| 1 | Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment. | Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show | 2020 | Cell Research2020,30,10: | 19 |
| 2 | Experimental studies of explosion energy output with different igniter mass显示文摘For study the energy output law of cylindrical charge with shell induced by different input energies,four different black powder masses were selected to ignite the main charge.Fragmentation degree of the shell was qualitatively analyzed by the area of holes on the witness plate and the recovered fragments mass.Through theoretical analysis,established the functional relationship between the average mass of fragments and the relative energy output of warhead,obtained how the relative energy output of charge changed with different initial energy input.The results showed that the change of input energy could lead to obvious variation in fragment characteristics,and could also control the output of charge.When the igniter mass increases from 1.55 g to 5.00 g,the relative energy output of the charge increases by 26.28%.Excessive initial input energy will destroy the shell confine in advance,resulting in a decrease in the relative energy output of charge. | Heyang Xu Weibing Li Wenbin Li Yajun Wang | 2019 | Defence Technology(防务技术)2019,15,5: | 1 |
| 3 | SIN3B promotes integrin αV subunit gene transcription and cell migration of hepatocellular carcinoma显示文摘Paired amphipathic helix protein (SIN3B) is a transcription corepressor for many genes. Here we show a different regulation mechanism of integrin aV gene expression by SIN3B in human hepatocellular carcinoma (HCC). We first observed a close relationship between Integrin aV and SIN3B expressions in HCC patients and tumor cell lines with different metastatic potentials. Overexpression of SIN3B significantly accelerated the cell migration rate of SMMC-7721, but failed when integrin αV expression was silenced. Interestingly, SIN3B stimulated integrin aV subunit promoter activity only in the presence of sulfatide. Importantly, SIN3B was identified in the complex with sulfatide by mass spectrometry. Fat blot assay indicated that SIN3B specifically interacted with sulfatide. Molecular modeling suggested that sulfatide induced the conformational change of SIN3B from compacted α-helices to a relaxed β-sheet in PAH2 domain. The data of immunoprecipitation and ChIP assay indicated that altered SIN3B lost the binding affinity with MAD1 and HDAC2, which reduced the recruitment of HDAC2 on integrin aV gene promoter and prevented the deacetylation of the histone 3. | 4 | Spatial changes and driving factors of lake water quality in Inner Mongolia, China显示文摘Lakes play important roles in sustaining the ecosystem and economic development in Inner Mongolia Autonomous Region of China,but the spatial patterns and driving mechanisms of water quality in lakes so far remain unclear.This study aimed to identify the spatial changes in water quality and the driving factors of seven lakes(Juyanhai Lake,Ulansuhai Lake,Hongjiannao Lake,Daihai Lake,Chagannaoer Lake,Hulun Lake,and Wulannuoer Lake)across the longitudinal axis(from the west to the east)of Inner Mongolia.Large-scale research was conducted using the comprehensive trophic level index(TLI(Σ)),multivariate statistics,and spatial analysis methods.The results showed that most lakes in Inner Mongolia were weakly alkaline.Total dissolved solids and salinity of lake water showed obvious zonation characteristics.Nitrogen and phosphorus were identified as the main pollutants in lakes,with high average concentrations of total nitrogen and total phosphorus being of 4.05 and 0.21 mg/L,respectively.The values of TLI(Σ)ranged from 49.14 to 71.77,indicating varying degrees of lake eutrophication,and phosphorus was the main driver of lake eutrophication.The lakes of Inner Mongolia could be categorized into lakes to the west of Daihai Lake and lakes to the east of Daihai Lake in terms of salinity and TLI(Σ).The salinity levels of lakes to the west of Daihai Lake exceeded those of lakes to the east of Daihai Lake,whereas the opposite trend was observed for lake trophic level.The intensity and mode of anthropogenic activities were the driving factors of the spatial patterns of lake water quality.It is recommended to control the impact of anthropogenic activities on the water quality of lakes in Inner Mongolia to improve lake ecological environment.These findings provide a more thorough understanding of the driving mechanism of the spatial patterns of water quality in lakes of Inner Mongolia,which can be used to develop strategies for lake ecosystem protection and water resources management in this region. | REN Xiaohui YU Ruihong LIU Xinyu SUN Heyang GENG Yue QI Zhen ZHANG Zhuangzhuang LI Xiangwei WANG Jun ZHU Penghang GUO Zhiwei WANG Lixin XU Jifei | 2023 | Journal of Arid Land2023,15,2: | 0 |
