| 3 | Stress Corrosion of X80 Pipeline Steel Welded Joints by Slow Strain Test in NACE H_2S Solutions显示文摘The X80 pipeline steel was welded with the way of submerged arc welding. The SST (slow strain test) of the welded joint samples in the air, NACE (National Association of Corrosion Engineers) solution (no H2S), NACE solution (saturated H2S) was performed to research the sensibility index of SCC (stress corrosion cracking) Iscc. The morphologies of the welded joint fractures and the fracture modes were observed with SEM (scanning electron microscope), and the fracture chemical compositions were analyzed with EDS (energy dispersive spectrometer), respectively. The fracture mechanisms of the welded joints were discussed. The results show that sensibility index of SCC in the air is not obvious, the fracture is dimple, and the mode of fracture is ductile fracture. The sensibility index of SCC in NACE solution (no H2S) is 13.21%, the stress corrosion is not obvious. The sample fracture shows quasi cleavage+dimple, and the fracture mode is toughness+brittle rupture. The sensibility index of SCC in NACE solution (saturated H2S) is 56.94%, the plastic loss is the most serious, appearing an obvious stress corrosion tendency, and there is no obvious necking phenomenon. | 6 | Chemical composition and source apportionment of the ambient PM_(2.5)in Hangzhou,China显示文摘To identify and apportion the sources of the ambient PM_(2.5) in the urban area of Hangzhou,China,PM_(2.5)samples were collected at three sites in the city from April 2004 to March 2005.Water-soluble ions,metal elements,and total carbon(TC) in PM_(2.5) samples were analyzed.The results indicated that the 24-h mean concentrations of PM_(2.5) ranged from 17.1 to 267.0 μg/m^3,with an annual average value of 108.2 μg/m^3.Moreover,the seasonal mean values for PM_(2.5) in spring,summer,autumn,and winter were 116,73.1,114.2,and 136.0 μg/m^3.respectively.According to the Chinese ambient quality standard,at least 70% of the monitoring data exceeded the limit value.The total contribution of water-soluble ions,including F^-,Cl^-,NO_3^-,SO_4^(2-).NH_4^+,K^+,and Na^+.to PM_(2.5) mass varied from 32.3% to 36.7%.SO_4^(2-),NO_3^-,and NH_4^+were the main constituents of the ions,with contributions to PM_(2.5) varying from 14.1% to 14.7%,6.0%to 7.8%,and 6.4% to 7.7%,respectively.In addition,the annual mean mass fraction of TC in PM_(2.5) was27.8%.The annual average total contribution of the group of elements of Zn,Pb,Cu,Mn,Cr,Ni,Se,Mo.Cd,Sb,and Ag to the aerosol was in the range of 1.7-2.0%.Furthermore,positive matrix factorization was applied to analyze the PM_(2.5) data collected from the central area,and five factors were identified.The factor contributions to PM_(2.5) mass were 12.8%,31.9%,10.1%,17.2%,and 27.9%,respectively.Iron/steel manufacturing and secondary aerosol were the main sources for the fine particles.These findings may have significance for controlling the atmospheric contamination in the city. | Gang Liu Jiuhai Li Dan Wu Hui Xu | 2015 | Particuology2015,13,1: | 26 |
| 7 | Indoor Allergen Levels and Household Distributions in Nine Cities Across China显示文摘Objective Chinese allergic subjects have high levels of sensitization to house dust mite(HDM) and other indoor allergens. This study quantifies common indoor allergen levels in Chinese households. Methods Dust samples were collected from nine cities. Major allergens Der p 1 and Der f 1 from Dermatophagoides pteronyssinus and D. farinae, and specific antigens of Blomia tropicalis, Tyrophagus putrescentiae, Acarus siro, and cockroach species Blattella germanica and Periplaneta americana were measured by ELISA. Results HDM allergens were found in dust samples from bedding in 95% of the Chinese households. The median levels varied from <0.006 to 9.2 μg/g of dust, depending on the city. The percentages of households having HDM allergen levels associated with the risk of developing allergy sensitization and asthma were 65% and 25%, respectively. Specific antigens of the storage mite and cockroach were only found in samples from the southern and tropical regions of China. Levels of mite allergens were generally higher in samples from bedding compared to samples from the living room, even for storage mites, whereas levels of cockroach antigens were higher in the living room samples. Conclusion HDM allergens are present in bedding dust samples from most Chinese households. Cities in southern and central China have relatively high levels of HDM major allergens compared to cities in northern and western China. | 8 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 19 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive.
