| 1 | GOLM1 exacerbates CD8^(+)T cell suppression in hepatocellular carcinoma by promoting exosomal PD-L1 transport into tumor-associated macrophages显示文摘The immunosuppressive microenvironment plays an important role in tumor progression and immunotherapy responses.Golgi membrane protein 1(GOLM1)is correlated to hepatocellular carcinoma(HCC)progression and metastasis.However,little is known about the role of GOLM1 in regulating the immunosuppressive environment and its impact on immunotherapeutic efficacy in HCC.In this study,GOLM1 was positively correlated with infiltrating tumor-associated macrophages(TAMs)expressed high levels of programmed death-ligand 1(PD-L1)and CD8^(+)T cell suppression in HCC tissues.Both gain-and loss-of-function studies determined a close correlation between GOLM1 and immunosuppression.In the mechanism,GOLM1 promoted COP9 signalosome 5-mediated PD-L1 deubiquitination in HCC cells and increased the transport of PD-L1 into exosomes via suppression of Rab27b expression.Furthermore,co-culture with exosomes derived from HCC cells upregulated the expression of PD-L1 on macrophages.Zoledronic acid in combination with anti-PD-L1 therapy reduced PD-L1^(+)TAMs infiltration and alleviated CD8^(+)T cell suppression,resulting in tumor growth inhibition in the mouse HCC model.Together,our study unveils a mechanism by which GOLM1 induces CD8^(+)T cells suppression through promoting PD-L1 stabilization and transporting PD-L1 into TAMs with exosome dependent.Targeting PD-L1^(+)TAM could be a novel strategy to enhance the efficacy of anti-PD-L1 therapy in HCC. | Jinhong Chen Zhifei Lin Lu Liu Rui Zhang Yan Geng Minghao Fan Wenwei Zhu Ming Lu Lu Lu Huliang Jia Jubo Zhang Lun-Xiu Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 5 |

| 3 | Real-time filter based on the quasi-accurate detection of gross errors显示文摘Because of the sensitivity of the Kalman framework to gross errors, proper techniques for detection of gross errors are necessary. By integrating the selection of quasi-accurate observations and the Kalman framework, a new filter called the quasi-accurate filter (QUAF) is developed. The expansibility and implementation scheme of the new algorithm are then discussed in detail, and the reliability matrix for the Kalman filter is proposed to analyze the reliability of the filters with different detection technologies. Finally, the experimental results from a real world case study are used to validate the conclusions. The QUAF carries out the preliminary selection of the quasi-accurate observations (QAOs) using the innovation of the Kalman filter, and use the check QAOs to determine reasonable observations. This causes the QUAF to handle more easily and possess wider expansibility. QUAF can be reformulated to the special cases of several common detection methods, such as the innovation method, robust estimation and quasi-accurate detection (QUAD). Since only reasonable observations are used, the QUAF has better detection accuracy and stronger avoidance of gross errors than the innovation method and robust estimation. | 4 | Efficacy of Carbamazepine Combined with Botulinum Toxin A in the Treatment of Blepharospasm and Hemifacial Spasm显示文摘Purpose:To observe the efficacy of the combined treatment of carbamazepine and botulinum toxin A for blepharospasm and hemifacial spasm. Methods:Fifty-eight patients with either blepharospasm or hemifacial spasm were randomly divided into treatment and control groups. In the treatment group, 30 patients were administered with local intramuscular injections of botulinum toxin A and oral carbamazepine 100 mg/time,3 times/day for 60 days. Twenty-eight subjects in the control group under-went local intramuscular injections of botulinum toxin A only. Results:After combined treatment, the complete remission rate was 90%, which was significantly higher than that of the of the control group (67.9% , P<0.05,χ2 =4.733). However, no statistical significance was noted regarding the duration of therapeutic effects between the treatment group (range 14~40 weeks; 19.2 weeks on average) and control group (range 12~36 weeks; 18 weeks on average). | 19 | A Global Analysis of the Relationship Between Urbanization and Fatalities in Earthquake-Prone Areas显示文摘Urbanization can be a challenge and an opportunity for earthquake risk mitigation.However,little is known about the changes in exposure(for example,population and urban land)to earthquakes in the context of global urbanization,and their impacts on fatalities in earthquake-prone areas.We present a global analysis of the changes in population size and urban land area in earthquake-prone areas from 1990 to 2015,and their impacts on earthquake-related fatalities.We found that more than two thirds of population growth(or 70%of total population in 2015)and nearly three quarters of earthquake-related deaths(or 307,918 deaths)in global earthquake-prone areas occurred in developing countries with an urbanization ratio(percentage of urban population to total population)between 20 and 60%.Holding other factors constant,population size was significantly and positively associated with earthquake fatalities,while the area of urban land was negatively related.The results suggest that fatalities increase for areas where the urbanization ratio is low,but after a ratio between 40 and 50%occurs,earthquake fatalities decline.This finding suggests that the resistance of building and infrastructure is greater in countries with higher urbanization ratios and highlights the need for further investigation.Our quantitative analysis is extended into the future using Shared Socioeconomic Pathways to reveal that by 2050,more than 50%of the population increase in global earthquake-prone areas will take place in a few developing countries(Pakistan,India,Afghanistan,and Bangladesh)that are particularly vulnerable to earthquakes.To reduce earthquake-induced fatalities,enhanced resilience of buildings and urban infrastructure generally in these few countries should be a priority. | Chunyang He Qingxu Huang Xuemei Bai Derek TRobinson Peijun Shi Yinyin Dou Bo Zhao Jubo Yan Qiang Zhang Fangjin Xu James Daniell | 2021 | International Journal of Disaster Risk Science2021,12,6: | 1 |
