| 1 | Tribological behaviors of some polymeric materials in sea water显示文摘Ocean tribology, a new research field of tribology, is being established and developing. The investigation on the tribology in sea water has important significance to ocean exploitation. In this paper, the tribological behaviors of UHMWPE, PTFE as well as Ekonol/PTFE, graphite/PTFE and CF/PTFE sliding against GCr15 steel and Ni-Cr-WC alloy in sea water were investigated, and compared with those in pure water. Results show that the main factors that affect the tribological behaviors of five polymeric materials in sea water are the corrosive effect and lubricating effect of sea water. When polymeric materials sliding against GCr15 in sea water, owing to serious corrosion of counterface by sea water, the surface roughness of the counterface increases largely. As a result, the lubricating effect of medium decreases sharply and the plowing effect of counterface increases greatly. So the friction coefficients and wear rates of five materials sliding against GCr15 in sea water are much larger than those under other conditions. Such wear model, depending on the corrosion of counterface by medium, can be called indirect corrosive wear. Under other three conditions, owing to the more superior lubricating effect of sea water than pure water and the better wettability of Ni-Cr-WC than GCr15, the friction coefficients and wear rates of five materials increase in the following order: sliding against Ni-Cr-WC in sea water < sliding against Ni-Cr-WC in pure water < sliding against GCr15 in pure water. | 4 | Design, synthesis and biological evaluation of chalcone analogues with novel dual antioxidant mechanisms as potential anti-ischemic stroke agents显示文摘Scavenging reactive oxygen species(ROS) by antioxidants is the important therapy to cerebral ischemia-reperfusion injury(CIRI) in stroke. The antioxidant with novel dual-antioxidant mechanism of directly scavenging ROS and indirectly through antioxidant pathway activation may be a promising CIRI therapeutic strategy. In our study, a series of chalcone analogues were designed and synthesized, and multiple potential chalcone analogues with dual antioxidant mechanisms were screened. Among these compounds, the most active 33 not only conferred cytoprotection of H2 O2-induced oxidative damage in PC12 cells through scavenging free radicals directly and activating NRF2/ARE antioxidant pathway at the same time, but also played an important role against ischemia/reperfusion-related brain injury in animals. More importantly, in comparison with mono-antioxidant mechanism compounds, 33 exhibited higher cytoprotective and neuroprotective potential in vitro and in vivo. | 5 | Correlation of Chlamydia pneumonias infection with primary biliary cirrhosis显示文摘AIM: To evaluate the association between Chlamydia pneumoniae (Cpn) infection and primary biliary cirrhosis (PBC).METHODS: Cpn IgG and IgM were determined by enzymelinked immunosorbent assay (ELISA) in 41 well-established PBC patients and two race-matched control groups (posthepatitis cirrhosis, n = 70; healthy controls, n = 57).RESULTS: The mean level and seroprevalence of Cpn IgG in PBC group and post-hepatitis cirrhosis (PHC) group were significantly higher than those in healthy controls (46.8±43.4 RU/mL, 49.5±45.2 RU/mL vs28.3±32.7 RU/mL;68.3%, 71.4%, 42.1%, respectively; P<0.05). There was a remarkably elevated seroprevalence of Cpn IgM in patients with PBC (22.0%) compared to the PHC and healthy control (HC) groups. For the PBC patients versus the HCs, the odds ratios (ORs) of the presence of Cpn IgG and IgM were 2.7 (95% CI 0.9-6.1) and 5.1 (95% CI 1.4-18.5), respectively. | 6 | The Full-length Genome Analysis of a Street Rabies Virus Strain Isolated in Yunnan Province of China显示文摘The epidemic of rabies has rapidly increased and expanded in Yunnan province in recent years. In order to further analyze and understand the etiological reasons for the rapid expansion of rabies in Yunnan, a strain of rabies virus CYN1009D in Yunnan was isolated, and the complete genomic sequencing was carried out, and the bioimfomative analysis on genes/encoded proteins and phylogeny with reference to sequences in GenBank was performed. The complete genome of CYN1009D was 11923 nt in length and belonged to genotype I. The genes encoding different structural proteins were all conserved in their lengths, in comparison to other strains in China. The amino acid sequence was conserved at different antigen sites of NP, but the variation was detected at the secondary phosphorylation site of position 375; variations were also detected in the phosphorylation sites at positions 63-63 and 162 of PP; the sites playing important roles in virus synthesis, budding and viral morphology in MP were conserved; two glycosylation sites were detected at Asn37 and Asn319 in GP, the neutralizing antigen sites in GP were conserved; the initial amino acid of LP (ML) was different from that of most of the strains in China (MM); the variations in G-L region in the intergenic region were significant. | 7 | RELATIONSHIP BETWEEN ENDOTHELIAL DYSFUNCTION AND SERUM