1Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 2Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. 7A new family of primitive mammal from the Mesozoic of western Liaoning,China显示文摘A fairly well-preserved specimen of a new primitive mammal, Repenomamus robustus gen. et sp. nov., has been described from the Yixian Formation (Lower Cretaceous), western Liaoning, China. This animal has several derived characteristics, such as well-developed den-tary/squamosal articulation, reduced number of tooth, differentiation of premolars and molars in postcanine teeth, presence of a dorsal process of the premaxilla that is not in contact with the nasal, closed medial wall of the orbit, and presence of fingerlike promontorium on the petrosal. It also retains some primitive reptile-like features. 8Reliability testing of a 3D encapsulated VHF MEMS resonator显示文摘The frequency stability of a three-dimensional(3D) vacuum encapsulated very high frequency(VHF)disk resonator is systematically investigated. For eliminating the parasitic effect caused by the parasitic capacitance of the printed circuit board(PCB), a negating capacitive compensation method was developed. The testing results implemented at 25 ℃ for 240 h for the long-term stability indicates that the resonant frequency variation remained within ±1 ppm and the noise floor derived from Allan Deviation was 26 ppb, which is competitive with the conventional quartz resonators. 10Middle Triassic(Anisian)Conodont Biostratigraphy at the Shaiwa Section,Guizhou,South China显示文摘The Nanpanjiang Basin is a key area for paleontological and biostratigraphical study of the Middle Triassic.Herein we studied Middle Triassic conodonts from a well-exposed section,the Shaiwa Section,which is located at the northwest end of the Nanpanjiang Basin.A total of six Anisian conodont zones are recognized;in ascending order,they are:the Nicoraella germanica Zone,the Nicoraella kockeli Zone,the Paragondolella bulgarica Zone,the Neogondolella constricta Zone,the Neogondolella cornuta Zone,and the Paragondolella excelsa Zone,respectively.The first occurrence of Nicoraella kockeli defines the Bithynian-Pelsonian boundary.The Pelsonian-Illyrian boundary is defined by the first occurrence of Neogondolella constricta.The Anisian-Ladinian boundary cannot be recognized at the Shaiwa Section due to the absence of conodont indicative of the Ladinian.However,the new conodont data indicate that the uppermost strata could be very close to the boundary.The abrasion of conodont surfaces provides evidence for demonstrating reworking at the Shaiwa Section,which makes some conodonts possess a longer stratigraphic range than previously recorded.The variation in relative abundance between blade-shaped conodonts and platform conodonts indicates that segminiplanate elements probably preferred deeper and oxygenated environments whereas a restricted marine environment is more suitable for segminate elements.Binxian Qin Martyn Lee Golding Haishui Jiang Yan Chen Muhui Zhang Li Kang Dacheng Wang Jinling Yuan 2021Journal of Earth Science2021,32,3:0
11Conodonts and Carbon Isotopes during the Permian-Triassic Transition on the Napo Platform, South China显示文摘Two Permian-Triassic boundary(PTB) sections(Pojue and Dala) are well exposed in an isolated carbonate platform(Napo Platform) on the southwestern margin of the Nanpanjiang Basin, South China. These sections provide an insight into the transition across the PTB and a detailed investigation of the conodont biostratigraphy and inorganic carbon isotopes is presented. The PTB at the Pojue Section is placed at the base of Bed 10 B(a dolomitized mudstone found below a microbialite horizon), defined by the first occurrence of Hindeodus parvus. At the Dala Section, four conodont zones occur. They are, in ascending order, the Hindeodus parvus Zone, Isarcicella staeschei Zone, Isarcicella isarcica Zone and Clarkina planata Zone. Comparison with the Pojue Section suggests the PTB at Dala also occurs at the base of dolomitized mudstone below a microbialite horizon, although the first occurrence of Hindeodus parvus is near the top of a microbialite bed: an occurrence that is also seen in other platform sections. The succeeding microbialite beds developed during the ongoing PTB mass extinction phase. 12Solidification and Crystal Growth on the SJ-10 Recoverable Scientific Experiment Satellite显示文摘The low-gravity environment aboard the space provides a unique platform for understanding crystal-growth-related phenomena that are masked by gravity on the Earth and for exploring new crystal growth techniques. We have characterized the wetting behavior of metal alloys and carried out melt growth of compound semiconductors under the support of materials science program in the SJ-10 recoverable satellite. We found that interfacial reaction plays a significant role in the interfacial evolution of Sn-based alloys. Detached growth of InAsSb was realized under microgravity, whereas during the terrestrial experiment the crystal and the crucible wall contact with each other.
