| 4 | Targeting Kindlin-2 in adipocytes increases bone mass through inhibiting FAS/PPARγ/FABP4 signaling in mice显示文摘Osteoporosis(OP)is a systemic skeletal disease that primarily affects the elderly population,which greatly increases the risk of fractures.Here we report that Kindlin-2 expression in adipose tissue increases during aging and high-fat diet fed and is accompanied by decreased bone mass.Kindlin-2 specific deletion(K2KO)controlled by Adipoq-Cre mice or adipose tissue-targeting AAV(AAV-Rec2-CasRx-sgK2)significantly increases bone mass.Mechanistically,Kindlin-2 promotes peroxisome proliferator-activated receptor gamma(PPARγ)activation and downstream fatty acid binding protein 4(FABP4)expression through stabilizing fatty acid synthase(FAS),and increased FABP4 inhibits insulin expression and decreases bone mass.Kindlin-2 inhibition results in accelerated FAS degradation,decreased PPARγactivation and FABP4 expression,and therefore increased insulin expression and bone mass.Interestingly,we find that FABP4 is increased while insulin is decreased in serum of OP patients.Increased FABP4 expression through PPARγactivation by rosiglitazone reverses the high bone mass phenotype of K2KO mice.Inhibition of FAS by C75 phenocopies the high bone mass phenotype of K2KO mice.Collectively,our study establishes a novel Kindlin-2/FAS/PPARγ/FABP4/insulin axis in adipose tissue modulating bone mass and strongly indicates that FAS and Kindlin-2 are new potential targets and C75 or AAV-Rec2-CasRx-sgK2 treatment are potential strategies for OP treatment. | Wanze Tang Zhen Ding Huanqing Gao Qinnan Yan Jingping Liu Yingying Han Xiaoting Hou Zhengwei Liu Litong Chen Dazhi Yang Guixing Ma Huiling Cao | 2023 | Acta Pharmaceutica Sinica B2023,13,11: | 0 |
| 5 | Transcriptome analysis reveals tumor antigen and immune subtypes of melanoma显示文摘Purpose:To screen potential tumor antigens for melanoma vaccine development and identify different immune subtypes.Methods:Transcriptional data(HTSEQ-FPKM)and clinical information of a 472 Melanoma cohort GDC TCGA Melanoma(SKCM)were downloaded from the UCSC XENA website(http://gffzz797b50e9334d4ed4hq0uuk5xqc6vv60cq.ffgz.tsg.suse.edu.cn/).Subsequently,transcriptome data and clinical information of 210 melanoma cohort GSE65904 were downloaded from Gene Expression Omnibus(GEO),a large global public database.All the transcriptome expression data matrices were log2 transformed for subsequent analysis.GEPIA,TIMER,and IMMPORT databases are also used for analysis.Cell function experiments were performed to validate the role of the IDO1 gene in melanoma cell line A375.Results:Our study provides potential tumor antigens for vaccine development in melanoma patients:GZMB,GBP4,CD79A,APOBEC3F,IDO1,JCHAIN,LAG3,PLA2G2D,XCL2.In addition,we divide melanoma patients into two immune subtypes that have significant differences in tumor immunity and may have different responses to vaccination.In view of the unclear role of IDO1 in melanoma,we selected IDO1 for cell assay validation.Cell function assay showed that IDO1 was significantly overexpressed in the melanoma A375 cell line.After IDO1 knockdown,the activity,invasion,migration and healing ability of A375 cell lines were significantly decreased.Conclusion:Our study could provide a reference for the development of vaccines for melanoma patients. | JIAHENG XIE MENGMENG OU PAN YU DAN WU QIKAI TANG YUAN CAO JING HANG LU YIN TINGHONG XIANG MING WANG JINGPING SHI | 2023 | Oncology Research2023,31,3: | 0 |

A design scheme of teaching quality evaluation method based on cloud model and D-S theory is proposed with the school teaching evaluation index. With the experiences and knowledge of experts, evaluation index was described by cloud model and judgment matrix of AHP was revised by D-S theory. The weight of evaluation indexes was determined reasonably and objectively. With transformation and development of universities, to cultivate a large number of technical talents who can make scientific and technological achievements into practice, an educating strategy of integrating enterprises into higher education is proposed. From universities’ perspective, the cooperation with enterprises was further strengthened by linking the Industry- University-Research cooperation. With university-enterprise cooperation to indepth integration, the talent supply was further improved. And an in-depth ecological mechanism of government-led, market-driven and common development between universities and enterprises was established.
