| 2 | Genetic basis and breeding application of clonal diversity and dual reproduction modes in polyploid Carassius auratus gibelio显示文摘A unisexual species is generally associated with polyploidy, and reproduced by a unisexual reproduction mode, such as gyno- genesis, hybridogenesis or parthenogenesis. Compared with other unisexual and polyploid species, gibel carp (Carassius au- ratus gibelio) has a higher ploidy level of hexaploid. It has undergone several successive rounds of genome polyploidy, and experienced an additional, more recent genome duplication event. More significantly, the dual reproduction modes, including gynogenesis and sexual reproduction, have been demonstrated to coexist in the polyploid gibel carp. This article reviews the genetic basis concerning polyploidy origin, clonal diversity and dual reproduction modes, and outlines the progress in new va- riety breeding and gene identification involved in the reproduction and early development. The data suggests that gibel carp are under an evolutionary trajectory of diploidization. | 3 | Molecular basis and genetic improvement of economically important traits in aquaculture animals显示文摘Aquaculture has been believed to be a major Chinese contribution to the world. In recent 20 years, genome and other genetic technologies have promoted significant advances in basic studies on molecular basis and genetic improvement of aquaculture animals, and complete genomes of some main aquaculture animals have been sequenced or announced to be sequenced since the beginning of this century. Here, we review some significant breakthrough progress of aquaculture genetic improvement technologies including genome technologies, somatic cell nuclear transfer and stem cell technologies, outline the molecular basis of several economically important traits including reproduction, sex, growth, disease resistance, cold tolerance and hypoxia tolerance, and present a series of candidate trait-related genes. | 4 | Clay minerals in surface sediments of the Pearl River drainage basin and their contribution to the South China Sea显示文摘Clay minerals have played a significant role in the study of the East Asian monsoon evolution in the South China Sea by being able to track oceanic current variations and to reveal contemporaneous pa- leoclimatic changes prevailing in continental source areas.As one of the most important rivers input- ting terrigenous matters to the northern South China Sea,the Pearl River was not previously paid at- tention to from the viewpoint of clay mineralogy.This paper presents a detailed study on clay minerals in surface sediments collected from the Pearl River drainage basin(including all three main channels, various branches,and the Lingdingyang in the estuary)by using the X-ray diffraction(XRD)method. The results indicate that the clay mineral assemblage consists dominantly of kaolinite(35%-65%), lesser abundance of chlorite(20%-35%)and illite(12%-42%),and very scare smectite occurrences (generally<5%).Their respective distribution does not present any obvious difference throughout the Pearl River drainage basin.However,downstream the Pearl River to the northern South China Sea,the clay mineral assemblage varies significantly:kaolinite decreases gradually,smectite and illite increase gradually.Additionally,illite chemistry index steps down and illite crystallinity steps up.These varia- tions indicate the contribution of major kaolinite,lesser illite and chlorite,and very scarce smectite to the northern South China Sea from the Pearl River drainage basin.The maximum contribution of clay minerals from the Pearl River is 72%to the northern margin and only 15%to the northern slope of the South China Sea.In both glacials and interglacials,kaolinite indicates that the ability of mechanical erosion occurred in the Pearl River drainage basin. | LIU ZhiFei Christophe COLIN HUANG Wei CHEN Zhong Alain TRENTESAUX CHEN JianFang | 2007 | Chinese Science Bulletin2007,52,8: | 33 |
| 5 | Detection of Grass Carp Hemorrhage Virus (GCHV) from Vietnam and Comparison with GCHV Strain from China显示文摘Grass carp plays an important role in small-scale aquaculture in Vietnam. However, a severe disease, known in Vietnam as 'Red Spot Disease', is causing significant economic loss in grass carp aquaculture. In this study, the tissue samples isolated from the grass carp with Red Spot Disease in Vietnam are investigated and comparied with the control GCHV isolated in China by experimental infection, culture cell infection, serological cross reactivity, and RT-PCR amplification. Infected grass carp exhibits hemorrhage symptoms about 5 days after experimental injection with GCHV-V (Vietnam) strain. The symptoms and lethality induced by the GCHV-V