1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3In vitro differentiation of adipose-derived stem cells and bone marrow-derived stromal stem cells into neuronal-like cells显示文摘Adipose-derived stem cells and bone marrow-derived stromal stem cells were co-cultured with untreated or Aβ1-40-treated PC12 cells, or grown in supernatant derived from untreated or Aβ1-40-treated PC12 cells. Analysis by western blot and quantitative real-time PCR showed that protein levels of Nanog, Oct4, and Sox2, and mRNA levels of miR/125a/3p were decreased, while expression of insulin-like growth factor-2 and neuron specific enolase was increased. In comparison, the generation of neuron specific enolase-positive cells was most successful when adipose-derived stem cells were co-cultured with Aβ1-40-treated PC12 cells. Our results demonstrate that adipose-derived stem cells and bone marrow-derived stromal stem cells exhibit trends of neuronal-like cell differentiation after co-culture with Aβ1-40-treated PC12 cells. 7Comparison of non-schistosomal rectosigmoid cancer and schistosomal rectosigmoid cancer显示文摘AIM: To compare the clinicopathological features of patients with non-schistosomal rectosigmoid cancer and schistosomal rectosigmoid cancer.METHODS: All the patients with rectosigmoid carcinoma who underwent laparoscopic radical surgical resection in the Shanghai Minimally Invasive Surgical Center at Ruijin Hospital affiliated to Shanghai JiaoTong University between October 2009 and October2013 were included in this study. Twenty-six cases of colonic schistosomiasis diagnosed through colonoscopy and pathological examinations were collected. Symptoms,endoscopic findings and clinicopathological characteristics were evaluated retrospectively.RESULTS: There were no significant differences between patients with and without schistosomiasis in gender, age, CEA, CA19-9, preoperative biopsy findings or postoperative pathology. Patients with rectosigmoid schistosomiasis had a significantly higher CA-125 level and a larger proportion of these patients were at an early tumor stage(P = 0.003). Various morphological characteristics of schistosomiasis combined with rectosigmoid cancer could be found by colonoscopic examination: 46% were fungating mass polyps, 23%were congestive and ulcerative polyps, 23% were cauliflower-like masses, 8% were annular masses.Only 27% of the patients were diagnosed with rectal carcinoma preoperatively after the biopsy. Computed tomography(CT) scans showed thickened intestinal walls combined with linear and tram-track calcifications in 26 patients.CONCLUSION: Rectosigmoid carcinoma combined with schistosomiasis is associated with higher CA-125 values and early tumor stages. 8The hypoxia-inducible factor-1α activates ectopic production of fibroblast growth factor 23 in tumor-induced osteomalacia显示文摘Tumor-induced osteomalacia(TIO) is a rare paraneoplastic syndrome in which ectopic production of fibroblast growth factor 23(FGF23) by non-malignant mesenchymal tumors causes phosphate wasting and bone fractures. Recent studies have implicated the hypoxia-inducible factor-1α(HIF-1α) in other phosphate wasting disorders caused by elevated FGF23, including X-linked hypophosphatemic rickets and autosomal dominant hypophosphatemia. Here we provide evidence that HIF-1α mediates aberrant FGF23 in TIO by transcriptionally activating its promoter. Immunohistochemical studies in phosphaturic mesenchymal tumors resected from patients with documented TIO showed that HIF-1α and FGF23 were co-localized in spindleshaped cells adjacent to blood vessels. Cultured tumor tissue produced high levels of intact FGF23 and demonstrated increased expression of HIF-1α protein. Transfection of MC3T3-E1 and Saos-2 cells with a HIF-1α expression construct induced the activity of a FGF23 reporter construct. Prior treatment of tumor organ cultures with HIF-1α inhibitors decreased HIF-1α and FGF23 protein accumulation and inhibited HIF-1α-induced luciferase reporter activity in transfected cells. Chromatin immunoprecipitation assays confirmed binding to a HIF-1α consensus sequence within the proximal FGF23 promoter, which was eliminated by treatment with a HIF-1α inhibitor. 