Th e role of ferroptosis in ionizing radiation-induced cell death and tumor suppression显示文摘Ferroptosis,a form of regulated cell death caused by lipid peroxidation, was recently identified as a natural tumor suppression mechanism. Here,we show that ionizing radiation(IR)induces ferroptosis in cancer cells.Mechanistically,IR induces not only reactive oxygen species(ROS)but also the expression of ACSL4,a lipid metabolism enzyme required for ferroptosis, resulting in elevated lipid peroxidation and ferroptosis.ACSL4 ablation largely abolishes IR-induced ferroptosis and promotes radioresistance.IR also induces the expression of ferroptosis inhibitors, including SLC7A11 and GPX4;as an adaptive response.IR-or KEAP1 deficiencyinduced SLC7A11 expression promotes radioresistance through inhibiting ferroptosis.Inactivating SLC7A11 or GPX4 with ferroptosis inducers(FINs)sensitizes radioresistant cancer cells and xenograft tumors to IR.Furthermore, radiotherapy induces ferroptosis in cancer patients, and increased ferroptosis correlates with better response and longer survival to radiotherapy in cancer patients. Our study reveals a previously unrecognized link between IR and ferroptosis and indicates that further exploration of the combination of radiotherapy and FINs in cancer treatment is warranted. | Guang Lei Yilei Zhang Pranavi Koppula Xiaoguang Liu Jie Zhang Steven HLin Jaffer AAjani Qin Xiao Zhongxing Liao Hui Wang Boyi Gan | 2020 | Cell Research2020,30,2: | 88
Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally. Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans. Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed. Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever, cough, and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation. One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients, with 99.8%to 99.9%nucleotide identities among the isolates. These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity, but is in a separate clade. Moreover, these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However, the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor. Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans. | Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang | 2020 | Chinese Medical Journal2020,,9: | 99

Traditional herbal medicine in preventing recurrence after resection of small hepatocellular carcinoma: a multicenter randomized controlled trial显示文摘BACKGROUND: Disease recurrence is a main challenge in treatment of hepatocellular carcinoma (HCC). There is no generally accepted method for preventing recurrence of HCC after resection. OBJECTIVE: To compare the efficacy of a traditional herbal medicine (THM) regimen and transarterial chemoembolization (TACE) in preventing recurrence in post-resection patients with small HCC. DESIGN, SETTING, PARTICIPANTS AND INTERVENTIONS: This is a multicenter, open-label, randomized, controlled study, which was undertaken in five centers of China. A total of 379 patients who met the eligibility criteria and underwent randomization were enrolled in this trial. One hundred and eighty-eight patients were assigned to the THM group and received Cinobufacini injection and Jiedu Granule, and the other 191 patients were assigned to the TACE group and received one single course of TACE. MAIN OUTCOME MEASURES: Primary outcome measures were the annual recurrence rate and the time to recurrence. Incidence of adverse events was regarded as the secondary outcome measure. RESULTS: Among the 364 patients who were included in the intention-to-treat analysis, 67 patients of the THM group and 87 of the TACE group had recurrence, with a hazard ratio of 0.695 (P = 0.048). Median recurrence-free survival of the patients in the THM and TACE groups was 46.89 and 34.49 months, respectively. Recurrence rates at 1, 2 and 3 years were 17.7%, 33.0% and 43.5% for the THM group, and 28.8%, 42.5% and 54.0% for the TACE group, respectively (P = 0.026). Multivariate analysis indicated that the THM regimen had a big advantage for prolonging the recurrence-free survival. Adverse events were mild and abnormality of laboratory indices of the two groups were similar. CONCLUSION: In comparison with TACE therapy, the THM regimen was associated with diminished risk of recurrence of small-sized HCC after resection, with comparable adverse events. TRIAL REGISTRTION IDENTIFIER: This trial was registered in the Chinese Clinical Trial Registry with the identifier ChiCTR-TRC-07000033. | Xiao-feng Zhai Zhe Chen Bai Li Feng Shen Jia Fan Wei-ping Zhou Yun-ke Yang Jing Xu Xiao Qin Le-qun Li Chang-quan Ling | 2013 | Journal of Integrative Medicine2013,11,2: | 49

