4Allyl isothiocyanate ameliorates lipid accumulation and inflammation in nonalcoholic fatty liver disease via the Sirt1/AMPK and NF-κB signaling pathways显示文摘BACKGROUND Allyl isothiocyanate(AITC),a classic anti-inflammatory and antitumorigenic agent,was recently identified as a potential treatment for obesity and insulin resistance.However,little is known about its direct impact on the liver.AIM To investigate the effect and underlying mechanism of AITC in nonalcoholic fatty liver disease(commonly referred to as NAFLD).METHODS To establish a mouse and cellular model of NAFLD,C57BL/6 mice were fed a high fat diet(HFD)for 8 wk,and AML-12 cells were treated with 200μM palmitate acid for 24 h.For AITC treatment,mice were administered AITC(100 mg/kg/d)orally and AML-12 cells were treated with AITC(20μmol/L).RESULTS AITC significantly ameliorated HFD-induced weight gain,hepatic lipid accumulation and inflammation in vivo.Furthermore,serum alanine aminotransferase and aspartate aminotransferase levels were markedly reduced in AITC-treated mice.Mechanistically,AITC significantly downregulated the protein levels of sterol regulatory elementbinding protein 1(SREBP1)and its lipogenesis target genes and upregulated the levels of proteins involved in fatty acidβ-oxidation,as well as the upstream mediators Sirtuin 1(Sirt1)and AMPactivated protein kinaseα(AMPKα),in the livers of HFD-fed mice.AITC also attenuated the nuclear factor kappa B(NF-κB)signaling pathway.Consistently,AITC relieved palmitate acid-induced lipid accumulation and inflammation in AML-12 cells in vitro through the Sirt1/AMPK and NF-κB signaling pathways.Importantly,further studies showed that the curative effect of AITC on lipid accumulation was abolished by siRNA-mediated knockdown of either Sirt1 or AMPKαin AML-12 cells.CONCLUSION AITC significantly ameliorates hepatic steatosis and inflammation by activating the Sirt1/AMPK pathway and inhibiting the NF-κB pathway.Therefore,AITC is a potential therapeutic agent for NAFLD.Chun-Xiao Li Jian-Guo Gao Xing-Yong Wan Yi Chen Cheng-Fu Xu Ze-Min Feng Hang Zeng Yi-Ming Lin Han Ma Ping Xu Chao-Hui Yu You-Ming Li 2019World Journal of Gastroenterology2019,25,34:14
5Magnetic resonance-based total liver volume and magnetic resonance-diffusion weighted imaging for staging liver fibrosis in mini-pigs显示文摘AIM:To determine whether and how magnetic resonance imaging(MRI)-based total liver volume(TLV) and diffusion weighted imaging(DWI) could predict liver fibrosis.METHODS:Sixteen experimental mature mini-pigs(6 males,10 females),weighing between 20.0 and 24.0 kg were prospectively used to model liver fibrosis induced by intraperitoneal injection of 40% CCl4 dissolved in fat emulsion twice a week for 16 wk,and by feeding 40% CCl4 mixed with maize flour twice daily for the subsequent 5 wk.All the survival animals underwent percutaneous liver biopsy and DWI using b = 300,500 and 800 s/mm2 followed by abdominal gadolinium-enhanced MRI at the 0,5th,9th,16th and 21st weekend after beginning of the modeling.TLV was obtained on enhanced MRI,and apparent diffusion coefficient(ADC) was obtained on DWI.Hepatic tissue specimens were stained with hematoxylin and Masson' s trichrome staining for staging liver fibrosis.Pathological specimens were scored using the human METAVIR classification system.Statistical analyses were performed to determine whether and how the TLV and ADC could be used to predict the stage of liver fibrosis.RESULTS:TLV increased from stage 0 to 2 and decreased from stage 3(r = 0.211;P < 0.001).There was a difference in TLV between stage 0-1 and 2-4(P = 0.03) whereas no difference between stage 0-2 and 3-4(P = 0.71).TLV could predict stage ≥ 2 [area under receiver operating characteristic curve(AUC) = 0.682].There was a decrease in ADC values with increasing stage of fibrosis for b = 300,500 and 800 s/mm2(r =-0.418,-0.535 and-0.622,respectively;all P < 0.001).Differences were found between stage 0-1 and 2-4 in ADC values for b = 300,500 and 800 s/mm2,and between stage 0-2 and 3-4 for b = 500 or 800 s/mm2(all P < 0.05).For predicting stage ≥ 2 and ≥ 3,AUC was 0.803 and 0.847 for b = 500 s/mm2,and 0.848 and 0.887 for b = 800 s/mm2,respectively.CONCLUSION:ADC for b = 500 or 800 s/mm2 could be better than TLV and ADC for b = 300 s/mm2 to pre-dict fibrosis stage ≥ 2 or ≥ 3.Hang Li Tian-Wu Chen Xiao-Li Chen Xiao-Ming Zhang Zhen-Lin Li Nan-Lin Zeng Li Zhou Li-Ying Wang Hong-Jie Tang Chun-Ping Li Li Li Xian-Yong Xie 2012World Journal of Gastroenterology2012,18,48:13