9Colonoscopic yield of colorectal neoplasia in daily clinical practice显示文摘AIM:To assess the prevalence and location of ad-vanced neoplasia in patients undergoing colonoscopy,and to compare the yield per indication.METHODS:In a multicenter colonoscopy survey (n = 18 hospitals) in the Amsterdam area (Northern Holland),data of all colonoscopies performed during a three month period in 2005 were analyzed. The location and the histological features of all colonic neoplasia were recorded. The prevalence and the distribution ofadvanced colorectal neoplasia and differences in yield between indication clusters were evaluated. Advanced neoplasm was defi ned as adenoma > 10 mm in size,with > 25% villous features or with high-grade dyspla-sia or cancer.RESULTS:A total of 4623 eligible patients underwent a total colonoscopy. The prevalence of advanced neo-plasia was 13%,with 281 (6%) adenocarcinomas and 342 (7%) advanced adenomas. Sixty-seven percent and 33% of advanced neoplasia were located in the distal and proximal colon,respectively. Of all patients with right-sided advanced neoplasia (n = 228),51% had a normal distal colon,whereas 27% had a syn-chronous distal adenoma. Ten percent of all colono-scopies were performed in asymptomatic patients,7% of whom had advanced neoplasia. In the respective procedure indication clusters,the prevalence of right-sided advanced neoplasia ranged from 11%-57%. CONCLUSION:One out of every 7-8 colonoscopies yielded an advanced colorectal neoplasm. 10Evaluation of three-dimensional biofilms on antibacterial bonding agents containing novel quaternary ammonium methacrylates显示文摘Antibacterial adhesives are promising to inhibit biofilms and secondary caries.The objectives of this study were to synthesize and incorporate quaternary ammonium methacrylates into adhesives,and investigate the alkyl chain length effects on three-dimensional biofilms adherent on adhesives for the first time.Six quaternary ammonium methacrylates with chain lengths of 3,6,9,12,16 and 18were synthesized and incorporated into Scotchbond Multi-Purpose.Streptococcus mutans bacteria were cultured on resin to form biofilms.Confocal laser scanning microscopy was used to measure biofilm thickness,live/dead volumes and live-bacteria percentage vs.distance from resin surface.Biofilm thickness was the greatest for Scotchbond control;it decreased with increasing chain length,reaching a minimum at chain length 16.Live-biofilm volume had a similar trend.Dead-biofilm volume increased with increasing chain length.The adhesive with chain length 9 had 37%live bacteria near resin surface,but close to 100%live bacteria in the biofilm top section.For chain length 16,there were nearly 0%live bacteria throughout the three-dimensional biofilm.In conclusion,strong antibacterial activity was achieved by adding quaternary ammonium into adhesive,with biofilm thickness and live-biofilm volume decreasing as chain length was increased from 3 to 16.Antibacterial adhesives typically only inhibited bacteria close to its surface;however,adhesive with chain length 16 had mostly dead bacteria in the entire three-dimensional biofilm.Antibacterial adhesive with chain length 16 is promising to inhibit biofilms at the margins and combat secondary caries.Han Zhou Michael D Weir Joseph M Antonucci Gary E Schumacher Xue-Dong Zhou Hockin H K Xu 2014International Journal of Oral Science2014,6,2:5
11Geophysical evidence for thrusting of crustal materials from orogenic belts over both sides of the Yangtze Block and its geological significance显示文摘Based on deep geophysical detections, we have reconstructed the crustal structure from the eastern margin of the Tibetan Plateau to the Jiangnan-Xuefeng orogenic belt. The results suggest that the Yangtze Block was overthrusted by crustal materials in its NW direction from the eastern Tibetan Plateau but in its SE direction from the Jiangnan orogen. These overthrusting effects control the crustal structure from the western Sichuan to the western area of the Jiangnan orogen-Xuefeng orogenic belt. The eastward extruded materials from the eastern Tibetan Plateau were blocked by the rigid basement in the Sichuan Basin, where upper-middle crust was overthrusted whereas the lower crust was underthrusted beneath the Sichuan Basin. The underthrusted unit was absorbed by crustal folding, shortening and thickening in the Yangtze Block, forming the Xiongpo and Longquan Mountains tectonic belts and resulting in the NW-directed thrusting of the Pujiang-Chengdu-Deyang fault, and the western hillsiden fault in the Longquan Mountain. These results provide resolution to the controversy where the eastward extrusion material from the Qinghai-Tibet Plateau had gone. Overall, that Yangtze Block was subjected to thrusting of the crustal materials from the orogenic belts over its both sides. 