| 1 | Multi-omics analyses of 398 foxtail millet accessions reveal genomic regions associated with domestication,metabolite traits,and antiinflammatory effects显示文摘Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor humans.To evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet accessions.We identified hundreds of common variants that influence numerous secondary metabolites.We observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail millet.Furthermore,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous phytohormones.Using CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand quality.Interestingly,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory effects.Taken together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite profile.The datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement. | Xukai Li Jianhua Gao Jingyi Song Kai Guo Siyu Hou Xingchun Wang Qiang He Yanyan Zhang Yakun Zhang Yulu Yang Jiaoyan Tang Hailang Wang Staffan Persson Mingquan Huang Lishuai Xu Linlin Zhong Dongqin Li Yongming Liu Hua Wu Xianmin Diao Peng Chen Xiaowen Wang Yuanhuai Han | 2022 | Molecular Plant2022,15,8: | 8 |
| 2 | The IL-33/ST2 axis affects tumor growth by regulating mitophagy in macrophages and reprogramming their polarization显示文摘Objective:Macrophages are a major component of the tumor microenvironment.M1 macrophages secrete pro-inflammatory factors that inhibit tumor growth and development,whereas tumor-associated macrophages(TAMs)mainly exhibit an M2 phenotype.Our previous studies have shown that the interleukin-33/ST2(IL-33/ST2)axis is essential for activation of the M1 phenotype.This study investigates the role of the IL-33/ST2 axis in TAMs,its effects on tumor growth,and whether it participates in the mutual conversion between the M1 and M2 phenotypes.Methods:Bone marrow-derived macrophages were extracted from wildtype,ST2 knockout(ST2-/-),and Il33-overexpressing mice and differentiated with IL-4.The mitochondrial and lysosomal number and location,and the expression of related proteins were used to analyze mitophagy.Oxygen consumption rates and glucose and lactate levels were measured to reveal metabolic changes.Results:The IL-33/ST2 axis was demonstrated to play an important role in the metabolic conversion of macrophages from OXPHOS to glycolysis by altering mitophagy levels.The IL-33/ST2 axis promoted enhanced cell oxidative phosphorylation,thereby further increasing M2 polarization gene expression and ultimately promoting tumor growth(P<0.05)(Figure 4).This metabolic shift was not due to mitochondrial damage,because the mitochondrial membrane potential was not significantly altered by IL-4 stimulation or ST2 knockout;however,it might be associated with the m TOR activity.Conclusions:These results clarify the interaction between the IL-33/ST2 pathway and macrophage polarization,and may pave the way to the development of new cancer immunotherapies targeting the IL-33/ST2 axis. | Huadan Xu Dong Li Jiaoyan Ma Yuanxin Zhao Long Xu Rui Tian Yanan Liu Liankun Sun Jing Su | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |
| 3 | Efficacy and safety of repetitive transcranial magnetic stimulation for generalised anxiety disorder: A metaanalysis显示文摘Background Pharmacological and conventional nonpharmacological treatments are only moderately effective in treatingeneralised anxiety disorder(GAD).Recently,repetitive transcranial magnetic stimulation(rTMS)has attracted interest because of its potential therapeutic value.Aim To investigate the efficacy and safety of rTMS treatment for GAD.Methods Literature studies published in English or Chinese were screened in 10 electronic databases up to 5 December 2018.The included studies'bias risk was assessed using Cochrane risk of bias assessment tool.Meta-analysis was performed to compute the standardised mean difference(SMD)and risk ratio(RR)along with its 95%Cis through using RevMan V.5.3.Heterogeneity was inspected by I2 and the χ2 test.We performed subgroup analysis and meta-regression to investigate heterogeneity.We used funnel plot to assess publication bias.We used the GRADE approach to assess the whole quality of evidence.Results Twenty-one studies,with a total sample size of 1481,were analysed.The risk of bias in most studies included is moderate,the majority of which are lacking of blinding methods of treatment allocation.The treatment had beneficial effects in the rTMS group compared with the control group in mean anxiety score(SMD=-0.68;95%Cl-0.89 to-0.46).None of the 21 studies included here reported severe adverse events.As for dropout rates,there are no statistically significant differences between the two groups(RR 1.14,95%Cl 0.72 to 1.82)or adverse events(RR 0.95,95%Cl 0.77 to 1.18).No particular influence on the heterogeneity of any variable was observed.The risk of publication bias was low.According to the GRADE approach,the evidence levels of primary outcome(treatment effects)and secondary outcomes(acceptability and safety)were rated as‘medium’.Conclusion The use of rTMS combined with medication treatment may have a significant positive anti-anxiety effect on patients with GAD.However,we should interpret the results cautiously due to the relatively high heterogeneity of the meta-analysis.Future high-quality clinical trials are needed to confirm our results. | Huiru Cui Lijuan Jiang Yanyan Wei Wei Li Hui Li Junjuan Zhu Jiaoyan Pang Jijun Wang Chunbo Li | 2019 | General Psychiatry2019,32,5: | 5 |

| 5 | Altered heartbeat perception sensitivity associated with brain structural alterations in generalised anxiety disorder显示文摘Background Palpitation is a common complaint in generalised anxiety disorder(GAD).Brain imaging studies have Investigated the neural mechanism of heartbeat perception in healthy volunteers.This study explored the neuroanatomical differences of altered heartbeat perception in patients with GAD using structural MRI.Aims Based on the strong somatic-interoceptive symptoms in GAD,we explored the regional structural brain abnormalities involved in heartbeat perception in patients with GAD.Methods This study was applied to the a priori regions using neuroanatomical theories of heartbeat perception,including the insula,anterior cingulate cortex,supplementary motor area and prefrontal cortex.A total of 19 patients with GAD and 19 healthy control subjects were enrolled.We used the FMRIB Software Library voxel-based morphometry software for estimating the grey matter volume of these regions of interest and analysed the correlation between heartbeat perception sensitivity and the volume of abnormal grey matter.Results Patients with GAD showed a significantly decreased volume of grey matter in their left medial prefrontal cortex,right orbital frontal cortex and anterior cingulate cortex.The grey matter volume of the left medial prefrontal cortex negatively correlated with heartbeat perception sensitivity in patients with GAD.Conclusions It should be the first study that shows heartbeat perception is associated with brain structure in GAD.Our findings suggest that the frontal region may play an important role in aberrant heartbeat perception processing in patients with GAD,and this may be an underlying mechanism resulting in the abnormal cardiovascular complaints in GAD.This is hypothesised as a'top-down'deficiency,especially in the medial prefrontal cortex.This will provide the foundation for a more targeted region for neuromodulation intervention in the future. | Hui Li Bin Zhang Qiang Hu Lanlan Zhang Yi Jin Jijun Wang Huiru Cui Jiaoyan Pang Chunbo Li | 2020 | General Psychiatry2020,33,1: | 2 |
