1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29
2Design strategies for two-dimensional material photodetectors to enhance device performance显示文摘Two-dimensional(2D)materials are intensively attractive for fabricating high sensitive photodetectors in terms of atomically thin flexible and ultrafast charge transport feature.Due to their atomically thin body,designing high performance detector requires new physical mechanisms and device structures.In this review,we classify design strategies and device structures into four categories depending on their physical mechanisms(photovoltaic effect,photoconductive effect,photothermoelectric effect or photobolometric effect,and surface plasma-wave-assisted effect),and summarize the device performances.Finally,future prospects and development direction for 2D material photodetectors are described.Those design strategies descriptions about photoelectronic detector provide a reference for high responsivity and fast response speed photodetector at broadband sensing in the future.Jun Wang Jiayue Han Xiaoqing Chen Xinran Wang 2019InfoMat2019,1,1:16
3Investigating the Effectiveness of Road-related Mitigation Measures under Semi-controlled Conditions:A Case Study on Asian Amphibians显示文摘Road traffic is the main factor causing the decline in amphibian populations worldwide. The proper design of an amphibian tunnel is one of the most efficient measures to mitigate the negative impacts of road traffic on amphibians. However, no study has investigated the effectiveness of amphibian tunnels under semi-controlled conditions in Asian amphibians. Here, we selected two representative amphibian species, the Chinese brown frog, Rana chensinensis, and the Asiatic toad, Bufo gargarizans, which suffer the most severe road mortality along the roads in Northeast China. We placed experimental arrays of culverts of various sizes(diameters of 1.5, 1, and 0.5 m for circular culverts; side lengths of 1.5, 1, and 0.5 m for box culverts), and substrate type(soil, concrete, and metal) to examine the preferences of both species during the migratory season between May and September in 2016 and 2017. The results revealed that the Chinese brown frog preferred mid-and large-sized culverts as well as soil culverts. 4Dose-dense paclitaxel plus carboplatin vs.epirubicin and cyclophosphamide with paclitaxel as adjuvant chemotherapy for high-risk triple-negative breast cancer显示文摘Objective:The objective of this open-label,randomized study was to compare dose-dense paclitaxel plus carboplatin(PCdd)with dose-dense epirubicin and cyclophosphamide followed by paclitaxel(ECdd-P)as an adjuvant chemotherapy for early triple-negative breast cancer(TNBC).Methods:We included Chinese patients with high recurrence risk TNBC who underwent primary breast cancer surgery.They were randomly assigned to receive PCdd[paclitaxel 150 mg/m2 on d 1 and carboplatin,the area under the curve,(AUC)=3 on d 2]or ECdd-P(epirubicin 80 mg/m2 divided in 2 d and cyclophosphamide 600 mg/m2 on d 1 for 4 cycles followed by paclitaxel 175 mg/m2 on d 1 for 4 cycles)every 2 weeks with granulocyte colony-stimulating factor(G-CSF)support.The primary endpoint was 3-year disease-free survival(DFS);the secondary endpoints were overall survival(OS)and safety.Results:The intent-to-treat population included 143 patients(70 in the PCdd arm and 73 in the ECdd-P arm).Compared with the ECdd-P arm,the PCdd arm had significantly higher 3-year DFS[93.9%vs.79.1%;hazard ratio(HR)=0.310;95%confidence interval(95%CI),0.137-0.704;log-rank,P=0.005]and OS(98.5%vs.92.9%;HR=0.142;95%CI,0.060-0.825;log-rank,P=0.028).Worse neutropenia(grade 3/4)was found in the ECdd-P than the PCdd arm(47.9%V5.21.4%,P=0.001).Conclusions:PCdd was superior to ECdd-P as an adjuvant chemotherapy for early TNBC with respect to improving the 3-year DFS and OS.PCdd also yielded lower hematological toxicity.Thus,PCdd might be a preferred regimen for early TNBC patients with a high recurrence risk.Qing Li Jiani Wang Yuxin Mu Tongtong Zhang Ying Han Jiayu Wang Qiao Li Yang Luo Fei Ma Ying Fan Pin Zhang Binghe Xulj 2020Chinese Journal of Cancer Research2020,32,4:4