6Activation of mTORC1 in subchondral bone preosteoblasts promotes osteoarthritis by stimulating bone sclerosis and secretion of CXCL12显示文摘Increasing evidences show that aberrant subchondral bone remodeling plays an important role in the development of osteoarthritis(OA).However,how subchondral bone formation is activated and the mechanism by which increased subchondral bone turnover promotes cartilage degeneration during OA remains unclear.Here,we show that the mechanistic target of rapamycin complex 1(mTORC1)pathway is activated in subchondral bone preosteoblasts(Osterix+)from OA patients and mice.Constitutive activation of mTORC1 in preosteoblasts by deletion of the mTORC1 upstream inhibitor,tuberous sclerosis 1,induced aberrant subchondral bone formation,and sclerosis with little-to-no effects on articular cartilage integrity,but accelerated posttraumatic OA development in mice.In contrast,inhibition of mTORC1 in preosteoblasts by disruption of Raptor(mTORC1-specific component)reduced subchondral bone formation and cartilage degeneration,and attenuated post-traumatic OA in mice.Mechanistically,mTORC1 activation promoted preosteoblast expansion and Cxcl12 secretion,which induced subchondral bone remodeling and cartilage degeneration during OA.A Cxcl12-neutralizing antibody reduced cartilage degeneration and alleviated OA in mice.Altogether,these findings demonstrate that mTORC1 activation in subchondral preosteoblasts is not sufficient to induce OA,but can induce aberrant subchondral bone formation and secrete of Cxcl12 to accelerate disease progression following surgical destabilization of the joint.Pharmaceutical inhibition of the pathway presents a promising therapeutic approach for OA treatment.Chuangxin Lin Liangliang Liu Chun Zeng Zhong-Kai Cui Yuhui Chen Pinling Lai Hong Wang Yan Shao Haiyan Zhang Rongkai Zhang Chang Zhao Hang Fang Daozhang Cai Xiaochun Bai 2019Bone Research2019,7,1:12
7Platelet count combined with right liver volume and spleen volume measured by magnetic resonance imaging for identifying cirrhosis and esophageal varices显示文摘AIM: To determine whether the combination of platelet count(PLT) with spleen volume parameters and right liver volume(RV) measured by magnetic resonance imaging(MRI) could predict the Child-Pugh class of liver cirrhosis and esophageal varices(EV).METHODS: Two hundred and five cirrhotic patients with hepatitis B and 40 healthy volunteers underwent abdominal triphasic-enhancement MRI and laboratory examination of PLT in 109/L. Cirrhotic patients underwent endoscopy for detecting EV. Spleen maximal width(W), thickness(T) and length(L) in mm together with spleen volume(SV) and RV in mm3 were measured by MRI, and spleen volume index(SI) in mm3 was obtained by W × T × L. SV/PLT, SI/PLT and RV × PLT/SV(RVPS) were calculated and statistically analyzed to assess cirrhosis and EV.RESULTS: SV/PLT(r = 0.676) and SI/PLT(r = 0.707) increased, and PLT(r =-0.626) and RVPS(r =-0.802) decreased with the progress of Child-Pugh class(P < 0.001 for all). All parameters could determine the presence of cirrhosis, distinguish between each class of Child-Pugh class, and identify the presence of EV [the areas under the curve(AUCs) = 0.661-0.973]. 