| 1 | Gα_s Relays Sphingosine-1-Phosphate Receptor 1 Signaling to Stabilize Vascular Endothelial-Cadherin at Endothelial Junctions to Control Mouse Embryonic Vascular Integrity显示文摘Sphingosine-1-phosphate receptor 1(S1PR1),a G protein-coupled receptor(GPCR),controls vascular stability by stabilizing vascular endothelial(VE)-cadherin junctional localization and inhibiting vascular endothelial growth factor receptor 2(VEGFR2) signaling.However,the molecular mechanisms that link S 1PR1 signaling to intracellular effectors remain unknown,In this study,we demonstrate that the heterotrimeric G protein subfamily member Gα_s,encoded by GNAS,acts as a relay mediator of S1PR1 signaling to control vascular integrity by stabilizing VE-cadherin at endothelial junctions.The endothelial cell-specific deletion of Gαs in mice causes early embryonic lethality with massive hemorrhage and a disorganized vasculature.The immunostalning results revealed that Gα_s deletion remarkably reduces the junctional localization of VE-cadherin,whereas the mural cell coverage of the vessels is not impaired.In addition,we found that Gαs depletion blocks the S1PRl-activation induced VE-cadherin stabilization at junctions,supporting that Gα_s acts downstream of S1PR1 signaling,Thus,our results demonstrate that Gα_s is an essential mediator to relay S1PR1 signaling and maintain vascular integrity. | Ximing Shao Ke Liu Yi Fan Zhihao Ding Min Chen Minyan Zhu Lee S.Weinstein Hongchang Li Huashun Li | 2015 | Journal of Genetics and Genomics2015,42,11: | 8 |
| 2 | Isolation and characterization of a heat-resistant strain with high yield of Pyropia yezoensis Ueda(Bangiales,Rhodophyta)显示文摘A heat-resistant Pyropia yezoensis strain YZ-4 was isolated from the regenerated blades of single somatic cells of the wild-type(WT)blades treated with 60Co-g-irradiation.When cultured at 23C and 24C,the survival and division rates of the YZ-4 conchospores were significantly higher than those of WT(P<0.01).After 16 days in culture the survival rates of YZ-4 conchospores were 71.2%and 60.8%at 23C and 24C,respectively,and the division rates of the surviving conchospores were 100%in both cases.Under the same culture conditions,the survival rates of WT conchospores were only 16.9%and 11.5%,and the division rate 99.8%and 81.8%respectively.The resistance of 50-day-aged F1 gametophytic blades of YZ-4 to the temperature rebound was also significantly higher than those of WT(P<0.01).After 30 days in culture at 18C,23C and 24C,the mean blade length increased by 8.5,4.3 and 2.3 times in YZ-4 and was only 3,0.6 and 0.4 times in WT,respectively.The mean blade weight increased 191.9,85.4 and 66.2 times in YZ-4,but only 78.2,7.8 and 4.6 times in WT.On the 15th day WT blades began to decay,but no decay was observed in YZ-4 blades even after 45 days in culture at 23C and 24C.The results obtained indicate that YZ-4 has a higher resistance to higher temperature insults than WT and it might be of value for commercial production of P.yezoensis. | Hongchang Ding Binglei Zhang Xinghong Yan | 2016 | Aquaculture and Fisheries2016,1,1: | 5 |
| 3 | Novel application of bergapten and quercetin with anti-bacterial,osteogenesis-potentiating,and anti-inflammation tri-effects显示文摘The bacteria-mediated inflammatory conditions adversely affect the osseointegration process of endosseous implants,which can even lead to implant malfunction or failure.Local drug delivery has been designed to exert anti-inflammatory and antibacterial activities,but whether this strategy has an effect on the compromised osseointegration under inflammation has rarely been studied.The present study focused on the osteoinductive efficacy of two known phytoestrogens[bergapten(BP)and quercetin(QE)]on implant sites under multiple bacteria-infected conditions in situ.Furthermore,the gene expression profiles of rat bone mesenchymal stem cells(rBMSCs)treated with BP and QE in the presence of Porphyromonas gingivalis-derived lipopolysaccharide were identified.The results showed that both drugs,especially QE,had significant potentiating effects on promoting osteogenic differentiation of rBMSCs,resisting multiple pathogens,and reducing inflammatory activity.Meanwhile,RNA sequencing analysis highlighted the enriched gene ontology terms and the differentially expressed genes(Vps25,Il1r2,Csf3,Efemp1,and Ccl20)that might play essential roles in regulating the above tri-effects,which provided the basis for the drug delivery system to be used as a novel therapeutic strategy for integrating peri-implant health.Overall,our study confirmed that QE appeared to outperform BP in osteogenesis and bacterial killing but not in anti-inflammation.Moreover,both drugs possess favorable tri-effects and can serve as the pivotal agents for the drug delivery system to boost osseointegration at inflammatory implant sites. | Jianxu Wei Xiaomeng Zhang Yuan Li Xinxin Ding Yi Zhang Xue Jiang Hongchang Lai Junyu Shi | 2021 | Acta Biochimica et Biophysica Sinica2021,53,6: | 4 |