5Matrix stiffness exacerbates the proinflammatory responses of vascular smooth muscle cell through the DDR1-DNMT1 mechanotransduction axis显示文摘Vascular smooth muscle cell (vSMC) is highly plastic as its phenotype can change in response to mechanical cues inherent to the extracellular matrix (ECM). VSMC may be activated from its quiescent contractile phenotype to a proinflammatory phenotype, whereby the cell secretes chemotactic and inflammatory cytokines, e.g. MCP1 and IL6, to functionally regulate monocyte and macrophage infiltration during the development of various vascular diseases including arteriosclerosis. Here, by culturing vSMCs on polyacrylamide (PA) substrates with variable elastic moduli, we discovered a role of discoidin domain receptor 1 (DDR1), a receptor tyrosine kinase that binds collagens, in mediating the mechanical regulation of vSMC gene expression, phenotype, and proinflammatory responses. We found that ECM stiffness induced DDR1 phosphorylation, oligomerization, and endocytosis to repress the expression of DNA methyltransferase 1 (DNMT1), very likely in a collagen-independent manner. The DDR1-to-DNMT1 signaling was sequentially mediated by the extracellular signal-regulated kinases (ERKs) and p53 pathways. ECM stiffness primed vSMC to a proinflammatory phenotype and this regulation was diminished by DDR1 inhibition. In agreement with the in vitro findings, increased DDR1 phosphorylation was observed in human arterial stiffening. DDR1 inhibition in mouse attenuated the acute injury or adenine diet-induced vascular stiffening and inflammation. Furthermore, mouse vasculature with SMC-specific deletion of Dnmt1 exhibited proinflammatory and stiffening phenotypes. 6An H^m-conforming spectral element method on multi-dimensional domain and its application to transmission eigenvalues显示文摘We develop an Hm-conforming(m 1) spectral element method on multi-dimensional domain associated with the partition into multi-dimensional rectangles. We construct a set of basis functions on the interval [-1, 1] that are made up of the generalized Jacobi polynomials(GJPs) and the nodal basis functions.So the basis functions on multi-dimensional rectangles consist of the tensorial product of the basis functions on the interval [-1, 1]. Then we construct the spectral element interpolation operator and prove the associated interpolation error estimates. 8SOD1^(G93A) Induces a Unique PSAP-Dependent Mitochondrial Apoptosis Pathway via Bax-Bak Interaction显示文摘Amyotrophic lateral syndrome(ALS)is a progressive degenerative disorder characterized by motor neuron death and axon degeneration.Mitochondrial dysfunction plays a key role in the pathogenesis of ALS,the mechanism of which remains poorly understood.The B-cell lymphoma-2(Bcl-2)family of proteins that control and mediate mitochondrial function and apoptosis,including the pro-apoptotic members Bcl2-Associated X(Bax),are involved in ALS development.The death receptor 6(DR6)regulates motor neuron death in ALS,and DR6 antibodies can prevent axon degeneration and motor neuron damage by blocking DR6.Previous studies demonstrated that PSAP localized to mitochondria and was required for DR6-induced apoptosis.In this study,SOD1^(G93A) was transfected into the motor neuron cell line NSC-34 to serve as an ALS cell model in vitro.The data assessed the role of PSAP in SOD1^(G93A)induced apoptosis and demonstrated that the overexpression of SOD1^(G93A),but not wtSOD1,induced PARP cleavage,caspase-3 activation,cytochrome c release,and Bax translocation.PSAP,Bax,and Bak were necessary for SOD1^(G93A)induced apoptosis,as silencing PSAP inhibited SOD1^(G93A)-mediated cell death that was dependent on Bax-Bak interaction.HAN NIU XIN CHEN XUEQI FU JINGTIAN ZHANG GUODONG LI YUXIANG WANG JIAYUE SONG XUETING MA CHEN HU XUEMIN XU FUQIANG ZHANG LINLIN ZENG 2021BIOCELL2021,45,4:2

