1Effect of artificial liver support system on patients with severe vira hepatitis:A study of four hundred cases显示文摘AIM: To assess the effect of artificial liver support system(ALSS) on patients with severe viral hepatitis, who were divided into treatment group and control group. METHODS: Four hundred in-hospital patients enrolled during 1995-2003 who received ALSS therapy were studied as the treatment group. Four hundred in-hospital patients enrolled during 1986-1994 who received other medical therapies served as the control group. The methods of ALSS used included plasma exchange, hemoperfusion, hemofiltration, continuous hemodiafiltration (CHDF). The effect of ALSS treatment was studied in patients at different stages of the disease.RESULTS: The cure rate of acute and subacute severe hepatitis in the treatment group was 78.9% (30/38), and was 11.9% (5/42) in the control group. The improved rate of chronic severe hepatitis in the treatment group was 43.4% (157/362), and was 15.4% (55/358) in the control group. We found that patients treated with ALSS in the early or middle stage of the disease had much higher survival rates than patients in the end stage of the disease. 2Molecular epidemiological study on pre-X region of hepatitis B virus and identification of hepatocyte proteins interacting with whole-X protein by yeast two-hybrid显示文摘AIM: To identify the pre-X region in hepatitis B virus (HBV)genome and to study the relationship between the genotype and the pre-X region. To investigate the biological function of whole-X (pre-X plus X) protein, we performed yeast two-hybrid to screen proteins in liver interacting with whole-X protein.METHODS: The pre-X region of HBV was amplified by polymerase chain reaction (PCR) method, and was cloned to pGEM Teasy vector. After the target region was sequenced, Vector 8.0 software was used to analyze the sequences. The whole-X bait plasmid was constructed by using yeast two-hybrid system 3. Yeast strain AH109 was transformed. After expression of the whole-X protein in AH109 yeast strains was proved, yeast two-hybrid screening was performed by mating AH109 with Y187 containing liver cDNA library plasmid. The mated yeast was plated on quadruple dropout medium and assayed for α-gal activity. The interaction between whole-X protein and the protein obtained from positive colonies was further confirmed by repeating yeast two-hybrid. After extracting and sequencing of plasmid from blue colonies, we carried out analysis by bioinformatics. RESULTS: After sequencing, 27 of 45 clones (60%) were found encoding the pre-X peptide. Eighteen of twenty-seven clones (66.7%) of pre-X coding sequences were found from genotype C. Five positive colonies that interacted with whole-X protein were obtained and sequenced; namely, fetuin B, UDP glycosyltransferase 1 family-polypeptide A9, mannose-P-dolichol utilization defect 1, fibrinogen-B beta polypeptide, transmembrane 4 superfamily member 4CD81 (TM4SF4).CONCLUSION: The pre-X gene exists in HBV genome.Genes of proteins interacting with whole-X protein in hepatocytes were successfully cloned. 3Transactivating effect of hepatitis C virus core protein: A suppression subtractive hybridization study显示文摘AIM: To investigate the transactivating effect of hepatitis C virus (HCV) core protein and to screen genes transactivated by HCV core protein. METHODS: pcDNA3.1(-)-core containing full-length HCV core gene was constructed by insertion of HCV core gene into EcoRI/BarnHI site. HepG2 cells were cotransfected with pcDNA3.1(-)-core and pSV-lacZ. After 48 h, cells were collected and detected for the expression of β-gal by an enzyme-linked immunosorbent assay (ELISA) kit. HepG2 cell swere transiently transfected with pcDNA3.1(-)-core using Upofectamine reagent. Cells were collected and total mRNA was isolated. A subtracted cDNA library was generated and constructed into a pGEM-Teasy vector. The library was amplified with E. coil strain JM109. The cDNAs were sequenced and analyzed in GenBank with BLAST search after polymerase chain reaction (PCR). RESULTS: The core mRNA and protein could be detected in HepG2 cell lysate which was transfected by the pcDNA3.1(-)-core. The activity of β-galactosidase in HepG2 cells transfected by the pcDNA3.1(-)-core was 5.4 times higher than that of HepG2 cells transfected by control plasmid. The subtractive library of genes transactivated by HCV core protein was constructed successfully. The amplified library contained 233 positive clones. Colony PCR showed that 213 clones contained 100-1 000 bp inserts. Sequence analysis was performed in 63 clones. Six of the sequences were unknown genes. The full length sequences were obtained with bioinformatics method, accepted by Genl3ank. It was suggested that six novel cDNA sequences might be target genes transactivated by HCV core protein. CONCLUSION: The core protein of HCV has transactivating effects on SV40 early promoter/enhancer. A total of 63 clones from cDNA library were randomly chosen and sequenced. Using the BLAST program at the National Center for Biotechnology Information, six of the sequences were unknown genes. 