1Functional analysis of the larval serum protein gene promoter from silkworm,Bombyx mori.显示文摘The regulation region of larval serum protein gene, Bombyx mori. (BmLSP), consisting of the first intron, the first exon, the central promoter region and 5′-upstream region, is cloned from genomic DNA from the silkworm va-riety of Suju譓inghu. Using PCR and restriction endonu-clease methods, a series of luciferase reporter plasmids, driven by different length of BmLSP promoters, are con-structed. Via the transient expression system in BmN cells, the effects of the regulation elements and foreign insect hor-mones on the BmLSP promoter activity are investigated. The results demonstrate that the promoter activity of BmLSP is 5.8- or 4.4-fold higher than that of BmLSPs whose first in-tron or the element in 5′-upstream region harboring the homologous sequence with the first intron of light-chain fib-roin gene (EHIF) is deleted, respectively, suggesting that both the first intron and EHIF contain the main positive cis-acting elements. However, the inactive mariner transposable ele-ment (MTE) in 5′-upstream region presents a negative effect. Furthermore, the effects of juvenile hormone analogue (JHA) on the BmLSP promoter activity show a typical dose-dependent manner, that is, low concentration treat-ments increase the BmLSP promoter activity and high con-centration treatments decrease it. 4A new type of ERGIC–ERES membrane contact mediated by TMED9 and SEC12 is required for autophagosome biogenesis显示文摘Under stress,the endomembrane system undergoes reorganization to support autophagosome biogenesis,which is a central step in autophagy.How the endomembrane system remodels has been poorly understood.Here we identify a new type of membrane contact formed between the ER-Golgi intermediate compartment(ERGIC)and the ER-exit site(ERES)in the ER-Golgi system,which is essential for promoting autophagosome biogenesis induced by different stress stimuli.The ERGIC-ERES contact is established by the interaction between TMED9 and SEC12 which generates a short distance opposition(as close as 2-5 nm)between the two compartments.The tight membrane contact allows the ERES-located SEC12 to transactivate COPII assembly on the ERGIC.In addition,a portion of SEC12 also relocates to the ERGIC.Through both mechanisms,the ERGIC-ERES contact promotes formation of the ERGIC-derived COPII vesicle,a membrane precursor of the autophagosome.The ERGIC-ERES contact is physically and functionally different from the TFG-mediated ERGIC-ERES adjunction involved in secretory protein transport,and therefore defines a unique endomembrane structure generated upon stress conditions for autophagic membrane formation.Shulin Li Rui Yan Jialu Xu Shiqun Zhao Xinyu Ma Qiming Sun Min Zhang Ying Li Jun-Jie Gogo Liu Liangyi Chen Sai Li Ke Xu Liang Ge 2022Cell Research2022,32,2:3
5A theoretic study of a distance-based regression model显示文摘The distance-based regression model has many applications in analysis of multivariate response regression in various ?elds, such as ecology, genomics, genetics, human microbiomics, and neuroimaging. It yields a pseudo F test statistic that assesses the relation between the distance(dissimilarity) of the subjects and the predictors of interest. Despite its popularity in recent decades, the statistical properties of the pseudo F test statistic have not been revealed to our knowledge. This study derives the asymptotic properties of the pseudo F test statistic using spectral decomposition under the matrix normal assumption, when the utilized dissimilarity measure is the Euclidean or Mahalanobis distance. The pseudo F test statistic with the Euclidean distance has the same distribution as the quotient of two Chi-squared-type mixtures. The denominator and numerator of the quotient are approximated using a random variable of the form ξχ_d^2+ η, and the approximate error bound is given. The pseudo F test statistic with the Mahalanobis distance follows an F distribution.In simulation studies, the approximated distribution well matched the 'exact' distribution obtained by the permutation procedure. 