12G-protein-coupled estrogen receptor as a new therapeutic target for treating coronary artery disease显示文摘Coronary heart disease(CHD) continues to be the greatest mortality risk factor in the developed world. Estrogens are recognized to have great therapeutic potential to treat CHD and other cardiovascular diseases; however,a significant array of potentially debilitating side effects continues to limit their use. Moreover,recent clinical trials have indicated that long-term postmenopausal estrogen therapy may actually be detrimental to cardiovascular health. An exciting new development is the finding that the more recently discovered G-protein-coupled estrogen receptor(GPER) is expressed in coronary arteries-both in coronary endothelium and in smooth muscle within the vascular wall. Accumulating evidence indicates that GPER activation dilates coronary arteries and can also inhibit the prolif-eration and migration of coronary smooth muscle cells. Thus,selective GPER activation has the potential to increase coronary blood flow and possibly limit the debilitating consequences of coronary atherosclerotic disease. This review will highlight what is currently known regarding the impact of GPER activation on coronary arteries and the potential signaling mechanisms stimulated by GPER agonists in these vessels. 13Neuronal differentiation of adipose-derived stem cells and their transplantation for cerebral ischemia显示文摘OBJECTIVE:To review published data on the biological characteristics,differentiation and applications of adipose-derived stem cells in ischemic diseases.DATA RETRIEVAL:A computer-based online search of reports published from January 2005 to June 2012 related to the development of adipose-derived stem cells and their transplantation for treatment of cerebral ischemia was performed in Web of Science using the key words 'adipose-derived stem cells','neural-like cells','transplantation','stroke',and 'cerebral ischemia'.SELECTION CRITERIA:The documents associated with the development of adipose-derived stem cells and their transplantation for treatment of cerebral ischemia were selected,and those published in the last 3-5 years or in authoritative journals were preferred in the same field.Totally 89 articles were obtained in the initial retrieval,of which 53 were chosen based on the inclusion criteria.MAIN OUTCOME MEASURES:Biological characteristics and induced differentiation of adipose-derived stem cells and cell transplantation for disease treatment as well as the underlying mechanism of clinical application.RESULTS:The advantages of adipose-derived stem cells include their ease of procurement,wide availability,rapid expansion,low tumorigenesis,low immunogenicity,and absence of ethical constraints.Preclinical experiments have demonstrated that transplanted adipose-derived stem cells can improve neurological functions,reduce small regions of cerebral infarction,promote angiogenesis,and express neuron-specific markers.The improvement of neurological functions was demonstrated in experiments using different methods and time courses of adipose-derived stem cell transplantation,but the mechanisms remain unclear.CONCLUSION:Further research into the treatment of ischemic disease by adipose-derived stem cell transplantation is needed to determine their mechanism of action.Guoping Tian Jin Zhou Jing'e Wang Bing Xu Li Li Feng Zhu Jian Han Jianping Li Siyang Zhang Xiaoguang Luo 2012Neural Regeneration Research2012,7,25:2
14Successful operation of a cooperative SLR station of China and Argentina in San Juan显示文摘We introduced the observations and researches using a Satellite Laser Ranging (SLR) system with high precision, which was designed and made in China and installed in the Observatory of San Juan Uni- versity, Argentina, the capability of the system and the achievement of the cooperative procedure be- tween China and Argentina. The SLR station in San Juan, set up by China and Argentina, is quite sig- nificant for improving the distribution of SLR stations and enhancing the orbital coverage of the whole earth. Since the SLR system started to work in the Observatory of San Juan University in the beginning of 2006, the operation is rather good, and rich data with high precision have been obtained. 