1Profiling and bioinformatics analysis of differentially expressed circular RNAs in human intervertebral disc degeneration显示文摘The functional changes of nucleus pulposus (NP) cells are considered to be the initiating factors of intervertebral disc degeneration (IDD), and the differentially expressed circRNAs in NP cells may play an important role in the process of IDD. To identify circular RNAs (circRNAs) associated with human IDD, we isolated the NP cells from human degenerated and non-degenerated intervertebral disc and identified NP cells by microscopy and cell proliferation. CircRNA microarray expression profiles were obtained from NP cells of degenerated and non-degenerated intervertebral disc and further validated by quantitative reverse transcription PCR (qRTPCR). The expression data were analyzed by bioinformatics. Microarray analysis identified 7294 circRNAs differentially expressed in degenerated human IDD NP cells. Among them, 3724 circRNAs were up-regulated and 3570 circRNAs were down-regulated by more than 2 folds. After validating by qRT-PCR, we predicted the possible miRNAs of the top dysregulated circRNAs using TargetScan, and miRanda. Furthermore, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that the most modulated circRNAs regulate the viability, degradation, apoptosis and oxidative stress in NP cells, and the possible mechanism underlying IDD was discussed. 2The joint long-term distribution of design environmental factors for offshore structures Multivariate probabilities and simulating method显示文摘With the development of ocean engineering, it is one of the most important factors which determine the structural safety, cost and suitable forms of engineerings to select the ocean environmental design criteria. Owing to the complexity , variation and randomness of ocean environmental conditions, the commonly used methods for determining design criteria cannot consider the joint occurring probabilities of several environmental factors ,therefore, lead to overestimate design criteria of them and result in an unnecessary overspend invest in engineering. 3Changes in compressed neurons from dogs with acute and severe cauda equina constrictions following intrathecal injection of brain-derived neurotrophic factor-conjugated polymer nanoparticles显示文摘This study established a dog model of acute multiple cauda equina constriction by experimental constriction injury (48 hours) of the lumbosacral central processes in dorsal root ganglia neurons. The repair effect of intrathecal injection of brain-derived neurotrophic factor with 15 mg encapsulated biodegradable poly(lactide-co-glycolide) nanoparticles on this injury was then analyzed. Dorsal root ganglion cells (L7) of all experimental dogs were analyzed using hematoxylin-eosin staining and immunohistochemistry at 1, 2 and 4 weeks following model induction. Intrathecal injection of brain-derived neurotrophic factor can relieve degeneration and inflammation, and elevate the expression of brain-derived neurotrophic factor in sensory neurons of compressed dorsal root ganglion. Simultaneously, intrathecal injection of brain-derived neurotrophic factor obviously improved neurological function in the dog model of acute multiple cauda equina constriction. 4Skp2 dictates cell cycle-dependent metabolic oscillation between glycolysis and TCA cycle显示文摘Whether glucose is predomi nantly metabolized via oxidative phosphorylati on or glycolysis differs between quiesce nt versus proliferating cells,iincluding tumor cells.However,how glucose metabolism is coordinated with cell cycle in mammalian cells remains elusive.Here,we report that mammalian cells predominantly utilize the tricarboxylic acid(TCA)cycle in G1 phase,but prefer glycolysis in S phase.Mechanistically,coupling cell cycle with metabolism is largely achieved by timely destruction of IDH1/2,key TCA cycle enzymes,in a Skp2-dependent manner.As such,depleting SKP2 abolishes cell cycle-dependent fluctuation of IDH1 protein abundance,leading to reduced glycolysis in S phase.Furthermore,elevated Skp2 abundance in prostate cancer cells destabilizes IDH1 to favor glycolysis and subsequent tumorigenesis.Therefore,our study reveals a mechanistic link between two cancer hallmarks,aberrant cell cycle and addiction to glycolysis,and provides the underlying mechanism for the coupling of metabolic fluctuation with periodic cell cycle in mammalian cells.Jing Liu Yunhua Peng Le Shi Lixin Wan Hiroyuki Inuzuka Jiangang Long Jianping Guo Jinfang Zhang Min Yuan Shuangxi Zhang Xun Wang Jing Gao Xiangpeng Dai Shozo Furumoto Lijun Jia Pier Paolo Pandolfic John M.Asara William G.Kaelin Jr. Jjankang Liu Wenyi Wei 2021Cell Research2021,31,1:2
5Analysis of Abnormal Characteristics of Regional Crustal Deformation before the Menyuan MS6.4 Earthquake by GPS Continuous Data显示文摘In order to study the characteristics of crustal deformation around the epicenter before the 2016 M_S6. 4 Menyuan earthquake,the GPS continuous stations of the period from 2010 to 2016 were selected according to the observation data of the tectonic environment monitoring network in Chinese Mainland. The deformation characteristics of the crust before the earthquake were discussed through inter-station baseline time series analysis and the strain time series analysis in the epicentral region. The results show that a trend turn of the baseline movement state around the epicenter region occurred after 2014,and the movement after 2014 reflects an obvious decreasing trend of compressional deformation.During this period,the stress field energy was in a certain accumulation state. Since the beginning of 2014,the EW-component linear strain and surface strain rate weakened gradually before the earthquake. It shows that there was an obvious deformation deficit at the epicentral area in the past two years,which indicates that the region accumulated a high degree of strain energy before the earthquake. Therefore,there was a significant background change in the area before the earthquake. 