6Immunogenicity and protective efficacy of a recombinant protein subunit vaccine and an inactivated vaccine against SARS-CoV-2 variants in non-human primates显示文摘Emerging SARS-CoV-2 variants and the gradually decreasing neutralizing antibodies over time post vaccination have led to an increase in incidents of breakthrough infection across the world.To investigate the potential protective effect of the recombinant protein subunit COVID-19 vaccine targeting receptor-binding domain(RBD)(PS-RBD)and whole inactivated virus particle vaccine(IV)against the variant strains,in this study,rhesus macaques were immunized with PS-RBD or IV vaccine,followed by a Beta variant(B.1.351)challenge.Although neutralizing activity against the Beta variant was reduced compared with that against the prototype,the decreased viral load in both upper and lower respiratory tracts,milder pathological changes,and downregulated inflammatory cytokine levels in lung tissues after challenge demonstrated that PS-RBD and IV still provided effective protection against the Beta variant in the macaque model.Furthermore,PS-RBD-induced macaque sera possessed general binding and neutralizing activity to Alpha,Beta,Delta,and Omicron variants in our study,though the neutralizing antibody(NAb)titers declined by varying degrees,demonstrating potential protection of PS-RBD against current circulating variants of concern(VOCs).Interestingly,although the IV vaccine-induced extremely low neutralizing antibody titers against the Beta variant,it still showed reduction for viral load and significantly alleviated pathological change.Other correlates of vaccine-induced protection(CoP)like antibody-dependent cellular cytotoxicity(ADCC)and immune memory were both confirmed to be existing in IV vaccinated group and possibly be involved in the protective mechanism.Qian He Qunying Mao Xiaozhong Peng Zhanlong He Shuaiyao Lu Jialu Zhang Fan Gao Lianlian Bian Chaoqiang An Wenhai Yu Fengmei Yang Yanan Zhou Yun Yang Yanyan Li Yadi Yuan Xujia Yan Jinghuan Yang Xing Wu Weijin Huang Changgui Li Junzhi Wang Zhenglun Liang Miao Xu 2022Signal Transduction and Targeted Therapy2022,7,4:2

8Research on the influence of equity pledge on stock price crash risk:based on financial shock of 2015 stock market crisis显示文摘The 2015 Chinese stock market crisis triggered liquidation because of equity pledge so that the leverage effect of the small probability event with severe results got intensive attention from investors.It is found that the effects of equity pledge on stock price crash risk reversed significantly before and after the 2015 stock market crisis.In the mechanism analysis,we further find that the equity pledge influenced the stock price crash risk by longer suspension and greater price fluctuation.The shareholding ratio of institutional investors and information environment also had a significant moderating effect on the influence of equity pledge on stock price crash risk.Alternative interpretation tests excluded the tunnel effect and pressure effect by shareholders and incentive effect by management.This study by analysing empirical data provides evidence on the change of investors’risk recognition,which is caused by financial shock,in the Chinese capital market.Rong Xu Jialu Chang Conggang Li Wenlan Wang 2019Economic and Political Studies2019,7,4:2

10Visible-Light-Sensitive SrCO3/AgI Hybrids for Tetracycline Degradation显示文摘The SrCO3/AgI photocatalysts were prepared via a co-precipitation method by using SrCO3 as a co-photocatalyst and AgI as a photo sensitizer.X-ray diffraction,field emission scanning electron microscope,X-ray photoelectron spectrometer,UV-vis diffuse reflectance spectroscopy and electrochemical impedance spectroscope were used to analyze the structure,micro-morphology,chemical compositions,optical properties and photo-generated carrier behaviors of the as-prepared samples,respectively.The photocatalytic degradation mechanism of the as-developed composites was also proposed.Analysis results show SrCO3,an insulator,can improve the photocatalytic performances and recyclability of AgI for degrading tetracycline under visible light.As the theoretical molar ratio of Sr(NO3)2 to AgNO3 increases,the degradation efficiency of the hybrids first increases and then descends.When the theoretical molar ratio of that is 1:1,it acquires the maximum of 66.6% within 8 min.This is higher than 32.0% of pure AgI and 34.0% of SrCO3.Moreover,after three times degradations it is 63.0%,which is higher than 13.6% of AgI.The improvement of the photocatalytic performance of the sample is attributed to the construction of hybrids.The main activated species in catalysis process are superoxide radicals.贾云宁 吴湘锋 LI Hui ZHANG Weiguang WANG Hui CHANG Tianlong FU Yunxuan LIU Xutao GUO Yudong SHANG Jialu 2020Journal of Wuhan University of Technology(Materials Science)2020,35,5:1
