Early-onset colorectal cancer:A review of current knowledge显示文摘Colorectal cancer(CRC)is one of the most prevalent malignancies worldwide.Although most prevalent among older people, its incidence above 50 years old has been decreasing globally in the last decades, probably as a result of better screening.Paradoxically, its incidence in patients below 50 years old[early-onset CRC(EO-CRC)]has been increasing, for reasons not yet fully understood.EOCRC’s increasing incidence is genre independent but shows racial disparities and has been described to occur worldwide. It follows a birth-cohort effect which probably reflects a change in exposure to CRC risk factors. Its incidence is predicted to double until 2030,which makes EO-CRC a serious public health issue. Both modifiable and non-modifiable risk factors have been identified-some are potential targets for preventive measures.EO-CRC is often diagnosed at advanced stages and histological features associated with poor prognosis have been described.EO-CRC presents some distinctive features:Microsatellite instability is common, but another subtype of tumours, both microsatellite and chromosome stable also seems relevant. There are no age-specific treatment protocols and studies on EO-CRC survival rates have shown conflicting data.Due to the higher germline pathological mutations found in EO-CRC patients,an accurate genetic risk evaluation should be performed. In this review,we summarize the current evidence on epidemiological, clinical, histopathological and molecular features of EO-CRC and discuss the contribution of genetics and lifestyle risk factors. We further comment on screening strategies and specific dimensions to consider when dealing with a younger cancer patient.Margarida R Saraiva Isadora Rosa Isabel Claro 2023World Journal of Gastroenterology2023,29,8:3
Therapeutic effectiveness of a single exercise session combined with WalkAide functional electrical stimulation in post-stroke patients:a crossover design study显示文摘A growing body of evidence has suggested that the imbalance of epigenetic markers and oxidative stress appears to be involved in the pathophysiology and progression of stroke. Thus, strategies that modulate these biomarkers might be considered targets for neuroprotection and novel therapeutic opportunities for these patients.Physical exercise has been reported to induce changes in these epigenetic markers and improve clinical outcomes in different populations. However, little is reported on this in post-stroke patients. The purpose of this study was to investigate the effect of a single exercise session with WalkAide functional electrical stimulation(FES)on cognitive performance, clinical functional parameters, oxidative stress and epigenetic modulation in post-stroke individuals. In this crossover design study,12 post-stroke individuals aged 54–72 years of either sexes were included and subjected to a single session of exercise(45 minutes)without WalkAide functional electrical stimulation(EXE alone group),followed by another single session of exercise(45 minutes)with WalkAide functional electrical stimulation(EXE+FES group). The clinical functional outcome measures, cognitive performance and blood collections for biomarker measurements were assessed pre-and post-intervention.After intervention, higher Berg Balance Scale scores were obtained in the EXE+FES group than in the EXE alone group. There was no significant difference in the Timed Up and Go test results post-intervention between EXE alone and EXE+FES groups. After intervention,a better cognitive performance was found in both groups compared with before the intervention. After intervention, the Timed Up and Go test scores were higher in the EXE+FES group than in the EXE alone group. In addition, the intervention induced lower levels of lipid peroxidation. After intervention, carbonyl level was lower, superoxide dismutase activity and superoxide dismutase/catalase activity ratio were higher in the EXE+FES group, compared with the EXE group alone. In each group, both histone deacetylase(HDAC2)and histone acetyltransferase activities were increased after intervention compared with before the intervention. These findings suggest that a single exercise session with WalkAide FES is more effective on balance ability and cognitive performance compared with conventional exercise alone in post-stroke patients. This is likely to be related to the regulation of oxidative stress markers. The present study was approved by the Research Ethics Committee of the Methodist University Center-IPA(approval No.2.423.376)on December 7,2017 and registered in the Brazilian Registry of Clinical Trials—ReBEC(RBR-9phj2q)on February 11,2019.Viviane Rostirola Elsner Lucieli Trevizol Isadora de Leon Marcos da Silva ThaynáWeiss Milena Braga Daniela Pochmann Amanda Stolzenberg Blembeel Caroline Dani Elenice Boggio 2021Neural Regeneration Research2021,16,5:3
