1The emergence of pandemic influenza viruses显示文摘Pandemic influenza has posed an increasing threat to public health worldwide in the last decade.In the 20th century,three human pandemic influenza outbreaks occurred in 1918,1957 and 1968,causing significant mortality.A number of hypotheses have been proposed for the emergence and development of pandemic viruses,including direct introduction into humans from an avian origin and reassortment between avian and previously circulating human viruses,either directly in humans or via an intermediate mammalian host.However,the evolutionary history of the pandemic viruses has been controversial,largely due to the lack of background genetic information and rigorous phylogenetic analyses.The pandemic that emerged in early April 2009 in North America provides a unique opportunity to investigate its emergence and development both in human and animal aspects.Recent genetic analyses of data accumulated through long-term influenza surveillance provided insights into the emergence of this novel pandemic virus.In this review,we summarise the recent literature that describes the evolutionary pathway of the pandemic viruses.We also discuss the implications of these findings on the early detection and control of future pandemics.Yi Guan Dhanasekaran Vijaykrishna Justin Bahl Huachen Zhu Jia Wang Gavin J.D.Smith 2010Protein & Cell2010,1,1:16
2Expression of e-cadherin and β-catenin in human esophageal squamous cell carcinoma: relationships with prognosis显示文摘AIM: To elucidate the expression of E-cadherin and β-catenincorrelating with its clinical outcome in patients withesophageal squamous cell carcinoma (ESCC), by analyzingtheir interrelationship with clinicopathological variables andtheir effects on progress and prognosis.METHODS: Expression of E-cadherin and β-catenin wasdetermined by SP immunohistochemical technique inpatients with ESCC consecutively, their correlation withclinical characteristics was evaluated and analyzed bymultivariate analysis.RESULTS: The rate of expression of E-cadherin decreasedto 66.03 % (70/106) in ESCC and the protein level wasnegative correlated with histologic grade, tumor size, clinicalstaging, lymph node metastasis and venous invasion.Whereas the expression rate of β-catenin was reduced to69.8 % (74/106) and the level of protein expressioncorrelated only with histologic grade. There obviously existedinverse correlation between level of E-cadherin protein andsurvival, especially in .stage I, IIa, IIb (P=0.0033), Patientswith low-expressing tumors for β-catenin and non-expressingtumors for E-cadherin/β-catenin had lower survival periodthan those with normal-expressing ones (P=0.0501 andP=0.0080, respectively). Patients with diminished expressionof E-cadherin as grade Ⅱ or Ⅲ had shorter survival periodthan those with normally expressing and grade Ⅰ, nosignificance existed between grade I and grade Ⅱ or Ⅲwith respect to different status of E-cadherin expression.Furthermore, Correlation analysis showed level of E-cadherincorrelated with that of β-catenin (P=0.005). 3Relationship between expression of CD105 and growth factors in malignant tumors of gastrointestinal tract and its significance显示文摘AIM: Angiogenesis is an important step in the growth of solid malignant tumors. A number of angiogenic factors have been found such as transforming growth factorβ1 (TGF-β1)and vascular endothelial growth factor (VEGF). However,the roles of TGFβ1 and VEGF in gastrointestinal carcinogenesis are still unclear. This study was to investigate the expressions of TGF-β1 and VEGF in gastrointestinal tract malignant tumors, as well as their association with microvessel density (MVD). At the same time, we also observed the localization of TGF-β1 and its receptor CD105 in gastric malignant tumors.METHODS: The expressions of TGF-β1 and CDL05 were detected in 55 fresh specimens of gastric carcinoma and VEGF and CD105 in 44 fresh specimens of colorectal carcinoma by immunohistochemical staining (S-ABC). TGF-β1 and CD105 in 55 gastric carcinoma tissues on the same slide were detected by using double-stain Tmmunohistochemistry (DS-ABC).RESULTS: Among the 55 cases of gastric carcinoma tissues,30 were positive for TGF-β1 (54.55 %). The MVD of TGF-β1 strong positive group (++~+++ 23.22±5.8) was significantly higher than that of weak positive group (+17.56±7.2) and negative group (- 17.46±3.9) (q=4.5, q=5.3207, respectively,P<0.01). In the areas of high expression of TGF-β1, MVD and the expression of CD105 were also high. Among the 44 cases of colonic carcinoma tissues, 26 were positive for VEGF (59.1%). The expressions of both VEGF and CD105 (MVD)were related with the depth of invasion (F=5.438, P<0.05;F=4.168, P=0.05), lymph node metastasis (F=10.311, P<0.01;F=20.282, P<0.01) and Dukes stage (F=6.196, P<0.01;F=10.274, P<0.01), but not with histological grade (F=0.487,P>0.05). There was a significant correlation between the expression of VEGF and CD105 (MVD) (r=0.720, P<0.01).CONCLUSION: Over-expression of TGF-β1 and VEGF acts as stimulating factors of angiogenesis in gastrointestinal tumors.CD105, as a receptor of TGF-β1, can regulate the biological effect of TGF-β1 in tumor angiogenesis. 