HOMOCYSTEINE IN PATIENTS WITH CORONARY LESIONS显示文摘Objective To investigate the relationship between vascular endothelial dysfunction and serum homocysteine (HCY) level in patients with coronary lesions. Methods Serum HCY, serum nitric oxide (NO), plasma endothelin-1 (ET-1), and circulation endothelial cell (CEC) were measured in 76 patients who received coronary angiography. Fifty-four patients with a stenosis of 50% or more at least in one coronary atery were as coronary artery disease (CAD) group. Other 22 cases with no recognizable plaque and/or stenosis were as control group. HCY level was detected using an enzyme immunoassay kit. NO concentration was measured using a nitrate reductase kit. Radio-immunoassay was applied to analyse the ET-1 level, and CEC was measured by flow cytometry. Results The levels of HCY, ET-1, and CEC in patients with coronary lesions were significantly increased in comparison with control group (P < 0.01), while NO level in CAD group was significantly lower compared with that in control (P < 0.01). Using a multivariate stepwise regression analysis, HCY level had a positive correlation with ET-1 level (r = 0.420, P < 0.05) and CECs number (r = 0.423, P < 0.05); and had a negative correlation with NO/ET-1 (r = -0.403, P < 0.05). But there was no significant correlation between HCY and NO levels. Conclusions HCY might lead to endothelial cell injury, which would provide a plausible mechanism for the relationship between hyperhomocysteinemia and development of coronary artery disease. | 9 | Genesis and Implications of the Composition and Sedimentary Structure of Fine-Grained Carbonate Rocks in the Shulu Sag显示文摘Fine-grained carbonate rocks, which extensively occur in the Eocene strata in the Shulu sag, Bohai Bay Basin, North China, represent an unconventional, fine-grained carbonate reservoir. However, previous studies have ignored the complexity of the lithofacies components and their formation mechanisms. Fine-grained carbonate rocks are typical reservoirs in which hydrocarbons form and gather. A better understanding of the nature of these rocks is extremely important for evaluating the quality of unconventional, fine-grained carbonate reservoirs. Various lithofacies components were discriminated in this study with a combination of petrographic observations and carbon isotope analyses. These finegrained carbonate rocks comprise terrigenous, biogenic and diagenetic materials. Terrigenous input and biologically induced precipitation are the main sources of the materials in the lake. Five lithofacies were identified based on the observations of sedimentary features(core and thin section) and mineralogical data:(1) varve-like laminated calcilutite,(2) graded laminated calcilutite,(3) interlaminated calcisiltitecalcilutite,(4) massive calcilutite, and(5) massive calcisiltite-calcarenite. Their origins were recorded by various lithofacies components, which are controlled by the interactions of physical, chemical and biological processes. This study indicated that the lithology of the bedrocks was the key factor controlling carbonate accumulation. The tectonics and climate can influence the weathering and types of lithofacies. Primary productivity controlled the precipitation of the endogenic calcite. The discrete element method (DEM) was used for the simulations. The terms stratification and penetration are defined and the change in fine particle concentration is discussed. Mathematical models relating fine particle ratio to time are established using the least squares method. The effect of structural parameters on fine particle ratio is analyzed. Stratification and penetration rate are discussed by considering the time derivative of the fine particle ratio. | 12 | Tribocorrosion behavior of nickel-aluminium bronze sliding against alumina under the lubrication by seawater with different halide concentrations显示文摘The tribocorrosion failure mechanism of nickel-aluminium bronze(NAB) in different halide concentrations of seawater was studied using a pin-on-disc tribometer that was modified to conduct in-situ electrochemical detection during the sliding process. It has been reported that high-halide-concentration seawater provided a good lubricating effect, and thus reduced the coefficient of friction and wear rate of NAB during the tribocorrosion process. However, the existence of halide ions corroded the passive film and hindered the repassivation of the damaged areas in the wear track, resulting in an increased corrosion rate. In addition, the morphology of the wear scar revealed the occurrence of abrasive, delamination, and adhesive wear of NAB in seawater. For the whole range of halide concentration values, a positive synergy between wear and corrosion was proven, and the quantification of this synergy was discussed in detail. | 13 | Molecular epidemiological study on pre-X region of hepatitis B virus and identification of hepatocyte proteins interacting with whole-X protein by yeast two-hybrid显示文摘AIM: To identify the pre-X region in hepatitis B virus (HBV)genome and to study the relationship between the genotype and the pre-X region. To investigate