| 2 | A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18 |

| 4 | Biological soil crust distribution in Artemisia ordosica communities along a grazing pressure gradient in Mu Us Sandy Land, Northern China显示文摘This study investigated the distribution pattern of biological soil crust (BSC) in Artemisia ordosica communities in Mu Us Sandy Land. Three experimental sites were selected according to grazing pressure gradient. In each experimental site, the total vegetation cover, A. ordosica cover, BSC cover, litter-fall cover, BSC degree of fragmentation, BSC thickness and soil properties were investigated in both fixed and semi-fixed sand dunes and simultaneously analyzed in the laboratory. The results showed that at the same grazing pressure, BSC cover and composition were significantly affected by the fixation degree of sand dunes. In addition, BSC cover in the fixed sand dunes was 83.74% on average, whereas it is proportionally dominated by 28% mosses, 21% lichens, and 51% algae. Meanwhile, BSC cover in the semi-fixed sand dunes was 23.54% on average, which is proportionally dominated by 6.3% mosses, 2.5% lichens, and 91.2% algae. Fine sand, organic matter, and total nitrogen (N) contents in the fixed sand dunes were all significantly higher than those in the semi-fixed sand dunes. Litter-fall cover decreased along the grazing pressure gradient, whereas BSC fragmentation degree increased. Fine sand content decreased along with the increase of grazing pressure, whereas medium sand content increased in both fixed and semi-fixed dunes. The organic matter and total N contents in the no grazing site were significantly higher than those in light and normal grazing sites. However, there were no significant differences between the light and normal grazing sites. In addition, there were also no significant differences in BSC thickness between the light and normal grazing sites in the fixed sand dunes. However, a significant decrease was observed in both BSC cover and thickness in the normal grazing site. | 6 | In-situ lunar dust deposition amount induced by lander landing in Chang’E-3 mission显示文摘China first in-situ lunar dust experiment is performed by a lunar dust detector in Chang’E-3 mission. The existed dust(less than 20 μm in diameter) properties, such as levitation, transportation and adhesion, are critical constraints for future lunar exploration program and even manned lunar exploration. Based on the problems discussed above, the in-situ lunar dust detector is originally designed to characterize dust deposition properties induced by lander landing as a function of environmental temperature, solar incident angle and orbit short circuit current on the northern Mare Imbrium, aiming to study lunar dust deposition properties induced by lander landing in depth. This paper begins with a brief of introduction of Chang’E-3 lunar dust detector design,followed by a series of experimental analysis of this instrument under different influencing factors, and concludes with lunar dust mass density deposition amount observed on the first lunar day is about 0.83 mg/cm^2, which is less than that observed in Apollo 11 mission because the landing site of Chang’E-3 has the youngest mare basalts comparing with previous Apollo and lunar landing sites. | 9 | Symptoms of long-term exposure to moxa smoke in acupuncturists: a correlation analysis显示文摘OBJECTIVE: To investigate the effects of long-term exposure to moxa smoke on acupuncturists and to consider the association between physical symptoms and gender, age, secondhand smoke(SHS) exposure, and the duration of exposure.METHODS: A self-report, web-based questionnaire was used to evaluate the effects of moxa smoke onacupuncturists. Logistic regression was used to analyze the relationship between physical symptoms and possible correlative factors.RESULTS: A total of 858 questionnaires were analyzed. The data showed that 42.42% of acupuncturists had been exposed to moxa smoke for 5-10 years. The most frequent symptom was tearing(32.98%) and the least frequent symptom was asthma(5.24%). Logistic regression analysis showed that female acupuncturists were slightly more susceptible to cough and tearing than males(cough:OR: 1.583, 95% CI: 1.079, 2.321, P = 0.019; tearing:OR: 1.519, 95% CI: 1.094, 2.108, P = 0.013). Acupuncturists aged over 25 years reported a slightly lower incidence of eye itchiness than those aged under25 years(26-40 years: OR: 0.604, 95% CI: 0.379,0.965, P = 0.035; over 40 years: OR: 0.330, 95% CI:0.114, 0.958, P = 0.042). Ophthalmodynia occurred less in acupuncturists aged 26-40 years(OR: 0.591,95% CI: 0.359, 0.970, P = 0.038) than in those younger than 25 years. The only association between prevalence of symptoms and tobacco use and SHS exposure was that smokers had a lower occurrence of phlegm in the throat(OR: 0.579, 95% CI: 0.392,0.856, P = 0.006). Shortness of breath was less frequent in participants exposed to moxa smoke for more than 5 