| 3 | Metabolic reprogramming and redox adaptation in sorafenib-resistant leukemia cells:detected by untargeted metabolomics and stable isotope tracing analysis显示文摘Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMS-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors,such as sorafenib,are limited due to the emergence of drug resistance.While investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be created.This study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 inhibitor.Methods:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations,namely the murine BaF3/ITD-R and the human MV4-11-R cell lines.We performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic resistance.Results:The resistant cells displayed fundamentally rewired metabolic profiles,characterized by a higher demand for glucose,accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress,accompanied by an enhanced glutathione synthesis.We demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant cells.Further experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative arm.In addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells,which was accompanied by decreased cell proliferation and an enhanced antioxidant response.Conclusions:Our data regarding comparative metabolomics characterized a distinct metabolic and redox adaptation that may contribute to sorafenib resistance in FLT3/ITD-mutated leukemia cells. | Xin You Weiye Jiang Wenhua Lu Hui Zhang Tiantian Yu Jingyu Tian Shijun Wen Guillermo Garcia-Manero Peng Huang Yumin Hu | 2019 | Cancer Communications2019,39,1: | 1 |

| 9 | ZIF-8 coated gold nanospheres: A multi-responsive drug delivery system promoting the killing effect of photothermal therapy against osteosarcoma cells显示文摘Photothermal therapy(PTT)has been widely used in the treatment of tumors,but its efficacy is greatly limited by the inability of precise drug delivery and the increase of heat shock proteins(HSPs)caused by high temperature.This article describes a therapeutic strategy to enhance PTT with starvation therapy in conjunction with ferroptosis mechanism.A nanoparticle platform ZIF-8@GA was constructed by wrapping together glucose oxidase(GOX)and gold nanospheres(AuNPs)loaded with dihydroartemisinin(DHA)with zeolitic imidazolate framework-8(ZIF-8).This platform can take advantage of the micro-environment of osteosarcoma(OS)cells,featuring low pH and high reactive oxygen species(ROS),for precision drug delivery.GOX can effectively catalyze glucose to produce gluconic acid and H_(2)O_(2),and DHA can also induce ROS production in OS cells.ROS produced by GOX and DHA can further generate lipid peroxidation(LPO)and lead to ferroptosis of OS cells.At the same time,ROS and LPO produced can inhibit the expression of HSPs,thereby increasing the therapeutic effect of PTT.In vitro experiments show that the nanoparticles are pH and ROS responsive.1μg/mL GOX combined with 0.2μg/mL DHA promotes ferroptosis of OS cells,and increases the killing effect of near-infrared(NIR)on OS cells.Further in vivo experiments showed that the nano drug-delivery platform combined with PTT can effectively inhibit the growth of OS cells.Meanwhile,this study provides a new idea for the treatment of OS with biomaterials combined with various treatment methods. | Miao Lei Weiye Cai Luetao Zou Bin Yu Lin Chen Yang Cao Shanlin Xiang Chao Song Jiandu Lei Wei Jiang Zhenming Hu | 2024 | Nano Research2024,17,3: | 0 |
| 10 | Field validation of UHPC layer in negative moment region of steel-concrete composite continuous girder bridge显示文摘Improving the cracking resistance of steel-normal concrete(NC)composite beams in the negative moment region is one of the main tasks in designing continuous composite beam(CCB)bridges due to the low tensile strength of the NC deck at pier supports.This study proposed an innovative structural configuration for the negative bending moment region in a steel-concrete CCB bridge with the aid of ultrahigh performance concrete(UHPC)layer.In order to investigate the feasibility and effectiveness of this new UHPC jointed structure in the negative bending moment region,field load testing was conducted on a newly built full-scale bridge.The newly designed structural configuration was described in detail regarding the structural characteristics(cracking resistance,economy,durability,and constructability).In the field investigation,strains on the surface of the concrete bridge deck,rebar,and steel beam in the negative bending moment region,as well as mid-span deflection,were measured under different load cases.Also,a finite element model for the four-span superstructure of the full-scale bridge was established and validated by the field test results.The simulated results in terms of strains and mid-span deflection showed moderate consistency with the test results.This field test and the finite element model results demonstrated that the new configuration with the UHPC layer provided an effective alternative for the negative bending moment region of the composite beam. | Minghong QIU Xudong SHAO Weiye HU Yanping ZHU Husam H.HUSSEIN Yaobei HE Qiongwei LIU | 2022 | Frontiers of Structural and Civil Engineering2022,16,6: | 0 |