8Water quality,natural chemical weathering and ecological risk assessment of the contaminated area of vanadium ore in Yinhua River,China:Evidence from major ions and trace elements显示文摘There are abundant vanadium ores in the Cambrian strata in southern Shaanxi,China.Many years of mining activities and surface leaching have polluted the surface water to a certain extent,but the researches on the water quality characteristics and pollution degree are relatively weak.This contribution was organized to investigate the surface water quality by general parameters,including TDS,Eh,pH,DO,TOC,COD,and EC,in the vanadium ore belt(Yinhua River basin).Major ions were determined to detect the water type and natural chemical weathering,while trace elements were used to illustrate their geochemical characteristics and ecological risk assessment of heavy metals.The study found that the surface water was weakly alkaline and mainly dominated from normal to pool grade.The particle size with1000–10,000 nm of suspended particles was the main carrier of organic matter.The concentration of HCO_(3)^(-)and SO_(4)^(2-)in the anions and Ca^(2+)and Mg^(2+)ions in the cations were relatively high,and the water type was Ca-Mg-HCO_(3)-SO_(4) type.Rock weathering had a great influence on surface water,and the weathering products were mainly silicate and carbonate.Compared with the Type river,the contents of V elements showed an obvious positive anomaly,which may be affected by mining activities of vanadium ore and the annual leaching of the tailings pond.As and Cd in the surface water was polluted seriously.The integrated pollution index suggested that the surface water pollution was serious,and the main stream was more serious with the increasing tailings ponds.Delu Li Yong Xu Xiaotuan Zhang Zhidong Yang Shaofei Wang Qianyang He Zhe Jia 2022Acta Geochimica2022,41,1:0

10Cloning and identification of NS5A TP2gene and its spliced variant transactivated by hepatitis C virus non-structural protein 5A显示文摘AIM: To clone, identify and study new NSSATP2 gene and its spliced variant transactivated by hepatitis C virus nonstructural protein 5A. METHODS: On the basis of subtractive cDNA library of genes transactivated by NS5A protein of hepatitis C virus, the coding sequence of new gene and its spliced variant were obtained by bioinformatics method. Polymerase chain reaction (PCR) was conducted to amplify/VSSATP2 gene. RESULTS: The coding sequence of a new gene and its spliced variant were cloned and identified successfully. CONCLUSION: A new gene has been recognized as the new target transactivated by HCV NS5A protein. 11Jadomycin B,an Aurora-B kinase inhibitor discovered through virtual screening显示文摘Aurora kinases are clearly implicated in tumorgenesis and have emerged as promising targets for cancer therapy in recent years.In a virtual screening attempt,22 compounds were identified from nearly 15,000 microbial natural products as potential small-molecule inhib- itors of human Aurora-B kinase.When tested in yeast models,2 compounds(one is Jado- mycin B)showed preferential inhibition of ipl1-321(yeast Aurora kinase temperature sen- sitive mutant)than wild type yeast cell,suggesting these compounds are true Aurora kinase inhibitors.Further in vitro biochemical assay using purified recombinant Aurora-B kinase showed Jadomycin B inhibits Aurora-B activity in a dose-dependent fashion while two Jadomycin congeners,Jadomycin S and T,showed no activity.The marked differences in activities of Jadomycins were explained by docking analyses.Our results also showed that Jadomycin B competes with ATP for the kinase active center,which is consistent with our docking prediction.Like other Aurora kinase inhibitors,Jadomycin B also reduced the phos- phorylation of histone H3 on Ser10 in vivo.Cell culture based tests of Jadomycin B activity confirmed previous results that Jadomycin B could induce apoptosis of tumor cells without blocking cell cycle.Above results indicate that Jadomyicn B is a new Aurora B kinase inhibi- tor worthy of further investigation.The SAR(structure and activity relationship)analysis in- dicated that groups attached to the oxazolone ring of Jadomycin were important for its ability to enter the catalytic cleft and occupy the active site to inhibit Aurora-B kinase.Jadomycin offers an ideal scaffold to generate novel derivatives with a hydrophobic side chain on their oxazolone rings,from which we may find Aurora-B inhibitors with improved activity.Da Huafu Jing Zhang Jian Tingzheng Yan Li Ke Qianyang Yan Changwang Shu Yisi 2008癌症进展2008,6,3:0
