| 1 | Targets and molecular mechanisms of triptolide in cancer therapy显示文摘Triptolide(TPL/TL) is a natural drug with novel anticancer effects. Preclinical studies indicated that TPL inhibits cell proliferation, induces cell apoptosis, inhibits tumor metastasis and enhances the effect of other therapeutic methods in various cancer cell lines. Multiple molecules and signaling pathways, such as caspases, heat-shock proteins, NF-κB, and deoxyribonucleic acid(DNA) repair-associated factors, are associated with the anti-cancer effect. TPL also improves chemoradiosensitivity in cancer therapy. Phase I trials indicate the potential clinical value of TPL use. | 2 | MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple. | Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen | 2019 | Horticulture Research2019,6,1: | 16 |

| 9 | Bioresponsive cisplatin crosslinked albumin hydrogel served for efficient cancer combination therapy显示文摘Combination therapy is one of the potential strategies for tackling complicated tumor treatments like drug resistance.In this work,we have generated a therapeutic cisplatin-crosslinked albumin hydrogel(BC-Gel)that allows the local release of L-Buthionine-sulfoximine(BSO),cisplatin,and glucose oxidase(GOx)with distinct release kinetics.The BC-Gel with favorable biostimuli degradability and injectability could release therapeutic agents in a programmed manner within the tumor microenvironment(TME).The preferentially released BSO significantly suppressed the glutathione(GSH)-related cisplatin resistance and sensitized the tumor cells to cisplatin by inhibiting theγ-glutamylcysteine synthetase.Meanwhile,cisplatin achieved a sequential release and long-term treatment following the bioresponsive gel degradation under the combined action of chloride ions(Cl−)and proteinase in the body.In addition,the overproduced H_(2)O_(2)of GOx-catalyzed glucose oxidation accelerated the depletion of existed GSH within cells and further weakened the cisplatin resistance,achieving enhanced tumor treatment together with a strong cell-killing effect.The above sequential drug release strategy based on the dual GSH depletion effect breaks the balance of the GSH-mediated redox TME and enhances the sensitivity of A549 cells to cisplatin forcefully,and provides a promising way for temporal control of drug release as well as efficient cancer combination therapy. | An Yan Zherui Zhang Jiamei Gu Xiaoran Ding Yongchen Chen Jingjing Du Shu Wei Hongcheng Sun Jiayun Xu Shuangjiang Yu Junqiu Liu | 2023 | Nano Research2023,16,2: | 1 |

Methods: The expression of CEA, STAT3 mRNA and it's protein in human lung adenocarcinoma cell A549 and normal human lung cells MRC-5 were tested by immunohistochemistry staining (PV) and quantitative real time fluorescent PCR. The correlation between STAT3 and CEA in human lung adenocarcinoma cell A549 was analyzed. Results: The protein and mRNA levels of STAT3, CEA in lung adenocarcinoma cell A549 were apparently higher than those in normal human lung cells MRC-5. The levels of STAT3 mRNA and it's protein were positively correlated with CEA in lung adenocarcinoma cell A549. | 12 | Report of China's innovation increase and research growth in radiation oncology显示文摘Aims: To investigate the research status of radiation oncology in China through survey of literature in international radiation oncology journals and retrospectively compare the outputs of radiation oncology articles of the three major regions of China—Mainland(ML), Taiwan(TW) and Hong Kong(HK).Methods: Radiation oncology journals were selected from 'oncology' and 'radiology, nuclear & medical image' category from Science Citation Index Expand(SCIE). Articles from the ML, TW and HK were retrieved from MEDLINE. The number of total articles, clinical trials, case reports, impact factors(IF), institutions and articles published in each journals were conducted for quantity and quality comparisons.Results: A total 818 articles from 13 radiation oncology journals were searched, of which 427 are from ML, 259 from TW, and 132 from HK. Ninety-seven clinical trials and 5 case reports are reported in China. Accumulated IF of articles from ML(1,417.11) was much higher than that of TW(1,003.093) and HK(544.711), while the average IF of articles from ML is the lowest.Conclusions: The total number of articles from China especially ML increased significantly in the last decade. The number of articles published from the ML has exceeded those from TW and HK. | 14 | Low-Energy Protons in Strong-Field Dissociation of H_(2)^(+) via Dipole-Transitions at Large Bond Lengths显示文摘More than ten years ago,the observation of the low-energy structure in the photoelectron energy spectrum,regarded as an“ionization surprise,”has overthrown our understanding of strong-field physics.However,the similar low-energy nuclear fragment generation from dissociating molecules upon the photon energy absorption,one of the well-observed phenomena in light-molecule interaction,still lacks an unambiguous mechanism and remains mysterious.Here,we introduce a time-energy-resolved manner using a multicycle near-infrared femtosecond laser pulse to identify the physical origin of the light-induced ultrafast dynamics of molecules.By simultaneously measuring the bond-stretching times and photon numbers involved in the dissociation of H_(2)^(+) driven by a polarization-skewed laser pulse,we reveal that the low-energy protons(below 0.7 eV)are produced via dipole-transitions at large bond lengths.The observed low-energy protons originate from strong-field dissociation of high vibrational states rather than the low ones of H_(2)^(+) cation,which is distinct from the well-accepted bond-softening picture.Further numerical simulation of the time-dependent Schrödinger equation unveils that the electronic states are periodically distorted by the strong laser field,and the energy gap between the field-dressed transient electronic states may favor the one-or three-photon transitions at the internuclear distance larger than 5 a.u.The time-dependent scenario and our time-energy-resolved approach presented here can be extended to other molecules to understand the complex ultrafast dynamics. | Shengzhe Pan Chenxi Hu Zhaohan Zhang Peifen Lu Chenxu Lu Lianrong Zhou Jiawei Wang Fenghao Sun Junjie Qiang Hui Li Hongcheng Ni Xiaochun Gong Feng He Jian Wu | 2022 | Ultrafast Science2022,,2: | 0 |
