| 2 | Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. | 3 | The “Traditional Chinese medicine regulating liver regeneration” treatment plan for reducing mortality of patients with hepatitis B-related liver failure based on real-world clinical data显示文摘On the basis of real-world clinical data,the study aimed to explore the effect and mechanisms of the treatment plan of“traditional Chinese medicine(TCM)regulating liver regeneration.”A total of 457 patients with HBV-related liver failure were retrospectively collected.The patients were divided into three groups:the modern medicine control group(MMC group),patients treated with routine medical treatment;the control group combining traditional Chinese and Western medicine(CTW),patients treated with routine medical treatment plus the common TCM formula;and the treatment group of“TCM regulating liver regeneration”(RLR),patients treated with both routine medical treatment and the special TCM formula of RLR.After 8 weeks of treatment,the mortality of patients in the RLR group(12.31%)was significantly lower than those in the MMC(50%)and CTW(29.11%)groups.Total bilirubin level significantly decreased and albumin increased in the RLR group when compared with the MMC and CTW groups(P<0.05).In addition,there were significant differences in the expression of several cytokines related to liver regeneration in the RLR group compared with the MMC group.RLR treatment can decrease jaundice,improve liver function,and significantly reduce the mortality in patients with HBV-related liver failure.The mechanism may be related to the role of RLR treatment in influencing cytokines related to liver regeneration. | Ling Dai Xiang Gao Zhihua Ye Hanmin Li Xin Yao Dingbo Lu Na Wu | 2021 | Frontiers of Medicine2021，15，3： | 4 |

| 5 | Monitoring fire regimes and assessing their driving factors in Central Asia显示文摘Relatively little is known about fire regimes in grassland and cropland in Central Asia.In this study,eleven variables of fire regimes were measured from 2001 to 2019 by utilizing the burned area and active fire product,which was obtained and processed from the GEE(Google Earth Engine)platform,to describe the incidence,inter-annual variability,peak month and size of fire in four land cover types(forest,grassland,cropland and bare land).Then all variables were clustered to define clusters of fire regimes with unique fire attributes using the K-means algorithm.Results showed that Kazakhstan(KAZ)was the most affected by fire in Central Asia.Fire regimes in cropland in KAZ had the frequent,large and intense characters,which covered large burned areas and had a long duration.Fires in grassland mainly occurred in central KAZ and had the small scale and high-intensity characters with different quarterly frequencies.Fires in forest were mainly distributed in northern KAZ and eastern KAZ.Although fires in grassland underwent a shift from more to less frequent from 2001 to 2019 in Central Asia,vigilance is needed because most fires in grassland occur suddenly and cause harm to humans and livestock. | YIN Hanmin Jiapaer GULI JIANG Liangliang YU Tao Jeanine UMUHOZA LI Xu | 2021 | Journal of Arid Land2021，13，5： | 1 |
