| 1 | Towards enriching and isolation of uncultivated archaea from marine sediments using a refined combination of conventional microbial cultivation methods显示文摘The archaea that can be readily cultivated in the laboratory are only a small fraction of the total diversity that exists in nature.Although molecular ecology methods,such as metagenomic sequencing,can provide valuable information independent of cell cultivation,it is only through cultivation-based experiments that they may be fully characterized,both for their physiological and ecological properties.Here,we report our efforts towards enriching and isolation of uncultivated archaea from marine sediments using a refined combination of conventional microbial cultivation methods.Initially,cells were retrieved from the sediment samples through a cell extraction procedure and the sediment-free mixed cells were then divided into different size-range fractions by successive filtration through 0.8µm,0.6µm and 0.2µm membranes.Archaeal 16S rRNA gene analyses indicated noticeable retention of different archaeal groups in different fractions.For each fraction,supplementation with a variety of defined substrates(e.g.,methane,sulfate,and lignin)and stepwise dilutions led to highly active enrichment cultures of several archaeal groups with Bathyarchaeota most prominently enriched.Finally,using a roll-bottle technique,three co-cultures consisting of Bathyarchaeota(subgroup-8)and a bacterial species affiliated with either Pseudomonas or Glutamicibacter were obtained.Our results demonstrate that a combination of cell extraction,size fractionation,and roll-bottle isolation methods could be a useful protocol for the successful enrichment and isolation of numerous slow-growing archaeal groups from marine sediments. | Haining Hu Vengadesh Perumal Natarajan Fengping Wang | 2021 | Marine Life Science & Technology2021,3,2: | 2 |

| 3 | Widespread Bathyarchaeia encode a novel methyltransferase utilizing lignin-derived aromatics显示文摘Lignin degradation is a major process in the global carbon cycle across both terrestrial and marine ecosystems.Bathyarchaeia,which are among the most abundant microorganisms in marine sediment,have been proposed to mediate anaerobic lignin degradation.However,the mechanism of bathyarchaeial lignin degradation remains unclear.Here,we report an enrichment culture of Bathy-archaeia,named Candidatus Baizosediminiarchaeum ligniniphilus DL1YTT001(Ca.B.ligniniphilus),from coastal sediments that can grow with lignin as the sole organic carbon source under mesophilic anoxic conditions.Ca.B.ligniniphilus possesses and highly expresses novel methyltransferase 1(MT1,mtgB)for transferring methoxyl groups from lignin monomers to cob(I)alamin.MtgBs have no homology with known microbial methyltransferases and are present only in bathyarchaeial lineages.Heterologous expression of the mtgB gene confirmed O-demethylation activity.The mtgB genes were identified in metagenomic data sets from a wide range of coastal sediments,and they were highly expressed in coastal sediments from the East China Sea.These findings suggest that Bathyarchaeia,capable of O-demethylation via their novel and specific methyltransferases,are ubiquitous in coastal sediments. | Tiantian Yu Haining Hu Xianhong Zeng Yinzhao Wang Donald Pan Longhui Deng Lewen Liang Jialin Hou Fengping Wang | 2023 | mLife2023,2,3: | 0 |
| 4 | Enzymatic pretreatment mitigates the dissemination of antibiotic resistance genes via regulating microbial populations and gene expressions during food waste fermentation显示文摘Food waste(FW)has been recognized as essential reservoir for resource recovery via anaerobic fermentation,which could also bring the potential risk of antibiotic resistance genes(ARGs)dissemination.Although the structural deficiency of FW could be stimulated by enzymatic pretreatment to enhance fermentation efficiency,the influences of enzymatic pretreatment on ARGs fate and microbial metabolic pathways involved in ARGs dissemination have rarely been reported.This work proved that enzymatic pretreatment could effectively decrease the total abundance