8Underwater image quality enhancement of sea cucumbers based on improved histogram equalization and wavelet transform显示文摘Sea cucumbers usually live in an environment where lighting and visibility are generally not controllable,which cause the underwater image of sea cucumbers to be distorted,blurred,and severely attenuated.Therefore,the valuable information from such an image cannot be fully extracted for further processing.To solve the problems mentioned above and improve the quality of the underwater images of sea cucumbers,pre-processing of a sea cucumber image is attracting increasing interest.This paper presents a newmethod based on contrast limited adaptive histogram equalization and wavelet transform(CLAHE-WT)to enhance the sea cucumber image quality.CLAHE was used to process the underwater image for increasing contrast based on the Rayleigh distribution,and WTwas used for de-noising based on a soft threshold.Qualitative analysis indicated that the proposed method exhibited better performance in enhancing the quality and retaining the image details.For quantitative analysis,the test with 120 underwater images showed that for the proposed method,the mean square error(MSE),peak signal to noise ratio(PSNR),and entropy were 49.2098,13.3909,and 6.6815,respectively.The proposed method outperformed three established methods in enhancing the visual quality of sea cucumber underwater gray image.Xi Qiao Jianhua Bao Hang Zhang Lihua Zeng Daoliang Li 2017Information Processing in Agriculture2017,4,3:9
9Daytime HONO formation in the suburban area of the megacity Beijing, China显示文摘Nitrous acid(HONO),as a primary precursor of OH radicals,has been considered one of the most important nitrogencontaining species in the atmosphere.Up to 30%of primary OH radical production is attributed to the photolysis of HONO.However,the major HONO formation mechanisms are still under discussion.During the Campaigns of Air Quality Research in Beijing and Surrounding Region(CAREBeijing2006)campaign,comprehensive measurements were carried out in the megacity Beijing,where the chemical budget of HONO was fully constrained.The average diurnal HONO concentration varied from 0.33 to 1.2 ppbv.The net OH production rate from HONO,POH(HONO)net,was on average(from 05:00 to 19:00)7.1×106 molecule/(cm3 s),2.7 times higher than from O3 photolysis.This production rate demonstrates the important role of HONO in the atmospheric chemistry of megacity Beijing.An unknown HONO source(Punknown)with an average of 7.3×106molecule/(cm3 s)was derived from the budget analysis during daytime.Punknown provided four times more HONO than the reaction of NO with OH did.The diurnal variation of Punknown showed an apparent photo-enhanced feature with a maximum around 12:00,which was consistent with previous studies at forest and rural sites.Laboratory studies proposed new mechanisms to recruit NO2 and J(NO2)in order to explain a photo-enhancement of of Punknown.In this study,these mechanisms were validated against the observation-constraint Punknown.The reaction of exited NO2 accounted for only 6%of Punknown,and Punknown poorly correlated with[NO2](R=0.26)and J(NO2)[NO2](R=0.35).These results challenged the role of NO2 as a major precursor of the missing HONO source.YANG Qiang SU Hang LI Xin CHENG YaFang LU KeDing CHENG Peng GU JianWei GUO Song HU Min ZENG LiMin ZHU Tong ZHANG YuanHang 2014Science China Chemistry2014,57,7:8
10Research on the carbon isotopic composition of organic matter from Lake Chenghai and Caohai Lake sediments显示文摘The carbon isotopic composition of organic matter from lake sediments has been extensively used to infer variations in productivity. In this paper, based on the study of the contents and δ13C values of organic matter in different types of lakes, it has been found that δ13C values of organic matter have different responses to lake productivity in different lakes. As to the lakes dominated by aqutic macrophytes such as Lake Caohai, organic matter becomes enriched in 13C with increasing productivity. As to the lakes dominated by aquatic algae such as Lake Chenghai, δ13C values of organic matter decrease with increasing productivity, and the degradation of aquatic algae is the main factor leading to the decrease of δ13C values of organic matter with increasing productivity. 