| 1 | Mechanistic and therapeutic advances in non-alcoholic fatty liver disease by targeting the gut microbiota显示文摘Non-alcoholic fatty liver disease (NAFLD)is one of the most common metabolic diseases currently in the context of obesity worldwide,which contains a spectrum of chronic liver diseases,including hepatic steatosis, non-alcoholic steatohepatitis and hepatic carcinoma.In addition to the classical 'Two-hit'theory,NAFLD has been recognized as a typical gut microbiota-related disease because of the intricate role of gut microbiota in maintaining human health and disease formation.Moreover,gut microbiota is even regarded as a 'metabolic organ'that play complementary roles to that of liver in many aspects.The mechanisms underlying gut microbiota-mediated development of NAFLD include modulation of host energy metabolism,insulin sensitivity, and bile acid and choline metabolism.As a result,gut microbiota have been emerging as a novel therapeutic target for NAFLD by manipulating it in various ways,including probiotics,prebiotics,synbiotics,antibiotics,fecal microbiota transplantation,and herbal components.In this review,we summarized the most recent advances in gut microbiota-mediated mechanisms,as well as gut microbiota-targeted therapies on NAFLD. | Ruiting Han Junli Ma Houkai Li | 2018 | Frontiers of Medicine2018,12,6: | 11 |
| 2 | Functional Metabolomics Reveals that Astragalus Polysaccharides Improve Lipids Metabolism through Microbial Metabolite 2-Hydroxybutyric Acid in Obese Mice显示文摘Polysaccharides are widely present in herbs with multiple activities,especially immunity regulation and metabolic benefits for metabolic disorders.However,the underlying mechanisms are not well under-stood.Functional metabolomics is increasingly used to investigate systemic effects on the host by iden-tifying metabolites with particular functions.This study explores the mechanisms underlying the metabolic benefits of Astragalus polysaccharides(APS)by adopting a functional metabolomics strategy.The effects of APS were determined in eight-week high-fat diet(HFD)-fed obese mice.Then,gas chromatography–time-of-flight mass spectrometry(GC–TOFMS)-based untargeted metabolomics was performed for an analysis of serum and liver tissues,and liquid chromatography–tandem mass spectrom-etry(LC–MS/MS)-based targeted metabolomics was performed.The potential functions of the metabo-lites were tested with in vitro and in vivo models of metabolic disorders.Our results first confirmed the metabolic benefits of APS in obese mice.Then,metabolomics analysis revealed that APS supplemen-tation reversed the HFD-induced metabolic changes,and identified 2-hydroxybutyric acid(2-HB)as a potential functional metabolite for APS activity that was significantly decreased by a HFD and reversed by APS.Further study indicated that 2-HB inhibited oleic acid(OA)-induced triglyceride(TG)accumula-tion.It was also found to stimulate the expression of proteins in lipid degradation in hepatocytes and TG lipolysis in 3T3-L1 cells.Moreover,it was found to reduce serum TG and regulate the proteins involved in lipid degradation in high-fat and high-sucrose(HFHS)-fed mice.In conclusion,our study demonstrates that the metabolic benefits of APS are at least partially due to 2-HB generation,which modulated lipid metabolism both in vitro and in vivo.Our results also highlight that functional metabolomics is practical for investigating the mechanism underlying the systemic benefits of plant polysaccharides. | Bingbing Li Ying Hong Yu Gu Shengjie Ye Kaili Hu Jian Yao Kan Ding Aihua Zhao Wei Jia Houkai Li | 2022 | Engineering2022,8,2: | 6 |
| 3 | China’s Urbanization Strategy and Policy During the 14th Five-Year Plan Period显示文摘During the 13th Five-Year Plan period,China’s urbanization construction has obtained remarkable achievements.Its urbanization continues to advance rapidly.The gap between different regions is constantly narrowed.City clusters have become the strategic core areas to promote urbanization and economic development.Currently,the key issue in China’s urbanization is not about the level or speed,but the quality.Tough challenges faced by China in its urbanization include:(1)granting permanent urban residency to people originally from rural areas(citizenization)is severely lagging-behind;(2)the key links in integrated urban–rural development system and mechanism are blocked and(3)the resources and environment costs in advancing urbanization are excessively high,etc.During the 14th Five-Year Plan period,China’s urbanization will continue to advance rapidly,but the speed will further slow down.By 2025,China’s urbanization rate is expected to reach about 65.5%,among which the eastern,central,western and northeastern regions will reach 73.0%,63.1%,61.2%and 66.7%,respectively.To this end,China’s