of ARGs(reduced by 13.8%-24.5%)during long-term FW fermentation.It was found that enzymatic pretreatment significantly reduced the ARGs belonging to the efflux pump,which might be ascribed to its ability to increase membrane permeability.Furthermore,enzymatic pretreatment was in favor of reducing microbial diversity and various potential ARGs host(e.g.,Methanosarcina,Clostridium,Prevotella,Parabacteroides).Also,this pretreatment remarkably up-regulated the genetic expressions involved in ABC transporter(e.g.,eryF and mntA)and down-regulated the genetic expressions that participated in DNA replication,two-component systems(e.g.,uphA and cckA),and quorum sensing(e.g.,rpfF and lsrG),thereby decreasing ARGs transmission.This study would expand the insight of the influences of pretreatment method on ARGs fate during FW fermentation,and offer practical guidance on the sustainable management of FW. | Yang Wu Wanying Hu Haining Huang Xiong Zheng Lei Dong Yinguang Chen | 2023 | Chinese Chemical Letters2023,34,8: | 0 |
| 5 | Polystyrene nanoplastics induce glycolipid metabolism disorder via NF-κB and MAPK signaling pathway in mice显示文摘Nanoplastics-induced developmental and reproductive toxicity,neurotoxicity and immunotoxicity are a focus of widespread attention.However,the effects of nanoplastics(NPs)on glycolipid metabolism and the precise underlying mechanisms are unclear at present.Here,we showed that oral administration of polystyrene nanoparticles(PS-NPs)disrupts glycolipid metabolism,with reactive oxygen species(ROS)identified as a potential key signaling molecule.After PS-NPs treatment,excessive production of ROS induced the infammatory response and activated the antioxidant pathway through nuclear factor-erythroid factor 2-related factor 2.The activation of nuclear factor-κB(NFκB)signaling pathway induced the phosphorylation of the mitogen-activated protein kinases(MAPK)signaling pathway,which induced the activation of extracellular regulated kinases(ERK)and p38.Constitutive activation of the MAPK signaling proteins induced high continued phosphorylation of insulin receptor substrate-1,in turn,leading to decreased protein kinase B(Akt)activity,which weakened the sensitivity of liver cells to insulin signals and induced insulin resistance.In parallel,phosphorylation of Akt led to loss of control of Fo XO1,a key gene of gluconeogenesis,activating transcription of glucose-6-phosphatase(G6PC)and phosphoenolpyruvate carboxykinase(PEPCK)in a manner dependent on PGC1α.Moreover,the activated ERK promoted lipid accumulation through ERK-PPARγcascades.Therefore,sterol regulatory element-binding protein-1 and levels of its downstream lipogenic enzymes,ACC-1,were up-regulated.Upon treatment with the antioxidant resveratrol,PS-NPs-induced glucose and lipid metabolic disorders were improved by inhibiting ROS-induced activation of NFκB and MAPK signaling pathway in mice.Based on above,PS-NPs exposure disrupts glycolipid metabolism in mice,with ROS identified as a potential key signaling molecule. | Xingpei Fan Xiaoyan Li Jiaxin Li Yuxia Zhang Xiangjuan Wei Hailong Hu Boya Zhang Haining Du Meimei Zhao Ruijiao Zhu Daqian Yang Yuri Oh Ning Gu | 2024 | Journal of Environmental Sciences2024,,3: | 0 |

| 8 | Expanding Asgard members in the domain of Archaea sheds new light on the origin of eukaryotes显示文摘The hypothesis that eukaryotes originated from within the domain Archaea has been strongly supported by recent phylogenomic analyses placing Heimdallarchaeota-Wukongarchaeota branch from the Asgard superphylum as the closest known archaeal sister-group to eukaryotes. However, our understanding is still limited in terms of the relationship between eukaryotes and archaea, as well as the evolution and ecological functions of the Asgard archaea. Here, we describe three previously unknown phylum-level Asgard archaeal lineages, tentatively named Sigyn-, Freyr-and Njordarchaeota. Additional members in Wukongarchaeota and Baldrarchaeota from distinct environments are also reported here, further expanding their ecological roles and metabolic capacities. Comprehensive phylogenomic