1Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. 3Warburg Effects in Cancer and Normal Proliferating Cells:Two Tales of the Same Name显示文摘It has been observed that both cancer tissue cells and normal proliferating cells(NPCs)have the Warburg effect.Our goal here is to demonstrate that they do this for different reasons.To accomplish this,we have analyzed the transcriptomic data of over 7000 cancer and control tissues of 14 cancer types in TCGA and data of five NPC types in GEO.Our analyses reveal that NPCs accumulate large quantities of ATPs produced by the respiration process before starting the Warburg effect,to raise the intracellular pH from 6.8 to 7.2 and to prepare for cell division energetically.Once cell cycle starts,the cells start to rely on glycolysis for ATP generation followed by ATP hydrolysis and lactic acid release,to maintain the elevated intracellular pH as needed by cell division since together the three processes are pH neutral.The cells go back to the normal respirationbased ATP production once the cell division phase ends.In comparison,cancer cells have reached their intracellular pH at 7.4 from top down as multiple acid-loading transporters are up-regulated and most acid-extruding ones except for lactic acid exporters are repressed.Cancer cells use continuous glycolysis for ATP production as way to acidify the intracellular space since the lactic acid secretion is decoupled from glycolysis-based ATP generation and is pH balanced by increased expressions of acid-loading transporters.Co-expression analyses suggest that lactic acid secretion is regulated by external,non-pH related signals.Overall,our data strongly suggest that the two cell types have the Warburg effect for very different reasons.Huiyan Sun Liang Chen Sha Cao Yanchun Liang Ying Xu 2019Genomics, Proteomics & Bioinformatics2019,17,3:4

9Effect of autologous bone marrow stem cells-scaffold transplantation on the ongoing pregnancy rate in intrauterine adhesion women:a randomized,controlled trial显示文摘Intrauterine adhesion is a major cause of female reproductive disorders.Although we and others uncontrolled pilot studies showed that treatment with autologous bone marrow stem cells made a few patients with severe intrauterine adhesion obtain live birth,no large sample randomized controlled studies on this therapeutic strategy in such patients have been reported so far.To verify if the therapy of autologous bone marrow stem cells-scaffold is superior to traditional treatment in moderate to severe intrauterine adhesion patients in increasing their ongoing pregnancy rate,we conducted this randomized controlled clinical trial.Totally 195 participants with moderate to severe intrauterine adhesion were screened and 152 of them were randomly assigned in a 1:1 ratio to either group with autologous bone marrow stem cells-scaffold plus Foley balloon catheter or group with only Foley balloon catheter(control group)from February 2016 to January 2020.The per-protocol analysis included 140 participants:72 in bone marrow stem cells-scaffold group and 68 in control group.The ongoing pregnancy occurred in 45/72(62.5%)participants in the bone marrow stem cells-scaffold group which was significantly higher than that in the control group(28/68,41.2%)(RR=1.52,95%CI 1.08–2.12,P=0.012).The situation was similar in live birth rate(bone marrow stem cells-scaffold group 56.9%(41/72)vs.control group 38.2%(26/68),RR=1.49,95%CI 1.04–2.14,P=0.027).Compared with control group,participants in bone marrow stem cells-scaffold group showed more menstrual blood volume in the 3rd and 6th cycles and maximal endometrial thickness in the 6th cycle after hysteroscopic adhesiolysis.The incidence of mild placenta accrete was increased in bone marrow stem cells-scaffold group and no severe adverse effects were observed.In conclusion,transplantation of bone marrow stem cells-scaffold into uterine cavities of the participants with moderate to severe intrauterine adhesion increased their ongoing pregnancy and live birth rates,and this therapy was relatively safe.Hui Zhu Taishun Li Peizhen Xu Lijun Ding Xianghong Zhu Bin Wang Xiaoqiu Tang Juan Li Pengfeng Zhu Huiyan Wang Chenyan Dai Haixiang Sun Jianwu Dai Yali Hu 2024Science China(Life Sciences)2024,67,1:0
10Gene regulatory network inference based on causal discovery integrating with graph neural network显示文摘Gene regulatory network (GRN) inference from gene expression data is asignificant approach to understanding aspects of the biological system.Compared with generalized correlation-based methods, causality-inspiredones seem more rational to infer regulatory relationships. We proposeGRINCD, a novel GRN inference framework empowered by graph representationlearning and causal asymmetric learning, considering both linearand non-linear regulatory relationships. First, high-quality representation ofeach gene is generated using graph neural network. Then, we apply theadditive noise model to predict the causal regulation of each regulator-targetpair. Additionally, we design two channels and finally assemble them forrobust prediction. Through comprehensive comparisons of our frameworkwith state-of-the-art methods based on different principles on numerousdatasets of diverse types and scales, the experimental results show that ourframework achieves superior or comparable performance under variousevaluation metrics. 13Energy-Efficient Deployment of Water Quality Sensor Networks显示文摘Water quality sensor networks are promising tools for the exploration of oceans.Some key areas need to be monitored effectively.Water quality sensors are deployed randomly or uniformly,however,and understanding how to deploy sensor nodes reasonably and realize effective monitoring of key areas on the basis of monitoring the whole area is an urgent problem to be solved.Additionally,energy is limited in water quality sensor networks.When moving sensor nodes,we should extend the life cycle of the sensor networks as much as possible.In this study,sensor nodes in non-key monitored areas are moved to key areas.First,we used the concentric circle method to determine the mobile sensor nodes and the target locations.Then,we determined the relationship between the mobile sensor nodes and the target locations according to the energy matrix.Finally,we calculated the shortest moving path according to the Floyd algorithm,which realizes the redeployment of the key monitored area.The simulation results showed that,compared with the method of direct movement,the proposed method can effectively reduce the energy consumption and save the network adjustment time based on the effective coverage of key areas.Qian Sun Zhiping Shen Jinglin Liang Xiaoyi Wang Jiping Xu Li Wang Huiyan Zhang Jiabin Yu Ning Cao Ruichao Wang 2021Computers, Materials & Continua2021,,9:0

