| 1 | Engineered targeting tLyp-1 exosomes as gene therapy vectors for efficient delivery of siRNA into lung cancer cells显示文摘Natural exosomes can express specific proteins and carbohydratemolecules on the surface and hence have demonstrated the great potentials for gene therapy of cancer.However,the use of natural exosomes is restricted by their low transfection efficiency.Here,we report a novel targeting tLyp-1 exosome by gene recombinant engineering for delivery of siRNA to cancer and cancer stem cells.To reach such a purpose,the engineered tLyp-1-lamp2b plasmids were constructed and amplified in Escherichia coli.The tLyp-1-lamp2b plasmids were further used to transfect HEK293T tool cells and the targeting tLyp-1 exosomes were isolated from secretion of the transfected HEK293T cells.Afterwards,the artificially synthesized siRNA was encapsulated into targeting tLyp-1 exosomes by electroporation technology.Finally,the targeting siRNA tLyp-1 exosomes were used to transfect cancer or cancer stem cells.Results showed that the engineered targeting tLyp-1 exosomes had a nanosized structure(approximately 100 nm)and high transfection efficiency into lung cancer and cancer stem cells.The function verifications demonstrated that the targeting siRNA tLyp-1 exosomes were able to knock-down the target gene of cancer cells and to reduce the stemness of cancer stem cells.In conclusion,the targeting tLyp-1 exosomes are successfully engineered,and can be used for gene therapy with a high transfection efficiency.Therefore,the engineered targeting tLyp-1 exosomes offer a promising gene delivery platform for future cancer therapy. | Jing Bai Jialun Duan Rui Liu Yafei Du Qian Luo Yinuo Cui Zhanbo Su Jiarui Xu Ying Xie Wanliang Lu | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,4: | 5 |

| 3 | RNA-binding protein QKI regulates contact inhibition via Yes-associate protein in ccRCC显示文摘Contact inhibition adjusts organ size to the proper size and ensures the cultured cells growing to a monolayer.By regulating the downstream coordinator YAP,the evolutionarily conserved Hippo transduction pathway attunes cell growth and death in response to cell contact inhibition,polarity,self-renewal,and differentiation.Dysregulation of this pathway is involved in various diseases such as cancer.RNA-binding protein QKI regulates cell proliferation,metabolism,division,and immunity in various cancer models,but its role in cancer cell contact inhibition remains unclear.In this study,we aimed to clarify the relationship between QKI and YAP,and the role of their interaction in cell contact inhibition.We found a lower QKI expression level in sparse condition,whereas a higher expression level in confluent condition by western blot analysis and immunofluorescence assay.QKI knockdown elevated cell proliferation and invasion both in vitro and in vivo.Strikingly,the results of CCK-8 assay,colony formation assay,and transwell assay showed that the phenomenon was in accord with the expression level of pYAP and reverse with YAP.Higher levels of Wnt3a and β-catenin were also found in xenografts of QKI-knockdown clear cell renal cell carcinoma (ccRCC) CAKI-1 cells by western blot analysis and immumohistochemical staining.Finally,a positive correlation between QKI and pYAP was found in clinical specimens by immunohistochemistry.Thus,as a negative regulator of YAP,QKI attuned the cell contact inhibition,leading to inhibition of cancer cell proliferation and invasion through Wnt and GPCR pathway. | Zheng Zhu Di Wei Xi'an Li Fuli Wang Fei Yan Zibao Xing Zhao Yan Huanyu Lu Dongsheng Zhai Ziehen Ye Geng Zhang Ping Meng Yu Zheng Jiarui Yuan Zifan Lu Jianlin Yuan | 2019 | Acta Biochimica et Biophysica Sinica2019,51,1: | 2 |

| 6 | Single-crystalline Mn-based oxide as a high-rate and long-life cathode material for potassium-ion battery显示文摘Mn-based oxides are promising cathode materials for potassium-ion batteries due to their high theoretical ca-pacity and abundant raw materials.However,the anisotropic properties of their conventional polycrystalline structures lead to insufficient rate capability and cycle life.Here,a single-crystal Mn-based layered oxide,P3′-type K_(0.35)Mn_(0.8)Fe_(0.1)Cu_(0.1)O_(2)(KMFCO),is designed and synthesized through a bimetallic co-induction effect and used as a cathode for potassium-ion battery.Benefiting from a unique single-crystal structure that is devoid of grain boundaries,it achieves a higher Kþtransport rate and a reduced volume change during the Kþintercalation/deintercalation process.Accordingly,the single-crystal P3′-type KMFCO delivers superior rate capability(52.9 mAh g^(-1) at 1000 mA g^(-1))and excellent cycling stability(91.1%capacity retention after 500 cycles at 500 mA g^(-1)).A full cell assembled with the P3′-type KMFCO cathode and a graphite anode also exhibits a high reversible capacity(81.2 mAh g^(-1) at 100 mA g^(-1))and excellent cycling performance(97%capacity retention after 300 cycles).The strategy of developing single-crystal materials may offer a new pathway for maintaining structural stability and improving the rate capability of layered manganese oxide cathodes and beyond. | Jiarui Lv Bin Wang Jiaxin Hao Hongbo Ding Ling Fan Renqian Tao Hongguan Yang Jiang Zhou Bingan Lu | 2023 | eScience2023,3,1: | 1 |

