1Loss of VAPB Regulates Autophagy in a Beclin 1-Dependent Manner显示文摘Autophagy is an evolutionarily-conserved selfdegradative process that maintains cellular homeostasis by eliminating protein aggregates and damaged organelles.Recently, vesicle-associated membrane protein-associated protein B(VAPB), which is associated with the familial form of amyotrophic lateral sclerosis, has been shown to regulate autophagy. In the present study, we demonstrated that knockdown of VAPB induced the up-regulation of beclin 1 expression, which promoted LC3(microtubuleassociated protein light chain 3) conversion and the formation of LC3 puncta, whereas overexpression of VAPB inhibited these processes. The regulation of beclin1 by VAPB was at the transcriptional level. Moreover,knockdown of VAPB increased autophagic flux, which promoted the degradation of the autophagy substrate p62 and neurodegenerative disease proteins. 2Familial Parkinson's Disease-Associated L166P Mutant DJ-1 is Cleaved by Mitochondrial Serine Protease Omi/HtrA2显示文摘Parkinson's disease(PD) is the most common neurodegenerative movement disorder. Mutations in the DJ-1, including L166 P, are responsible for recessive earlyonset PD. Many lines of evidence have shown that L166 P is not only a loss-of-function mutant, but also a proapoptotic-like protein that results in mitochondrial dysfunction. L166 P has been reported to be unstable and to mislocalize to mitochondria. However, the mechanisms underlying the instability of L166 P compared to wild-type DJ-1 remain largely unknown. Here, we showed that Omi/Htr A2, a mitochondrial serine protease that has also been linked to the pathogenesis of PD, contributed to L166 P instability. Omi directly interacted with and cleaved L166 P in mitochondria to decrease the L166 P level. However,Omi did not bind and cleave wild-type DJ-1. Moreover,Omi cleaved L166 P at both serine residues 3 and 121,while L166 P-induced cell death under H_2O_2 treatment was alleviated by over-expression of Omi. 3α-Synuclein aggregation and transmission in Parkinson’s disease: a link to mitochondria and lysosome显示文摘The presence of intraneuronal Lewy bodies(LBs) and Lewy neurites(LNs) in the substantia nigra(SN) composed of aggregatedα-synuclein(α-syn) has been recognized as a hallmark of pathological changes in Parkinson’s disease(PD). Numerous studies have shown that aggregated α-syn is necessary for neurotoxicity. Meanwhile, the mitochondrial and lysosomal dysfunctions are associated with α-syn pathogenicity. The hypothesis that α-syn transmission in the human brain contributes to the instigation and progression of PD has provided insights into PD pathology. 4Poly-PR in C9ORF72-Related Amyotrophic Lateral Sclerosis/Frontotemporal Dementia Causes Neurotoxicity by Clathrin-Dependent Endocytosis显示文摘GGGGCC repeat expansions in the C9 ORF72 gene are the most common cause of amyotrophic lateral sclerosis and frontotemporal dementia(c9 ALS/FTD). It has been reported that hexanucleotide repeat expansions in C9 ORF72 produce five dipeptide repeat(DPR) proteins by an unconventional repeat-associated non-ATG(RAN)translation. Within the five DPR proteins, poly-PR and poly-GR that contain arginine are more toxic than the other DPRs(poly-GA, poly-GP, and poly-PA). Here, we demonstrated that poly-PR peptides transferred into cells by endocytosis in a clathrin-dependent manner, leading to endoplasmic reticulum stress and cell death. In SH-SY5 Y cells and primary cortical neurons, poly-PR activated JUN amino-terminal kinase(JNK) and increased the levels of p53 and Bax. The uptake of poly-PR peptides by cells was significantly inhibited by knockdown of clathrin or by chlorpromazine, an inhibitor that blocks clathrin-mediated endocytosis. Inhibition of clathrin-dependent endocytosis by chlorpromazine significantly blocked the transfer of poly-PR peptides into cells, and attenuated poly-PRinduced JNK activation and cell death. 6Regulation of autophagic flux by CHIP显示文摘Autophagy is a major degradation system which processes substrates through the steps of autophagosome formation, autophagosome-lysosome fusion, and substrate degradation. Aberrant autophagic flux is present in many pathological conditions including neurodegeneration and tumors. CHIP/STUB1, an E3 ligase, plays an important role in neurodegeneration. In this study, we identified the regulation of autophagic flux by CHIP(carboxy-terminus of Hsc70-interacting protein). Knockdown of CHIP induced autophagosome formation through increasing the PTEN protein level and decreasing the AKT/m TOR activity as well as decreasing phosphorylation of ULK1 on Ser757. However, degradation of the autophagic substrate p62 was disturbed by knockdown of CHIP, suggesting an abnormality of autophagic flux. Furthermore, knockdown of CHIP increased the susceptibility of cells to autophagic cell death induced by bafilomycin A1. 8Association of Glial Activation and α-Synuclein Pathology in Parkinson's Disease显示文摘The accumulation of pathological α-synuclein(α-syn)in the central nervous system and the progressive loss of dopaminergic neurons in the substantia nigra pars compacta are the neuropathological features of Parkinson's disease(PD).Recently,the findings of prion-like transmission of α-syn pathology have expanded our understanding of the region-specific distribution ofα-syn in PD patients.Accumulating evidence suggests that α-syn aggregates are released from neurons and endocytosed by glial cells,which contributes to the clearance of α-syn.However,the activation of glial cells by α-syn species produces pro-inflammatory factors that decrease the uptake of α-syn aggregates by glial cells and promote the transmission of α-syn between neurons,which promotes the spread of α-syn pathology.In this article,we provide an overview of current knowledge on the role of glia and α-syn pathology in PD pathogenesis,highlighting the relationships between glial responses and the spread ofα-syn pathology.Rui Wang Haigang Ren Elena Kaznacheyeva Xiaojun Lu Guanghui Wang 2023Neuroscience Bulletin2023,39,3:1