urbanization should be people-centered and led by high-quality integrated urban and rural development;thus,the urbanization quality can be improved in an all-round way.It should be a path of highquality urbanization with Chinese characteristics featuring moderate and reasonable advancing speed,effectively improved quality in citizenization,balanced and orderly urbanization pattern,deeply integrated urban and rural development,and green,healthy and sustainable development.In terms of specific measures,the government should strengthen the leading,demonstrating and radiating role of central cities and promote the formation of four-tier central city system,namely,the global central cities,national central cities,regional central cities and local central cities;plan and build 34 high-end metropolitan areas at the national level,making them the core areas of advancing new-type urbanization in the new era;accelerate the process of establishing cities,encourage the transformation of large towns into cities,and strictly control the transformation of counties into urban districts;implement differentiated policies for expanding,stable and shrinking cities and towns,respectively;connect the channels between homesteads exiting the market and collective construction land for commercial use entering the market. | WEI Houkai LI Le NIAN Meng | 2021 | Chinese Journal of Urban and Environmental Studies2021,9,1: | 1 |
| 4 | Vancomycin pretreatment attenuates acetaminophen-induced liver injury through 2-hydroxybutyric acid显示文摘Liver injury caused by acetaminophen(AP)overdose is a leading public health problem.Although APinduced liver injury is well recognized as the formation of N-acetyl-p-benzoquinone(NAPQI),a toxic metabolite of AP,resulting in cell damage,emerging evidence indicates that AP-induced liver injury is also associated with gut microbiota.However,the gut microbiota-involved mechanism remains largely unknown.In our study,we found that vancomycin(Vac)pretreatment(100 mg/kg,twice a day for 4 days)attenuated AP-induced liver injury,altered the composition of gut microbiota,and changed serum metabolic profile.Moreover,we identified Vac pretreatment elevated cecum and serum 2-hydroxybutyric acid(2-HB),which ameliorated AP-induced cell damage and liver injury in mice by reducing AP bioavailability and elevating GSH levels.Our current results revealed the novel role of 2-HB in protecting AP-induced liver injury and add new evidence for gut microbiota in affecting AP toxicity. | Ningning Zheng Yu Gu Ying Hong Lili Sheng Linlin Chen Feng Zhang Jie Hou Weidong Zhang Zean Zhang Wei Jia Houkai Li | 2020 | Journal of Pharmaceutical Analysis2020,10,6: | 1 |

| 8 | China’s Strategies and Policies for Regional Development During the Period of the 14th Five-Year Plan显示文摘During the 13th Five-Year Plan period, China’s regional development strategies and policies have positively contributed to the economic transformation and upgrading in the eastern China, sound economic growth momentum in the central and western China, faster economic growth in old revolutionary areas, ethnic minority areas, border areas, and poor areas, and more coordinated development among regions. Despite its remarkable achievements, China’s coordinated regional development still faces problems such as unbalanced economic growth between the southern and northern China, great gaps in innovation capacity among regions, difficulties in ensuring equitable access to basic public services, and slow growth of Blue Economy. To pursue regional development in China, the most important task at present is to promote coordinated and high-quality regional development and create a new pattern that can help such development become better in terms of quality, efficiency, fairness, and sustainability. The shift from questing for coordinated regional development to high-quality coordinated regional development not only reflects the transformation of regional development concept but also adapts to the requirement for regional development transformation in today’s China. During the 14th Five-Year Plan period, to promote high-quality coordinated regional development, China should continue to make overall planning for the four major regions and coordinate the development of belts and zones with the four major regions as the basis and the key belts and zones as the framework. It should continue to deepen and improve the “4 + X” master strategy for regional development and facilitate the formation of a national system for regional development strategies. It should further improve the governance system for China’s national space, with an emphasis on creating the main framework for the development of national space that consists of three horizontal axes and three vertical axes,stretches over the eastern, central and western