11LB100 ameliorates nonalcoholic fatty liver disease via the AMPK/Sirt1 pathway显示文摘BACKGROUND It is well known that nonalcoholic fatty liver disease(NAFLD)is associated with insulin resistance(IR).LB100,a serine/threonine protein phosphatase 2A(PP2A)inhibitor,is closely related to IR.However,there is little data regarding its direct influence on NAFLD.AIM To elucidate the effect and underlying mechanism of LB100 in NAFLD.METHODS After 10 wk of high fat diet(HFD)feeding,male C57BL/6 mice were injected intraperitoneally with vehicle or LB100 for an additional 6 wk(three times a week).The L02 cell line was treated with LB100 and free fatty acids(FFAs)for 24 h.Hematoxylin and eosin and oil red O staining were performed for histological examination.Western blot analysis was used to detect the protein expression of Sirtuin 1(Sirt1),total and phosphorylated AMP-activated protein kinaseα(AMPKα),and the proteins involved in lipogenesis and fatty acid oxidation.The mRNA levels were determined by qPCR.Pharmacological inhibition of AMPK was performed to further examine the exact mechanism of LB100 in NAFLD.RESULTS LB100 significantly ameliorated HFD-induced obesity,hepatic lipid accumulation and hepatic injury in mice.In addition,LB100 significantly downregulated the protein levels of acetyl-CoA carboxylase,sterol regulatory element-binding protein 1 and its lipogenesis target genes,including stearoyl-CoA desaturase-1 and fatty acid synthase,and upregulated the levels of proteins involved in fatty acidβ-oxidation,such as peroxisome proliferator-activated receptorα(PPARα),peroxisome proliferator-activated receptor gamma coactivator-1α(PGC-1α),carnitine palmitoyltransferase 1α,acyl-CoA oxidase 1 and uncoupling protein 2,as well as the upstream mediators Sirt1 and AMPKαin the livers of HFD-fed mice.In vitro,LB100 alleviated FFA-induced lipid accumulation in L02 cells through the AMPK/Sirt1 signaling pathway.Further studies showed that the curative effect of LB100 on lipid accumulation was abolished by inhibiting AMPKαin L02 cells.CONCLUSION PP2A inhibition by LB100 significantly ameliorates hepatic steatosis by regulating hepatic lipogenesis and fatty acid oxidation via the AMPK/Sirt1 pathway.LB100 may be a potential therapeutic agent for NAFLD.Xue-Yang Chen Chang-Zhou Cai Meng-Li Yu Ze-Min Feng Yu-Wei Zhang Pei-Hao Liu Hang Zeng Chao-Hui Yu 2019World Journal of Gastroenterology2019,25,45:7
12An online monitoring system for atmospheric nitrous acid(HONO) based on stripping coil and ion chromatography显示文摘A new instrument for measuring atmospheric nitrous acid(HONO) was developed,consisting of a double-wall glass stripping coil sampler coupled with ion chromatography(SC-IC).SC-IC is featured by small size(50 × 35 × 25 cm) and modular construction,including three independent parts:the sampling unit,the transfer and supporting unit,and the detection unit.High collection efficiency(> 99%) was achieved with 25 μmol/L Na2CO3 as absorption solution even in the presence of highly acidic compounds.This instrument has a detection limit of 8 pptv at 15 min time resolution,with a measurement uncertainty of 7%.Potential interferences from NOx,NO2+SO2,NO2+VOCs,HONO+O3,HNO3,peroxyacetyl nitrite(PAN) and particle nitrite were quantified in laboratory studies and were found to be insignificant under typical atmospheric conditions.Within the framework of the 3C-STAR project,inter-comparison between the SC-IC and LOPAP(long path liquid absorption photometer) was conducted at a rural site in the Pearl River Delta.Good agreement was achieved between the two instruments over three weeks.Both instruments determined a clear diurnal profile of ambient HONO concentrations from 0.1 to 2.5 ppbv.However,deviations were found for low ambient HONO concentrations(i.e.< 0.3 ppbv),which cannot be explained by previous investigated interference species.To accurately determine the HONO budget under illuminated conditions,more intercomparison of HONO measurement techniques is still needed in future studies,especially at low HONO concentrations.Peng Cheng Yafang Cheng Keding Lu Hang Su Qiang Yang Yikan Zou Yanran Zhao Huabing Dong Limin Zeng Yuanhang Zhang 2013Journal of Environmental Sciences2013,25,5:5
