| 3 | Peroxisome proliferator-activated receptor-γ is essential in the pathogenesis of gastric carcinoma显示文摘AIM:To investigate whether peroxisome proliferator-activated receptor γ (PPAR-γ) is expressed in human gastric carcinoma and whether PPAR-γ is a potential target for gastric carcinoma therapy.METHODS: PPAR-γ protein in gastric carcinoma was examined by immunohistochemistry. In the gastric carcinoma cell line MGC803, PPAR-γ, survivin, Skp2 and p27 protein and mRNA were examined by Western blotting and real-time reverse transcription-polymerase chain reaction, respectively; proliferation was examined by MTT; apoptosis was examined by chromatin staining with Hoechst 33342 and fluorescence activated cell sorting (FACS). and cell cycle was examined by FACS; the knockdown of PPAR-γ was done by RNA interference.RESULTS: A high level of expression of PPAR-γ was observed in human gastric carcinoma and in a human gastric carcinoma cell line MGC803. The PPAR-γ agonist 15-deoxy-12,14-prostaglandin J2 (15d-PGJ2) inhibited growth, and induced apoptosis and G1/G0 cell cycle arrest in MGC803 cells in a concentration-dependent and time-dependent manner. The effect of 15d-PGJ2 on MGC803 cells was not reversed by the selective and irreversible antagonist GW9662 for PPAR-γ. Furthermore, survivin and Skp2 expression were decreased, whereasp27 expression was enhanced following 15d-PGJ2 treatment in a dose-dependent manner in MGC803 cells. Interestingly, we also found that small interfering RNA for PPAR-γ inhibited growth and induced apoptosis in MGC803 cells. | 5 | Real-time Fluorescence PCR Method for Detection of Burkholderia glumae from Rice显示文摘Burkholderia glumae causing seedling rot and grain rot of rice was listed as a plant quarantine disease of China in 2007. | 7 | Systems pharmacology dissection of action mechanisms for herbs in osteoporosis treatment显示文摘Objective:Osteoporosis has become the biggest cause of non-fatal health issue.Currently,the limitations of traditional anti-osteoporosis drugs such as long-term ill-effects and drug resistance,have raised concerns toward complementary and alternative therapies,particularly herbal medicines and their natural active compounds.Thus,this study aimed to provide an integrative analysis of active chemicals,drug targets and interacting pathways of the herbs for osteoporosis treatment.Methods:Here,we introduced a systematic pharmacology model,combining the absorption,distribution,metabolism,and excretion(ADME)screening model,drug targeting and network pharmacology,to probe into the therapeutic mechanisms of herbs in osteoporosis.Results:We obtained 86 natural compounds with favorable pharmacokinetic profiles and their 58 targets from seven osteoporosis-related herbs.Network analysis revealed that they probably synergistically work through multiple mechanisms,such as suppressing inflammatory response,maintaining bone metabolism or improving organism immunity,to benefit patients with osteoporosis.Furthermore,experimental results showed that all the five compounds(calycosin,asperosaponin VI,hederagenin,betulinic acid and luteolin)enhanced osteoblast proliferation and differentiation in vitro,which corroborated the validity of this system pharmacology approach.Notably,gentisin and aureusidin among the identified compounds were first predicted to be associated with osteoporosis.Conclusion:Herbs and their natural compounds,being characterized as the classical combination therapies,might be engaged in multiple mechanisms to coordinately improve the osteoporosis symptoms.This work may contribute to offer novel strategies and clues for the therapy and drug discovery of osteoporosis and other complex diseases. | Ying Huai Wen-juan Zhang Wei Wang Kai Dang Shan-feng Jiang Dan-ming Li Meng Li Qiang Hao Zhi-ping Miao Yu Li Ai-rong Qian | 2021 | Chinese Herbal Medicines2021,13,3: | 3 |

| 9 | 3D MHD simulation of the double-gradient instability of the magnetotail current sheet显示文摘Flapping motion of the current sheet(CS) is an important physical process in the Earth's magnetotail. The magnetic doublegradient model, which includes both the instability and wave modes, offers a reasonable explanation for the exciting and propagation of the flapping wave. In this paper, we apply an advanced numerical magnetohydrodynamic(MHD) scheme(conservation element and solution element(CESE)-MHD) to simulate the magnetic double-gradient instability in an idealized current sheet that mimics the magnetotail configuration. We initialize the simulations with a numerically relaxed magnetotail equilibrium, in which the normal component of the magnetic field has a tailward gradient. It is confirmed in our simulation that the instability develops in the current layer. The growth rate of the instability yielded from the simulation is very close to the prediction of theory, with a relative deviation of only ten percent. Semiconducting ferroelectrics that are stable at room temperature afford many possibilities for the assembly of various high-performance heterostructures and fabricating multifuntional devices.Herein, we report the synthesis of a stable van der Waals(vd W) single-crystal semiconductor α-In2 Se3.Piezoresponse force microscopy(PFM) measurements demonstrated the out-of-plane ferroelectricity in ~15 layers α-In2 Se3 at room temperature. Both ferroelectric domains with opposite polarization and the tested amplitude and phase curve proved that this semiconductor exhibits hysteresis behavior during polarization.In the α-In2Se3/WSe2 vertical heterostructure device, the switchable diode effect and nonvolatile memory phenomenon showed a high on/off ratio and a small switching voltage. The distinct resistance switches were further analyzed by band alignment of the heterostructure under different polarizations by first principle calculations. Each cDNA fragment has been amplified by RT-PCR from the anticoagulant blood of strain Shimen infected pig. The PCR products have been cloned respectively and sequenced. Results show that the cDNA library of strain Shimen and its nucleotide sequence have been obtained. The genomic RNA of strain Shimen is 12 298 nucleotides in length, containing a 5’ and a 3’ noncoding region 373 and 231 nt long respectively. The center of genome is a single large open reading frame of 11 697 nt which encodes a polyprotein of 3 898 amino acids. | 19 | Molecular genetic analysis of phosphomannomutase genes in Triticum monococcum显示文摘In higher plants, phosphomannomutase(PMM) is essential for synthesizing the antioxidant ascorbic acid through the Smirnoff–Wheeler pathway. Previously, we characterized six PMM genes(Ta PMM-A1, A2, B1, B2, D1 and D2) in common wheat(Triticum aestivum, AABBDD).Here, we report a molecular genetic analysis of PMM genes in Triticum monococcum(AmAm), a diploid wheat species whose Amgenome is closely related to the A genome of common wheat. Two distinct PMM genes, Tm PMM-1 and Tm PMM-2, were found in T. monococcum. The coding region of Tm PMM-1 was intact and highly conserved. In contrast, two main Tm PMM-2 alleles were identified, with Tm PMM-2a possessing an intact coding sequence and Tm PMM-2b being a pseudogene. The transcript level of Tm PMM-2a was much higher than that of Tm PMM-2b, and a bacterially expressed Tm PMM-2a recombinant protein displayed relatively high PMM activity. In general, the total transcript level of PMM was substantially higher in accessions carrying Tm PMM-1 and Tm PMM-2a than those harboring Tm PMM-1 and Tm PMM-2b. However, total PMM protein and activity levels did not differ drastically between the two genotypes.
