In situ cryogenic Raman spectroscopic studies on the synthetic fluid inclusions in the systems H_(2)O and NaCl-H_(2)O显示文摘Salt-hydrates have diagnostic cryogenic Raman spectra, which can reflect the composition of the parent solutions. As analogue to the natural fluid inclusions, the synthetic inclusions can be used to validate numerous assumptions related to fluid inclu- sion research. They can also be used to test the feasibility of application of laser Raman spectroscopy to individual fluid inclusion analysis. Using the tech- nique proposed by Sterner and Bodnar(1984), syn- thetic inclusions of the systems H2O and NaCl-H2O (with NaCl as 5.12 wt%, 9.06 wt%, 16.6 wt% and 25 wt%) were formed under the pressures from 50Mpa to 100Mpa and at the temperatures from 500℃ to 600℃. In situ cryogenic Raman spectra were col- lected at about –180℃ by combined use of freezing- heating stage and Laser Raman Microscopy. It is shown that hydrohalite (NaCl?2H2O), the salt-hydrate of NaCl in the fluid inclusions has the specific Raman spectrum and can be used as the standard to verify the existence of NaCl in the aqueous inclusions. The crystalline ice other than amorphous ice (glasses) formed from the aqueous phase whthin the synthetic inclusions during the initial freezing, but hydrohalite did not form. Subsequent warming of these inclusions induced a phase change, typically between ap- proximately –40 and –22℃, that represents the hy- drohalite crystallization event but not a eutectic melting event. So, for fluid inclusions in the system NaCl-H2O, interpretation of phase behavior below the eutectic temperature (–20.8℃) should be made with caution. The ratios of the relative intensity and the area of Raman spectra between 3423 cm–1 peak of hydrohalite and 3098 cm–1 peak of ice show positivecorrelations to the salinities in aqueous inclusions, which can be used to determine the salinity of NaCl- H2O system inclusions. | NI Pei DING Junying RAO Bing | 2006 | Chinese Science Bulletin2006,51,1: | 30
Necking of Q&P Steel during Uniaxial Tensile Test with the Aid of DIC Technique显示文摘A lot of research has been focused on the necking process during the plastic deformation of sheet metals, but the localized necking is rarely distinguished form diffused necking by experiments, due to the limit of measurement equipment and method. Quenching and Partitioning (Q&P) steel is a 3rd generation advanced high strength steel (AHSS). Its good combination of high strength and ductility ensures potential application in automobile industry. Uniaxial tensile tests of QP980 steel sheet at five strain rates are performed to investigate the necking process and the effect of strain rate on necking behavior of Q&P steel. Digital image correlation (DIC) method is applied during tensile tests, and evolutions of major strain, minor strain and normal strain distributions along gauge section of the tensile specimens are obtained. The diffused and localized necking strains are determined according to SWIFT necking theory and HILL necking theory respectively. The test results indicate that with the increasing of strain rate in the investigated range, the diffused necking strain decreases from 0.152 to 0.120 and localized necking strain decreases from 0.245 to 0.137. Meanwhile, the difference of the two strains decreases form 0.096 to 0.017. Thus it can be concluded that strain rate has an influence on both necking strains during the deformation of QP980 steel sheet. Diffused and localized necking strains are determined by uniaxial tensile tests with the aid of DIC technique and the effect of strain rate on necking strains is evaluated. | DING Lei LIN Jianping MIN Junying PANG Zheng YE You | 2013 | Chinese Journal of Mechanical Engineering2013,26,3: | 11
BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays. | Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y. | 4 | Joint frequency,2-D DOA,and polarization estimation using parallel factor analysis显示文摘This paper proposes a new algorithm for joint frequency, two-dimensional (2-D) directions-of-arrival (DOA), and polarization estimation using parallel factor (PARAFAC) analysis model and cumulant. The proposed algorithm designs a new array configuration, and extends the PARAFAC analysis model from the common data-domain and subspace-domain to the cumulant one, and forms three-way arrays by using the three cumulant matrices obtained from the properly chosen dipole outputs, and analyzes the uniqueness of low-rank decomposition of the three-way arrays, and then jointly estimates the source parameters via the low-rank decomposition of the constructed PARAFAC model. In comparison with the conventional methods, the proposed method alleviates the aperture loss, and avoids pairing parameter. Finally, the simulation results are presented to validate the performance of the proposed method. | LIANG JunLi LIU Ding ZHANG JunYing | 2009 | Science in China(Series F)2009,52,10: | 4
Mitotic motor CENP-E cooperates with PRC1 in temporal control of central spindle assembly显示文摘Error-free cell division depends on the accurate assembly of the spindle midzone from dynamic spindle microtubules to ensure chromatid segregation during metaphase-anaphase transition. However, the mechanism underlying the key transition from the mitotic spindle to central spindle before anaphase onset remains elusive.Given the prevalence of chromosome instability phenotype in gastric tumorigenesis,we developed a strategy to model context-dependent cell division using a combination of light sheet microscope and 3D gastric organoids.Light sheet microscopic image analyses of 3D organoids showed that CENP-E inhibited cells undergoing aberrant metaphase-anaphase transition and exhibiting chromosome segregation errors during mitosis.Highresolution real-time imaging analyses of 2D cell culture revealed that CENP-E inhibited cells undergoing central spindle splitting and chromosome instability phenotype.Using biotinylated syntelin as an affinity matrix,we found that CENP-E forms a complex with PRC1 in mitotic cells.Chemical inhibition of CENP-E in metaphase by syntelin prevented accurate central spindle assembly by perturbing temporal assembly of PRC1 to the midzone.Thus,CENP-E-mediated PRC1 assembly to the central spindle constitutes a temporal switch to organize dynamic kinetochore microtubules into stable midzone arrays. These findings reveal a previously uncharacterized role of CENP-E in temporal control of central spindle assembly.Since CENP-E is absent from yeast,we reasoned that metazoans evolved an elaborate central spindle organization machinery to ensure accurate sister chromatid segregation during anaphase and cytokinesis. | Xu Liu Leilei Xu Junying Li Phil Y.Yao Wanjuan Wang Hazrat Ismail Haowei Wang Bryce Liao Zhihong Yang Tarsha Ward Ke Ruan Jianchun Zhang Quan Wu Ping He Xia Ding Dongmei Wang Chuanhai Fu Zhen Dou Feng Yan Wenwen Wang Xing Liu Xuebiao Yao | 2020 | Journal of Molecular Cell Biology2020,12,8: | 3
An iron-dependent form of non-canonical ferroptosis induced by labile iron显示文摘Ferroptosis is a recently identified iron-dependent form of nonapoptotic cell death characterized by reactive oxygen species(ROS) generation and lipid peroxidation. Here,we report a novel iron-dependent form of ferroptosis induced by labile iron and investigate the mechanism underlying this process. We find that labile iron-induced ferroptosis is distinct from canonical ferroptosis and is linked to the mitochondrial pathway.Specifically, the mitochondrial calcium uniporter mediates the ferroptosis induced by labile iron.Interestingly, cells undergoing labile iron-induced ferroptosis exhibit cytoplasmic features of oncosis and nuclear features of apoptosis.Furthermore, labile iron-induced ferroptosis involves a unique set of genes. Finally, labile ironinduced ferroptosis was observed in liver subjected to acute iron overload in vivo. Our study reveals a novel form of ferroptosis that may be implicated in diseases caused by acute injury. | Yanmeng Li Qin Ouyang Wei Chen Ke Liu Bei Zhang Jingyi Yao Song Zhang Junying Ding Min Cong Anjian Xu | 2023 | Science China(Life Sciences)2023,66,3: | 1