the biological function of whole-X (pre-X plus X) protein, we performed yeast two-hybrid to screen proteins in liver interacting with whole-X protein.METHODS: The pre-X region of HBV was amplified by polymerase chain reaction (PCR) method, and was cloned to pGEM Teasy vector. After the target region was sequenced, Vector 8.0 software was used to analyze the sequences. The whole-X bait plasmid was constructed by using yeast two-hybrid system 3. Yeast strain AH109 was transformed. After expression of the whole-X protein in AH109 yeast strains was proved, yeast two-hybrid screening was performed by mating AH109 with Y187 containing liver cDNA library plasmid. The mated yeast was plated on quadruple dropout medium and assayed for α-gal activity. The interaction between whole-X protein and the protein obtained from positive colonies was further confirmed by repeating yeast two-hybrid. After extracting and sequencing of plasmid from blue colonies, we carried out analysis by bioinformatics. RESULTS: After sequencing, 27 of 45 clones (60%) were found encoding the pre-X peptide. Eighteen of twenty-seven clones (66.7%) of pre-X coding sequences were found from genotype C. Five positive colonies that interacted with whole-X protein were obtained and sequenced; namely, fetuin B, UDP glycosyltransferase 1 family-polypeptide A9, mannose-P-dolichol utilization defect 1, fibrinogen-B beta polypeptide, transmembrane 4 superfamily member 4CD81 (TM4SF4).CONCLUSION: The pre-X gene exists in HBV genome.Genes of proteins interacting with whole-X protein in hepatocytes were successfully cloned. | 14 | Transactivating effect of hepatitis C virus core protein: A suppression subtractive hybridization study显示文摘AIM: To investigate the transactivating effect of hepatitis C virus (HCV) core protein and to screen genes transactivated by HCV core protein. METHODS: pcDNA3.1(-)-core containing full-length HCV core gene was constructed by insertion of HCV core gene into EcoRI/BarnHI site. HepG2 cells were cotransfected with pcDNA3.1(-)-core and pSV-lacZ. After 48 h, cells were collected and detected for the expression of β-gal by an enzyme-linked immunosorbent assay (ELISA) kit. HepG2 cell swere transiently transfected with pcDNA3.1(-)-core using Upofectamine reagent. Cells were collected and total mRNA was isolated. A subtracted cDNA library was generated and constructed into a pGEM-Teasy vector. The library was amplified with E. coil strain JM109. The cDNAs were sequenced and analyzed in GenBank with BLAST search after polymerase chain reaction (PCR). RESULTS: The core mRNA and protein could be detected in HepG2 cell lysate which was transfected by the pcDNA3.1(-)-core. The activity of β-galactosidase in HepG2 cells transfected by the pcDNA3.1(-)-core was 5.4 times higher than that of HepG2 cells transfected by control plasmid. The subtractive library of genes transactivated by HCV core protein was constructed successfully. The amplified library contained 233 positive clones. Colony PCR showed that 213 clones contained 100-1 000 bp inserts. Sequence analysis was performed in 63 clones. Six of the sequences were unknown genes. The full length sequences were obtained with bioinformatics method, accepted by Genl3ank. It was suggested that six novel cDNA sequences might be target genes transactivated by HCV core protein. CONCLUSION: The core protein of HCV has transactivating effects on SV40 early promoter/enhancer. A total of 63 clones from cDNA library were randomly chosen and sequenced. Using the BLAST program at the National Center for Biotechnology Information, six of the sequences were unknown genes. | 15 | Human Prolactin Improves Engraftment and Reconstitution of Human Peripheral Blood Lymphocytes in SCID Mice显示文摘Recombinant human prolactin (rhPRL) was administered to huPBL-SCID mice to determine its effects on human immunologic reconsfitution and function. The huPBL-SCID mice were given 10 μg I.p. Injection of rhPRL every other day for a total of 10 injections after huPBL were transferred. The results demonstrated that rhPRL improved the engraftment of lymphocytes into thymus, lymph nodes and spleens, showing that the cellularities of these organs increased although the cellularities tended to vary depending on the donor. The amounts of human T cells (HLA-ABC+/CD3+) increased greatly in thymus (14.2 folds), spleen (4.16 folds) and lymph nodes (40.18 folds) after rhPRL injections. The amounts of human B cells (HLA-ABC+/CD19+) also increased greatly in lymph nodes (42.5 folds) and spleen (5.78 folds). The lymph node cells from the rhPRL-treated huPBL-SCID mice were more sensitive to PHA stimulation ([3H] thymidine incorporation). The supernatant of PHA-stimulated PBL from rhPRL-treated huPBL/SCID chimerism contained more cytokines (IFN-γ and IL-2). The natural cytotoxicity against human sensitive target cells, K562 cells, from spleen and bone marrow of hPBL/SCID chimerism was significantly enhanced by rhPRL administration. The lymph node cells were stimulated with LPS in vitro for 3 days and the lymphocytes from the rhPRL-treated huPBL-SCID mice were more sensitive to mitogen stimulation. Both