| 1 | N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. | 9 | Extraction of flocculants from a strain of Bacillus thuringiensis and analysis of their properties显示文摘In a preliminary screening study,our laboratory isolated from the biofloc in aquaculture waters a strain of Bacillus thuringiensis,which produced highly efficient bio-flocculants.In the present study,we extracted the crude flocculants from this strain and analyzed their properties.Distribution analysis indicated that the flocculants were mainly distributed in the supernatant of the fermentation liquid.The flocculants were extracted using an ethanol extraction method,and the chemical compositions and morphology of the crude flocculants were analyzed using the Molish reaction,Fehling reaction,ninhydrin reaction,biuret reaction,phenol-sulfuric acid assay,Coomassie brilliant blue staining,ultraviolet scanning,infrared scanning and scanning electron microscopy.The carbohydrate composition of the polysaccharides in the flocculants was analyzed with thin layer chromatography.The results indicated that flocculants were solid substances with an ivory white color and their texture was loose and soft.Visualization under scanning electron microscopy revealed that their ultra-morphology consisted of small,long and fiber-like shapes.Chemical and physical analyses indicated that polysaccharides accounted for 34.5%of the components in the crude flocculants.The monosaccharides present in crude flocculants included mainly glucose,galactose and mannitose. | Jingrong Wu Guangjun Wang Zhifei Li Ermeng Yu Jun Xie Zonglin Zheng | 2017 | Aquaculture and Fisheries2017,2,4: | 1 |

| 12 | AutoGDeterm: Automatic Geometry Determination for Electron Tomography显示文摘Electron Tomography(ET) is an important method for studying cell ultrastructure in three-dimensional(3 D) space. By combining cryo-electron tomography of frozen-hydrated samples(cryo-ET) and a sub-tomogram averaging approach, ET has recently reached sub-nanometer resolution, thereby realizing the capability for gaining direct insights into function and mechanism. To obtain a high-resolution 3 D ET reconstruction, alignment and geometry determination of the ET tilt series are necessary. However, typical methods for determining geometry require human intervention, which is not only subjective and easily introduces errors, but is also labor intensive for high-throughput tomographic reconstructions. To overcome these problems, we have developed an automatic geometry-determination method, called Auto GDeterm. By taking advantage of the high-contrast re-projections of the Iterative Compressed-sensing Optimized Non-Uniform Fast Fourier Transform(NUFFT) reconstruction(ICON)and a series of numerical analysis methods, Auto GDeterm achieves high-precision fully automated geometry determination. Experimental results on simulated and resin-embedded datasets show that the accuracy of Auto GDeterm is high and comparable to that of the typical 'manual positioning' method.