16Sustained local delivery of insulin for potential improvement of peri-implant bone formation in diabetes显示文摘Dental implantation is an effective standard treatment modality to restore missing teeth and maxillofacial defects. However, in diabetics there is an increased risk for implant failure due to impaired peri-implant osseous healing. Early topical insulin treat- ment was recently shown to normalize diabetic bone healing by rectifying impairments in osteoblastic activities. In this study, insulin/poly(lactic-co-glycolic acid) (PLGA) microspheres were prepared by a double-emulsion solvent evaporation method. Microspheres were then incorporated in fibrin gel to develop a local drug delivery system for diabetic patients requiring im- plant treatment. In vitro release of insulin from fibrin gel loaded with these microspheres was assessed, and sustained prolonged insulin release over 21 days ascertained. To assess the bioactivity of released insulin and determine whether slow release might improve impaired diabetic bone formation, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), alkaline phosphatase (ALP) activity, mineralized nodule formation, and ELISA (enzyme-linked immunosorbent assay) assays were performed. The insulin released from the drug delivery system stimulated cell growth in previously inhibited cells, and ameliorated the impaired bone-forming ability of human MG-63 cells under high glucose conditions. 18A molecular framework for seasonal growth-dormancy regulation in perennial plants显示文摘The timing of the onset and release of dormancy impacts the survival,productivity and spatial distribution of temperate horticultural and forestry perennials and is mediated by at least three main regulatory programs involving signal perception and processing by phytochromes(PHYs)and PHY-interacting transcription factors(PIFs).PIF4 functions as a key regulator of plant growth in response to both external and internal signals.In poplar,the expression of PIF4 and PIF3-LIKE1 is upregulated in response to short days,while PHYA and PHYB are not regulated at the transcriptional level.Integration of light and environmental signals is achieved by gating the expression and transcriptional activity of PIF4.During this annual cycle,auxin promotes the degradation of Aux/IAA transcriptional repressors through the SKP–Cullin-F–boxTIR1 complex,relieving the repression of auxin-responsive genes by allowing auxin response factors(ARFs)to activate the transcription of auxin-responsive genes involved in growth responses.Analyses of transcriptome changes during dormancy transitions have identified MADS-box transcription factors associated with endodormancy induction.Previous studies show that poplar dormancy-associated MADS-box(DAM)genes PtMADS7 and PtMADS21 are differentially regulated during the growth-dormancy cycle.Endodormancy may be regulated by internal factors,which are specifically localized in buds.PtMADS7/PtMADS21 may function as an internal regulator in poplar.The control of flowering time shares certain regulatory hierarchies with control of the dormancy/growth cycle.However,the particularities of different stages of the dormancy/growth cycle warrant comprehensive approaches to identify the causative genes for the entire cycle.A growing body of knowledge also indicates epigenetic regulation plays a role in these processes in perennial horticultural and forestry plants.The increased knowledge contributes to better understanding of the dormancy process and consequently to precise manipulation of dormancy-related horticultural traits,such as flowering time.Donghwan Shim Jae-Heung Ko Won-Chan Kim Qijun Wang Daniel E Keathley Kyung-Hwan Han 2014Horticulture Research2014,1,1:2
19Chemiluminescence Determination of Tetracycline and Oxytetracycline in Pharmaceutical Preparations Using Ru(bipy)_3^(2+)-Cerium(Ⅳ) System显示文摘A new chemiluminescence (CL) method for the determination of tetracycline and oxytetracycline is developed, hased on the CL reaction of tetracycline and oxytetracycline with Ru(bipy)32+ and Ce(IV). In sulfuric acid medium, the CL emission is generated upon continuous oxidation of Ru(bipy)32+ by cerium (IV ). The emission intensity is greatly enhanced when tetracycline and oxytetracycline are introduced into the reaction system after acid degradation. Under the optimum conditions, the calibration curves are linear over the range of 8. 0 X 10-8-4. 0 X 10-6 mol/L for tetracycline and of 2. 0 X 10-74. 0 X 10-5 mol/L for oxytetracycline, with the detection limits are 4. 2 X 10-8 mol/L for tetracycline and 1. 5 X 10-7 mol/L for oxytetracycline, respectively.