analyses further supported the origin of eukaryotes within Asgard archaea and a new lineage Njordarchaeota was supposed as the known closest branch with the eukaryotic nuclear host lineage. Metabolic reconstruction suggests that Njordarchaeota may have a heterotrophic lifestyle with capability of peptides and amino acids utilization, while Sigynarchaeota and Freyrarchaeota also have the potentials to fix inorganic carbon via the Wood-Ljungdahl pathway and degrade organic matters. Additionally, the Ack/Pta pathway for homoacetogenesis and de novo anaerobic cobalamin biosynthesis pathway were found in Freyrarchaeota and Wukongrarchaeota,respectively. Some previously unidentified eukaryotic signature proteins for intracellular membrane trafficking system, and the homologue of mu/sigma subunit of adaptor protein complex, were identified in Freyrarchaeota. | 9 | Impacts of corridor design:An investigation on occupant perception of corridor forms in elderly facilities显示文摘The study addresses the issue of monotonous and lengthy corridors with a single spatial form commonly found in Chinese elderly facilities.We aim to assess the influence of the built spatial environment on the emotions of the elderly quantitatively.To do this,Virtual Reality(VR)technology was employed to construct a digital twin model featuring three corridor forms:straight,arc,and folded,respectively.Forty participants experienced walking in each corridor form for 144 s,while electroencephalogram(EEG)data and subjective questionnaires were collected to explore the psychological and physiological effects caused by different corridors.The results of the questionnaire indicate that arc corridor yielded the highest satisfaction,followed by straight corridor,and the satisfaction for folded corridor is the lowest.EEG results show that folded corridor has 26.0%higher average power than arc corridor and 6.6%higher than straight corridor.The participants consume less energy and are more satisfied in arc corridor,compared to straight and folded corridors.The results establish a correlation between spatial form and occupant perception,suggesting that moderate spatial variations contribute to a better experience of the elderly.Moreover,this study provides quantitative cross-referencing information to optimize the design methods of public space in elderly facilities. | Keming Hou Xinyue Liu Zhe Kong Haining Wang Mingli Lu Songtao Hu | 2023 | Frontiers of Architectural Research2023,12,6: | 0 |
| 10 | Mechanistic investigation on Hg^(0) capture over MnO_(x) adsorbents:effects of the synthesis methods显示文摘This study demonstrated the impacts of the synthesis methods on the textural structures,chemical properties,and Hg^(0)capture capability of the MnO_(x)system.Compared with the samples synthesized using the precipitation(PR)and hydrothermal(HT)methods,the adsorbent prepared via the sol-gel(SG)technique gave the best performance.At 150℃,ca.90%Hg^(0)removal efficiency was reached after 7.5 h for MnO_(x)prepared by the SG method,ca.40%higher than that of the other two methods.The specific surface area of the adsorbent synthesized via the SG technique(23 m^(2)/g)was almost double that of the adsorbent prepared by the HT method(12 m^(2)/g)and three times that of the one prepared by the PR method(7 m^(2)/g).The presence of plentiful acid sites from the SG method facilitated the physisorption of Hg^(0),making more Hg^(0)available to be oxidized to HgO by the redox sites and thus giving the adsorbent prepared by the SG method the highest Hg^(0)removal efficiency.The strong oxidative ability accelerated the oxidation of the physically adsorbed Hg^(0)to HgO,which explained the higher Hg^(0)removal efficiency of the sample prepared using the HT method than that of the one synthesized by the PR technique.During the whole Hg^(0)removal cycles,chemisorption dominated,with the initial adsorption stage and the external mass-transfer process playing important roles. | Dong YE Yongjin HU Zhichang JIANG Xin LIU Haining WANG | 2023 | Journal of Zhejiang University-Science A(Applied Physics & Engineering)2023,24,1: | 0 |
