CRISPR-Cas13a mediated nanosystem for attomolar detection of canine parvovirus type 2显示文摘Canine parvovirus type 2(CPV-2) infection is the most lethal disease of dogs with higher mortality in puppies worldwide. In today’s world, dogs are an integral part of our communities as well as dogs breeding and rearing has become a lucrative business. Therefore,a fast, accurate, portable, and costeffective CPV-2 detection method with the ability for on-site detection is highly desired. In this study,we for the first time proposed a nanosystem for CPV-2 DNA detection with RNA-guided RNA endonuclease Cas13 a,which upon activation results in collateral RNA degradation. We expressed LwCasl3 a in prokaryotic expression system and purified it through nickel column.Activity of Cas13 a was verified by RNA-bound fluorescent group while using a quenched fluorescent probe as signals.Further Cas13 a was combined with Recombinase polymerase amplification(RPA) and T7 transcription to establish molecular detection system termed specific high-sensitivity enzymatic reporter un-locking(SHERLOCK) for sensitive detection of CPV-2 DNA.This nanosystem can detect 100 amol/L CPV-2 DNA within 30 min.The proposed nanosystem exhibited high specificity when tested for CPV-2 and other dog viruses. This CRISPR-Cas13 a mediated sensitive detection approach can be of formidable advantage during CPV-2 outbreaks because it is time-efficient,less laborious and does not involve the use of sophisticated instruments.Haroon Khan Adeel Khan Yufeng Liu Su Wang Sumaira Bibi Hongpan Xu Yuan Liu Samran Durrani Lian Jin Nongyue He Tao Xiong 2019Chinese Chemical Letters2019,30,12:8

Evaluating effectiveness of frequency ratio, fuzzy logic and logistic regression models in assessing landslide susceptibility: a case from Rudraprayag district, India显示文摘Rudraprayag in Garhwal Himalayan division is one of the most vulnerable districts to landslides in India. Heavy rainfall, steep slope and developmental activities are important factors for the occurrence of landslides in the district. Therefore, specific assessment of landslide susceptibility and its accuracy at regional level is essential for disaster management and proper land use planning. The article evaluates effectiveness of frequency ratio, fuzzy logic and logistic regression models for assessing landslide susceptibility in Rudraprayag district of Uttarakhand state, India. A landslide inventory map was prepared and verified by field data. Fourteen landslide parameters and generated inventory map were utilized to prepare landslide susceptibility maps through frequency ratio, fuzzy logic and logistic regression models. Landslide susceptibility maps generated through these models were classified into very high, high, medium, low and very low categories using natural breaks classification. Receiver operating characteristics(ROC) curve, spatially agreed area approach and seed cell area index(SCAI) method were used to validate the landslide models. Validation results revealed that fuzzy logic model was found to be more effective in assessing landslide susceptibility in the study area. The landslide susceptibility map generated through fuzzy logic model can be best utilized for landslide disaster management and effective land use planning.Mehebub SAHANA Haroon SAJJAD 2017Journal of Mountain Science2017,14,11:6
Antiglycation and antioxidation properties of juglans regia and calendula officinalis:possible role in reducing diabetic complications and slowing down ageing显示文摘OBJECTIVE:Accumulation of advanced glycation end products(AGEs) in the body due to the non-enzymatic glycation of proteins and oxidation is associated with aging and diabetes mellitus. In this study we wanted to investigate the antiglycation and antioxidation potential of two medicinal plants:Juglans regia and Calendula officinalis.METHODS:In-vitro investigation was carried out to discover the antiglycation and antioxidation potential of J.regia and C.officinalis.Using an Ultraviolet Double-beam Spectrophotometer,we evaluated the antiglycation property of the crude methanolic extracts of J.regia and C.officinalis by assessing their ability to inhibit the Maillard reaction.Employing the same instrument we also measured the antioxidation potential of these plant extracts using the nitric oxide(NO) free radical-scavenging assay.RESULTS:J.regia had greater antiglycation ability, with a minimum inhibitory concentration(MIC 50) of 28 μg/mL as compared with that of C.officinalis(270 μg/mL).C.officinalis had greater antioxidation potential(26.10,22.07 and 16.06% at 0.5 mg,0.25 mg and 0.125 mg,respectively,as compared with 18.15,16.50 and 16.06% of J.regia,respectively).CONCLUSION:J.regia and C.officinalis inhibited the Maillard reaction and prevented oxidation in-vitro.Hence,the extracts of these plants could have therapeutic uses in curbing chronic diabetic complications and slowing down aging.Haroon Ahmad Ibrar Khan Abdul Wahid 2012Journal of Traditional Chinese Medicine2012,32,3:5