8EFFECTS OF CALDESMON, CALPONIN, AND TROPOMYOSIN ON THE MG^(2+)-ATPase ACTIVITIES OF SMOOTH MUSCLE MYOSIN显示文摘Objective To test whether in the absence of actin, actin-binding proteins such as caldesmon, calponin, and tropomyosin interact with the myosin of unphosphorylation, Ca 2+ -dependent phosphorylation (CDP), and Ca 2+ -independent phosphorylati-on (CIP) and stimulate myosin Mg 2+ -ATPase activities. Methods Mg 2+ -ATPase activities were measured to evaluate the effects of caldesmon, calponin, and tropomyosin on the myosin in unphosphorylation, CDP by myosin light chain kinase (MLCK), and CIP by MLCK. Results (1) At different incubation-time, i.e., 5, 10, 20, 40, and 60 minutes, the highest Mg 2+ -ATPase activity was ob-served when myosin was in the state of CDP, the medium was CIP of myosin, and the lowest was the unphosphorylated myosin. (2) In the absence of caldesmon, calponin, and tropomyosin, the Mg 2+ -ATPase activities from high to low were in the following order: CDP, CIP, and unphosphorylated myosin. However, in the presence of caldesmon, calponin, and tropo-myosin, the order of relative value of Mg 2+ -ATPase activities from high to low was unphosphorylated, CIP, and CDP of myosin respectively compared to the corresponding controls. Conclusions The results propose that caldesmon, calponin, and tropomyosin are capable of stimulating Mg 2+ -ATPase activity of smooth muscle myosin in Ca 2+ -independent manner, since Ca 2+ is not obligating for the stimulating effects of the three proteins. 10Frequency-Domain Multi-Scale Early-Arrival Waveform Tomography with a Time-Domain Wavefield Modeling Engine显示文摘Early-arrival waveform tomography(EWT) is one of the most promising techniques for building near-surface velocity model. Based on finite-frequency wave equation, EWT estimates velocities by matching calculated early-arrival waveforms with the observed ones. However, the objective function of EWT can easily converge to local minimum because of the cycle-skipping phenomenon. In order to reduce the cycle-skipping problem, a hybrid-domain early-arrival waveform tomography(HEWT) is proposed in this paper. The forward modeling of HEWT is realized in the time domain where early-arrival waveforms are easier to be selected from seismic data and less memory is needed than they are in the frequency domain. The inversion is implemented in the frequency domain where multi-scale strategy is more convenient to be realized than that in the time domain. Discrete Fourier transformation(DFT) is used to transform the time-domain wavefield to the frequency-domain wavefield. 11Identification of key taste components in Baccaurea ramiflora Lour.fruit using non-targeted metabolomics显示文摘Among the numerous fruit species of Baccaurea ramiflora Lour.,‘LR’(white flesh)and‘BR’(pink flesh)are two kinds of local strains with high edibility.In order to study the metabolic causes of taste differences,we performed the non-targeted metabonomics analysis of‘LR’and‘BR’using LC-MS/MS.541 metabolites were totally identified,and 45 kinds of metabolites(carbohydrates,fatty acids,flavonoids and terpenoids,etc.)were different between the two strains.The results indicate L-sorbose,D-(+)-glucose,citric acid,L-phenylalanine and oleamide,α-eleostearic acid were the main primary metabolites.The significant difference existed in pathways of unsaturated fatty acids between the studied two strains by pathway enrichment analysis.The results demonstrate that the different in composition,as well as the abundance of primary and secondary metabolites may be the potential causes of taste differences,which provides a new insight into the possible metabolic factors setting off the changing taste of B.ramiflora.Jie Chen Fengnian Wu Huachen Wang Chunce Guo Wengen Zhang Peisi Luo Jing Zhou Wenwen Hao Guangyao Yang Jianjian Huang 2023Food Science and Human Wellness2023,12,1:1

13Oncolytic Activity of Wild-type Newcastle Disease Virus HK84 Against Hepatocellular Carcinoma Associated with Activation of Type I Interferon Signaling显示文摘Background and Aims:Hepatocellular carcinoma(HCC)is listed as one of the most common causes of cancer-related death.Oncolytic therapy has become a promising treatment because of novel immunotherapies and gene editing technology,but biosafety concerns remain the biggest limitation for clinical application.We studied the the antitumor activity and biosafety of the wild-type Newcastle disease virus HK84 strain(NDV/HK84)and 10 other NDV strains.Meth-ods:Cell proliferation and apoptosis were determined by cell counting Kit-8 and fluorescein isothiocyanate Annexin V apoptosis assays.Colony formation,wound healing,and a xenograft mouse model were used to evaluate in vivo and in vitro oncolytic effectiveness.The safety of NDV/HK84 was tested in nude mice by an in vivo luciferase imaging system.The replication kinetics of NDV/HK84 in normal tis-sues and tumors were evaluated by infectious-dose assays in eggs.RNA sequencing analysis was performed to explore NDV/HK84 activity and was validated by quantitative real-time PCR.Results:The cell counting Kit-8 assays of vi-ability found that the oncolytic activity of the NDV strains differed with the multiplicity of infection(MOI).At an MOI of 20,the oncolytic activity of all NDV strains except the DK/JX/21358/08 strain was>80%.The oncolytic activities of the NDV/HK84 and DK/JX/8224/04 strains were>80%at both MOI=20 and MOI=2.Only NDV/HK84 had>80%oncolytic activities at both MOI=20 and MOI=2.We chose NDV/HK84 as the candidate virus to test the oncolytic effect of NDV in HCC in the in vitro and in vivo experiments.NDV/HK84 killed human SK-HEP-1 HCC cells without affecting healthy cells.Conclusions:Intratumor infection with NDV/HK84 strains compared with vehicle controls or positive controls indicated that NDV/HK84 strain specifically inhib-ited HCC without affecting healthy mice.High-throughput RNA sequencing showed that the oncolytic activity of NDV/HK84 was dependent on the activation of type I interferon signaling.Liming Chen Yongdong Niu Jiating Sun Hong Lin Guoxi Liang Min Xiao Dongmei Shi Jia Wang Huachen Zhu Yi Guan 2022Journal of Clinical and Translational Hepatology2022,10,2:1

It is difficult to control the GPCM system because the system is discrete. With consideration of the stability and speediness of the GPCM position servo control system, a control strategy is developed through the theoretical and ex-perimental analyses. The control strategy integrates the merits of Bang-Bang control, PID control and fuzzy control.