13Pharmacological Activation of RXR-a Promotes Hematoma Absorption via a PPAR-y-dependent Pathway After Intracerebral Hemorrhage显示文摘Endogenously eliminating the hematoma is a favorable strategy in addressing intracerebral hemorrhage(ICH).This study sought to determine the role of retinoid X receptor-ot(RXR-a)in the context of hematoma absorption after ICH.Our results showed that pharmacologically activating RXR-a with bexarotene significantly accelerated hematoma clearance and alleviated neurological dysfunction after ICH.RXR-ot was expressed in microglia/macro-phages,neurons,and astrocytes.Mechanistically,bexarotene promoted the nuclear translocation of RXR-a and PPAR-y,as well as reducing neuroinflammation by modulating microglia/macrophage reprograming from the Ml into the M2 phenotype.Furthermore,all the beneficial effects of RXR-a in ICH were reversed by the PPAR-y inhibitor GW9662.In conclusion,the pharmacological activation of RXR-a confers robust neuroprotection against ICH by accelerating hematoma clearance and repolarizing microglia/macrophages towards the M2 phenotype through PPAR-y-related mechanisms.Our data support the notion that RXR-ot might be a promising therapeutic target for ICH.Chaoran Xu Huaijun Chen Shengjun Zhou Chenjun Sun Xiaolong Xia Yucong Peng Jianfeng Zhuang Xiongjie Fu Hanhai Zeng Hang Zhou Yang Cao Qian Yu Yin Li Libin Hu Guoyang Zhou Feng Yan Gao Chen Jianru Li 2021Neuroscience Bulletin2021,37,10:4
14Combined amendment reduces soil Cd availability and rice Cd accumulation in three consecutive rice planting seasons显示文摘The scientific application of stabilized materials has been considered an effective method for the in situ remediation of Cd-contaminated soil.This study aimed to investigate the persistence of the effect of a combined amendment of limestone and sepiolite(LS)on soil Cd availability and accumulation in rice grown in a mildly Cd-contaminated paddy field(0.45 mg/kg of Cd)over three consecutive rice seasons.1125–4500 kg/ha of LS was applied to the soil before the first rice planting season and 562.5–2250 kg/ha of LS was supplemented before the third rice planting season.The application of LS(1125–4500 kg/ha)increased the soil p H by 0.44–1.09,0.18–0.53,and 0.42–0.68 in the first,second,and third season,respectively,and decreased the soil acid-extractable Cd content by 18.2–36.4%,17.7–33.5%,and9.6–17.6%.LS application significantly decreased the Cd contents in the rice tissues.The application of 4500 kg/ha of LS decreased the Cd content in brown rice to below the National Food Limit Standard of 0.2 mg/kg(GB 2762-2017)in the three consecutive rice seasons.However,the effect of LS on the soil-rice system was significantly weakened in the third season.The supplementary application of 562.5–2250 kg/ha of LS further decreased the Cd content in brown rice by 26.1–56.5%and decreased the health risk index by 23.7–43.8%.Therefore,it was recommended to apply 4500 kg/ha of LS in the first season and to supplement2250 kg/ha of LS in the third season to effectively guarantee the clean production of rice in three consecutive rice seasons.Wenjun Yang Shilong Wang Hang Zhou Min Zeng Jingyi Zhang Fang Huang Shiping Shan Zhaohui Guo Hongwei Yi Zhiguang Sun Jiaofeng Gu Bohan Liao 2022Journal of Environmental Sciences2022,34,1:4
15Glomerular Disease Associated with Takayasu Arteritis:6 Cases Analysis and Review of the Literature显示文摘Objective To evaluate the clinical features,renal histopathology and therapeutic response to glucocorticoid and immunosuppressive agents in patients with glomerular disease associated with Takayasu arteritis(TA).Methods Patients with TA and renal biopsy-confirmed glomerular disease were investigated retrospectively.None of them had renal artery stenosis or occlusive changes.Results Six patients with glomerulopathy,accounting for 3.75% of the 160 TA patients admitted to our hospital at the same period,were analyzed.All of them were females