regions, and connects the northern region with the southern region, setting up a network of growth poles mainly supported by city clusters and metropolitan areas, and putting in place a negative list system for national space development.Besides, China should continue to accelerate the development of C-shaped open economic belts in border areas, further strengthen the opening up of inland, and spare no effort to foster the three major marine economic zones, thus working toward all-around opening up that coordinates land and marine development as well as the development of China’s coastal, border and inland areas.On that basis, China should also actively steer the northeastern region out of difficulty and help it develop vigorously, redouble efforts to cultivate advanced manufacturing centers in the central and western regions, formulate and implement support policies for relatively poor areas, and expedite the integration of modern infrastructure. | WEI Houkai NIAN Meng LI Le | 2020 | Chinese Journal of Urban and Environmental Studies2020,8,2: | 0 |

| 10 | Microbial-host-isozyme:a new territory for understanding personalized responses towards drug therapy显示文摘Inter-individual responses to medicinal drugs vary widely in clinic,which drives the urgent need of precision medicine for minimizing adverse reactions and maximalizing therapeutic effects among patients[1].In addition to genetic factors,increasing evidence suggests that gut microbiome plays a crucial role in affecting either pharmacodynamics or pharmacokinetics[2].In recent years,at least two ways have been demonstrated through which gut microbiome jointly affects drug metabolism or efficacy with host.One is the microbial metabolism on drugs.Zimmermann et al.[3]systematically studied the direct interaction between microorganisms and drugs.They found that 2/3 of the 271 selected orally administered drugs could be metabolized by at least one strain.They also verified that 30 enzymes encoded by microorganisms have the ability to transform 20 drugs into 59 candidate metabolites.The direct interaction between gut microbiota and drugs,including chemical modification(such as sulfasalazine[4]),inactivation(such as digoxin[5])or changes in toxicity(such as irinotecan[6]).Another way is the bioaccumulation of drugs in gut bacteria that alters drug availability and metabolism.Klünemann et al.[7]investigated the depletion of 15 drugs with different structures by 25 representative strains.This study reveals 70 interactions between bacteria and drugs,of which 29 have not been reported in previous studies,and more than half of the new interactions can be attributed to bioaccumulation. | LI Mingxiao LI Houkai | 2023 | Chinese Journal of Natural Medicines2023,21,8: | 0 |
| 11 | Cold exposure promotes coronavirus infection by altering the gut microbiota and lipid metabolism to reduce host immunity显示文摘Objective:Cold exposure has been suggested to be advantageous for the spread and infection of the coronavirus,and the gut microbiota influences the severity of the infection by modulating host inflammatory and immune responses.However,it remains unclear whether the promotion of viral infection through cold exposure is linked to the gut microbiota.Methods:In this study,we performed an unbiased analysis of gut microbiota,serum,and lung tissue metabolome changes in cold-exposed and virus-infected mice,alongside the assessment of immune-inflammatory indicators in serum and lung tissue.Results:The results revealed that both cold exposure and viral infection significantly decreased the percentage of peripheral blood lymphocytes(CD4^(+)T cells,CD8^(+)T cells,and B cell)and increased the expression of inflammatory factors(IL-6,IL-1β,TNF-α,and IFN-γ).Meanwhile,cold exposure disrupted the homeostasis of gut microbiota,elevating the abundance of pathogenic bacteria(Staphylococcus)and diminishing the abundance of beneficial bacteria(Alistipes).Notably,in virus-infected mice exposed to a cold environment,the reduction in the abundance of beneficial bacteria Alistipes was more pronounced than in cases of single virus infection and cold exposure.Analysis of altered serum and lung tissue metabolites highlighted glycerophospholipids,fatty acids,and eicosanoids as the most affected metabolites by cold exposure.These metabolites,closely associated with virus infection,exhibited a significant correlation with immune-inflammatory indicators.Conclusion:These findings establish a mechanistic connection between cold exposure and virus infection,suggesting that cold exposure-induced dysregulation of gut microbiota and lipid metabolism diminishes host immunity,promoting virus infection. | Gaosong Wu Yuhao Zhang Ningning Zheng Saisai Tian Jingyu Liao Wanqi Le Houkai Li Weidong Zhang | 2023 | Frigid Zone Medicine2023,3,4: | 0 |
