1Study on the Thermal Ignition of Gasoline-air Mixture in Underground Oil Depots based on Experiment and Numerical Simulation显示文摘The study on the special phenomenon,occurrence process and control mechanism of gasoline-air mixture thermal ignition in underground oil depots is of important academic and applied value for enriching scientific theories of explosion safety,developing protective technology against fire and decreasing the number of fire accidents.In this paper,the research on thermal ignition process of gasoline-air mixture in model underground oil depots tunnel has been carried out by using experiment and numerical simulation methods.The calculation result has been demonstrated by the experiment data.The five stages of thermal ignition course,which are slow oxidation stage,rapid oxidation stage,fire stage,flameout stage and quench stage,have been firstly defined and accurately descried.According to the magnitude order of concentration,the species have been divided into six categories,which lay the foundation for explosion-proof design based on the role of different species.The influence of space scale on thermal ignition in small-scale space has been found,and the mechanism for not easy to fire is that the wall reflection causes the reflux of fluids and changes the distribution of heat and mass,so that the progress of chemical reactions in the whole space are also changed.The novel mathematical model on the basis of unification chemical kinetics and thermodynamics established in this paper provides supplementary means for the analysis of process and mechanism of thermal ignition.Ou Yihong Du Yang Jiang Xingsheng Wang dong Liang Jianjun 2010Journal of Thermal Science2010,19,2:6

4WIP1 promotes cancer stem cell properties by inhibiting p38 MAPK in NSCLC显示文摘Cancer stem cells(CSCs)are a small population of stem cell-like cancer cells that can initiate tumors in vivo,and are the major source of cancer initiation,relapse,and drug resistance.We previously reported that the p38 MAPK,through its downstream effectors MK2 and HSP27,suppressed CSC properties by downregulating the expression of transcription factors that mediate stemness in non-small-cell lung cancer(NSCLC)cells,and that despite unaltered total expression of total p38 proteins,the levels of activated p38 were reduced in NSCLC tissues.However,the mechanism underlying the reduced levels of activated p38 in NSCLC is unknown.In this study,we identified WIP1,a p38 phosphatase frequently overexpressed in cancer,as a suppressor of p38 in a pathway that regulates CSC properties in NSCLC.Increased WIP1 expression correlated with reduced levels of activated p38,and with increased levels of a CSC marker in NSCLC tissues.Further investigation revealed that WIP1 promoted stemness-related protein expression and CSC properties by inhibiting p38 activity in NSCLC cells.WIP1 inhibitors are currently under development as anticancer drugs based on their ability to reactivate p53.We found that a WIP1 inhibitor suppressed stemness-related protein expression and CSC properties by activating p38 in NSCLC cells in vitro and in vivo.These studies have identified the WIP1–p38–MK2–HSP27 cascade as a novel signaling pathway that,when altered,promotes CSC properties in NSCLC development,and have defined novel mechanisms underlying the oncogenic activity of WIP1 and the anticancer efficacy of WIP1 inhibitors.Kaiyuan Deng Liang Liu Xiaoming Tan Zhen Zhang Jianjun Li Yang Ou Xin Wang Shuang Yang Rong Xiang Peiqing Sun 2020Signal Transduction and Targeted Therapy2020,5,1:1

8Supplemental N-acyl homoserine lactonase alleviates intestinal disruption and improves gut microbiota in broilers challenged by Salmonella Typhimurium显示文摘Background Salmonella Typhimurium challenge causes a huge detriment to chicken production.N-acyl homoserine lactonase(AHLase),a quorum quenching enzyme,potentially inhibits the growth and virulence of Gram-negative bacteria.However,it is unknown whether AHLase can protect chickens against S.Typhimurium challenge.This study aimed to evaluate the effects of AHLase on growth performance and intestinal health in broilers challenged by S.Typhimurium.A total of 240 one-day-old female crossbred broilers(817C)were randomly divided into 5 groups(6 replicates/group):negative control(NC),positive control(PC),and PC group supplemented with 5,10 or 20 U/g AHLase.All birds except those in NC were challenged with S.Typhimurium from 7 to 9 days of age.All parameters related to growth and intestinal health were determined on d 10 and 14.Results The reductions(P<0.05)in body weight(BW)and average daily gain(ADG)in challenged birds were alleviated by AHLase addition especially at 10 U/g.Thus,samples from NC,PC and PC plus 10 U/g AHLase group were selected for further analysis.S.Typhimurium challenge impaired(P<0.05)intestinal morphology,elevated(P<0.05)ileal inflammatory cytokines(IL-1βand IL-8)expression,and increased(P<0.05)serum diamine oxidase(DAO)activity on d 10.However,AHLase addition normalized these changes.Gut microbiota analysis on d 10 showed that AHLase reversed the reductions(P<0.05)in several beneficial bacteria(e.g.Bacilli,Bacillales and Lactobacillales),along with increases(P<0.05)in certain harmful