2Activation of Toll-like receptors signaling in non-small cell lung cancer cell line induced by tumor-associated macrophages显示文摘Background:Inflammation is often linked with the progress and poor outcome of lung cancer.The understanding of the relationship between tumor-associated macrophages(TAMs) and lung cancer cells involves in the underlying mechanism of inflammatory cytokine production.Toll-like receptors(TLRs) are engaged in promoting the production of pro-inflammatory cytokines and play an important role in tumor immunology.Methods:To investigate the mechanisms by which TAMs influence the production of pro-inflammatory cytokines in lung cancer cells,we established an in vitro coculture system using TAMs and human nonsmall cell lung cancer(NSCLC) cell line SPC-A1.Levels of interleukin(IL)-1β,IL-6 and IL-8 in SPC-A1 were evaluated by RT-PCR and cytometric bead array assay after being cocultured with TAMs.Expression changes of TLRs and TLRs signaling pathway proteins in SPC-A1 were further confirmed by RT-PCR and western blot.The level changes of IL-1β,IL-6 and IL-8 in SPC-A1 were also detected after the stimulation of TLRs agonists.Results:We found that the phenotype markers of TAMs were highly expressed after stimulating human monocyte cell line THP-1 by phorbol-12-myristate-13-acetate(PMA).Higher mRNA and supernate secretion levels of IL-1β,IL-6 and IL-8 were detected in SPC-A1 after being cocultured with TAMs.We also found that TLR1,TLR6 and TLR7 were up-regulated in SPC-A1 in the coculture system with TAMs.Meanwhile,TLRs signaling pathway proteins were also significantly activated.Moreover,pre-treatment with agonist ligands for TLR1,TLR6 and TLR7 could dramatically promote inductions of IL-1β,IL-6 and IL-8.Conclusions:These findings demonstrated that TAMs may enhance IL-1β,IL-6 and IL-8 expressions via TLRs signaling pathway.We conclude that TAMs contribute to maintain the inflammation microenvironment and ultimately promote the development and progression of lung cancer.Xing Ke Meng Wu Jianfang Lou Shuping Zhang Peijun Huang Ruihong Sun Lei Huang Erfu Xie Fang Wang Bing Gu 2015Chinese Journal of Cancer Research2015,27,2:6
3Machine learning enabled prediction and process optimization of VFA production from riboflavin-mediated sludge fermentation显示文摘Riboflavin is a redox mediator that promotes volatile fatty acids(VFAs)production from waste activated sludge(WAS)and is a promising method for WAS reuse.However,time-and labor-consuming experiments challenge obtaining optimal operating conditions for maximal VFA production.In this study,three machine learning(ML)models were developed to predict the VFAs production from riboflavin-mediated WAS fermentation systems.Among the three tested ML algorithms,eXtreme Gradient Boosting(XGBoost)presented the best prediction performance and excellent generalization ability,with the highest testing coefficient of determination(R^(2)of 0.93)and lowest root mean square error(RMSE of 0.070).Feature importance analysis and their interactions using the Shepley Additive Explanations(SHAP)method indicated that pH and soluble protein were the top two input features for the modeling.The intrinsic correlations between input features and microbial communities corroborated this deduction.On the optimized ML model,genetic algorithm(GA)and particle swarm optimization(PSO)solved the optimal solution of VFA output,predicting the maximum VFA output as 650 mg COD/g VSS.This study provided a data-driven approach to predict and optimize VFA production from riboflavin-mediated WAS fermentation.Weishuai Li Jingang Huang Zhuoer Shi Wei Han Ting Lü Yuanyuan Lin Jianfang Meng Xiaobing Xu Pingzhi Hou 2023Frontiers of Environmental Science & Engineering2023,17,11:0
4A Large-Scale Group Decision Making Model Based on Trust Relationship and Social Network Updating显示文摘With the development of big data and social computing,large-scale group decisionmaking(LGDM)is nowmerging with social networks.Using social network analysis(SNA),this study proposes an LGDM consensus model that considers the trust relationship among decisionmakers(DMs).In the process of consensusmeasurement:the social network is constructed according to the social relationship among DMs,and the Louvain method is introduced to classify social networks to form subgroups.In this study,the weights of each decision maker and each subgroup are computed by comprehensive network weights and trust weights.In the process of consensus improvement:A feedback mechanism with four identification and two direction rules is designed to guide the consensus of the improvement process.Based on the trust relationship among DMs,the preferences are modified,and the corresponding social network is updated to accelerate the consensus.Compared with the previous research,the proposedmodel not only allows the subgroups to be reconstructed and updated during the adjustment process,but also improves the accuracy of the adjustment by the feedbackmechanism.Finally,an example analysis is conducted to verify the effectiveness and flexibility of the proposed method.Moreover,compared with previous studies,the superiority of the proposed method in solving the LGDM problem is highlighted.Rongrong Ren Luyang Su Xinyu Meng Jianfang Wang Meng Zhao 2024Computer Modeling in Engineering & Sciences2024,138,1:0
