| 1 | HBV cccDNA in patients' sera as an indicator for HBV reactivation and an early signal of liver damage显示文摘AIM: To evaluate the covalently closed circle DNA (cccDNA)level of hepatitis B virus (HBV) in patients' liver and sera. METHODS: HBV DNA was isolated from patients' liver biopsies and sera. A sensitive real-time PCR method, which is capable of differentiation of HBV viral genomic DNA and cccDNA, was used to quantify the total HBV cccDNA. The total HBV viral DNA was quantitated by real-time PCR using a HBV diagnostic kit (PG Biotech, LTD, Shenzhen, China)described previously. RESULTS: For the first time, we measured the level of HBV DNA and cccDNA isolated from ten HBV patients' liver biopsies and sera. In the liver biopsies, cccDNA was detected from all the biopsy samples. The copy number of cccDNA ranged from from 0.03 to 173.1 per ceil, the copy number of total HBV DNA ranged from 0.08 to 3 717 per cell. The ratio of total HBV DNA to cccDNA ranged from 1 to 3 406. In the sera,cccDNA was only detected from six samples whereas HBV viral DNA was detected from all ten samples. The ratio of cccDNA to total HBV DNA ranged from 0 to 1.77%. To further investigate the reason why cccDNA could only be detected in some patients' sera, we performed longitudinal studies. The cccDNA was detected from the patients' sera with HBV reactivation but not from the patients' sera without HBV reactivation. The level of cccDNA in the sera was correlated with ALT and viral load in the HBV reactivation patients. CONCLUSION: HBV cccDNA is actively transcribed and replicated in some patients' hepatocytes, which is reflected by a high ratio of HBV total DNA vs cccDNA. Detection of cccDNA in the liver biopsy will provide an end-point for the anti-HBV therapy. | 2 | Influence of South China Sea SST and the ENSO on Winter Rainfall over South China显示文摘The present study investigates the influence of South China Sea (SCS) SST and ENSO on winter (January-February-March; JFM) rainfall over South China and its dynamic processes by using station observations for the period 1951-2003, Met Office Hadley Center SST data for the period 1900-2008, and ERA-40 reanalysis data for the period 1958-2002. It is found that JFM rainfall over South China has a sig- nificant correlation with Nio-3 and SCS SST. Analyses show that in El Nio or positive SCS SST anomaly years, southwesterly anomalies at 700 hPa dominate over the South China Sea, which in turn transports more moisture into South China and favors increased rainfall. A partial regression analysis indicates that the independent ENSO influence on winter rainfall occurs mainly over South China, whereas SCS SST has a larger independent influence on winter rainfall in northern part of South China. The temperature over South China shows an obvious decrease at 300 hPa and an increase near the surface, with the former induced by Nio-3 and the latter SCS SST anomalies. | 4 | Interannual and Interdecadal Variability of Winter Precipitation over China in Relation to Global Sea Level Pressure Anomalies显示文摘Based on the method of rotated principal component (RPC) analysis and wavelet transforms, the win-ter precipitation from 36 stations over China for the period 1881-1993 is examined. The results show thatthe three leading space-time modes correspond, in sequence, to winter rainfall anomalies over the reaches ofthe Yangtze River, the bend of the Yellow River, and the northeastern region of China. The three modes ex-hibit interannual oscillations with quasi-biennial and 8-year periods as well as interdecadal oscillationswith 16- and 32-year periods. The interannual oscillation (< 10 years) occurs in phase over the differentareas, and its maximum amplitude migrates northward considerably with prominent interdecadal variations.However, the interdecadal oscillations (10-32 years) are out of phase over the different regions, and theamplitude variations have the characteristics of stationary waves.The rainfall anomalies appear to be closely re lated to the anti-phase changes of mean sea-level pres-sure (SLP) over the Asian mainland and the North Pacific. When the SLP rises over the North Pacific anddecreases over the Asian mainland, the precipitation over East China increases noticeably. | 7 | Circulating inflammatory factors associated with worse long-term prognosis in colorectal cancer显示文摘AIM To investigate association of circulating inflammatory factors at the time of colorectal cancer(CRC) surgery with survival.METHODS Plasma levels from 174 CRC patients(69 females and 105 men), with median age 70 years(range 29-90), localized in the colon(n = 105) or