bacteria(e.g.Proteobacteria,Gammaproteobacteria,Enterobacteriaceae and Escherichia/Shigel a)in PC group.Furthermore,AHLase-induced increased beneficial bacteria and decreased harmful bacteria were basically negatively correlated(P<0.05)with the reductions of ileal IL-1βand IL-8 expression and serum DAO activity,but positively correlated(P<0.05)with the increased BW and ADG.Functional prediction revealed that AHLase abolished S.Typhimurium-induced upregulations(P<0.05)of certain pathogenicity-related pathways such as lipopolysaccharide biosynthesis,shigellosis,bacterial invasion of epithelial cells and pathogenic Escherichia coli infection of gut microbiota.Conclusions Supplemental AHLase attenuated S.Typhimurium-induced growth retardation and intestinal disruption in broilers,which could be associated with the observed recovery of gut microbiota dysbiosis.Weiwei Wang Jingseng Ou Hui Ye Qingyun Cao Changming Zhang Zemin Dong Dingyuan Feng Jianjun Zuo 2023Journal of Animal Science and Biotechnology2023,14,4:1

10Measurement method and results of divergence angle of laser-controlled solid propellants used in space propulsion显示文摘The plume divergence angle is an important reference index for evaluating the thrust efficiency and propellant utilization of space propulsion systems.However,the characteristics of the dynamic variation of plume divergence angle over time cannot be measured using current methods.This paper utilizes high-speed photography and image processing methods to develop a strategy that can give a quick,non-destructive and real-time detection of the divergence angle.Effectiveness of the strategy is verified,and the characteristics of plume divergence angles of different lasercontrolled solid propellants were further analyzed and fitted.The experimental results indicate that graphene could effectively reduce the divergence angle,while oxide-doped samples had larger divergence angles than alloy-doped and carbon-doped samples.Yang OU Jianjun WU Yuqiang CHENG Yu ZHANG Yuqi LI 2023Chinese Journal of Aeronautics2023,36,9:0
11Rare inherited missense variants of POGZ associate with autism risk and disrupt neuronal development显示文摘Excess de novo likely gene-disruptive and missense variants within dozens of genes have been identified in autism spectrum disorder(ASD)and other neurodevelopmental disorders.However,many rare inherited missense variants of these high-risk genes have not been thoroughly evaluated.In this study,we analyzed the rare missense variant burden of POGZ in a large cohort of ASD patients from the Autism Clinical and Genetic Resources in China(ACGC)and further dissected the functional effect of diseaseassociated missense variants on neuronal development.Our results showed a significant burden of rare missense variants in ASD patients compared to the control population(P=4.6×10-5,OR=3.96),and missense variants in ASD patients showed more severe predicted functional outcomes than those in controls.Furthermore,by leveraging published large-scale sequencing data of neurodevelopmental disorders(NDDs)and sporadic case reports,we identified 8 de novo missense variants of POGZ in NDD patients.Functional analysis revealed that two inherited,but not de novo,missense variants influenced the cellular localization of POGZ and failed to rescue the defects in neurite and dendritic spine development caused by Pogz knockdown in cultured mouse primary cortical neurons.Significantly,L1CAM,an autism candidate risk gene,is differentially expressed in POGZ deficient cell lines.Reduced expression of L1cam was able to partially rescue the neurite length defects caused by Pogz knockdown.Our study showed the important roles of rare inherited missense variants of POGZ in ASD risk and neuronal development and identified the potential downstream targets of POGZ,which are important for further molecular mechanism studies.Wenjing Zhao Jieqiong Tan Tengfei Zhu Jianjun Ou Ying Li Lu Shen Huidan Wu Lin Han Yanling Liu Xiangbin Jia Ting Bai Honghui Li Xiaoyan Ke Jingping Zhao Xiaobing Zou Zhengmao Hu Hui Guo Kun Xia 2019Journal of Genetics and Genomics2019,46,5:0
12Diagnostic Quantification of the Cloud Water Resource in China during 2000–2019显示文摘By using the diagnostic quantification method for cloud water resource(CWR),the three-dimensional(3D)cloud fields of 1°×1°resolution during 2000-2019 in China are firstly obtained based on the NCEP reanalysis data and related satellite data.Then,combined with the Global Precipitation Climatology Project(GPCP)products,a 1°×1°gridded CWR dataset of China in recent 20 years is established.On this basis,the monthly and annual CWR and related variables in China and its six weather modification operation sub-regions are obtained,and the CWR characteristics in different regions are analyzed finally.The results show that in the past 20 years,the annual total amount of atmospheric hydrometeors(GM_(h))and water vapor(GM_(v))in the Chinese mainland are about 838.1 and 3835.9 mm,respectively.After deducting the annual mean precipitation of China(P_(s),661.7 mm),the annual CWR is about 176.4 mm.Among the six sub-regions,the southeast region has the largest amount of cloud condensation(C_(vh))and precipitation,leading to the largest GM_(h) and CWR there.In contrast,the annual P_(s),GM_(h),and