11Superoxide-like Cu/GO single-atom catalysts nanozyme with high specificity and activity for removing superoxide free radicals显示文摘Although nanozyme has become an emerging area of research attracting extensive attention recently,the activity and specificity of currently reported nanozymes are generally lower than those of natural enzymes.Developing highly active and specific nanozymes is therefore extremely necessary and also remains a great challenge.Superoxide dismutase(SOD)catalyzes the disproportionation of cytotoxic O_(2)·^(−)into hydrogen peroxide and oxygen,and plays an important role in reducing human oxidative stress.In this work,we prepare Cu single-atom catalysts(Cu/GO SACs,GO=graphene oxide)through a simple and low-cost strategy at room temperature using Cu foam and graphene oxide.Cu/GO SACs can maintain excellent catalytic activity under harsh environment.Compared with the natural enzyme,SOD-like Cu/GO SAC nanozyme has higher catalytic activity and meanwhile,it does not possess the common properties of other mimic enzymes often existing in nanomaterials.Based on the excellent SOD-like enzyme activity of Cu/GO SACs,it successfully eliminates the active oxygen in cigarette smoke.This work not only provides a new idea for the design and synthesis of nanozymes with excellent SOD mimetic properties,but also is promising in the treatment of lung injury and inflammatory diseases related to free radical production.Mingju Lu Jialu Wang Guoyuan Ren Fengjuan Qin Zhiqiang Zhao Kai Li Wenxing Chen Yuqing Lin 2022Nano Research2022,15,10:1
12Identification of two migratory colon ILC2 populations differentially expressing IL-17A and IL-5/IL-13显示文摘Group 2 innate lymphoid cells(ILC2s)play important tissue resident roles in anti-parasite immunity,allergic immune response,tissue homeostasis,and tumor immunity.ILC2s are considered tissue resident cells with little proliferation at steady state.Recent studies have shown that a subset of small intestinal ILC2s could leave their residing tissues,circulate and migrate to different organs,including lung,liver,mesenteric LN and spleen,upon activation.However,it remains unknown whether other ILC populations with migratory behavior exist.In this study,we find two major colon ILC2 populations with potential to migrate to the lung in response to IL-25 stimulation.One subset expresses IL-17A and resembles inflammatory ILC2s(iILC2s)but lacks CD27 expression,whereas the other expresses CD27 but not IL-17A.In addition,the IL-17A^(+)ILC2s express lower levels of CD127,CD25,and ST2 than CD27^(+)ILC2s,which express higher levels of IL-5 and IL-13.Surprisingly,we found that both colon ILC2 populations still maintained their colonic features of preferential expression of IL-17A and CD27,IL-5/IL-13,respectively.Together,our study identifies two migratory colon ILC2 subsets with unique surface markers and cytokine profiles which are critical in regulating lung and colon immunity and homeostasis.Hongzhi Liu Liang Li Yanyun Hao Jialu Li Zhaoyuan Liu Jingjing Qi Jingjing Zhang Ningbo Wu Dandan Wu Caixia Gao Lei Chen Lei Shen Jinke Cheng Bing Su 2023Science China(Life Sciences)2023,66,1:1

15Tenglong Buzhong granules(藤龙补中颗粒) inhibits the growth of SW620 human colon cancer in vivo显示文摘OBJECTIVE: To observe the anticancer effects of the granular preparation of Tenglong Buzhong decoction(藤龙补中汤,TBD), i.e Tenglong Buzhong granules(藤龙补中颗粒, TBG), in human SW620 colon cancer. METHODS: BALB/c nude mice were subcutaneously transplanted with SW620 cells, and treated with TBG(2.56 g/kg, once per day) and/or 5-Fu(104 mg/kg, once per week) for 21 d. Apoptosis, Caspase activities and cellular senescence were measured by commercial kits. The protein expression and phosphorylation were detected by Western blot or immunohistochemistry. RESULTS: TBG and 5-Fu inhibited tumor growth. The tumor inhibition rate of the TBG, 5-Fu, and TBG+5-Fu groups was 42.25%, 51.58%, and 76.08%, respectively. Combination of TBG and 5-Fu showed synergetic anticancer effects. TBG and 5-Fu induced apoptosis, activated caspase-3,-8, and-9, increased SMAC expression, inhibited XIAP expression. TBG induced cellular senescence, upregulated cyclin-dependent kinase inhibitor 1a(CDKN1a) and cyclin-dependent kinase inhibitor 2a(CDKN2a) expression, and inhibited phosphorylation of retinoblastoma-associated protein(RB) and expression of cyclin E1(CCNE1) and cyclindependent kinases(CDK) 2. TBG also inhibited angiogenesis accompanied by downregulation of vascular endothelial growth factor(VEGF) and hypoxiainducible factor-1α(HIF-1α).