| 5 | Developments of crawler steering gearbox for combine harvester straight forward and steering in situ显示文摘Soil compaction and accumulation are caused by crawler steering.Soil characteristics would affect the ability of crawler straight forward and steering in situ.This paper studied the principle of straight forward steering and steering in situ mode of traditional unilateral brake steering gearbox of a crawler combine harvester.Then a positive and negative steering gearbox model was developed and processed.The performance of positive and negative steering gearbox was analyzed by performing two steering modes on soil compaction effect.Results showed that the positive and negative steering gearboxes can achieve three operating modes,such as straight forward,unilateral brake steering,and positive and negative steering in situ.The positive and negative steering in situ was smooth and without soil accumulation phenomenon.When the crawlers center distance,crawler grounding length,crawler width were 1150 mm,1620 mm and 450 mm respectively,the rolling area and the scratch free area of positive and negative steering gearbox were 5.37 m2 and 0.36 m2 respectively.However,the rolling area and the scratch free area produced by unilateral brake steering gearbox were 13.92 m2 and 1.88 m2 respectively,which was 8.55 m2 larger than that of positive and negative steering gearbox.The crawler with positive and negative steering gearbox has minor damage to paddy soil.This research can provide a theoretical basis for the smooth steering in situ and structural optimization of the crawler steering gearbox. | Zhong Tang Haotian Zhang Hongchang Li Yaoming Li Zhao Ding Jinsong Chen | 2020 | International Journal of Agricultural and Biological Engineering2020,13,1: | 1 |
| 6 | Single somatic cell development and differentiation in the gametiphytic blades of Pyropia suborbiculata (Bangiales, Rhodophyta)显示文摘Conchospores released from a wild-type strain of Pyropia suborbiculata were cultured in the laboratory for 20-80 days.Mother blades of different ages were enzymatically isolated to obtain single cells,which then developed into different types of regenerative plants within a liquid medium.These regenerative plant types includes:normal blades,abnormal blades,cell-masses,and sexual cell-masses.The age of mother blades distinctly affected the number and proportion of each type of regenerative plants.When the age of mother blades increased from 20 to 80 days,the proportion of normal blades in the isolated cells sharply decreased,the percentage of abnormal blades increased,and the proportion of cell-masses was relatively unchanged.After 40 days of cultivation,sexual cell-masses appeared in regenerated plants derived from the single blade cells of the PS-WT strain,and gradually increased with mother cell age.In addition,the proportion of normal blades decreased in regenerated plants sourced from the basal to apical parts of the same alga,while the proportion of abnormal blades and cell-masses increased.In summary,the isolated cells of the gametophytic blade of P.suborbiculata developed into different types of regenerated plantlets in vivo due to the different stages of cell differentiation.We preliminarily concluded that the differentiation from conchospores to the sexual cells could be divided into at least seven different groups. | Wenxia Guo Hongchang Ding Xinghong Yan | 2021 | Aquaculture and Fisheries2021,6,5: | 0 |
| 7 | Modeling and Control Strategy of Built-in Skin Effect Electric Tracing System显示文摘In order to ensure the safety of fluid flow in deep-water submarine pipelines,a safe and energy-saving built-in skin effect electric heat tracing technology was adopted as the thermal management strategy.The magnetic field distribution of built-in skin effect heating system is analyzed based on the mechanism of built-in skin effect heating system,so as to obtain the equivalent circuit model of built-in skin effect electric heating system.Meanwhile,heating power is introduced as an intermediate variable to establish the relationship between power supply frequency and built-in skin effect heating temperature.Aiming at the skin effect electric heating system,an Active Disturbance Rejection Control(ADRC)method is proposed macroscopically based on Hammerstein model.Firstly,the parameters of Hammerstein model are identified and optimized using the auxiliary model and standard particle swarm optimization algorithm.Then,the ADRC controller of linear link is designed,and the required heating temperature is used to solve the intermediate variable heating power.Finally,inversion calculation is applied in the nonlinear link to solve the required power frequency,so as to achieve the purpose of efficient heating and verify the feasibility and effectiveness of control strategy through simulation. | Li Ding Jiasheng Zhang Hongchang Sun | 2018 | Computer Modeling in Engineering & Sciences2018,,11: | 0 |