Analysing the relationship between drought and soil erosion using vegetation health index and RUSLE models in Godavari middle sub-basin,India显示文摘Drought is a natural phenomenon posing severe implications for soil, groundwater and agricultural yield. It has been recognized as one of the most pervasive global change drivers to affect the soil.Soil being a weakly renewable resource takes a long time to form, but it takes no time to degrade. However, the response of soil to drought conditions as soil loss is not manifested in the existing literature. Thus, this study makes a concerted effort to analyze the relationship between drought conditions and soil erosion in the middle sub-basin of the Godavari River in India.MODIS remote sensing data was utilized for driving drought indices during 2000-2019.Firstly,we constricted Temperature condition index(TCI)and Vegetation Condition Index(VCI)from Land Surface Temperature(LST)and Enhanced Vegetation Index(EVI)derived from MODIS data.TCI and VCI were then integrated to determine the Vegetation Health Index(VHI).Revised Universal Soil Loss Equation(RUSLE)was utilized for estimating soil loss. The relationship between drought condition and vegetation was ascertained using the Pearson correlation. Most of the northern and southern watersheds experienced severe drought condition in the sub-basin during2000-2019.The mean frequency of the drought occurrence was 7.95 months. The average soil erosion in the sub-basin was estimated to be 9.88 t ha^(-1)year^(-1).A positive relationship was observed between drought indices and soil erosion values(r value being 0.35).However,wide variations were observed in the distribution of spatial correlation.Among various factors, the slope length and steepness were found to be the main drivers of soil erosion in the sub-basin.Thus,the study calls for policy measures to lessen the impact of drought and soil erosion.Md Masroor Haroon Sajjad Sufia Rehman Roshani Singh Md Hibjur Rahaman Mehebub Sahana Raihan Ahmed Ram Avtar 2022Geoscience Frontiers2022,13,2:4
Oxidative desulfurization of dibenzothiophene over Fe promoted Co–Mo/Al_2O_3 and Ni–Mo/Al_2O_3 catalysts using hydrogen peroxide and formic acid as oxidants显示文摘This work reports the enhancing effect of a highly cost effective and efficient metal, Fe, incorporation to Co or Ni based Mo/Al_2O_3 catalysts in the oxidative desulfurization(ODS) of dibenzothiophene(DBT) using H_2O_2 and formic acid as oxidants. The influence of operating parameters i.e. reaction time, catalyst dose, reaction temperature and oxidant amount on oxidation process was investigated. Results revealed that 99% DBT conversion was achieved at 60 °C and 150 min reaction time over Fe–Ni–Mo/Al_2O_3. Fe tremendously enhanced the ODS activity of Co or Ni based Mo/Al_2O_3 catalysts following the activity order: Fe–Ni–Mo/Al_2O_3 NFe–Co–Mo/Al_2O_3 NNi–Mo/Al_2O_3 NCo–Mo/Al_2O_3, while H_2O_2 exhibited higher oxidation activity than formic acid over all catalyst systems. Insight about the surface morphology and textural properties of fresh and spent catalysts were achieved using scanning electron microscopy(SEM), X-ray diffraction(XRD), energy dispersive X-ray(EDX)analysis, Atomic Absorption Spectroscopy(AAS) and BET surface area analysis, which helped in the interpretation of experimental data. The present study can be deemed as an effective approach on industrial level for ODS of fuel oils crediting to its high efficiency, low process/catalyst cost, safety and mild operating condition.Yaseen Muhammad Ayesha Shoukat Ata Ur Rahman Haroon Ur Rashid Waqas Ahmad 2018Chinese Journal of Chemical Engineering2018,26,3:3
Luminous composite ultrathin films of CdTe quantum dots/silk fibroin co-assembled with layered doubled hydroxide:Enhanced photoluminescence and biosensor application显示文摘Quantum dots(QDs)luminescent films are extensively applied to optoelectronics and optical devices. However,QDs aggregation results in the quenching of their fluorescence property which limits their practical applications to a greater extent. In order to resolve this issue,3-mercaptopropionic acid(3-MPA)functionalized Cadmium Tellurium(CdTe)QDs were stabilized by silk fibroin(SB)and coassembled with layered doubled hydroxide(LDH)to form(QDs@SF/LDH)_(n)ultrathin films(UTFs)via the layer-by-layer(LBL)technique.UV-Vis absorption and fluorescence spectroscopy showed a stepwise and normal growth of the films upon increasing the number of deposition cycles.XRD and AFM studies confirmed the formation of a periodic layered structure and regular surface morphology of the thin films. As compared to(CdTe QDs/LDH)_(n)UTFs,the(CdTe QDs@SF/LDH)_(n)UTFs displayed fluorescence enhancement and longer fluorescent lifetime, both in solid states and aqueous solutions.Furthermore compared with the solution state, the fluorescence enhancement of SF-RC and SF-β are,respectively,7 times and 17 times in the(CdTe QDs@SF/LDH)_(n)UTFs,indicating that the LDH nanosheets favor the fluorescence enhancement effect on the CdTe QDs@SF.The fabricated materials displayed fluorescence response to a biological molecule such as immune globulin,lgG.Thus, the(CdTe QDs@SF/LDH)_(n)UTFs has a potential to be used as biosensor.Muhammad Sohail Haroone Ling Li Aftab Ahmad Yaping Huang Ruili Ma Ping Zhang Yuehua Hu Qari Muhammad Kaleem Jun Lu 2018Journal of Materiomics2018,4,2:3