Height-to-diameter ratios with temporal and dendro/morphometric variables for Brazilian pine in south Brazil显示文摘Height-to-diameter ratios(HD)are an important measure of the stability, density and competition of forest stands. It refl ects the vertical growth of the trees, the vulnerability of the forest canopy structure and infl uences volumetric production.HD ratios vary according to tree size, availability of resources for growth, stand density and species composition.Data were taken from 210 trees and a regression technique of generalized linear models for the HD ratio applicable for forest structure conservation was developed. The objective of this study was to model the HD ratios of dominant and co-dominant trees of Araucaria angustifolia according to morphometric, dendrometric, annual diameter increment, stand density, and age variables in three sites in southern Brazil. The results show that the HD ratio decreases with increasing age, crown area and basal area, and increases with stand density and annual diameter increment.Accuracy of the developed equations was demonstrated by the values of deviation,Bayesian and Akaike criteria. The results are of interest to forest managers since they make decisions about silvicultural operations.Growth continuity and forest production indicate that any intervention should be directed at younger trees of smaller sizes, and that one of the main management factors for stand stability and growth is the formation of the stand and its capture of light.Andre Felipe Hess Myrcia Minatti Emanuel Arnoni Costa Luis Paulo Baldissera Schorr Gabriel Teixeira da Rosa Isadora de Arruda Souza Geedre Adriano Borsoi Veraldo Liesenberg Thiago Floriani Stepka Roberta Abatti 2021Journal of Forestry Research2021,32,1:2

Potential use of lactate for the treatment of neonatal hypoxic-ischemic encephalopathy显示文摘Function of lactate:Lactate is a three-carbon molecule produced by glycolytic metabolism that is a metabolic waste product with no known use in clinical therapy.Conversely,it is a metabolite that the body should quickly guarantee the clearance. However, lactate is now recognized as a potential energy substrate,as well as an anti-inflammatory signaling molecule. These actions were first reported in adult animal models with a brain injury, including a traumatic brain injury and cerebral ischemia, and have also been observed in human patients(Magistretti and Allaman,2018).Recently,however,two studies by independent research groups described promising n euro p rotective results from the use of lactate in animal models with neonatal hypoxia-ischemia(Roumes et al.,2021;Tassinari et al.,2020).Isadora D’Ávila Tassinari Luciano Stürmer de Fraga 2022Neural Regeneration Research2022,17,4:1
Geoctenologies Applied to Evolution in Urban Design in a City in Southern Brazil显示文摘The evolution process of urban design is the result of transformations due to population increase along the expansion of cities on a global scale.Geotechnologies applied to forms of mapping make it possible to understand the changes that have occurred in urban spaces in relation to temporal space. The general objective of this study is to analyze the change that occurred from the year 2010 to 2021,in the evolution of urban design,in the city of Marau(southern Brazil). The identification of urban evolution in the years 2010 and 2021 was carried out. These data were modeled using the QGIS software,in the SIRGAS(Geocentric Reference System for the Americas)database, related to the evolution of urban design in Marau. The results demonstrate the need to apply geotechnologies not only in the city of Marau, but in other cities worldwide, because city halls need precision maps, avoiding the fragmentation of the urban fabric,a result of population expansion not projected in the built environment.Michele Bianchini Alcindo Neckel Maria Eduarda Cargnin Bade Laura Pasa Cambrussi Emanuela Brandelero Siqueira Julian Grub Luana Pasquetti Paloma Toscan Henrique Aniceto Kujawa Isadora Cezar Caino 2021Journal of Civil Engineering and Architecture2021,15,10:0
Temporal pattern of humoral immune response in mild cases of COVID-19显示文摘BACKGROUND Understanding the humoral response pattern of coronavirus disease 2019(COVID-19)is one of the essential factors to better characterize the immune memory of patients, which allows understanding the temporality of reinfection, provides answers about the efficacy and durability of protection against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),and consequently helps in global public health and vaccination strategy.Among the patients who became infected with SARS-CoV-2,the majority who did not progress to death were those who developed the mild COVID-19,so understanding the pattern and temporality of the antibody response of these patients is certainly relevant.AIM To investigate the temporal pattern of humoral response of specific immunoglobulin G(IgG)in mild cases of COVID-19.METHODS Blood samples from 191 COVID-19 real-time reverse transcriptase-polymerase chain reaction(RT-qPCR)-positive volunteers from the municipality of Toledo/Paraná/Brazil,underwent two distinct serological tests,enzyme-linked immunosorbent assay, and detection of anti-nucleocapsid IgG.Blood samples and clinicoepidemiological data of the volunteers were collected between November 2020 and February 2021.All assays were performed in duplicate and the manufacturers'recommendations were strictly followed. The data were statistically analyzed using multiple logistic regression;the variables were selected by applying the P<0.05 criterion.RESULTS Serological tests to detect specific IgG were performed on serum samples from volunteers who were diagnosed as being positive by RT-qPCR for COVID-19 or had disease onset in the time interval from less than 1 