strain are identical to that induced by the Chinese GCHV-9014 strain. The Chinese GCHV-873 strain induces typical cytopathogenic effects in 4 cell lines, such as CIK, CAB, FHM and GCO, from the 6 fish cell lines examined. No cytopathogenic effects are observed in all the 6 examined cell lines, including CAB, FHM, CIK, EPC, CCO and GCO, infected by the GCHV-V strain and GCHV-9014 strain. Immunodiffusion assays demonstrate an obvious cross-reactivity among three GCHV strains. Precipitin lines are clearly observed not only between the anti-GCHV-873 serum and the two strains GCHV-873 and GCHV-9014, but also between the anti-GCHV-873 serum and the GCHV-V strain. GCHV can be detected by immunodiffusion assays after three generations of blind propagations in the cell lines inoculated by GCHV-V strain. This implicates that GCHV-V viruses have been replicated and amplified despite there being no cytopathogenic effects observed in these examined cell lines. Three genome segments of GCHV, including S8, S9 and S10, are amplified by three sets of PCR primers designed according to the segment sequences published recently. The Q8fp and Q8rp primer set specific for genome segment S8 amplifies a 955 bp fragment from the extracted sample of diseased fish with Red Spot Disease, and the fragment size is identical to that amplified by the same primer set from control GCHV-873 strain. Simultaneously, the Q9fp and Q9rp primer set specific for genome segment S9 generates a same 635 bp product, and the Q10fp and Q10rp primer set specific for genome segment S10 produces a same 697 bp fragment from both template samples of diseased fish with Red Spot Disease and control GCHV-873 strain. The RT-PCR amplification and corresponding size comparison data indicate that the three GCHV-V genome segments extracted from the diseased grass carp with Red Spot Disease in Vietnam should be identical to that in control GCHV-873 strain from China. | 6 | Estimations of primary production and export production in the South China Sea based on sediment trap experiments显示文摘The results of time series sediment trap experiments in the South China Sea show that particulate organic carbon (POC) fluxes are influenced by the monsoons. The increase of productivity in the northern South China Sea is mainly due to northeast monsoon while in the central South China Sea the influence of southwest monsoon becomes more prominent. The annual primary production and export production calculated based on POC fluxes are 53 0-63 4 and 10 32-12 93 gC·m -2·a -1, respectively. | 11 | Recognition on the forming-vanishing process and underlying mechanisms of the hypoxia off the Yangtze River estuary显示文摘On the basis of compiled multidisciplinary historical data in 2006–2007 and incorporation of relevant simulation results and remote sensing data, we performed an in-depth study of the generation and dissipation process of the hypoxic zone and its distribution morphology and structure off the Yangtze River estuary. Based on the hydrological circulation dynamics, reproduction of phytoplankton(leading to the decomposition of organic matter), and other factors, we comprehensively and systematically investigated the generation and dissipation of the hypoxic zone and underlying mechanisms for the seasonal variation in its position, explored the multi-factorial synergistic reactions during the generation and dissipation process of the hypoxic zone, and revealed the controlled mechanism for the morphology and structure of the hypoxic zone's distribution. Our studies indicate that in the winter and spring seasons, the hydrological environment off the Yangtze River estuary provides a water body with relatively low contents of dissolved oxygen(DO), which is the background for the formation of a hypoxic zone. After entering into the summer season, the hypoxic zone gradually develops towards the north and becomes mature. Because of the impact of the terrain, local decomposition of organic matter, and upwelling of the Kuroshio subsurface water in July–August, the hypoxic zone off the Yangtze River estuary exhibits the characteristics of discontinuous distribution in space and has a south and north 'dual-core' structure in the inner continental shelf. In addition, there is a hypoxic core in the eastern outer continental shelf. The degrees of hypoxia vary for different areas; they are strongest overall in the north, next strongest in the south; they are weakest on the outer continental shelf. In summer, the hypoxic zone in the north is related to the northward differentiation of the southern hypoxic zone and results from local development and intensification. In August, the hypoxic zone in the north reaches its peak, and after September, it rapidly