years(5-10 years: OR: 0.400, 95% CI:0.204, 0.785, P = 0.008; 11-20 years: OR: 0.392, 95%CI: 0.178, 0.864, P = 0.02), but a slightly higher incidence of eye itchiness was found in those with over20 years of moxa exposure(OR: 4.200, 95% CI:1.344, 3.128, P = 0.014).CONCLUSION: The most frequent symptom of moxa smoke exposure in acupuncturists was tear-ing. | 11 | FRUCTOKINASE-LIKE PROTEIN 1 interacts with TRXz to regulate chloroplast development in rice显示文摘Chloroplast genes are transcribed by the plastidencoded RNA polymerase(PEP) or nucleus-encoded RNA polymerase. FRUCTOKINASE-LIKE PROTEINS(FLNs) are phosphofructokinase-B(Pfk B)-type carbohydrate kinases that act as part of the PEP complex; however, the molecular mechanisms underlying FLN activity in rice remain elusive.Previously, we identified and characterized a heat-stress sensitive albino(hsa_1) mutant in rice. Map-based cloning revealed that HSA_1 encodes a putative OsFLN_2. Here, we further demonstrated that knockdown or knockout of the OsFLN_1, a close homolog of HSA_1/OsFLN_2, considerably inhibits chloroplast biogenesis and the fln_1 knockout mutants, created by clustered regularly interspaced short palindromic repeats(CRISPR) and CRISPR-associate protein_9, exhibit severe albino phenotype and seedling lethality. Moreover, OsFLN_1 localizes to the chloroplast.Yeast two-hybrid, pull-down and bimolecular fluorescencecomplementation experiments revealed that OsFLN_1 and HSA_1/OsFLN_2 interact with THIOREDOXINZ(OsTRXz) to regulate chloroplast development. In agreement with this,knockout of OsTRXz resulted in a similar albino and seedling lethality phenotype to that of the fln_1 mutants. Quantitative reverse transcription polymerase chain reaction and immunoblot analysis revealed that the transcription and translation of PEP-dependent genes were strongly inhibited in fln_1 and trxz mutants, indicating that loss of OsFLN_1, HSA_1/OsFLN_2, or OsTRXz function perturbs the stability of the transcriptionally active chromosome complex and PEP activity. | 12 | Characterization of a LAL-type regulator NemR in nemadectin biosynthesis and its application for increasing nemadectin production in Streptomyces cyaneogriseus显示文摘Nemadectin, a macrocyclic lactone antibiotic, is produced by Streptomyces cyaneogriseus ssp. noncyanogenus. A methoxime derivative of nemadectin, moxdectin, has been widely used to control insect and helminth in animal health. Despite the importance of nemadectin, little attention has been paid to the regulation of nemadectin biosynthesis, which has hindered efforts to improve nemadectin production via genetic manipulation of regulatory genes. Here, we characterize the function of nemR, the cluster-situated regulatory gene encoding a LAL-family transcriptional regulator, in the nemadectin biosynthesis gene cluster of S. cyaneogriseus ssp. noncyanogenus NMWT1. NemR is shown to be essential for nemadectin production and found to directly activate the transcription of nemA1-1/A1-2/A2, nemC and nemA4/A3/E/D operons, but indirectly activate that of nemG and nemF. A highly conserved sequence 5′-TGGGGTGKATAGGGGGTA-3′(K=T/G) is verified to be essential for NemR binding.Moreover, four novel targets of NemR, including genes encoding an SsgA-like protein(TU94_12730), a methylmalonyl-CoA mutase(TU94_19950), a thioesterase of oligomycin biosynthesis(TU94_22425) and a MFS family transporter(TU94_24835)are identified. | 13 | Ocean surface wave measurements from fully polarimetric SAR imagery显示文摘A new method for the retrieval of ocean wave parameters from SAR imagery is developed,based on the shape-from-shading(SFS)technique.Previously,the SFS technique has been used in the reconstruction of 3D landform information from SAR images,in order to generate elevation maps of topography for land surfaces.Here,in order to retrieve ocean wave characteristics,we apply the SFS methodology,together with a method to orient the angular measurements of the azimuth slope and range slope,in the measurement of ocean surface waves.This method is applied to high resolution fine-quad polarization mode(HH,VV,VH and HV)C-band RADARSAT-2 SAR imagery,in order to retrieve ocean wave spectra and extract wave parameters.Collocated in situ buoy measurements are used to validate the reliability of this method.Results show that the method can reliably estimate wave height,dominant wave period,dominant wave length and dominant wave direction from C-band SAR images.The advantage of this method is that it does not depend on modulation transfer functions(MTFs),in order to measure ocean surface waves.This method can be used in monitoring ocean surface wave propagation through open water areas into ice-covered areas,especially the marginal ice zone(MIZ)in polar oceans. | XIE Tao William PERRIE HE YiJun LI HaiYan FANG He ZHAO ShangZhuo YU WenJin | 2015 | Science China Earth Sciences2015,58,10: | 6 |