with a mean age of 35.5 ± 10.0 years.Four cases presented with lower extremity edema.Laboratory tests showed that one was nephrotic syndrome,three were nephrotic range proteinuria,and two of them had mild renal dysfunction.The other two patients were asymptomatic microscopic hematuria and proteinuria.Renal pathology revealed mild immunoglobulin A nephropathy in two cases,mild mesangial proliferative glomerulonephritis(GN),membranoproliferative GN,minimal change disease,and fibrillary GN in one case respectively.Five cases received glucocorticoids and cyclophosphamide therapy.Proteinuria and microscopic hematuria disappeared in 2 to 4 weeks after the initiation of therapy in three cases.The patient with membranoproliferative GN also reached complete remission of proteinuria and recovered renal function 6 months after the treatment.Conclusions TA may induce glomerular disease as a part of its histological spectrum.Apart from ischemic glomerular disease,glomerular disease should be suspected when TA patients have microscopic hematuria or proteinuria,that may be therapeutically responsive to glucocorticoids and immunosuppressive agent in relative early phase.Xue-mei Li Wen-ling Ye Yu-bing Wen Hang Li Li-meng Chen Dong-yan Liu Xue-jun Zeng Xue-wang Li 2009Chinese Medical Sciences Journal2009,24,2:4
16Curative effect of assisted reproduction technology by Traditional Chinese Medicine multi-channel interventional therapy on 95 cases of in vitro fertilization and embryo transfer failure显示文摘OBJECTIVE: To evaluate the curative effect of Traditional Chinese Medicine(TCM) multi-channel interventional therapy on women with Assisted Reproductive Technology(ART) failure, to compare the curative effect of the dual therapy and triple therapy on women with ART Failure, and to choose the best TCM interventional therapeutic plan.METHODS: The 95 cases with ART Failure from West China second University Hospital of Sichuan University meeting the inclusion criteria were randomly divided into three groups:dual therapy group(31 cases), triple therapy group(33 cases)and control group(31 cases). According to the intervene treatment of in vitro Fertilization and Embryo Transfer(IVF-ET) long cycle scheme, the control group wait naturally for 3 months before IVFET. The dual therapy group take TCM prescriptionⅡ of cultivating emotion and assisting reproduction and auricular acupoint therapy for 3 months before IVF-ET, then Western Medicine treatment progestin supporting as well as auricular application and Antai Recipe after IVF-ET transplantation.The triple therapy group take TCM prescription Ⅱof cultivated emotion and assisted reproduction,auricular acupoint therapy and retention enema of TCM, and combination treatment the same as dual therapy group after transplantation. The natural pregnancy number, the period condition of secondary IVF-ET and the improvement of the kidney deficiency, liver depression and blood stasis syndrome among those three groups were compared.RESULTS: It was showed from analysis in 95 cases with ART failure that the number of natural preg-nancy was as followings: 3 patients from the dual therapy group, 10 patients from the triple therapy group, and no patient from the control group. The comparison among three groups have statistical significance(P < 0.05). The treatment group is superior to the control group, while the triple therapy is superior to the dual therapy. The comparison of the condition of the fertility rate, clinical pregnancy rate, biochemical pregnancy rate and early abortion rate during the period of secondary IVF-ET between pre-therapy and post-treatment of both the dual therapy group and the triple therapy group have statistical significance(P < 0.05). The comparison of the improvement of the kidney deficiency,liver depression and blood stasis syndrome between pre-therapy and post-treatment of both the dual therapy group and the triple therapy group have statistical significance(P < 0.05). The comparison between three groups after treatment have statistical significance(P < 0.05).CONCLUSION: TCM multi-channel interventional therapy can increase the natural pregnancy rate of patients with ART Failure(the triple therapy is superior to the dual therapy); it can increase the fertility rate, clinical pregnancy rate, and decrease the early abortion rate during the period of secondary IVF-ET; it can improve syndromes of kidney deficiency, liver depression and blood stasis.Xia Wanting Zhong Yanmei Zhang Ying Xue Huarong Huang Jinzhu Wang Zhen Zhou Hang Ma Qianhong Zeng Qian 2017Journal of Traditional Chinese Medicine2017,37,5:4