mo to 7 mo.The time periods when the highest number of participants with detectable IgG was observed were 1,2 and 3 mo.It was observed that 9.42%of participants no longer had detectable IgG antibodies 1 mo only after being infected with SARS-CoV-2 and 1.57%were also IgG negative at less than 1 mo.At 5 mo,3.14%of volunteers were IgG negative, and at 6 or 7 mo,1 volunteer(0.52%)had no detectable IgG.During the period between diagnosis by RT-qPCR/symptoms onset and the date of collection for the study,no statistical significance was observed for any association analyzed. Moreover, considering the age category between 31 and 59 years as the exposed group, the P value was 0.11 for the category 31 to 59 years and 0.32 for the category 60 years or older, showing that in both age categories there was no association between the pair of variables analyzed.Regarding chronic disease, the exposure group consisted of the participants without any comorbidity,so the P value of 0.07 for the category of those with at least one chronic disease showed no association between the two variables.CONCLUSION A temporal pattern of IgG response was not observed, but it is suggested that immunological memory is weak and there is no association between IgG production and age or chronic disease in mild COVID-19.Isadora Maria Pilati Campos Milena Marques Gabrielle Caroline Peiter Ana Paula Carneiro Brandalize Mauricio Bedim dos Santos Fabrício Freire de Melo Kádima Nayara Teixeira 2023World Journal of Biological Chemistry2023,14,2:0
Quo vadis? Bioengineered polysaccharide-based hydrogel scaffolds for damaged central nervous system recovery and regeneration显示文摘Impairments in the central nervous system(CNS)are a prevalent cause of life-long disabilities worldwide, representing serious health, social, and economic concerns(Doblado et al.,2021).During the last decades, with the population eldering and the increase in the life span,we have experienced an increase in CNS-related disorders, like stroke and neurodegenerative diseases(Jarrin et al.,2021).Isadora C.Carvalho Herman S.Mansur 2022Neural Regeneration Research2022,17,7:0
Syndemic aspects between COVID-19 pandemic and social inequalities显示文摘Although the coronavirus disease 2019(COVID-19)pandemic has reached all over the world population,it has demonstrated a heterogeneous impact on different populations. The most vulnerable communities which coexist daily with the social inequalities like low access to hygiene and personal protection products, crowded residences, and higher levels of chronic diseases have a higher risk of contact and the spread of infection, beyond unfavorable clinical outcomes. The elevation of the risk of infection exposure can be related to gender due to the presence of a larger contingent of women in essential services,as well as frontline and cleaning professionals who regardless of gender have the greatest exposure to the virus.Such exposures can contribute to the development of fear of contaminating themselves or their family members associated also with the work stress, both of which are related to the emergence of mental disturbances in these populations.Furthermore, conditions of unsanitary living and low socioeconomic status, populations at war,pre-existing social barriers, and ethnicity have contributed to more impact of the pandemic both in the exposure to the virus and access to health services,COVID-19 management, and management of other pathologies. At the same time, factors such as the closing of non-essential services, the loss of jobs, and the increase in household spending aggravated the social vulnerabilities and impacted the family economy.Lastly, the COVID-19 pandemic contributed still more to the impact on women's health since it propitiated a favorable environment for increasing domestic violence rates, through the segregation of women from social life, and increasing the time of the victims with their aggressors.Jonathan Santos Apolonio Ronaldo Teixeira da Silva Júnior Beatriz Rocha Cuzzuol Glauber Rocha Lima Araújo Hanna Santos Marques Isadora de Souza Barcelos Luana Kauany de SáSantos Luciano Hasimoto Malheiro Vinícius Lima de Souza Gonçalves Fabrício Freire de Melo 2022World Journal of Methodology2022,12,5:0
Ionic liquid-loaded microcapsules doped into dental resin infiltrants显示文摘Resin infiltrants have been effectively applied in dentistry to manage non-cavitated carious lesions in proximal dental surfaces. However, the common formulations are composed of inert methacrylate monomers. In this study,we developed a novel resin infiltrant with microcapsules loaded with an ionic liquid(MC-IL),and analyzed the physical properties and cytotoxicity of the dental resin.First, the ionic liquid 1-n-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)imide(BMI.NTf2)was synthesized.BMI.NTf2 has previously shown antibacterial activity in a dental resin.Then,MC-IL were synthesized by the deposition of a preformed polymer. The MC-IL were analyzed for particle size and de-agglomeration effect via laser diffraction analysis and shape via scanning electron microscopy(SEM).The infiltrants were formulated, and the MC-IL were incorporated at 2.5%,5%,and 10 wt%.A group without MC-IL was used as a control. The infiltrants were evaluated for ultimate tensile strength(UTS),contact angle, surface free energy(SFE),and cytotoxicity. The MC-IL showed