. Our findings therefore provide key new insights into the basic mechanisms involved in the therapeutic role of iTregs in T1D.Li Zhou Xuemin He Peihong Cai Ting Li Rongdong Peng Junlong Dang Yue Li Haicheng Li Feng Huang Guojun Shi Chichu Xie Yan Lu Yanming Chen 2021Cellular & Molecular Immunology2021,18,3:3
Nicotinamide mononucleotide improves spermatogenic function in streptozotocin-induced diabetic mice via modulating the glycolysis pathway显示文摘Spermatogenic dysfunction is one of the major secondary complications of diabetes;however, the underlying mechanisms remain ill-defined,and there is no available drug or strategy for the radical treatment of diabetic spermatogenic dysfunction. Therefore, the objective of this study is to investigate the protective effects of nicotinamide mononucleotide(NMN)on testicular spermatogenic function in streptozotocin(STZ)-induced diabetic mice. The results show that oral administration of NMN significantly increases the body and testis weight and the number of sperms. Moreover, the abnormal sperm count and the rate of sperm malformation are significantly decreased compared with the saline-treated diabetic mice.Histological analysis reveals that NMN treatment significantly increases the area and diameter of seminiferous tubules, accompanied by an increased number of spermatogenic cells and sperms.Immunohistochemistry and qRT-PCR results show that NMN increases Bcl-2 expression and decreases Bax expression in the testis.NMN also increases the protein expression of Vimentin and the mRNA expressions of WT1 and GATA4.In addition,qRT-PCR,western blot analysis and immunohistochemistry results also show that NMN increases the expressions of glycolysis-related rate-limiting enzymes including HK2,PKM2,and LDHA.In summary, this study demonstrates the protective effects of NMN on the testis in an STZ-induced diabetic mice model.NMN exerts its protective effects via reducing spermatogenic cell apoptosis by regulating glycolysis of Sertoli cells in diabetic mice. This study provides an experimental basis for the future clinical application of NMN in diabetes-induced spermatogenicdysfunction.Duo Ma Linlin Hu Jinyuan Wang Min Luo Aihong Liang Xiaocan Lei Biyun Liao Meixiang Li Ming Xie Haicheng Li Yiwei Gong Dan Zi Xiangrun Li Xi Chen Xucai Liao 2022Acta Biochimica et Biophysica Sinica2022,54,9:1

Enabling dendrite-free charging for lithium batteries based on transport-reaction competition mechanism in CHAIN framework显示文摘Worldwide trends in mobile electrification will skyrocket demands for lithium-based battery production, driven by the popularity of electric vehicles. However, both lithium metal batteries and lithium ion batteries face severe safety issues due to dendrite nucleation and growth process.Li deposition is significantly influenced by interfacial factors and charging conditions. In this paper,an electrochemical model considering the internal and external factors is proposed based on Monte Carlo method. The influence of internal solid electrolyte interphase(SEI)porosity, thickness and the external conditions on dendrite growth process is systematically described. The simulation results support that the three factors investigated in this model could synergistically regulate the dendrite growth process.Three competition mechanisms are proposed to tailor lithium deposition for Li-based batteries and numerical solutions for variation pattern of dendrite growth with time are fitted.A three-step process describing kinetic process of lithium deposition is proposed. To achieve dendrite-free charging process, charging strategies and emerging materials design should be considered, including physicochemical materials engineering, artificial SEI,and design for dynamic safety boundary. This work could contribute to the foundation for insights of Li deposition mechanism, which is promising to provide guidelines for next-generation high-energy-density and safe batteries in CHAIN framework.Lisheng Zhang Siyan Chen Wentao Wang Hanqing Yu Haicheng Xie Huizhi Wang Shichun Yang Cheng Zhang Xinhua Liu 2022Journal of Energy Chemistry2022,,12:1