Impact of community-based interventions on HIV knowledge, attitudes, and transmission显示文摘In 2012,an estimated 35.3 million people lived with HIV,while approximately two million new HIV infections were reported.Community-based interventions(CBIs)for the prevention and control of HIV allow increased access and ease availability of medical care to population at risk,or already infected with,HIV.This paper evaluates the impact of CBIs on HIV knowledge, attitudes, and transmission. We included 39 studies on educational activities, counseling sessions, home visits, mentoring, women’s groups, peer leadership, and street outreach activities in community settings that aimed to increase awareness on HIV/AIDS risk factors and ensure treatment adherence. Our review findings suggest that CBIs to increase HIV awareness and risk reduction are effective in improving knowledge, attitudes, and practice outcomes as evidenced by the increased knowledge scores for HIV/AIDS(SMD:0.66,95%CI:0.25,1.07),protected sexual encounters(RR:1.19,95%CI:1.13,1.25),condom use(SMD:0.96,95%CI:0.03,1.58),and decreased frequency of sexual intercourse(RR:0.76,95%CI:0.61,0.96).Analysis shows that CBIs did not have any significant impact on scores for self-efficacy and communication. We found very limited evidence on community-based management for HIV infected population and prevention of mother-to-child transmission(MTCT)for HIV-infected pregnant women.Qualitative synthesis suggests that establishment of community support at the onset of HIV prevention programs leads to community acceptance and engagement.School-based delivery of HIV prevention education and contraceptive distribution have also been advocated as potential strategies to target high-risk youth group.Future studies should focus on evaluating the effectiveness of community delivery platforms for prevention of MTCT,and various emerging models of care to improve morbidity and mortality outcomes.Rehana A Salam Sarah Haroon Hashim H Ahmed Jai K Das Zulfiqar A Bhutta 2014Infectious Diseases of Poverty2014,3,1:3
Why more research needs to be done on echinococcosis in Pakistan显示文摘Background:Echinococcosis has a worldwide geographical distribution with endemic foci on every inhabited continent.Due to the frequent outbreaks in different parts of Pakistan in the recent past, echinococcosis is being described as a neglected tropical disease and is considered one of the most neglected parasitic diseases in the country. In endemic regions, predominantly settings with limited resources, there are high numbers of echinococcosis patients,as these communities do not have access to appropriate treatment. In Pakistan, there are limited reports on echinococcosis. The disease is prevalent in human and livestock, but this has not been sufficiently explored yet.Pakistan is an agricultural country and due to the disease’s zoonotic mode of transmission, there is a dire need of future research on it.The present paper is an effort to highlight the importance of echinococcosis in Pakistan.Discussion:There is a dire need for future research on echinococcosis in Pakistan as very few investigations had been carried out on this topic thus far.The prevalence of the disease in neighbouring countries highlights that Pakistan might be at severe risk of this zoonotic infection and further supports the need for more research. In Pakistan, the majority of the population lives in rural areas with limited acess to proper hygienic/sanitary facilities. These conditions favour the outbreak of diseases such as echinococcosis. The limited available data could result in higher outbreaks in the future, and thus cause the already weak healthcare system to overburden. The country has a meagre annual budget for health, which is being spent on known infections such as polio, dengue fever and hepatic viral infections.A proper surveillance system for echinococcosis is required across the country as treatment is usually expensive, complicated and may require extensive surgery and/or prolonged drug therapy.Development of new/novel drugs and other treatment modalities receives very little,if any, attention.Prognostic awareness programmes against this infection involve deworming of the infected animals, improved food inspection and slaughterhouse hygiene, and public education campaigns. Conclusion:Future research on echinococcosis is anticipated to demonstrate whether the epidemiology, diagnosis and recombinant vaccines/antibodies relating to echinococcosis can meet the quality standards(purity, potency, safety and efficacy)defined by the World Health Organization.Research work should be carried out on the epidemiology and serodiagnosis of echinocossis in the different areas of Pakistan, which will be useful for the proper eradication of echinococcosis in this region. The health department should implement awareness-raising campaigns for the general public in order to reduce the burden of disease.Haroon Ahmed Shahzad Ali Muhammad Sohail Afzal Abid Ali Khan Hamid Raza Zaheer Hussain Shah Sami Simsek 2017Infectious Diseases of Poverty2017,6,1:2