6Application of Monte-Carlo statistical experiments in design of ocean engineering - Estimating the parameters, models and probabilities显示文摘Recently, some results have been acquired with the Monte- Carlo statistical experiments in the design of ocean en gineering. 11Reductase activity of human thioredoxin-like protein and its effects on cell apoptosis显示文摘Thioredoxin is a ubiquitous dithiol oxidoreductase found in many organisms and involved in numerous biochemical processes. Human thioredoxin-like protein (hTRXL) is differentially expressed at different development stages of human fetal cerebrum and belongs to an expanding family of thioredoxins. Recombinant hTRXL and truncated hTRXLs corresponding to the N-terminal (hTRXL-N) and C-terminal (hTRXL-C) domains are expressed and purified. In insulin disulfide reduction assay, both full-length hTRXL and hTRXL-N show reducing activity for the insulin disulfide bonds. As expected, the hTRXL-C failed to reduce insulin. 14Influence of inherent iron and oxygen concentrations on selenite sorption process using bentonite显示文摘The oxygen concentration and inherent Fe in bentonite have a significant influence on the Se(IV) sorption process.In this study,the sorption of selenite on natural bentonite was investigated using a batch experiment method,and the distribution coefficient(K_d)values were obtained in the pH range from 2.0 to 10.0 under oxic/anoxic conditions.The K_d values always reached a maximum value at a pH of 4 under oxic conditions and at a pH of 8 under anoxic conditions;meanwhile,the K_d value under anoxic conditions was larger than the value under oxic conditions,especially in regard to the maximum K_d values.The oxygen conditions have a significant influence on the ratio of redox-sensitive Fe^(2+)/Fe^(3+),which was closely related to the difference in the K_d values under both oxic/anoxic conditions.Ferric selenite and green rust could be responsible for the maximum K_d values under oxic/anoxic conditions.In the leaching experiments,we found that the Fe^(2+) in bentonite could replace Mg^(2+) and Al^(3+) in the octahedral sheet.Spectroscopy methods,such as Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD) and X-ray absorption spectroscopy(XAS) were used to characterize the surface properties of the samples after reaction.Overall,this study shows that the addition of Fe^(2+)-containing materials into backfill/buffer materials under anoxic condition could enhance the sorption of ^(79)Se(IV).Jiangang He Xueling Qiao Yanlin Shi Yao Li Xiaoyu Yang Wanqiang Zhou Chunli Liu 2017Science China Chemistry2017,60,9:0
15Stability of selenium and its speciation analysis in water using automatic system separation and HR-ICP-MS measurement显示文摘A simple and convenient method has been developed for the pre-concentration and separation of inorganic selenium species from environmental water samples using anion exchange chromatographic column combined with high resolution inductively coupled plasma mass spectrometry(HR-ICP-MS)measurement.^(75)Se(Ⅳ) and ^(75)Se(Ⅵ)were prepared and used as tracers during the experiments.The volatility of selenium during solution evaporation was investigated to establish a reliable water samples pretreatment procedure.The parameters which affect the uptake of Se(Ⅳ)and Se(Ⅵ)on Dowex1×8 resin was optimized and the procedure for Se(Ⅳ)and Se(Ⅵ)separation was proposed.Both Se(Ⅳ)and Se(Ⅵ)are retained on the column in natural or alkaline solution with high distribution coefficient.The successive gradient elution of pre-concentrated species of selenium with HNO_(3)solution allows to differentiate between them.Se(Ⅳ)and Se(Ⅵ)finally were eluted with 0.05 mol/L HNO_(3) and 5.0 mol/L HNO_(3),respectively.The proposed method has been successfully verified using the certified reference materials(CRMs)of real water samples,and spiked recoveries for real samples were 98%-104%with 5%relative standard deviations(RSDs).The developed procedure is proved to be reliable and can be used for the rapid determination of selenium species in environmental water samples.Junqiang Yang Yawen Chen Keliang Shi Kesheng Hu Run Li Xiaoqing Gao Qian Wang Weibo Zhang Yun Zhou Yanyun Wang Jiangang He Tonghuan Liu Xiaolin Hou 2022Chinese Chemical Letters2022,33,7:0
16Tannic acid coated single-wall carbon nanotube membranes for the recovery of Au from trace-level solutions显示文摘The efficient recovery of gold from industrial sewage is important for saving precious metals and remains a big challenge.We report the extraction of gold ions from a trace-level aqueous solution using a tannic acid(TA)coated single-wall carbon nanotube(SWCNT)film.The TA has many redox ligands that efficiently adsorb Au(III)from the solution and reduce them to Au particles.The interwoven SWCNTs not only act as a framework to improve the mechanical stability of the hybrid membrane,but also provide abundant paths for H_(2)O transport,and facilitate the full exposure of the TA.As a result,the hybrid membrane has an excellent ability to capture gold ions from solution with a high flux of 157 L/(m^(2)·h·bar),and an ultra-high adsorption capacity of 2095 mg/g from solutions with an extremely low gold concentration of 20 ppm.The adsorbed gold ions are reduced to Au particles,which can be easily collected by oxidation.The recovered Au nanoparticles on the TA–SWCNT hybrid film had a remarkable surface-enhanced Raman scattering effect that enabled the sensitive detection of rhodamine 6G.Chunmei Wang Peng-Xiang Hou Yiming Zhao Chao Shi Jiangang Zhang An-Ping Wu Chang Liu Hui-Ming Cheng 2023Nano Research2023,16,8:0