rectum(n = 69), with stage Ⅰ(n = 24), stage Ⅱ(n = 54), stage Ⅲ(n = 67) and stage Ⅳ(n = 29) were measured using commercially available Bio-Plex Pro? Human Chemokine Panel 40-Plex, including 40 different chemokines, cytokines and interleukins. The prognostic association of each inflammatory factor was analysed as CRC-specific and total mortality.RESULTS Out of 174 patients, 66 died during the follow-up, 40 because of CRC specific mortality. High tertile levels of 8 factors were significantly associated with increased CRC-specific mortality, of which CCL1, CCL20, CCL24, CX3CL1, IL-4 and TNF-α remained significant in a multivariate Cox regression analysis. High tertile levels of 14 factors were associated with increased total mortality, of which CCL1, CCL15, CCL20, CX3CL1, CXCL13, IFN-γ, IL-2, IL-4 and IL-10 remained significant after adjustment for clinical covariates. For most of the inflammatory factors the association between higher tertile levels and an increased mortality in general appeared two years after surgery. High tertile levels of TNF-α and CCL24 were exclusively associated with CRC-specific mortality. | 9 | Mesoscale Vortex Generation and Merging Process: A Case Study Associated with a Post-Landfall Tropical Depression显示文摘An observational analysis of satellite blackbody temperature (TBB) data and radar images suggests that the mesoscale vortex generation and merging process appeared to be essential for a tropical-depression-related heavy rain event in Shanghai, China. A numerical simulation reproduced the observed mesoscale vortex generation and merging process and the corresponding rain pattern, and then the model outputs were used to study the related dynamics through diagnosing the potential vorticity (PV) equation. The tropical depression (TD) was found to weaken first at lower levels and then at upper levels due to negative horizontal PV advection and diabatic heating effects. The meso-vortices developed gradually, also from the lower to the upper levels, as a result of positive horizontal PV advection and diabatic heating effects in the downshear left quadrant of the TD. One of these newly-generated vortices, V1, replaced the TD ultimately, while the other two, V2 and V3, merged due to the horizontal PV advection process. | 12 | The Effect of Three-Dimensional Variational Data Assimilation of QuikSCAT Data on the Numerical Simulation of Typhoon Track and Intensity显示文摘In this paper, the three-dimensional variational data assimilation scheme (3DVAR) in the mesoscale model version 5 (MM5) of the US Pennsylvania State University/National Center for Atmospheric Research is used to study the effect of assimilating the sea-wind data from QuikSCAT on the prediction of typhoon track and intensity. The case of Typhoon Dujuan (2003) is first tested and the results show appreciable improvements. Twelve other cases in 2003 are then evaluated. The assimilation of the QuikSCAT data produces significant impacts on the structure of Dujuan in terms of the horizontal and vertical winds, sealevel pressure and temperature at the initial time. With the assimilation, the 24-h (48-h) track prediction of 11 (10) out of the 12 typhoons is improved. The 24-h (48-h) prediction of typhoon intensity is also improved in 10 (9) of the 12 cases. | 16 | Sensitivity of the Simulation of Tropical Cyclone Size to Microphysics Schemes显示文摘The sensitivity of the simulation of tropical cyclone(TC) size to microphysics schemes is studied using the Advanced Hurricane Weather Research and Forecasting Model(WRF). Six TCs during the 2013 western North Pacific typhoon season and three mainstream microphysics schemes–Ferrier(FER), WRF Single-Moment 5-class(WSM5) and WRF Single-Moment6-class(WSM6)–are investigated. The results consistently show that the simulated TC track is not sensitive to the choice of microphysics scheme in the early simulation, especially in the open ocean. However, the sensitivity is much greater for TC intensity and inner-core size. The TC intensity and size simulated using the WSM5 and WSM6 schemes are respectively higher and larger than those using the FER scheme in general, which likely results from more diabatic heating being generated outside the eyewall in rainbands. More diabatic heating in rainbands gives higher inflow in the lower troposphere and higher outflow in the upper troposphere, with higher upward motion outside the eyewall. The lower-tropospheric inflow would transport absolute angular momentum inward to spin up tangential wind predominantly near the eyewall, leading to the increment in TC intensity and size(the inner-core size, especially).