9Energy Efficiency Analysis of Oxy-Fuel Circulating Fluidized Bed Combustion System with High Oxygen Concentration显示文摘The low net efficiency of oxy-fuel circulating fluidized bed(CFB)combustion is mainly due to the addition of air separation unit(ASU)and carbon dioxide compression and purification unit(CPU).High oxygen concentration is one of the effective methods to improve the net efficiency of oxy-fuel combustion technology in CFB.In this research,a series of calculation and simulation were carried out based on Aspen Plus platform to provide valuable information for further investigation on the CFB oxy-fuel combustion system with high oxygen concentration(40%,50%).A CFB oxy-fuel combustion system model with high oxygen concentration was established including ASU,CPU and CFB oxy-fuel combustion and heat exchange unit.Based on the simulation data,energy and exergy efficiency were analyzed to obtain the following results.The cross-sectional area of furnace and tail flue of 50%CFB oxy-fuel combustion boiler are 43%and 56%of the original size respectively,reducing the construction and investment cost effectively.With the increase of oxygen concentration,the net efficiency of power generation increased significantly,reaching 24.85%and increasing by 6.09%under the condition of 50%oxy-fuel combustion.The total exergy loss increases with the increase of oxygen concentration.In addition,the exergy loss of radiation heat transfer is far higher than convection heat transfer.KONG Runjuan LI Wei WANG Haigang REN Qiangqiang 2023Journal of Thermal Science2023,32,5:0
10Role of the C9ORF72 Gene in the Pathogenesis of Amyotrophic Lateral Sclerosis and Frontotemporal Dementia显示文摘Since the discovery of the C9ORF72 gene in2011,great advances have been achieved in its genetics and in identifying its role in disease models and pathological mechanisms;it is the most common genetic cause of amyotrophic lateral sclerosis(ALS) and frontotemporal dementia(FTD).ALS patients with C9ORF72 expansion show heterogeneous symptoms.Those who are C9ORF72 expansion carriers have shorter survival after disease onset than non-C9ORF72 expansion patients.Pathological and clinical features of C9ORF72 patients have been well mimicked via several models,including induced pluripotent stem cell-derived neurons and transgenic mice that were embedded with bacterial artificial chromosome construct and that overexpressing dipeptide repeat proteins.The mechanisms implicated in C9ORF72 pathology include DNA damage,changes of RNA metabolism,alteration of phase separation,and impairment of nucleocytoplasmic transport,which may underlie C9ORF72 expansion-related ALS/FTD and provide insight into nonC9ORF72 expansion-related ALS,FTD,and other neurodegenerative diseases.Zongbing Hao Rui Wang Haigang Ren Guanghui Wang 2020Neuroscience Bulletin2020,36,9:0
11Effects of Soil C/N Ratio on Apple Growth and Nitrogen Utilization,Residue and Loss显示文摘Soil C /N ratio is an important influencing factor in soil nitrogen cycling. Two-year old apple trees( Borkh. cv. ‘Fuji'/Malus hupehensis) were used to understand the effect of soil C/N ratio [6. 52( CK),10,15,20,25,30,35 and 40]on apple growth and nitrogen utilization and loss by using15N trace technique. The results showed that,with the increasing of soil C/N ratio,apple shoot length and fresh weight increased at first,and then decreased; the higher apple shoot length and fresh weight appeared in C/N = 15,20 and 25 treatments,and there were no significant differences among these three treatments,but significantly higher than the other treatments. Statistical analysis revealed that there was significant difference in nitrogen utilization rate between the different treatments,the highest N utilization rate was occurred in soil C/N = 25 treatment which value was 22. 87%,and there was no significant difference between soil C/N = 25 and C/N = 20 treatments,but both the two treatments were significantly higher than the other treatments; Soil C/N = 40 had the lowest N utilization rate which value was 15. 43%,and this value was less than CK( 16. 65%). The proportion of plant absorption nitrogen from fertilizer was much higher when the value of soil C/N ratio in the range of 15- 25,but the percentage of plant absorption nitrogen from soil was much higher when the soil C/N ratio was too low( < 15) or high( < 25). Amount of residual nitrogen in soil increased gradually with the soil C/N ratio increasing,the amount of residual nitrogen in C/N = 40 treatment was 1. 32 times than that in CK. With the increasing of soil C/N ratio,fertilizer nitrogen loss decreased at first,and then increased,fertilizer nitrogen loss was the minimum in C/N = 25 treatments( 49. 87%) and the maximum were occurred in CK( 61. 54%).