| 14 | A thinned lithosphere beneath coastal area of southeastern China as evidenced by seismic receiver functions显示文摘During Mesozoic to Cenozoic time, the large-scale tectono-magmatism had strongly modified the lithosphere beneath the southeastern continent of China, leaving the present-day lithosphere as a new one evolving from the ancient lithosphere that was largely removed and replaced. But this model proposed from geochemical and petrological research is urgently in need of support from seismic observational evidence. In this paper, based on the dataset recorded by the dense stations of two NE oriented broadband seismic profiles deployed in the coastal area of southeastern China(SE China), both P-wave(P-RF) and S-wave(S-RF) receiver functions were isolated. We identified Pls phase converted from the Lithosphere-Asthenosphere Boundary(LAB) in P-RFs of individual stations. Migrated Pls phase indicated a depth of 60–70 km for LAB. Inversions/comparisons of P-RF(Pls phase) and S-RF(Slp phase) waveforms together with Ps and Sp imaging for the crust and upper mantle structure further confirmed this result. P-RF and S-RF migrated images exhibit that a flat LAB is positioned at the depth of 60–70 km spreading along the profile, whereas a distinct structural change of lithospheric base appears at the Min River estuary. Both Ps and PpPs migrated images of P-RFs present an abrupt Moho drop across the Min River fault from south to north, which is consistent with previous result obtained from deep seismic sounding. A strain of transfected HeLa Tet-off cell HaCb, which contains antisense CENP-B expressing vector pBI-EGFP-as-CenpB, was prepared. Northern blot and Western blot were used to analyze the repression of internal CENP-B in transfected cells. According to the growth curve, the proliferation of HeLa (Tet-off) is repressed by antisense CENP-B, and the multiplication time is prolonged for 32.81 h. The analysis of flow cytometry revealed that, compared with HeLa (Tet-off), the G1 cell population of HaCb is increased (AG1=9%) while S fraction is decreased (AS=11%), but the G2/M phase is nearly unchanged (AG2/M=3%). | 17 | Regulation of mitochondrial NAD pool via NAD^+ transporter 2 is essential for matrix NADH homeostasis and ROS production in Arabidopsis显示文摘Reactive oxygen species(ROS) play a crucial role in numerous biological processes in plants, including development, responses to environmental stimuli, and programmed cell death(PCD). Deficiency in MOSAIC DEATH 1(MOD1), a plastid-localized enoyl-ACP reductase essential for de novo fatty acid biosynthesis in Arabidopsis thaliana, leads to the increased malate export from chloroplasts to mitochondria, and the subsequent accumulation of mitochondria-generated ROS and PCD. In this study, we report the identification and characterization of a mod1 suppressor, som592. SOM592 encodes mitochondrion-localized NAD^+ transporter 2(NDT2). We show that the mitochondrial NAD pool is elevated in the mod1 mutant. The som592 mutation fully suppressed mitochondrial NADH hyper-accumulation, ROS production, and PCD in the mod1 mutant, indicating a causal relationship between mitochondrial NAD accumulation and ROS/PCD phenotypes. We also show that in wild-type plants, the mitochondrial NAD+uptake is involved in the regulation of ROS production in response to continuous photoperiod. Elevation of the alternative respiration pathway can suppress ROS accumulation and PCD in mod1, but leads to growth restriction. | 18 | The thickness and structural characteristics of the crust across Tibetan plateau from active-sources seismic profiles显示文摘The Tibetan plateau as one of the youngest orogen on the Earth was considered as the result of continent-continent collision between the Eurasian and Indian plates.The thickness and structure of the crust beneath Tibetan plateau is essential to understand deformation behavior of the plateau.Active-source seismic profiling is most available geo-physical method for imaging the structure of the continental crust.The results from more than 25 active-sources seismic profiles carried out in the past twenty years were reviewed in this article.A preliminary cross crustal pattern of the Tibetan Pla-teau was presented and discussed.The Moho discontinuity buries at the range of 60-80 km on average and have steep ramps located roughly beneath the sutures that are compatible with the successive stacking/accretion of the former Cenozoic blocks northeastward.The deepest Moho(near 80 km) appears closely near IYS and the crustal scale thrust system beneath southern margin of Tibetan plateau suggests strong dependence on collision and non-distributed deformation there.However,the ~20 km order of Moho offsets hardly reappears in the inline section across northern Tibetan plateau.Without a universally accepted,convincing dynamic explanation model accommodated the all of the facts seen in controlled seismic sections,but vertical thickening and northeastern shorten of the crust is quite evident and interpretable to a certain