Genetic effects conferring heat tolerance in upland cotton(Gossypium hirsutum L.)显示文摘Background:Climate change and particularly global warming has emerged as an alarming threat to the crop productivity of field crops and exerted drastic effects on the cropping patterns.Production of cotton has been dropped down to one million bales from 1.4 million bales since 2015 in Pakistan due to the increase in temperature at critical growth stages,i.e.,flowering and boll formation.Keeping in view the importance of cotton in the country, this study was conducted to investigate the genetic effects conferring heat tolerance in six populations(P1,P2,F1,F2,BC1 and BC2)developed from cross-1 and cross-2,i.e.,VH-282×FH-142 and DNH-40×VH-259.Results:The results revealed that cross-1 performed better in heat stress as compared with cross-2 for majority of the traits recorded.Boll weight and ginning outturn(GO-T)were highly effected under heat stress and had negative correlation with Relative cell injury(RCI).Boll weight, fiber length, fiber strength and fiber fineness were under the control of non-additive gene action, whereas RCI was controlled by additive gene effects.Lower values of genetic advance coupled with higher values of broad sense heritability for these traits except RCI confirmed the role of non-additive genetic effects.Duplicate types of epistasis were recorded for fiber strength in cross-1 in normal condition. However, complementary type of non-allelic interaction was recorded for fiber strength under normal condition, fiber fineness and RCI under heat stressed condition in cross-1.Likewise,boll weight,GOT and fiber length in populations derived from cross-2 in normal condition were also under the influence of complementary type of non-allelic interaction.Significant differences among values of mid parent and better parent heterosis for boll weight in both normal and heat stress condition provided the opportunity to cotton breeders for utilization of this germplasm for improvement of this trait through exploitation of heterosis breeding.Conclusion:Cross-1 performed better in heat stress and could be utilized for development of heat tolerant cultivar.RCI was under the influence of additive gene action,so one can rely on this trait for screening of large number of accessions of cotton for heat stress. While other traits were predominantly controlled by non-additive gene action and selection based on these should be delayed in later generations.SALMAN Muhammad ZIA Zia Ullah RANA Iqrar Ahmad MAQSOOD Rana Haroon AHMAD Saghir BAKHSH Ali AZHAR Muhammad Tehseen 2019Journal of Cotton Research2019,2,1:2

Impacts of center pivot irrigation system uniformity on growth of potato crop and residual soil nitrogen显示文摘Maintaining the homogeneity of soil nitrogen(N)and plant vigor across agricultural fields is a major concern for farmers and agricultural scheme planners, particularly fields that are irrigated through pressurized systems, such as center pivots. Therefore, this study was carried out on a 30 hm2 potato field located 650 km south of Riyadh, the capital city of the Kingdom of Saudi Arabia,to investigate the impacts of the center pivot irrigation distribution uniformity on the crop development and the spatial distribution of residual soil N.Irrigation performance test was designed to investigate water application rate and distribution uniformities. The overall water application uniformity coefficients(Cu),determined through Christiansen(Cud)and Heerman(CuH)methods, were determined at 81.29%and 80.64%,respectively.However,the overall water distribution uniformity(Du)was determined at 70%.A considerable variability in the distribution uniformity of irrigation water was observed across the experimental field(a Du value of 67%over the medium spans compared to a Du value of 88%over the inner spans). Results of this study showed a linear correlation between the irrigation water distribution uniformity and the soil N(R^(2)=0.88).On the other hand, the vegetation cover distribution, indicated by the Cumulative Normalized Difference Vegetation Index(CNDVI),was not found to be much responsive to the irrigation distribution uniformity(R^(2)=0.11).A time series of successive NDVI maps extracted throughout the potato crop growth stages showed a consistent trend in the distribution of NDVI across the field, with R2 values that ranged between 0.25-0.73.Khalid A.Al-Gaadi Abdalhaleem A.Hassaballa ElKamil Tola Ahmed G.Kayad Rangaswamy Madugundu Fahad Assiri Mohamed Edrris Ahmed Alameen Haroon Edrees 2019International Journal of Agricultural and Biological Engineering2019,12,1:2