Cryospheric Science: research framework and disciplinary system显示文摘Cryospheric changes and their impacts have received increasing concern, extending the research to include the interactions between earth spheres and the impacts of and social adaptation to cryospheric changes. As a result, Cryospheric Science is rapidly developing. However, the research framework of Cryospheric Science has not completely been set up, and the intension and extension of Cryospheric Science are unclear.Following a literature review, the research framework for Cryospheric Science and the discipline components are analysed in this paper. We consider that Cryospheric Science is an inevitable product of international research on the earth and environmental changes as well as on human sustainable development. It will not only strengthen the linkage between the cryosphere and other earth spheres by deepening traditional research areas, but will also bring more focus on the critical roles of the cryosphere in interactions between earth spheres, and will enhance the relationship between cryospheric change and climate, ecology, hydrology, surface environment and sustainable development. The disciplinary tree of Cryospheric Science is developed following the mainstream of change-impacts adaptation. The disciplines of Cryospheric Science were teased out against characteristics of traditional branches and the developing interdisciplinary branches of cryosphere elements, which will provide reference to Cryospheric Science as it systematically grows to maturity. | Dahe Qin Yongjian Ding Cunde Xiao Shichang Kang Jianwen Ren Jianping Yang Shiqiang Zhang | 2018 | National Science Review2018,5,2: | 42
Developing the Science Product Algorithm Testbed for Chinese Next-Generation Geostationary Meteorological Satellites:Fengyun-4 Series显示文摘Fengyun-4A(FY-4A), the first of the Chinese next-generation geostationary meteorological satellites, launched in2016, offers several advances over the FY-2: more spectral bands, faster imaging, and infrared hyperspectral measurements. To support the major objective of developing the prototypes of FY-4 science algorithms, two science product algorithm testbeds for imagers and sounders have been developed by the scientists in the FY-4 Algorithm Working Group(AWG). Both testbeds, written in FORTRAN and C programming languages for Linux or UNIX systems, have been tested successfully by using Intel/g compilers. Some important FY-4 science products, including cloud mask, cloud properties, and temperature profiles, have been retrieved successfully through using a proxy imager, Himawari-8/Advanced Himawari Imager(AHI), and sounder data, obtained from the Atmospheric Infra Red Sounder, thus demonstrating their robustness. In addition, in early 2016, the FY-4 AWG was developed based on the imager testbed—a near real-time processing system for Himawari-8/AHI data for use by Chinese weather forecasters.Consequently, robust and flexible science product algorithm testbeds have provided essential and productive tools for popularizing FY-4 data and developing substantial improvements in FY-4 products. | Min MIN Chunqiang WU Chuan LI Hui LIU Na XU Xiao WU Lin CHEN Fu WANG Fenglin SUN Danyu QIN Xi WANG Bo LI Zhaojun ZHENG Guangzhen CAO Lixin DONG | 2017 | Journal of Meteorological Research2017,31,4: | 35
Microbial treatment in chronic constipation显示文摘Chronic functional constipation is a kind of common intestinal disease that occurs in children, adults and elderly people. This disease not only causes great influence to physiological function, but also results in varying degrees of psychological barriers. At present, constipation treatments continue to rely on traditional methods such as purgative therapy and surgery. However, these approaches can disrupt intestinal function. Recent research between intestinal diseases and gut microbiota has gradually revealed a connection between constipation and intestinal flora disturbance, providing a theoretical basis for microbial treatment in chronic constipation. Microbial