15Optimal Deployment of Heterogeneous Nodes to Enhance Network Invulnerability显示文摘Wireless sensor networks(WSN)can be used in many fields.In wireless sensor networks,sensor nodes transmit data in multi hop mode.The large number of hops required by data transmission will lead to unbalanced energy consumption and large data transmission delay of the whole network,which greatly affects the invulnerability of the network.Therefore,an optimal deployment of heterogeneous nodes(ODHN)algorithm is proposed to enhance the invulnerability of the wireless sensor networks.The algorithm combines the advantages of DEEC(design of distributed energy efficient clustering)clustering algorithm and BAS(beetle antenna search)optimization algorithm to find the globally optimal deployment locations of heterogeneous nodes.Then,establish a shortcut to communicate with sink nodes through heterogeneous nodes.Besides,considering the practical deployment operation,we set the threshold of the mobile location of heterogeneous nodes,which greatly simplifies the deployment difficulty.Simulation results show that compared with traditional routing protocols,the proposed algorithm can make the network load more evenly,and effectively improve energy-utilization and the fault tolerance of the whole network,which can greatly improve the invulnerability of the wireless sensor networks.Qian Sun Fengbo Yang Xiaoyi Wang Jiping Xu Huiyan Zhang Li Wang Jiabin Yu Xiao Peng Ruichao Wang 2022Computers, Materials & Continua2022,,11:0
16Suppression of cell pyroptosis by omeprazole through PDE4-mediated autophagy in gastric epithelial cells显示文摘Helicobacter pylori is a risk factor for the development of peptic ulcers with autophagy dysfunction.Omeprazole was widely known as the first-line regimen for H.pylori-associated gastritis.Objectives:The objective of this work was to assess the role of omeprazole on cell pyroptosis and autophagy.Methods:The clinical samples were collected.Quantitative polymerase chain reaction,western blotting,enzyme linked immunosorbent assay,and immunofluorescence(IF)analysis were conducted to reveal the mechanism of omeprazole on cell pyroptosis and autophagy.Results:The results revealed that omeprazole could decrease cell pyroptosis,which was attributed to the downregulation of cleaved caspase-1 expression,resulting in the inhibition of gasdermin E and interleukin-18/1βmaturation and secretion as well as the resolution of inflammation.Mechanistically,omeprazole treatment led to drastic downregulation of mammalian target of rapamycin(mTOR)activity was observed in BGC823 cells,leading to enhanced autophagy characterized by increased LC3II expression,which further reduced cell pyroptosis.This omeprazole-mediated phenomenon was enhanced after phosphodiesterase-4(PDE4)inhibitor dipyridamole(DIP)treatment.In addition,activation of mTOR by MHY1485 could rescue the suppression of cell pyroptosis induced by omeprazole.Most importantly,IF analysis suggested that phosphorylation of mTOR and PDE4 activity and caspase-1 were enhanced in H.pylori-infected gastric mucosa.Conclusion:These findings indicate that omeprazole suppresses cell pyroptosis through PDE4-mediated autophagy in gastric epithelial cells,and DIP enhanced the omeprazole-mediated inhibition of cell pyroptosis,implying that DIP is an alternative combined therapy strategy in improving the treatment of patients with H.pylori infection.LIPING YE HUIYAN SUN XINHUA LIANG WENXU PAN LI XIANG WENJUN DU LANLAN GENG WANFU XU SITANG GONG 2023BIOCELL2023,47,12:0

18Molecular movements of trehalose inside a single network enabling a rapidly-recoverable tough hydrogel显示文摘It remains a challenge to achieve rapidly recoverable hydrogels by molecular hydrogen-bonding interaction because of its slow inter-action kinetics.This work for the first time reports a trehalose(Tre)-based molecular movement mechanism inside a single network of polyacrylamide(PAM)that accelerates the kinetics of hydrogenbonding interaction,and thereby endows the hydrogel with high toughness and rapid shape and mechanical recoverability.The resultant PAM@Tre hydrogel is capable of full shape recovery after 10,000 loading/unloading cycles at a strain of 500%.Even after being stretched at a strain of 2500%,it can recover to its original shape within 10 seconds.Moreover,the molecular movement of trehalose also endows the PAM@Tre hydrogel with fracture energy and toughness as high as~9000 J m^(-2) and~1600 kJ m^(-3),respec-tively,leading to strong resistance to both static and dynamic piercing.The PAM@Tre hydrogel is thus believed to have enormous potentials in protection devices,bionic skin,soft actuator,and stretchable electronics.Xiaowen Huang Jimin Fu Huiyan Tan Yan Miu Mengda Xu Qiuhua Zhao Yujie Xie Shengtong Sun Haimin Yao Lidong Zhang 2022International Journal of Smart and Nano Materials2022,13,4:0
