| 1 | Induced regulatory T cells suppress Tel cells through TGF-β signaling to ameliorate STZ-induced type 1 diabetes mellitus显示文摘Type 1 diabetes mellitus(T1D)is a chronic autoimmune condition in which the immune system destroys insulin-producing pancreatic β cells.In addition to well-established pathogenic effector T cells,regulatory T cells(Tregs)have also been shown to be defective in T1D.Thus,an increasing number of therapeutic approaches are being developed to target Tregs.However,the role and mechanisms of TGF-β-induced Tregs(iTregs)in T1D remain poorly understood.Here,using a streptozotocin(STZ)-induced preclinical T1D mouse model,we found that iTregs could ameliorate the development of T1D and preserve β cell function.The preventive effect was associated with the inhibition of type 1 cytotoxic T(Tel)cell function and rebalancing the Treg/Tc1 cell ratio in recipients.Furthermore,we showed that the underlying mechanisms were due to the TGF-β-mediated combinatorial actions of mTOR and TCF1.In addition to the preventive role,the therapeutic effects of iTregs on the established STZ-T1D and nonobese diabetic(NOD)mouse models were tested,which revealed improved β cell function.Our findings therefore provide key new insights into the basic mechanisms involved in the therapeutic role of iTregs in T1D. | Li Zhou Xuemin He Peihong Cai Ting Li Rongdong Peng Junlong Dang Yue Li Haicheng Li Feng Huang Guojun Shi Chichu Xie Yan Lu Yanming Chen | 2021 | Cellular & Molecular Immunology2021,18,3: | 3 |
| 2 | Validation of novel hub genes and molecular mechanisms in acute lung injury using an integrative bioinformatics approach显示文摘Acute lung injury(ALI)is an inflammatory pulmonary disease that can easily develop into serious acute respiratory distress syndrome,which has high morbidity and mortality.However,the molecular mechanism of ALI remains unclear,and few molecular biomarkers for diagnosis and treatment have been identified.In this study,we aimed to identify novel molecular biomarkers using a bioinformatics approach.Gene expression data were obtained from the Gene Expression Omnibus database,co-expressed differentially expressed genes(CoDEGs)were identified using R software,and further functional enrichment analyses were conducted using the online tool Database for Annotation,Visualization,and Integrated Discovery.A protein-protein interaction network was established using the STRING database and Cytoscape software.Lipopolysaccharide(LPS)-induced ALI mouse model was constructed and verified.The hub genes were screened and validated in vivo.The transcription factors(TFs)and miRNAs associated with the hub genes were predicted using the NetworkAnalyst database.In total,71 CoDEGs were screened and found to be mainly involved in the cytokine-cytokine receptor interactions,and the tumor necrosis factor and malaria signaling pathways.Animal experiments showed that the lung injury score,bronchoalveolar lavage fluid protein concentration,and wet-to-dry weight ratio were higher in the LPS group than those in the control group.Real-time polymerase chain reaction analysis indicated that most of the hub genes such as colony-stimulating factor 2(Csf2)were overexpressed in the LPS group.A total of 20 TFs including nuclear respiratory factor 1(NRF1)and two miRNAs were predicted to be regulators of the hub genes.In summary,Csf2 may serve as a novel diagnostic and therapeutic target for ALI.NRF1 and mmu-mir-122-5p may be key regulators in the development of ALI. | Qingchun Liang Qin Zhou Jinhe Li Zhugui Chen Zhihao Zhang Ruimeng Liu Haicheng Huang Zhiyong Peng Youtan Liu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,3: | 1 |