treatment mainly includes probiotic preparations such as probiotics, prebiotics, synbiotics and fecal microbiota transplantation(FMT). Due to its safety, convenience and curative effect, probiotic preparations have been widely accepted, especially gradually developed FMT with higher curative effects. Microbial treatment improves clinical symptoms, promotes the recovery of intestinal flora, and has no complications during the treatment process. Compared with traditional treatments, microbial treatment in chronic constipation has advantages, and is worthy of further promotion from clinical research to clinical application. | Linsheng Huang Qi Zhu Xiao Qu Huanlong Qin | 2018 | Science China(Life Sciences)2018,61,7: | 40
New incompatible pair of TCM:Epimedii Folium combined with Psoraleae Fructus induces idiosyncratic hepatotoxicity under immunological stress conditions显示文摘Epimedii Folium(EF)combined with Psoraleae Fructus(PF)is a common modern preparation, but liver injury caused by Chinese patent medicine preparations containing EF and PF has been frequently reported in recent years.Zhuangguguanjiewan pills(ZGW),which contain EF and PF,could induce immune idiosyncratic liver injury according to clinical case reports and a nonhepatotoxic dose of lipopolysaccharide(LPS)model. This present study evaluated the liver injury induced by EF or PF alone or in combination and investigated the related mechanism by using the LPS model.Liver function indexes and pathological results showed that either EF or PF alone or in combination led to liver injury in normal rats;however,EF or PF alone could lead to liver injury in LPS-treated rats. Moreover,EF combined with PF could induce a greater degree of injury than that caused by EF or PF alone in LPS-treated rats.Furthermore,EF or PF alone or in combination enhanced the LPS-stimulated inflammatory cytokine production, implying that IL-1β,which is processed and released by activating the NLRP3 inflammasome,is a specific indicator of EF-induced immune idiosyncratic hepatotoxicity. Thus,EF may induce liver injury through enhancing the LPS-mediated proinflammatory cytokine production and activating the NLRP3 inflammasome. In addition, the metabolomics analysis results showed that PF affected more metabolites in glycerophospholipid and sphingolipid metabolic pathways compared with EF in LPS model, suggesting that PF increased the responsiveness of the liver to LPS or other inflammatory mediators via modulation of multiple metabolic pathways. Therefore,EF and PF combination indicates traditional Chinese medicine incompatibility, considering that it induces idiosyncratic hepatotoxicity under immunological stress conditions. | Yuan Gao Zhilei Wang Jinfa Tang Xiaoyi Liu Wei Shi Nan Qin Xiaoyan Wang Yu Pang Ruisheng Li Yaming Zhang Jiabo Wang Ming Niu Zhaofang Bai Xiaohe Xiao | 2020 | Frontiers of Medicine2020,14,1: | 32
Sample maturation calculated using Raman spectroscopic parameters for solid organics: Methodology and geological applications显示文摘The shape and shift of Raman peak of solid organics prove to be capable of revealing atomic and molecular level vibration information of aromatic ring structure and its relationship with sample maturation. Raman 'D' peaks and 'G' peaks of a series of continuously maturated coal samples were measured, and the inter-peak intervals (G-D) and peak height ratios (Dh/Gh) were derived and correlated with the vitrinite reflectance (vRo%) of standard coal samples. As a result, two formulae were established by using the two Raman indices for calculation of Raman reflectance (RmcRo%), which is equivalent to vitrinite reflectance. The formula for calculating Raman reflectance indicative of organic maturation using Raman shift inter-peak interval (G-D) is RmcRo%=0.0537d(G-D)-11.21, which is mainly applicable to matured to highly matured carbonized samples of solid organics; The formula for calculating Raman reflectance indicative of organic maturation using Raman peak height ratio (Dh/Gh) is RmcRo%=1.1659h (Dh/Gh)+2.7588, which is mainly applicable to carbonized samples of solid organics that are over matured or going to be turned into granulated graphite. Preliminary applications indicate that Raman reflectance 'RmcRo%' calculated based on results of Raman spectral analysis of solid organics can be used to characterize sample maturation at molecular level, so enjoying extensive prospects in geological applications. | LIU DeHan XIAO XianMing TIAN Hui MIN YuShun ZHOU Qin CHENG Peng SHEN JiaGui | 2013 | Chinese Science Bulletin2013,58,11: | 30