10Light-induced irreversible structural phase transition in trilayer graphene显示文摘A crystal structure has a profound influence on the physical properties of the corresponding material.By synthesizing crystals with particular symmetries,one can strongly tune their properties,even for the same chemical configuration(compare graphite and diamond,for instance).Even more interesting opportunities arise when the structural phases of crystals can be changed dynamically through external stimulations.Such abilities,though rare,lead to a number of exciting phenomena,such as phase-change memory effects.In the case of trilayer graphene,there are two common stacking configurations(ABA and ABC)that have distinct electronic band structures and exhibit very different behaviors.Domain walls exist in the trilayer graphene with both stacking orders,showing fascinating new physics such as the quantum valley Hall effect.Extensive efforts have been dedicated to the phase engineering of trilayer graphene.However,the manipulation of domain walls to achieve precise control of local structures and properties remains a considerable challenge.Here,we experimentally demonstrate that we can switch from one structural phase to another by laser irradiation,creating domains of different shapes in trilayer graphene.The ability to control the position and orientation of the domain walls leads to fine control of the local structural phases and properties of graphene,offering a simple but effective approach to create artificial two-dimensional materials with designed atomic structures and electronic and optical properties.Jianyu Zhang Jinsen Han Gang Peng Xi Yang Xiaoming Yuan Yongjun Li Jianing Chen Wei Xu Ken Liu Zhihong Zhu Weiqi Cao Zheng Han Jiayu Dai Mengjian Zhu Shiqiao Qin Kostya S.Novoselov 2020Light(Science & Applications)2020,9,1:2

14Light-modulated vertical heterojunction phototransistors with distinct logical photocurrents显示文摘The intriguing carrier dynamics in graphene heterojunctions have stimulated great interest in modulating the optoelectronic features to realize high-performance photodetectors.However,for most phototransistors,the photoresponse characteristics are modulated with an electrical gate or a static field.In this paper,we demonstrate a graphene/C_(60)/pentacene vertical phototransistor to tune both the photoresponse time and photocurrent based on light modulation.By exploiting the power-dependent multiple states of the photocurrent,remarkable logical photocurrent switching under infrared light modulation occurs in a thick C_(60) layer(11 nm)device,which implies competition of the photogenerated carriers between graphene/C_(60) and C_(60)/pentacene.Meanwhile,we observe a complete positive-negative alternating process under continuous 405 nm irradiation.Furthermore,infrared light modulation of a thin C_(60)(5 nm)device results in a photoresponsivity improvement from 3425 A/W up to 7673 A/W,and we clearly probe the primary reason for the distinct modulation results between the 5 and 11 nm C_(60) devices.In addition,the tuneable bandwidth of the infrared response from 10 to 3×10^(3) Hz under visible light modulation is explored.Such distinct types of optical modulation phenomena and logical photocurrent inversion characteristics pave the way for future tuneable logical photocurrent switching devices and high-performance phototransistors with vertical graphene heterojunction structures.Jiayue Han Meiyu He Ming Yang Qi Han Fang Wang Fang Zhong Mengjian Xu Qing Li He Zhu Chongxin Shan Weida Hu Xiaoqing Chen Xinran Wang Jun Gou Zhiming Wu Jun Wang 2020Light(Science & Applications)2020,9,1:1

