1BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays.Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y. 2Engineering Ruthenium-Based Electrocatalysts for Effective Hydrogen Evolution Reaction显示文摘The investigation of highly effective,durable,and cost-effective electrocatalysts for the hydrogen evolution reaction(HER)is a prerequisite for the upcoming hydrogen energy society.To establish a new hydrogen energy system and gradually replace the traditional fossil-based energy,electrochemical water-splitting is considered the most promising,environmentally friendly,and efficient way to produce pure hydrogen.Compared with the commonly used platinum(Pt)-based catalysts,ruthenium(Ru)is expected to be a good alternative because of its similar hydrogen bonding energy,lower water decomposition barrier,and considerably lower price.Analyzing and revealing the HER mechanisms,as well as identifying a rational design of Ru-based HER catalysts with desirable activity and stability is indispensable.In this review,the research progress on HER electrocatalysts and the relevant describing parameters for HER performance are briefly introduced.Moreover,four major strategies to improve the performance of Ru-based electrocatalysts,including electronic effect modulation,support engineering,structure design,and maximum utilization(single atom)are discussed.Finally,the challenges,solutions and prospects are highlighted to prompt the practical applications of Rubased electrocatalysts for HER.Yingjie Yang Yanhui Yu Jing Li Qingrong Chen Yanlian Du Peng Rao Ruisong Li Chunman Jia Zhenye Kang Peilin Deng Yijun Shen Xinlong Tian 2021Nano-Micro Letters2021,13,10:5

5Microstructures and Mechanical Properties of a New Multi-functional 460 MPa Grade Construction Structural Steel显示文摘This article reports a new generation of Q460 multi-functional construction structural steel,which has high strength(yield strength larger than 460 MPa),excellent toughness(higher than 110 J/cm^(2) at-60 ℃),lower yield ratio(lower than 0.8),good fire resistance(yield strength at 600 ℃ larger than two-thirds of its room-temperature yield strength)and better corrosion resistance.The eff ects of finish cooling temperature(FCT)on the microstructure and properties were studied by scanning electron microscopy(SEM),transmission electron microscopy(TEM),emission electron probe micro-analysis(EPMA),electron backscattering diff raction(EBSD),tensile tester,impact tester,periodic immersion cycle acceleration test and electrochemical experiment.The results show that the strength and toughness are simultaneously improved by decreasing the FCT due to more lath-like bainite with large number of dislocations,refined martensite/austenite(M/A)with higher carbon concentration and increased high angle boundaries.In addition,the fire resistance of the newly developed Q460 steel is obviously better than the conventional one,which is mainly due to non-recrystallized lath-like bainite with high dislocation density at elevated temperature.The corrosion resistance of the new Q460 steel is also improved due to the addition of Cu and Cr.Zhenye Chen Zhangguo Lin Jianjun Qi Yang Feng Liqing Chen Guodong Wang 2022Acta Metallurgica Sinica(English Letters)2022,35,7:1
6Towards Realizing Sign Language-to-Speech Conversion by Combining Deep Learning and Statistical Parametric Speech Synthesis显示文摘This paper realizes a sign language-to-speech conversion system to solve the communication problem between healthy people and speech disorders. 30 kinds of different static sign languages are firstly recognized by combining the support vector machine (SVM) with a restricted Boltzmann machine (RBM) based regulation and a feedback fine-tuning of the deep model. The text of sign language is then obtained from the recognition results. A context-dependent label is generated from the recognized text of sign language by a text analyzer. Meanwhile,a hiddenMarkov model (HMM) basedMandarin-Tibetan bilingual speech synthesis system is developed by using speaker adaptive training.The Mandarin speech or Tibetan speech is then naturally synthesized by using context-dependent label generated from the recognized sign language. Tests show that the static sign language recognition rate of the designed system achieves 93.6%. 7Towards Realizing Mandarin-Tibetan Bi-lingual Emotional Speech Synthesis with Mandarin Emotional Training Corpus显示文摘This paper presents a method of hidden Markov model (HMM)-based Mandarin-Tibetan bi-lingual emotional speech synthesis by speaker adaptive training with a Mandarin emotional speech corpus.A one-speaker Tibetan neutral speech corpus, a multi-speaker Mandarin neutral speech corpus and a multi-speaker Mandarin emotional speech corpus are firstly employed to train a set of mixed language average acoustic models of target emotion by using speaker adaptive training.Then a one-speaker Mandarin neutral speech corpus or a one-speaker Tibetan neutral speech corpus is adopted to obtain a set of speaker dependent acoustic models of target emotion by using the speaker adap-tation transformation. The Mandarin emotional speech or the Tibetan emotional speech is finally synthesized from Mandarin speaker depen-dent acoustic models of target emotion or Tibetan speaker dependent acoustic models of target emotion. Subjective tests show that the aver-age emotional mean opinion score is 4.14 for Tibetan and 4.26 for Mandarin. The average mean opinion score is 4.16 for Tibetan and 4.28 for Mandarin. The average degradation opinion score is 4.28 for Tibetan and 4.24 for Mandarin.