| 10 | Programming the lifestyles of engineered bacteria for cancer therapy显示文摘Bacteria can be genetically engineered to act as therapeutic delivery vehicles in the treatment of tumors,killing cancer cells or activating the immune system.This is known as bacteria-mediated cancer therapy(BMCT).Tumor invasion,colonization and tumor regression are major biological events,which are directly associated with antitumor effects and are uncontrollable due to the influence of tumor microenvironments during the BMCT process.Here,we developed a genetic circuit for dynamically programming bacterial lifestyles(planktonic,biofilm or lysis),to precisely manipulate the process of bacterial adhesion,colonization and drug release in the BMCT process,via hierarchical modulation of the lighting power density of near-infrared(NIR)light.The deep tissue penetration of NIR offers us a modality for spatio-temporal and non-invasive control of bacterial genetic circuits in vivo.By combining computational modeling with a high-throughput characterization device,we optimized the genetic circuits in engineered bacteria to program the process of bacterial lifestyle transitions by altering the illumination scheme of NIR.Our results showed that programming intratumoral bacterial lifestyle transitions allows precise control of multiple key steps throughout the BMCT process and therapeutic efficacy can be greatly improved by controlling the localization and dosage of therapeutic agents via optimizing the illumination scheme. | Shengwei Fu Rongrong Zhang Yanmei Gao Jiarui Xiong Ye Li Lu Pu Aiguo Xia Fan Jin | 2023 | National Science Review2023,10,5: | 0 |

| 15 | Identification of Bulbocodin D and C as novel STAT3 inhibitors and their anticancer activities in lung cancer cells显示文摘Cancer stands as one of the predominant causes of mortality globally, necessitating ongoing efforts to develop innovative therapeutics. Historically, natural products have been foundational in the quest for anticancer agents. Bulbocodin D (BD) and Bulbocodin C (BC), two bibenzyls derived from Pleione bulbocodioides (Franch.) Rolfe, have demonstrated notable in vitro anticancer activity. In human lung cancer A549 cells, the IC50s for BD and BC were 11.63 and 11.71 μmol·L^(−1), respectively. BD triggered apoptosis, as evidenced by an upsurge in Annexin V-positive cells and elevated protein expression of cleaved-PARP in cancer cells. Furthermore, BD and BC markedly inhibited the migratory and invasive potentials of A549 cells. The altered genes identified through RNA-sequencing analysis were integrated into the CMap dataset, suggesting BD’s role as a potential signal transducer and activator of transcription 3 (STAT3) inhibitor. SwissDock and MOE analyses further revealed that both BD and BC exhibited a commendable binding affinity with STAT3. Additionally, a surface plasmon resonance assay confirmed the direct binding affinity between these compounds and STAT3. Notably, treatment with either BD or BC led to a significant reduction in p-STAT3 (Tyr 705) protein levels, regardless of interleukin-6 stimulation in A549 cells. In addition, the extracellular signal-regulated kinase (ERK) was activated after BD or BC treatment. An enhancement in cancer cell mortality was observed upon combined treatment of BD and U0126, the MEK1/2 inhibitor. | 16 | Switchable nanoparticles complexing cisplatin for circumventing glutathione depletion in breast cancer chemotherapy显示文摘Cisplatin is broad-spectrum chemotherapeutic agent that has been widely used for the treatment of a variety of malignant tumors including breast cancer.However,the cisplatin chemoresistance,which derives from the inactivation by glutathione(GSH)depletion,remains a scientific issue to solve.Here,we report a novel type of smart disulfide switchable nanoparticles complexing cisplatin(switch NPs-cisplatin)that is rationally designed,and engineered by synthesizing a hyaluronic acid disulfide bonded polyaspartic acid(HA-ss-Pasp)and complexing cisplatin.The results showed that the switch NPs-cisplatin had a nanoscale of particle size(150 nm),higher drug encapsulation efficiency(>90%),and suitable drug release profile.They demonstrated evident pH responsiveness and GSH responsiveness,and targeting effect in the resistant breast cancer cells.Furthermore,they were able to block the cisplatin depletion by GSH in the resistant cancer cells,thereby circumventing the chemoresistance.Consequently,switch NPs-cisplatin displayed a remarkable killing effect in the resistant breast cancer cells in vitro,and in the resistant breast cancer-bearing mice.In conclusion,switch NPs-cisplatin could be used as a smart formulation of cisplatin for overcoming the chemoresistance of breast cancer.The present study also offers a universal drug delivery carrier platform for highly efficient but low systemic toxic chemotherapy. | Ming Chen Ying Xie Qian Luo Jiarui Xu Yuxin Ren Rui Liu Huihui Zhao Yuling Chen Hexuan Feng Yafei Du Jianwei Li Guiling Wang Wanliang Lu | 2023 | Chinese Chemical Letters2023,34,5: | 0 |