a mean particle size of 1.64(±0.08)μm,shriveled aspect, and a de-agglomeration profile suggestive of nanoparticles’presence in the synthesized powder. There were no differences in UTS among groups(p>0.05).The incorporation of 10 wt%of MC-IL increased the contact angle(p<0.05),while the addition from 5 wt%reduced the SFE in comparison to the control group(p<0.05).The human cell viability was above 90%for all groups(p>0.05).The incorporation of microcapsules as a drug-delivery system for ionic liquids may be a promising strategy to improve dental restorative materials.Marla Cuppini Isadora Martini Garcia Virgínia Serra de Souza Kelly Cristine Zatta Fernanda Visioli Vicente Castelo Branco Leitune Sílvia Stanisçuazki Guterres Jackson Damiani Scholten Fabrício Mezzomo Collares 2021Bioactive Materials2021,6,9:0
Colorectal cancer carcinogenesis:From bench to bedside显示文摘Colorectal cancer(CRC)remains one of the main causes of cancer death in developed countries.Yet,it is potentially preventable,by removing the precursor lesions-adenomas or serrated lesions.Several studies proved that this intervention reduces CRC mortality and that the first colonoscopy’s results can guide surveillance strategies.More recently,it became clear that several carcinogenesis pathways may lead to sporadic CRC.CRC is a heterogeneous disease, characterized by multiple molecular subtypes.Three main pathways have been implicated in the development of CRC:Chromosomal instability, microsatellite instability, and the“serrated”pathways, with overlapping features between them. This and other molecular and genetic based CRC classifications are known to have clinical implications, spanning from familial risk assessment to therapy choices. The authors review basic science data and provide insight on current implications for the management of patients with CRC.Pedro Currais Isadora Rosa Isabel Claro 2022World Journal of Gastrointestinal Oncology2022,14,3:0
Elite, transformable haploid inducers in maize显示文摘The introduction of alleles into commercial crop breeding pipelines is both time consuming and costly. Two technologies that are disrupting traditional breeding processes are doubled haploid(DH)breeding and genome editing(GE).Recently, these techniques were combined into a GE trait delivery system called HI-Edit(Haploid Inducer-Edit).In HI-Edit,the pollen of a haploid inducer line is reprogrammed to deliver GE traits to any variety, obviating recurrent selection. For HI-Edit to operate at scale,an efficient transformable HI line is needed, but most maize varieties are recalcitrant to transformation, and haploid inducers are especially difficult to transform given their aberrant reproductive behaviors.Leveraging marker assisted selection and a three-tiered testing scheme,we report the development of new Iodent and Stiff Stalk maize germplasm that are transformable, have high haploid induction rates, and exhibit a robust,genetically-dominant anthocyanin native trait that may be used for rapid haploid identification. We show that transformation of these elite‘‘HI-Edit”lines is enhanced using the BABYBOOM and WUSCHEL morphogenetic factors. Finally,we evaluate the HI-Edit performance of one of the lines against both Stiff Stalk and non-Stiff Stalk testers. The strategy and results of this study should facilitate the development of commercially scalable HI-Edit systems in diverse crops.Brent Delzer Dawei Liang David Szwerdszarf Isadora Rodriguez Gonzalo Mardones Sivamani Elumalai Francine Johnson Samson Nalapalli Rachel Egger Erin Burch Kerry Meier Juan Wei Xiujuan Zhang Huaping Gui Huaibing Jin Huan Guo Kun Yu Yubo Liu Becky Breitinger Ana Poets Jason Nichols Wan Shi David Skibbe Qiudeng Que Timothy Kelliher 2024The Crop Journal2024,12,1:0
Effects of nutrients on immunomodulation in patients with severe COVID-19:Current knowledge显示文摘Recent research has demonstrated that critically ill patients with coronavirus disease 2019(COVID-19)show significant immune system dysregulation.Due to that, some nutrients that influence immunomodulation have been suggested as a form of treatment against the infection. This review collected the information on the impact of vitamins on the prognosis of COVID-19,with the intention of facilitating treatment and prevention of the disease risk status in patients. The collected information was obtained using the PubMed electronic database by searching for articles that relate COVID-19 and the mechanisms/effects of the nutrients:Proteins, glucose, lipids, vitamin B12,vitamin D,calcium, iron, copper, zinc, and magnesium, including prospective, retrospective, and support articles. The findings reveal an optimal response related mainly to omega-3,eicosapentaenoic acid, docosahexaenoic acid, calcium, and iron that might represent benefits in the treatment of critically ill patients. However, nutrient supplementation should be done with caution due to the limited availability of randomized controlled studies.Bruna Teixeira da Costa Glauber Rocha Lima Araújo Ronaldo Teixeira da Silva Júnior Luana Kauany de SáSantos Vinícius Lima de Souza Gonçalves Daniel Bastos Alves Lima Beatriz Rocha Cuzzuol Jonathan Santos Apolonio Lorena Sousa de Carvalho Hanna Santos Marques Camilo Santana Silva Isadora de Souza Barcelos Márcio Vasconcelos Oliveira Fabrício Freire de Melo 2022World Journal of Critical Care Medicine2022,11,4:0
