| 1 | Euphorbia factor L2 induces apoptosis in A549 cells through the mitochondrial pathway显示文摘Euphorbia factor L2, a lathyrane diterpenoid isolated from caper euphorbia seed(the seeds of Euphorbia lathyris L.), has been traditionally applied to treat cancer. This article focuses on the cytotoxic activity of Euphorbia factor L2 against lung carcinoma A549 cells and the mechanism by which apoptosis is induced. We analyzed the cytotoxicity and related mechanism of Euphorbia factor L2 with an MTT assay, an annexin V-FITC/PI test, a colorimetric assay, and immunoblotting. Euphorbia factor L2 showed potent cytotoxicity to A549 cells. Euphorbia factor L2 led to an increase in reactive oxygen species(ROS) generation,a loss of mitochondrial electrochemical potential, release of cytochrome c, activation of caspase-9 and caspase-3, and cleavage of poly(ADP-ribose) polymerase, suggesting that Euphorbia factor L2 induced apoptosis through a mitochondrial pathway. | 2 | Simulation research on a minimum root-mean-square error rotation-fitting algorithm for gravity matching navigation显示文摘Gravity/inertial combination navigation is a leading issue in realizing passive navigation onboard a submarine. A new rotation-fitting gravity matching algorithm, based on the Terrain Contour Matching (TERCOM) algorithm, is proposed in this paper. The algorithm is based on the principle of least mean-square-error criterion, and searches for a certain matched trajectory that runs parallel to a trace indicated by an inertial navigation system on a gravity base map. A rotation is then made clockwise or counterclockwise through a certain angle around the matched trajectory to look for an optimal matched trajectory within a certain angle span range, and through weighted fitting with another eight suboptimal matched trajectories, the endpoint of the fitted trajectory is considered the optimal matched position. In analysis of the algorithm reliability and matching error, the results from simulation indicate that the optimal position can be obtained effectively in real time, and the positioning accuracy improves by 35% and up to 1.05 nautical miles using the proposed algorithm compared with using the widely employed TERCOM and SITAN methods. | 4 | Exosomes with low miR-34c-3p expression promote invasion and migration of non-small cell lung cancer by upregulating integrinα2β1显示文摘Exosomes play critical roles in regulating various physiological and pathological processes,including immune stimulation,immune suppression,cardiovascular diseases,and cancers.Recent studies show that exosomes that transport specific microRNAs(miRNAs)are involved in tumor development.However,the molecular mechanism by which tumor invasion and migration are regulated by exosomes from non-small cell lung cancer(NSCLC)is not well understood.Here,we show that exosomes shuttling low levels of miR-34c-3p are involved in NSCLC progression.Our results showed that exosomes derived from NSCLC cells carrying low levels of miR-34c-3p could be transported into the cytoplasm of NSCLC cells and accelerate NSCLC invasion and migration by upregulating integrinα2β1.A luciferase assay revealed that integrinα2β1 was the direct target of miR-34c-3p,and overexpression of integrinα2β1 could promote the invasion and migration of NSCLC cells.The analysis of exosomes derived from clinical serum samples indicated that the expression of miR-34c-3p was significantly downregulated in exosomes from NSCLC patients compared with that of normal controls.A549-derived exosomes promoted NSCLC cells lung metastases in vivo.Exosomes shuttling low levels of miR-34c-3p were associated with the progression of NSCLC in vitro and in vivo.Our data demonstrate that exosomes shuttling low levels of miR-34c-3p can accelerate the invasion and migration of NSCLC by upregulating integrinα2β1.MiR-34c-3p can be a diagnostic and prognostic marker for NSCLC.High expression of integrinα2β1 is positively related to the migration and metastasis of NSCLC cells. | Wenjing Huang Yanyan Yan Yun Liu Minting Lin Jinxiang Ma Wei Zhang Jianwei Dai Jiajun Li Qiaoru Guo Hubiao Chen Bolat Makabel Hong Liu Chaoyue Su Hong Bi Jianye Zhang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 5 |
| 5 | Tissue-based metabolite profiling and qualitative comparison of two species of Achyranthes roots by use of UHPLC-QTOF MS and laser micro-dissection显示文摘Achyranthes bidentata and Achyranthes aspera are saponin and steroid rich medicinal plants, used extensively for therapeutic treatments in Traditional Chinese Medicine(TCM) and Ayurveda. A. bidentata is reported to be one of the rare and extensively exploited medicinal plant species that face the issue of being endangered.Finding qualitative substitute with identical phyto-constituents contributing to similar composition and pharmacological benefits will help in reducing the burden of exploitation of the natural habitats of such plants.In the present study, a comparative metabolite analysis of the whole drug and specific tissues isolated by laser micro-dissection(LMD) was carried out for both the selected species, by use of ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UHPLC-QTOF MS). The results of the study indicate that the cortex and the medullary ray tissues are rich in their content of steroidal and saponin constituents such as(25 S)-inokosterone-20,22-acetonide, ginsenoside Ro, bidentatoside II and achyranthoside B.Metabolite profiling of the whole tissues of both the species indicates presence of identical constituents. | 19 | Study on Saccharification of Saccharum spontaneum L. by Sulphuric Acid Hydrolysis显示文摘In order to improve the saccharification rate by acid hydrolysis in the technique for production of ethanol from biomass,in this study,Saccharum spontaneum L. was used as the experimental material to investigate the rules and technological conditions for two-step acid-hydrolysis saccharification by single factor experiment and orthogonal experiment. According to the results,concentrated sulfuric acid hydrolysis was the first step,with the liquid-solid ratio of 42∶ 1,sulfuric acid concentration of 70%,hydrolysis time of 20 min,and hydrolysis temperature of 55 ℃; dilute sulphuric acid hydrolysis was the second step,with the liquidsolid ratio of 115∶ 1,sulfuric acid concentration of 5. 5%,hydrolysis time of 155 min,and hydrolysis temperature of 100 ℃. In two-step acid hydrolysis process,the yield of reducing sugar was 48. 78%. | 20 | Nutritional Value Evaluation of Saccharum spontaneum L. Germplasm Resources显示文摘The contents of dry matter, crude fat, crude protein, neutral detergent fiber, acid detergent fiber, crude ash, Ca and P in 43 Saccharum spontaneum L.germplasms during the vegetative period were determined. Among them, the crude protein, crude fat, neutral detergent fiber, acid detergent fiber, P and Ca contents were treated as the judging indicators. The nutritional value evaluation was carried out with the analytic hierarchy process(AHP). The results showed A49(Guangxi)had the highest nutritional value. Its dry matter, crude protein, crude fat, crude ash,neutral detergent fiber, acid detergent fiber, Ca and P contents were 32.35%,7.20%, 4.06%, 8.07%, 75.81%, 50.72%, 0.23% and 0.16% respectively. While A3(Guangdong) had the lowest nutritional value. Its dry matter, crude protein, crude fat, crude ash, neutral detergent fiber, acid detergent fiber, Ca and P contents were32.24%, 4.62%, 0.51%, 6.38%, 46.40%, 40.02%, 0.15% and 0.09% respectively.The crude protein content differed significantly among different germplasms. A48 had the highest crude protein content(9.11%), and A14 had the lowest crude protein content(3.72%). Based on the evaluation results, the 43 Saccharum spontaneum L.germplasms were divided into 3 groups: high-nutritional value type, moderate-nutritional value type and low-nutritional value type.