retreats southward and disappears because of weakening stratification. In the fall, there is hypoxic zone along the coast of Zhejiang in the south, and there is also a low-DO area to the southwest of Jeju Island, with both zones disappearing rapidly. In addition, the change of dynamic environment also causes the low-DO area of the outer continental shelf to move outward in the fall. The variation in the intensity of the stratification and its cumulative effects as a barrier of vertical DO transportation over long periods of time have a significant impact on the degree of hypoxia in the hypoxic zone. In addition, the seasonal variations in the size of the stratified region, intensity of each current system/water mass, upwelling, front, and high-value area of phytoplankton biomass jointly restrict the extension of the hypoxic zone in the inner continental shelf and latitudinal(south-north direction) movement of its location off the Yangtze River estuary. The combined effect of dynamic factors, such as that of the Kuroshio subsurface water, causes a low-DO core in the outer continental shelf. The bottom cold water to the north of the East China Sea is the dynamic basis for the formation of the low-DO area to the southwest of Jeju Island during the fall season. The special seabed topography and mud area distribution off the Yangtze River estuary have a certain degree of influence on the development of the hypoxic zone. | 12 | Roles of a sustained activation of NCED3 and the synergistic regulation of ABA biosynthesis and catabolism in ABA signal production in Arabidopsis显示文摘ABA, acting as a stress signal, plays crucial roles in plant resistance to water stress. Because ABA signal production is based on ABA biosynthesis, the regulation of NCED, a key enzyme in the ABA biosynthesis pathway, is normally thought of as the sole factor controlling ABA signal production. Here we demonstrate that ABA catabolism in combination with a synergistic regulation of ABA biosynthesis plays a crucial role in governing ABA signal production. Water stress induced a significant accumulation of ABA, which exhibited different patterns in detached and attached leaves. ABA catabolism followed a temporal trend of exponential decay for both basic and stress ABA, and there was little difference in the catabolic half-lives of basic ABA and stress ABA. Thus, the absolute rate of ABA catabolism, i.e. the amount of ABA catabolized per unit time, increases with increased ABA accumulation. From the dynamic processes of ABA biosynthesis and catabolism, it can be inferred that stress ABA accumulation may be governed by a synergistic regulation of all the steps in the ABA biosynthesis pathway. Moreover, to maintain an elevated level of stress ABA sustained activation of NCED3 should be required. This inference was supported by further findings that the genes encoding major enzymes in the ABA biosynthesis pathway, e.g. NCED3, AAO3 and ABA3 were all activated by water stress, and with ABA accumulation progressing, the expressions of NCED3, AAO3 and ABA3 remained activated. | 13 | Sedimentary records of eutrophication in the Changjiang Estuary upwelling area over last 100 a显示文摘The upwelling area in the Changjiang Estuary was selected to collect the core, where the red tide occurred frequently and hypoxic existed. The total organic carbon (TOC), total nitrogen (TN), biogenic silica (BSi) and stable organic carbon isotopic ratios(δ13 Corg) were determined on the 210Pb-dated sediment core. The concentrations of TOC, TN, BSi as well as their sedimentation fluxes have increased to some extent since the 1970s. TOC and TN fluxes increased about 45%, 36% respectively. The average δ13 Corg value in the core was -23.67×10-3 which remained nearly constant before the 20 century. The δ13 Corg values increased after the 1900s, two marked increases were observed from the 1950s and the 1970s. A simple δ13 Corg model was used to estimate the contribution of terrigenous and marine organic matter inputs for the sediment, which indicated the increase in accumulation since the 1970s has been almost exclusively marine. The increasing of marine organic matter accumulation (TOC, TN and BSi) was corresponding with the increasing of fertilizer consumption and the NO3-N budgets from the Changjiang River. The riverine runoff of fertilizers and nutrients stimulated the algae blooming. Enhanced primary production resulted in an enrichment of organic matter in the sediment. | 15 | Oceanic environmental changes of subarctic Bering Sea in recent 100 years: Evidence from molecular fossils显示文摘The core sample B2-9 from the seafloor of the subarctic Bering Sea was dated with 210 Pb to obtain a consecutive sequence of oceanic sedimentary environments