Metabolic dysregulation and emerging therapeutical targets for hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is an aggressive human cancer with increasing incidence worldwide.Multiple efforts have been made to explore pharmaceutical therapies to treat HCC,such as targeted tyrosine kinase inhibitors, immune based therapies and combination of chemotherapy. However, limitations exist in current strategies including chemoresistance for instance.Tumor initiation and progression is driven by reprogramming of metabolism,in particular during HCC development.Recently, metabolic associated fatty liver disease(MAFLD),a reappraisal of new nomenclature for nonalcoholic fatty liver disease(NAFLD),indicates growing appreciation of metabolism in the pathogenesis of liver disease, including HCC,thereby suggesting new strategies by targeting abnormal metabolism for HCC treatment. In this review,we introduce directions by highlighting the metabolic targets in glucose, fatty acid, amino acid and glutamine metabolism, which are suitable for HCC pharmaceutical intervention. We also summarize and discuss current pharmaceutical agents and studies targeting deregulated metabolism during HCC treatment.Furthermore, opportunities and challenges in the discovery and development of HCC therapy targeting metabolism are discussed. | Danyu Du Chan Liu Mengyao Qin Xiao Zhang Tao Xi Shengtao Yuan Haiping Hao Jing Xiong | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 27
Molecular mechanosensors in osteocytes显示文摘Osteocytes, the most abundant and long-lived cells in bone, are the master regulators of bone remodeling. In addition to their functions in endocrine regulation and calcium and phosphate metabolism, osteocytes are the major responsive cells in force adaptation due to mechanical stimulation. Mechanically induced bone formation and adaptation, disuse-induced bone loss and skeletal fragility are mediated by osteocytes, which sense local mechanical cues and respond to these cues in both direct and indirect ways. The mechanotransduction process in osteocytes is a complex but exquisite regulatory process between cells and their environment, between neighboring cells, and between different functional mechanosensors in individual cells. Over the past two decades, great efforts have focused on finding various mechanosensors in osteocytes that transmit extracellular mechanical signals into osteocytes and regulate responsive gene expression. The osteocyte cytoskeleton, dendritic processes, Integrin-based focal adhesions, connexin-based intercellular junctions, primary cilium, ion channels, and extracellular matrix are the major mechanosensors in osteocytes reported so far with evidence from both in vitro and in vitro studies. This review aims to give a systematic introduction to osteocyte mechanobiology, provide details of osteocyte mechanosensors, and discuss the roles of osteocyte mechanosensitive signaling pathways in the regulation of bone homeostasis. | Lei Qin Wen Liu Huiling Cao Guozhi Xiao | 2020 | Bone Research2020,8,2: | 24
A preliminary study of cryosphere service function and value evaluation显示文摘Cryosphere science research and development(R&D) has been strongly committed to public service, integrating natural sciences with socioeconomic impacts. Owing to the current shift from purely natural cryosphere scientific research to linking cryosphere science with socioeconomic and cultural science, cross-disciplinary research in this field is emerging, which advocates future cryosphere science research in this field. Utilizing the cryosphere service function(CSF), this study establishes CSF and its value evaluation system. Cryosphere service valuation can benefit the decisionmakers' and public's awareness of environmental protection. Implementing sustainable CSF utilization strategies and macroeconomic policymaking for global environmental protection will have profound and practical significance as well as avoid environmental degradation while pursuing short-term economic profits and achieving rapid economic development. | XIAO Cun-De WANG Shi-Jin QIN Da-He | 2015 | Advances in Climate Change Research2015,6,3: | 22