Dramatic response of hepatitis C patients chronically infected with hepatitis C virus genotype 3 to sofosbuvirbased therapies in Punjab, Pakistan: A prospective study显示文摘AIM To prospectively evaluate the efficacy of sofobuvir(SOF) in hepatitis C patients infected with hepatitis C virus(HCV) genotype 3 in Pakistan.METHODS The present study was performed with the coordination of gastroenterology and pathology departments of Shalamar Hospital Lahore from August 2014 to May 2016. The total number of patients included in this study was 1375 and all of them were infected with HCV genotype 3. On the basis of drug combinations, all the patients were separated into two groups. The first group of patients was treated for 24 wk with SOF(Sovaldi? by Gilead Sciences) plus ribavirin(RBV) [Ribazol? by Getz Pharma Pakistan(PVT) Ltd], while the patients of the second group were treated with SOF + RBV + pegylatedinterferon(peg IFN) alfa-2 a(Ropegra by Roach) for 12 wk. HCV genotyping and viral load measurement were performed on fully automated Abbott Real-Time PCR system(Abbott m24 sp automated nucleic acid extraction system and Abbott m2000 rt amplification system; abbott Molecular, Des Plaines, IL, United States). For the assessment of sustained virological response(SVR), all HCV RNA negative patients were followed for 12weeks after the treatment completion. Any patient with less than 12 IU/m L viral load after 12 wk of treatment completion was considered as a sustained virological responder(SVR-12).RESULTS A total of 1375 patients chronically infected with HCV genotype 3 were treated with two drug combinations SOF + RBV and SOF + RBV + peg IFN alfa-2 a. On the basis of these drug combinations, patients were divided into two groups(first and second). Overall SVR-12 was excellent in both groups(99.17% and 97.91%). Older patients(> 40 years) of second group showed lower SVR-12(93.46%) compared to first group older patients(98.79%), while in the younger patients of both groups, the SVR-12 rate was almost the same(99.54% in first group and 99.05% in second group). No such difference regarding SVR-12 rate was seen in males and females of first group patients(99.68% and 98.88%, respectively), while in second group the males were found to be better responders compared to females(98.96% and 95%). The SVR-12 rate in previously treated patients of first group was better(99.34%) than second group(93.70%), while na?ve patients of second group were marginally better responders(99.25%) than first group(97.80%). Rapid viral response at week-4 was found to be a very effective predictor for assessing the SVR rate at this stage of therapy in both groups. Headache, anemia and fatigue were common side effects in both groups either treated with SOF + RBV or SOF + RBV + peg IFN alfa-2 a, while the overall percentage of the side effects was higher in second group.CONCLUSION The remarkable SVR response rate of HCV genotype 3 infected patients to SOF provided a new way to look forward to eliminate hepatitis C from our region.Sajjad Iqbal Muhammad Haroon Yousuf Muhammad Iftikhar Yousaf 2017World Journal of Gastroenterology2017,23,44:2
A hybrid circuit breaker with fault current limiter circuit in a VSC-HVDC application显示文摘A conventional hybrid circuit breaker(HCB)is used to protect a voltage source converter-based high voltage direct current transmission system(VSC-HVDC)from a short circuit fault. With the increased converter capacity, the DC protection equipment also requires a regular upgrade. This paper adopts a novel type of HCB with a fault current limiter circuit(FCLC),and focuses on the responses of voltage and current during DC faults, which are associated with parameter selection.PSCAD/EMTDC based simulation of a three-terminal VSC-HVDC system confirms the effectiveness and value of HCB with FCLC,by using an equivalent circuit modelling approach.Laboratory experimental tests validate the simulation results. The peak fault current is reduced according to the current limiting inductor(CLI)increase, and can be isolated more quickly.By adopting parallel metal oxide arrester(MOA)with the main branch of HCB,voltage stresses across the breaker components decrease during transient and continuous operation, and less energy needs to be dissipated by the MOA.The remnant current for all cases is transmitted to power dissipating resis-tor(PDR)in the final stage, and the fault current is reduced to the lowest possible value. When the current from the main branch is transferred to the FCLC branch, transient voltage spikes occur, while smaller PDR is required to absorb current in the final stage.Muhammad Ahmad Chunyang Gong Muhammad Haroon Nadeem Hui Chen Zhixin Wang 2022Protection and Control of Modern Power Systems2022,7,1:1