Raman Micro-Spectroscopic Study of Sulfate Ion in the System Na_2SO_4-H_2O显示文摘This work aims to quantify sulfate ion concentrations in the system Na_2SO_4-H_2O using Raman micro-spectroscopy.Raman spectra of sodium sulfate solutions with known concentrations were collected at ambient temperature(293 K) and in the 500 cm^(_1)-4000 cm^(-1) spectral region. The results indicate that the intensity of the SO_4^(2-) band increases with increasing concentrations of sulfate ion.A linear correlation was found between the concentration of SO_4^(2-)(c) and parameter I_1,which represents the ratio of the area of the SO_4^(2-) band to that of the O-H stretching band of water(A_s/A_w):I_1=-0.00102+0.01538 c.Furthermore,we deconvoluted the O-H stretching band of water(2800 cm^(-1)-3800 cm^(-1)) at 3232 and 3430 cm^(-1) into two sub-Gaussian bands, and then defined Raman intensity of the two sub-bands as A_(Bi)(3232 cm^(-1)) and A_(B2)(3430 cm^(-1)),defined the full width of half maximum(FWHM) of the two sub-bands as W_(B1)(3232 cm^(-1)) and W_(B2)(3430 cm^(-1)).A linear correlation between the concentration of SO_4^(2-)(c) and parameter I_2,which represents the ratio of Raman intensity of SO_4^(2-)(A_s)(in 981 cm^(-1)) to(A_(B1)+A_(B2)),was also established:I_2=-0.0111+0.3653 c.However,no correlations were found between concentration of SO_4^(2-)(c) and FWHM ratios, which includes the ratio of FWHM of SO_4^(2-)(W_s) to W_(B1) W_(B2) and W_(B1+B2)(the sum of W_(B1) and W_(B2)),suggesting that FWHM is not suitable for quantitative studies of sulfate solutions with Raman spectroscopy.A comparison of Raman spectroscopic studies of mixed Na_2SO_4 and NaCI solutions with a constant SO_4^(2-) concentration and variable CI^- concentrations suggest that the I\ parameter is affected by CI^-,whereas the I_2 parameter was not. Therefore, even if the solution is not purely Na_2SO_4-H_2O,SO_4^(2-) concentrations can still be calculated from the Raman spectra if the H_2O band is deconvoluted into two sub-bands,making this method potentially applicable to analysis of natural fluid inclusions. | ZHU Dongliang ZHU Ziang PAN Junyi DING Junying NI Pei | 2015 | Acta Geologica Sinica(English Edition)2015,89,3: | 1
Analysis and implementation of cross sync period underwater acoustical positioning techniques for underwater targets显示文摘The sampling rate of an underwater acoustical synchronous positioning system for the track of an underwater target will be decreasing during the process of positioning while the target moves away, resulting in the reduction of raw data and insufficient use of the processing ability of the positioning system. For a long time, this problem has remained unsolved, and it is even pushed forward recently because of the rapid development of modern electronic tech- niques. Based on the thorough study and investigation of the problem, we developed a new synchronous positioning technique called 'Cross Sync Period' underwater acoustical positioning. It can increase the sampling rate of an underwater acoustical positioning system for the track of an underwater target at long range significantly Besides, a new algorithm specially designed for the detection of the propagation time delay of the positioning signals called 'Mod-ulo Algorithm' was also developed, which makes the implementation of the 'cross sync period'underwater acoustical positioning technique easier and more efficient. These techniques have been successfully applied in a real positioning system. The system can position 5 underwater targets at the maximum range of 6 km simultaneously without any ambiguity of target distances with the working period of 0.4 s. The 'cross sync period' underwater acoustical positioning technique applied in the system was performed in lake and searun tests, and satisfactory re-sults were obtained. | XU Xinsheng DING Shiqi LI Haisen and HUI Junying(Harbin Engineerng University Harbin 150001) | 1996 | Chinese Journal of Acoustics1996,15,4: | 0