extent as the result of continent-continent collision.Simultaneously,weak geophysical signature of the BNS suggests that convergence has been accommodated perhaps partially through pure-shear thickening accompanied by removal of lower crustal material by lateral escape.Recent years the result of Moho with ~7 km offset and long extend in south-dip angle beneath the east Kunlun oro-gen and a grand thrust fault at the northern margin of Qilian orogen has attract more attention to action from the northern blocks.The broad lower-velocity area in the upper-middle crust of the Lhasa block was once considered as resulted from partially melted rocks.However the low normal vP/vS ratio and the Moho stepwise rise fail to support significant partial melting in the middle-lower crust of the central-northern Tibetan plateau.Furthermore,the lower-velocity of crust occasionally disappears,and/or local thinned exhibits their non-stationary spatial distribution. | Qiusheng Li Rui Gao Zhanwu Lu Ye Guan Jisheng Zhang Pengwu Li Haiyan Wang Rizheng He Marianne Karplus | 2009 | Earthquake Science2009,22,1: | 5 |
| 19 | FUNCTIONAL CHARACTERIZATION OF CADMIUM-RESPONSIVE GARLIC GENE ASMT2B: A NEW MEMBER OF METALLOTHIONEIN FAMILY显示文摘A new gene of metallothionein (MT) family was cloned from garlic (Allium sativum) seed-lings using RACE method and designated AsMT2b. The full length of AsMT2b cDNA was 520 bp encod-ing 80 amino acids. The deduced amino acids of AsMT2b showed that AsMT2b contained the char-acteristic structure of type 2 MT proteins, but the number and arrangement of the cysteine residues in the N- and C-terminal domains was different from other type 2 MT proteins. Semi-quantitative reverse transcriptase-PCR showed that transcript levels of AsMT2b were enhanced only in response to higher concentrations or longer incubation time of Cd. Such an expression pattern of AsMT2b greatly differed from that of other type 2 MT genes. Yeast cells transformed with this gene had improved resistance to Cd. AsMT2b overexpressing Arabidopsis showed stronger Cd tolerance and higher Cd accumulation compared with wild-type plants. | 20 | KRAS-G12D mutation drives immune suppression and the primary resistance of anti-PD-1/PD-L1 immunotherapy in non-small cell lung cancer显示文摘Background:Although immune checkpoint inhibitors(ICIs)against programmed cell death protein 1(PD-1)and its ligand PD-L1 have demonstrated potency towards treating patients with non-small cell lung carcinoma(NSCLC),the potential association between Kirsten rat sarcoma viral oncogene homolog(KRAS)oncogene substitutions and the efficacy of ICIs remains unclear.In this study,we aimed to find point mutations in the KRAS gene resistant to ICIs and elucidate resistance mechanism.Methods:The association between KRAS variant status and the efficacy of ICIs was explored with a clinical cohort(n=74),and confirmed with a mouse model.In addition,the tumor immune microenvironment(TIME)of KRASmutant NSCLC,such as CD8+tumor-infiltrating lymphocytes(TILs)and PD-L1 level,was investigated.Cell lines expressing classic KRAS substitutions were used to explore signaling pathway activation involved in the formation of TIME.Furthermore,interventions that improved TIME were developed to increase responsiveness to ICIs.Results:We observed the inferior efficacy of ICIs in KRAS-G12D-mutant NSCLC.Based upon transcriptome data and immunostaining results from KRAS-mutant NSCLC,KRAS-G12D point mutation negatively correlated with PD-L1 level and secretion of chemokines CXCL10/CXCL11 that led to a decrease in CD8+TILs,which in turn yielded an immunosuppressive TIME.The analysis of cell lines overexpressing classic KRAS substitutions further revealed that KRAS-G12D mutation suppressed PD-L1 level via the P70S6K/PI3K/AKT axis and reduced CXCL10/CXCL11 levels by down-regulating high mobility group protein A2(HMGA2)level.Notably,paclitaxel,a chemotherapeutic agent,upregulated HMGA2 level,and in turn,stimulated the secretion of CXCL10/CXCL11.Moreover,PD-L1 blockade combined with paclitaxel significantly suppressed tumor growth compared with PD-L1 inhibitor monotherapy in a mouse model with KRAS-G12D-mutant lung adenocarcinoma.Further analyses revealed that the combined treatment significantly enhanced the recruitment of CD8+TILs via the up-regulation of CXCL10/CXCL11 levels.Results of clinical study also revealed the superior efficacy of chemo-immunotherapy in patients with KRAS-G12D-mutant NSCLC compared with ICI monotherapy.Conclusions:Our study elucidated the molecular mechanism by which KRASG12D mutation drives immunosuppression and enhances resistance of ICIs in NSCLC.Importantly,our findings demonstrate that ICIs in combination with chemotherapy may be more effective in patients with KRAS-G12D-mutant NSCLC. | Chengming Liu Sufei Zheng Zhanyu Wang Sihui Wang Xinfeng Wang Lu Yang Haiyan Xu Zheng Cao Xiaoli Feng Qi Xue Yan Wang Nan Sun Jie He | 2022 | Cancer Communications2022,42,9: | 4 |