17Influence of Arctic Sea-ice Concentration on Extended-range Forecasting of Cold Events in East Asia显示文摘Utilizing the Community Atmosphere Model,version 4,the influence of Arctic sea-ice concentration(SIC)on the extended-range prediction of three simulated cold events(CEs)in East Asia is investigated.Numerical results show that the Arctic SIC is crucial for the extended-range prediction of CEs in East Asia.The conditional nonlinear optimal perturbation approach is adopted to identify the optimal Arctic SIC perturbations with the largest influence on CE prediction on the extended-range time scale.It shows that the optimal SIC perturbations are more inclined to weaken the CEs and cause large prediction errors in the fourth pentad,as compared with random SIC perturbations under the same constraint.Further diagnosis reveals that the optimal SIC perturbations first modulate the local temperature through the diabatic process,and then influence the remote temperature by horizontal advection and vertical convection terms.Consequently,the optimal SIC perturbations trigger a warming center in East Asia through the propagation of Rossby wave trains,leading to the largest prediction uncertainty of the CEs in the fourth pentad.These results may provide scientific support for targeted observation of Arctic SIC to improve the extended-range CE prediction skill.Chunxiang LI Guokun DAI Mu MU Zhe HAN Xueying MA Zhina JIANG Jiayu ZHENG Mengbin ZHU 2023Advances in Atmospheric Sciences2023,40,12:0
18Application of three Ln(Ⅲ)-coordination polymers in fields of luminescence,antibacteria and detection of Fe^(3+) and 4-nitrophenol显示文摘Three lanthanide-based coordination polymers(Ln-CPs) with layered structure,{[La_(2)(L)_(3)(DMF)(H_(2)O)_(3)]·H_(2)O}_n(1),{[Eu_(2)(L)_(3)(DMF)_(2)(H_(2)O)]·(H_(2)O)}_n(2){Tb_(2)(L)_(3)(DMF)_(2)(H_(2)O)]·H_(2)O}n(3) were successfully synthesized from H_(2)L(N,N'-bis(4-carbozylbenzyl)aniline).Remarkably,compound 3 has extremely high sensitivity to 4-nitrophenol.When the concentration of 4-nitrophenol reaches 5.98 × 10^(-3) mmol/L,the fluorescence intensity of compound 3 is quenched by 59%.The calculated K_(SV) is 7.56 × 10~4 and limit of detection(LOD) is 4.73 μg/L.3 exhibits a highly sensitive detection towards Fe^(3+) and compound 2 shows outstanding antibacterial effects on Staphylococcus aureus,re spectively.The white light emission was also investigated by changing the mixing ratio of LaNO_(3),EuNO_(3) and TbNO_(3) during the preparation of coordination polymers.Lun Zhao Yue Han Guanlin Guo Helong Bai Zhexin Wang Hongwei Jing Guanying Song Ziyun Wang Jiayu Li Jia Li 2023Journal of Rare Earths2023,41,9:0
19Predictive biomarkers of response and survival following immunotherapy with a PD-L1 inhibitor benmelstobart(TQB2450)and antiangiogenic therapy with a VEGFR inhibitor anlotinib for pretreated advanced triple negative breast cancer显示文摘In our phase Ib trial(ClinialTrials.gov Identifier:NCT03855358),benmelstobart(TQB2450),a novel humanized IgG1 antibody against PD-L1,plus antiangiogenic multikinase inhibitor,anlotinib,demonstrated promising antitumor activities in pretreated triple negative breast cancer(TNBC)patients.We conducted explorative analyses of genomic biomarkers to explore the associations with treatment response and survival outcomes.Targeted next generation sequencing(NGS)was undertaken toward circulating tumor DNA(ctDNA)collected from peripheral blood samples prior to the start of treatment and after disease progression.A total of 31 patients received targeted NGS and functional driver mutations in 29 patients were analyzed.The most frequent mutations were TP53(72%),MLL3(28%),and PIK3CA(17%).At a blood-based tumor mutational burden(bTMB)cutoff of 6.7 mutations per megabase,patients with low bTMB showed better response to anlotinib plus TQB2450(50%vs.7%,P=0.015)and gained greater PFS benefits(7.3 vs.4.1 months,P=0.012)than those with high bTMB.At a maximum somatic allele frequency(MSAF)cutoff of 10%,a low MSAF indicated a better objective response(43%vs.20%)as well as a significantly longer median PFS(7.9 vs.2.7 months,P<0.001).Patients with both low MSAF and low bTMB showed a notably better objective response to anlotinib plus TQB2450(70%vs.11%,P<0.001)and a significantly longer median PFS(11.0 vs.2.9 months,P<0.001)than patients with other scenarios.Our findings support future studes and validation of MSAF and the combined bTMB-MSAF classification as predictive biomarkers of immune checkpoint inhibitor-based regimens in advanced TNBC patients.Yiqun Han Jiayu Wang Tao Sun Quchang Ouyang Jianwen Li Jie Yuan Binghe Xu 2023Signal Transduction and Targeted Therapy2023,8,12:0