at an interval of a decade during 1890—1999. A variety of molecular fossils were detected, including n-alkanes, isoprenoids, fatty acids, sterols, etc. By the characteristics of these fine molecules (C27, C28, and C29 sterols) and their molecular indices (Pr/Ph, ∑C+22/∑C?21, CPI and C18∶2/C18∶0) and in con- sideration of the variation of organic carbon content, the 100-year evolution history of subarctic sea paleoenvironment was reestablished. It is indicated that during the past 100 years in the Arctic, there were two events of strong climate warming (1920—1950 and 1980—1999), which resulted in an oxidated sediment environment owing to decreasing terrigenous organic matters and increasing marine-derived organic matters, and two events of transitory climate cooling (1910 and 1970—1980), which resulted in a slightly reduced sediment environment owing to increasing terrigenous organic matters and decreasing marine-derived organic matters. | 17 | Phenotypic and genetic variation between two populations of the Chinese yellow pond turtle, Mauremys mutica (Cantor, 1842)显示文摘Mauremys mutica(Cantor,1842)is an endangered species in China.Main phenotypic variations inbody color,body weight,body shape,clutch size,egg size,and hatchling size were revealed betweenthe southern and northern populations.Both populations have the phenomenon of'larger male'sexualsize dimorphism(SSD),especially in the southern population.Furthermore,genetic variations betweenthe two populations were analyzed by RAPD band patterns of 30 random individuals in each population.The average genetic distance was 0.299±0.108 among the samples of two populations.The average ge-netic distance between southern and northern populations was 0.305±0.046.Cluster analysis indicatedthat all the individuals from the southern and northern populations were clustered among themselves toform two distinct clades.A total of 20 population-specific RAPD fragments were scored from 16 primers,and could be used as RAPD markers for distinguishing the southern and the northern population.Basedon the nucleotide sequences of two RAPD markers,two pairs of SCAR primers(SC1-S and SC2-S)weredesigned,which could be used as SCAR markers for the southern population.According to the significantphenotypic and genetic variations,we suggested that the northern population and southern populationmight be considered as two separate taxa,the'northern taxon'and the'southern taxon',and the con-servation should be respectively conducted on the two taxa. | 朱新平 Zhou Li Chen Yongle Du Hejun Gui Jianfang | 2008 | High Technology Letters2008,14,1: | 10 |

| 19 | Remodeling the blood–brain barrier microenvironment by natural products for brain tumor therapy显示文摘Brain tumor incidence shows an upward trend in recent years; brain tumors account for 5% of adult tumors, while in children, this figure has increased to 70%. Moreover, 20%–30% of malignant tumors will eventually metastasize into the brain. Both benign and malignant tumors can cause an increase in intracranial pressure and brain tissue compression, leading to central nervous system(CNS) damage which endangers the patients' lives. Despite the many approaches to treating brain tumors and the progress that has been made, only modest gains in survival time of brain tumor patients have been achieved. At present, chemotherapy is the treatment of choice for many cancers, but the special structure of the blood–brain barrier(BBB) limits most chemotherapeutic agents from passing through the BBB and penetrating into tumors in the brain. The BBB microenvironment contains numerous cell types, including endothelial cells, astrocytes, peripheral cells and microglia, and extracellular matrix(ECM). Many chemical components of natural products are reported to regulate the BBB microenvironment near brain tumors and assist in their treatment. | 20 | Potential targets for protecting against hippocampal cell apoptosis after transient cerebral ischemiareperfusion injury in aged rats显示文摘Mitochondria play an important role in neuronal apoptosis caused by cerebral ischemia, and the role is mediated by the expression of mitochondrial proteins. This study investigated the involvement of mitochondrial proteins in hippocampal cell apoptosis after transient cerebral ischemia-reperfusion injury in aged rats using a comparative proteomics strategy. Our experimental results show that the aged rat brain is sensitive to ischemia-reperfusion injury and that transient ischemia led to cell apoptosis in the hippocampus and changes in memory and cognition of aged rats. Differential proteomics analysis suggested that this phenomenon may be mediated by mitochondrial proteins associated with energy metabolism and apoptosis in aged rats.