A Study on Genotype-by-Environment Interaction Analysis for Agronomic Traits of Maize Genotypes Across Huang-Huai-Hai Region in China显示文摘Facing the trend of increasing population, how to increase maize grain yield is a very important issue to ensure food security. In this study,28 nationally approved maize hybrids were evaluated across 24 different climatic conditions for two consecutive years(2018-2019).The purpose of this study was to select high-yield with stable genotypes and identify important agronomic traits for maize breeding program improvement. The results of this study showed that the genotypeenvironment interaction effects of the 12 evaluated agronomic traits was highly significant(P<0.001).We introduced a novel multi-trait genotype-ideotype distance index(MGIDI)to select genotypes based on multiple agronomic traits. The selection process exhibited by this method is unique and easy to understand,so the MGIDI index will have more and more important applications in future multi-environment trials(METs)research. The genotypes selected by the MGIDI index were G22,G10,G12 and G1 as the high yielding and stable genotypes. The parents of these selected genotypes have the ability to play a greater role as the basic germplasm in the breeding process.A new form of genotype(G)main effects and genotype(G)-by-environment(E)interaction(GGE)technician,genotypeyieldtrait(GYT)biplot,based on multiple traits for genotypes selection was also applied in this study. The GYT biplot ranked genotypes by combining grain yield with other evaluated agronomic traits, and displayed the distribution of their traits, namely strengths and weaknesses.Haiwang Yue Jianwei Wei Junliang Xie Shuping Chen Haicheng Peng Hongmei Cao Junzhou Bu Xuwen Jiang 2022Phyton-International Journal of Experimental Botany2022,91,1:1
Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images. Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases, comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance. Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. The average performance of all radiologists was also improved to varying degrees with MDCS assistance. Conclusions: MDCS demonstrated excellent performance in the detection and classification of breast lesions, and greatly enhanced the overall performance of radiologists.Yuqian Chen Zhen Hua Fan Lin Tiantian Zheng Heng Zhou Shijie Zhang Jing Gao Zhongyi Wang Huafei Shao Wenjuan Li Fengjie Liu Simin Wang Yan Zhang Feng Zhao Hao Liu Haizhu Xie Heng Ma Haicheng Zhang Ning Mao 2023Chinese Journal of Cancer Research2023,35,4:0

Toward a stellar population catalog in the Kilo Degree Survey:The impact of stellar recipes on stellar masses and star formation rates显示文摘The Kilo Degree Survey(Ki DS)is currently the only sky survey providing optical(ugri)plus near-infrared(NIR,ZY H JKS)seeing matched photometry over an area larger than 1000 deg2.This is obtained by incorporating the NIR data from the VISTA Kilo Degree Infrared Galaxy(VIKING)survey, covering the same Ki DS footprint. As such, the Ki DS multi-wavelength photometry represents a unique dataset to test the ability of stellar population models to return robust photometric stellar mass(M_(*))and star-formation rate(SFR)estimates. Here we use a spectroscopic sample of galaxies for which we possess ugri ZY JHK_(s)“gaussianized”magnitudes from Ki DS data release 4.We fit the spectral energy distribution from the 9-band photometry using:(1)three different popular libraries of stellar population templates,(2)single burst, simple and delayed exponential star-formation history models,and(3)a wide range of priors on age and metallicity. As template fitting codes we use two popular softwares:Le Phare and CIGALE.We investigate the variance of the stellar masses and the star-formation rates from the different combinations of templates, star formation recipes and codes to assess the stability of these estimates and define some“robust”median quantities to be included in the upcoming Ki DS data releases. As a science validation test,we derive the mass function, the star formation rate function, and the SFR-M_(*)relation for a low-redshift(z<0.5)sample of galaxies, that result in excellent agreement with previous literature data. The final catalog,containing~290000 galaxies with redshift 0.01Linghua Xie Nicola R.Napolitano Xiaotong Guo Crescenzo Tortora Haicheng Feng Antonios Katsianis Rui Li Sirui Wu Mario Radovich Leslie K.Hunt Yang Wang Lin Tang Baitian Tang Zhiqi Huang 2023Science China(Physics,Mechanics & Astronomy)2023,66,12:0