CWR are all the least in the northwest region.Furthermore,the monthly and interannual variation trends of P_(s),C_(vh),and GM_(h) in different regions are identical,and the evolution characteristics of CWR are also consistent with the hydrometeor inflow(Q_(hi)).For the north,northwest,and northeast regions,in spring and autumn the precipitation efficiency of hydrometeors(PEh)is not high(20%-60%),the renewal time of hydrometeors(RT_(h))is relatively long(5-25 h),and GM_(h) is relatively high.Therefore,there is great potential for the development of CWR through artificial precipitation enhancement(APE).For the central region,spring,autumn,and winter are suitable seasons for CWR development.For the southeast and southwest regions,P_(s) and PE_(h) in summer are so high that the development of CWR should be avoided.For different spatial scales,there are significant differences in the characteristics of CWR.Miao CAI Yuquan ZHOU Jianzhao LIU Yahui TANG Chao TAN Junjie ZHAO Jianjun OU 2022Journal of Meteorological Research2022,36,2:0
13USP51/PD-L1/ITGB1-deployed juxtacrine interaction plays a cell-intrinsic role in promoting chemoresistant phenotypes in non-small cell lung cancer显示文摘Background:Programmed death ligand 1(PD-L1)has been demonstrated to facilitate tumor progression and therapeutic resistance in an immuneindependent manner.Nevertheless,the function and underlying signaling network(s)of cancer cell-intrinsic PD-L1 action remain largely unknown.Herein,we sought to better understand how ubiquitin-specific peptidase 51(USP51)/PD-L1/integrin beta-1(ITGB1)signaling performs a cell-intrinsic role in mediating chemotherapeutic resistance in non-small cell lung cancer(NSCLC).Methods:Western blotting and flow cytometry were employed for PD-L1 detection in NSCLC cell lines.Coimmunoprecipitation and pulldown analyses,protein deubiquitination assay,tissue microarray,bioinformatic analysis and molecular biology methods were then used to determine the significance of PD-L1 in NSCLC chemoresistance and associated signaling pathways in several different cell lines,mouse models and patient tissue samples.Ubiquitin-7-amido-4-methylcoumarin(Ub-AMC)-based deubiquitinase activity,cellular thermal shift and surface plasmon resonance(SPR)analyses were performed to investigate the activity of USP51 inhibitors.Results:We provided evidence that cancer cell-intrinsic PD-L1 conferred the development of chemoresistance by directly binding to its membrane-bound receptor ITGB1 in NSCLC.At the molecular level,PD-L1/ITGB1 interaction subsequently activated the nuclear factor-kappa B(NF-κB)axis to elicit poor response to chemotherapy.We further determined USP51 as a bona fide deubiquitinase that targeted the deubiquitination and stabilization of the PD-L1 protein in chemoresistant NSCLC cells.Clinically,we found a significant direct relationship between the USP51,PD-L1 and ITGB1 contents in NSCLC patients with chemoresistant potency.The elevated USP51,PD-L1 and ITGB1 levels were strongly associated with worse patient prognosis.Of note,we identified that a flavonoid compound dihydromyricetin(DHM)acted as a potential USP51 inhibitor and rendered NSCLC cells more sensitive to chemotherapy by targeting USP51-dependent PD-L1 ubiquitination and degradation in vitro and in vivo.Conclusions:Together,our results demonstrated that the USP51/PD-L1/ITGB1 network potentially contributes to the malignant progression and therapeutic resistance in NSCLC.This knowledge is beneficial to the future design of advanced cancer therapy.Jianjun Li Xuechun Xiao Yang Ou Lixia Cao Min Guo Chunchun Qi Zhaoyang Wang Yuxin Liu Qiuying Shuai Hang Wang Peiqing Sun Yi Shi Guang Yang Shuang Yang 2023Cancer Communications2023,43,7:0

15CDK4/6 inhibition blocks cancer metastasis through a USP51-ZEB1-dependent deubiquitination mechanism显示文摘Tumor metastasis is the most common cause of cancer-related deaths,yet it remains poorly understood.The transcription factor zinc-finger E-box binding homeobox 1(ZEB1)is involved in the epithelial-to-mesenchymal transition(EMT)and plays a pivotal role in tumor metastasis.However,the underlying mechanisms of the posttranslational modification of ZEB1 remain largely unknown.Herein,we demonstrated that specific inhibition of CDK4/6 was able to block tumor metastasis of breast cancer by destabilizing the ZEB1 protein in vitro and in vivo.Mechanistically,we determined that the deubiquitinase USP51 is a bona fide target of CDK4/6.The phosphorylation and activation of USP51 by CDK4/6 is necessary to deubiquitinate and stabilize ZEB1.Moreover,we found a strong positive correlation between the expression of p-RB(an indicator of CDK4/6 activity),p-USP51 and ZEB1 in metastatic human breast cancer samples.Notably,the high expression of p-RB,p-USP51,and ZEB1 was significantly correlated with a poor clinical outcome.Taken together,our results provide evidence that the CDK4/6-USP51-ZEB1 axis plays a key role in breast cancer metastasis and could be a viable therapeutic target for the treatment of advanced human cancers.Zhen Zhang Jianjun Li Yang Ou Guang Yang Kaiyuan Deng Qiong Wang Zhaoyang Wang Wenhao Wang Quansheng Zhang Hang Wang Wei Sun Peiqing Sun Shuang Yang 2020Signal Transduction and Targeted Therapy2020,5,1:0