| 3 | A Study on Genotype-by-Environment Interaction Analysis for Agronomic Traits of Maize Genotypes Across Huang-Huai-Hai Region in China显示文摘Facing the trend of increasing population,how to increase maize grain yield is a very important issue to ensure food security.In this study,28 nationally approved maize hybrids were evaluated across 24 different climatic conditions for two consecutive years(2018-2019).The purpose of this study was to select high-yield with stable genotypes and identify important agronomic traits for maize breeding program improvement.The results of this study showed that the genotypeenvironment interaction effects of the 12 evaluated agronomic traits was highly significant(P<0.001).We introduced a novel multi-trait genotype-ideotype distance index(MGIDI)to select genotypes based on multiple agronomic traits.The selection process exhibited by this method is unique and easy to understand,so the MGIDI index will have more and more important applications in future multi-environment trials(METs)research.The genotypes selected by the MGIDI index were G22,G10,G12 and G1 as the high yielding and stable genotypes.The parents of these selected genotypes have the ability to play a greater role as the basic germplasm in the breeding process.A new form of genotype(G)main effects and genotype(G)-by-environment(E)interaction(GGE)technician,genotypeyieldtrait(GYT)biplot,based on multiple traits for genotypes selection was also applied in this study.The GYT biplot ranked genotypes by combining grain yield with other evaluated agronomic traits,and displayed the distribution of their traits,namely strengths and weaknesses. | Haiwang Yue Jianwei Wei Junliang Xie Shuping Chen Haicheng Peng Hongmei Cao Junzhou Bu Xuwen Jiang | 2022 | Phyton-International Journal of Experimental Botany2022,91,1: | 1 |
| 4 | Correlating the electronic structures of metallic/semiconducting MoTe_(2)interface to its atomic structures显示文摘Contact interface properties are important in determining the performances of devices that are based on atomically thin two-dimensional(2D)materials,especially for those with short channels.Understanding the contact interface is therefore important to design better devices.Herein,we use scanning transmission electron microscopy,electron energy loss spectroscopy,and first-principles calculations to reveal the electronic structures within the metallic(1T)-semiconducting(2H)MoTe_(2)coplanar phase boundary across a wide spectral range and correlate its properties to atomic structures.We find that the 2 H-MoTe_(2)excitonic peaks cross the phase boundary into the 1 T phase within a range of approximately 150 nm.The1 T-MoTe_(2)crystal field can penetrate the boundary and extend into the 2H phase by approximately two unit-cells.The plasmonic oscillations exhibit strong angle dependence,that is a red-shift ofπ+σ(approximately 0.3–1.2 e V)occurs within 4 nm at 1 T/2 H-MoTe_(2)boundaries with large tilt angles,but there is no shift at zero-tilted boundaries.These atomic-scale measurements reveal the structure–property relationships of the 1 T/2 H-MoTe_(2)boundary,providing useful information for phase boundary engineering and device development based on 2 D materials. | Bo Han Chen Yang Xiaolong Xu Yuehui Li Ruochen Shi Kaihui Liu Haicheng Wang Yu Ye Jing Lu Dapeng Yu Peng Gao | 2021 | National Science Review2021,8,2: | 0 |
| 5 | Pharmacognosy of Brazilian Ginseng ( Pfaffia paniculata)显示文摘[Objective] This study aimed to explore the pharmacognostical features of Brazilian ginseng( Pfaffia paniculata),and to provide a theoretical basis for its development and utilization. [Method] Morphological traits of Brazilian ginseng root were observed with traditional methods. Then,the root,stem and leaf of Brazilian ginseng were sliced,and observed under a microscope. The physical and chemical properties of root extract were detected. [Result] Brazilian ginseng root is elongated and cylindrical,with obvious longitudinal ridges and sparse root marks,some irregularly shaped lenticels on the surface. The cork layer of crosssection has five or more rows of cork cells. The abnormal vascular bundles are arranged in two to three rings. There are two to three central vascular bundles. The wood fibers in powder are long,with one bifurcate end. The vessels have bordered pits. The sand crystals are scattered or in the parenchyma surrounding the fibers.The mucilage cells are oval and widely exist. Physical and chemical experiments showed that the root of Brazilian ginseng contains sterols or triterpenes and saponins.