serum total IgG level and IgM level of rhPRL-treated huPBL/SCID chimerism were increased, and even without DT-rechallenge the base line of DT-specific IgG was elevated after rhPRL treatment in huPBL-SCID mice. | 16 | Key Elements Controlling Oil Accumulation within the Tight Sandstones显示文摘Tight oil sandstone reservoirs with low porosity and permeability, which are an unconventional petroleum resource, have been discovered in the Jurassic intervals of the central Junggar Basin, the northwestern China. To reveal the accumulation mechanism, a relatively comprehensive research was conducted, including oil-source correlation, porosity evolution, and hydrocarbon charging history. The results show that crude oil of these tight sandstone reservoirs were mainly from Permian source rocks with some contribution from Jurassic source rocks. The reservoirs were buried at shallow depth(<3 100 m) and exposed to weak diagenesis, and thus had high porosity(18.5%) when the Permian-sourced oil from Permian source rock was charging, indicating high GOI values(>5%). In contrast, the sandstone reservoir had already become tight and did not provide available space to accumulate oil due to severe compaction and cementation when hydrocarbon from Jurassic source rock filled, evidenced by low GOI values(<5%). Therefore, reservoir porosity controls the oil accumulation within tight sandstone. Whether tight sandstone reservoirs accumulate oil depends on the reservoir quality when hydrocarbons charge. | 17 | Transglutaminase 3 expression in C57BL/6J mouse embryo epidermis and the correlation with its differentiation显示文摘Epidermal-type transglutaminase 3 (TGM3) is involved in the cross-linking of structural proteins to form the cornifiedenvelope in the epidermis. In the present study, we detected the expression of TGM3 in the mouse embryo using RT-PCR.TGM3 mRNA is weakly presented from E11.5 to E14.5 and increases significantly from E15.5 to birth. Then wedetermined the spatial and temporal expression pattern of TGM3 in the skin and other organs by in situ hybridization. Wefound a deprivation of TGM3 in skin at E11.5, while a rich supply in periderm cells and a weak expression in basal cellsfrom E12.5 to E14.5. From the period of E15.5 to E16.5, after keratinization in the epidermis, TGM3 was expressed inthe granular and cornified layers. The electron microscopic observation of the C57BL/6J mouse limb bud skin develop-ment provided several morphological evidences for the epidermal differentiation. | 19 | Thermodynamic properties and mixing thermodynamic parameters of two-phase metallic melts显示文摘Based on the calculating model of metallic melts involving eutectic, the calculating equations of mixing thermodynamic parameters for two phase metallic melts have been formulated in the light of those equations of homogeneous solutions. Irrespective as to whether the activity deviation relative to Raoultian behavior is positive or negative, or the deviation is symmetrical or unsymmetrical, the evaluated results not only agree well with experimental values, but also strictly obey the mass action law. This testifies that these equations can authentically reflect the structural reality and mixing thermodynamic characteristics of two-phase metallic melts. | 20 | High-throughput sequencing analysis of the regulation of intestinal flora in giant pandas with indigestion using a probiotic agent LyPB显示文摘This study aimed to investigate the eff ect of LyPB on the intestinal microfl ora of giant pandas with indigestion,using high-throughput sequencing(HTS)technology.The species distribution and microfl oral density and diversity before and after administration of the LyPB probiotic agent were analyzed.LyPB evidently has the ability to adjust the fl oral imbalance in the panda’s intestine.To test the eff ects of LyPB on the microfl ora of the panda gut,fecal samples were taken from a healthy giant panda(Anan)without administration of LyPB and from a dyspeptic giant panda Yangyang before and after LyPB administration.Compared with the sample obtained from healthy Anan(anan-c)and that obtained from dyspeptic Yangyang before LyPB administration(yangyang1),the sample taken from Yangyang(yangyang2)after LyPB administration displayed a signifi cant increase in the operational taxonomic unit index.An increase in the Chao index indicated an increase in the microfl oral richness,while an increase in the Shannon index indicated an increase in microfl oral diversity.At phylum and genus levels,a signifi cant increase was observed in the density of probiotic bacteria of phylum fi rmicutes,genus Streptococcus,while a drastic reduction in the density of Escherichia coli/Escherichia coli Shigella/bacteria of genus Shigella was observed.Data obtained in this study shows that LyPB preparations successfully improve the microbial structure within the panda’s intestinal canal by signifi cantly increasing the eff ective microbial community and decreasing the number of pathogenic microbes. | Yu Zhang Zhi Zhang Jianzhang Ma Bo Luo Guiquan Zhang Guocai Zhang Kexin Yang Gang Wei | 2020 | Journal of Forestry Research2020,31,6: | 3 |