19Dihydromyricetin promotes apoptosis, suppresses proliferation and tumor necrosis factor-α-mediated nuclear factor kappa-B activation in nasopharyngeal carcinoma CNE-2 cells显示文摘OBJECTIVE: To investigate the efficacy of dihydromyricetin(DMY) on nasopharyngeal carcinoma(NPC) cell proliferation, apoptosis and to reveal the underlying mechanism in vitro experiments.METHODS: The CNE-2 cell line was treated with different concentrations of DMY and the effects of DMY on cell viability and proliferation were evaluated using cell counting kit-8(CCK-8) assay and plate colony formation assay. Cellular apoptosis was detected by flow cytometry following Annexin V fluorescein isothiocyanate/propidine iodide staining.Nuclei morphology was observed under a fluorescence microscope following Hoechst 333258 staining. The expression of phosphorylated inhibitor of nuclear factor kappa-B kinase subunit beta(p-IKKβ), phosphorylated inhibitor of nuclear factor kappa-B kinase subunit alpha(p-IKKα), inhibitor of nuclear factor kappa-B alpha(IκB-α), nuclear factor kappa-B(NF-κB)/p65 was examined by Western blot analysis and the nuclear translocation of NF-κB/p65 was observed using a confocal laser scanning microscopy.RESULTS: DMY inhibited the proliferative capability and colony formation of NPC CNE-2 cells. Meanwhile, DMY induced apoptosis of CNE-2 cells in a dose and time-dependent manner via upregulating B-cell lymphoma-2 associated X, but downregulating B-cell lymphoma-2 and pro-caspase-3. Importantly, we found that DMY suppressed tumor necrosis factor alpha(TNF-α)-mediated NF-κB activation via inhibiting p-IKKβ, p-IKKα and blocking NF-κB subunit p65.CONCLUSION: Our experiments demonstrated that DMY had significant antiproliferative and apoptosisinducing effects on CNE-2 cells. Additionally, DMY promoted inactivation of p-IKKβ, p-IKKα,and blocked the nuclear translocation of NF-κB subunit p65. 20Segmental ureteroileal conduit resection for the treatment of distal upper urinary tract recurrence of bladder cancer fol lowing cystectomy显示文摘Segmental ureterectomy is less invasive than radical nephroureterectomy and results in nephron preservation and satisfactory tumor control.This study was to determine the feasibility of segmental ureteroileal conduit resection(SUICR) for patients with distal upper urinary tract recurrence of bladder cancer following radical cystectomy.Four patients with high-grade distal upper urinary tract recurrence underwent SUICR 15-108 months after radical cystectomy.The surgical technique details of SUICR,operative results,and follow-up outcomes are reported.The median operation time was 280 min,and estimated blood loss was less than 100 ml One patient suffered from ileus 5 days after surgery and was managed conservatively.Histopathologic evaluation showed high-grade stages pTa-pT1 diseases for these patients,and ureteral margins were all negative.No patient suffered from tumor recurrence,with a median follow-up of 39 months.SUICR preserved the ipsilateral renal unit and conformed to oncological principles during surgery.The oncological outcome was satisfactory for these properly selected patients.This technique provides a valid alternative to nephroureterectomy for patients with imperative indications and high-grade upper urinary tract recurrence of bladder cancer following radical cystectomy.Shu‑Xiong Zeng Xin Lu Wei‑Dong Xu Zhen‑Sheng Zhang Hai‑Hang Li Ying‑Hao Sun Chuan‑Liang Xu 2016Chinese Journal of Cancer2016,35,3:3
