| 10 | A brain structural connectivity biomarker for autism spectrum disorder diagnosis in early childhood显示文摘Background:Autism spectrum disorder(ASD)is associated with altered brain development,but it is unclear which specific structural changes may serve as potential diagnostic markers,particularly in young children at the age when symptoms become fully estab-lished.Furthermore,such brain markers need to meet the requirements of precision medicine and be accurate in aiding diagnosis at an individual rather than only a group level.Objective:This study aimed to identify and model brain-wide differences in structural connectivity using diffusion tensor imaging(DTI)in young ASD and typically developing(TD)children.Methods:A discovery cohort including 93 ASD and 26 TD children and two independent validation cohorts including 12 ASD and 9 TD children from three different cities in China were included.Brain-wide(294 regions)structural connectivity was measured using DTI(fractional anisotropy,FA)together with symptom severity and cognitive development.A connection matrix was constructed for each child for comparisons between ASD and TD groups.Pattern classification was performed on the discovery dataset and the resulting model was tested on the two independent validation datasets.Results:Thirty-three structural connections showed increased FA in ASD compared to TD children and associated with both autistic symptom severity and impaired general cognitive development.The majority(29/33)involved the frontal lobe and comprised five different networks with functional relevance to default mode,motor control,social recognition,language and reward.Overall,clas-sification achieved very high accuracy of 96.77%in the discovery dataset,and 91.67%and 88.89%in the two independent validation datasets.Conclusions:Identified structural connectivity differences primarily involving the frontal cortex can very accurately distinguish novel individual ASD from TD children and may therefore represent a robust early brain biomarker which can address the requirements of precision medicine. | Xi Jiang Xiao-Jing Shou Zhongbo Zhao Yuzhong Chen Fan-Chao Meng Jiao Le Tian-Jia Song Xin-Jie Xu Weitong Guo Xiaoyan Ke Xiao-E Cai Weihua Zhao Juan Kou Ran Huo Ying Liu Hui-Shu Yuan Yan Xing Ji-Sheng Han Song-Ping Han Yun Li Hua Lai Lan Zhang Mei-Xiang Jia Jing Liu Xuan Liu Keith M.Kendrick Rong Zhang | 2023 | Psychoradiology2023,3,1: | 0 |
| 11 | Prediction of pandemic risk for animal-origin coronavirus using a deep learning method显示文摘Background:Coronaviruses can be isolated from bats,civets,pangolins,birds and other wild animals.As an animalorigin pathogen,coronavirus can cross species barrier and cause pandemic in humans.In this study,a deep learning model for early prediction of pandemic risk was proposed based on the sequences of viral genomes.Methods:A total of 3257 genomes were downloaded from the Coronavirus Genome Resource Library.We present a deep learning model of cross-species coronavirus infection that combines a bidirectional gated recurrent unit network with a one-dimensional convolution.The genome sequence of animal-origin coronavirus was directly input to extract features and predict pandemic risk.The best performances were explored with the use of pre-trained DNA vector and attention mechanism.The area under the receiver operating characteristic curve(AUROC)and the area under precision-recall curve(AUPR)were used to evaluate the predictive models.Results:The six specifc models achieved good performances for the corresponding virus groups(1 for AUROC and 1 for AUPR).The general model with pre-training vector and attention mechanism provided excellent predictions for all virus groups(1 for AUROC and 1 for AUPR)while those without pre-training vector or attention mechanism had obvi‑ously reduction of performance(about 5–25%).Re-training experiments showed that the general model has good capabilities of transfer learning(average for six groups:0.968 for AUROC and 0.942 for AUPR)and should give reason‑able prediction for potential pathogen of next pandemic.The artifcial negative data with the replacement of the coding region of the spike protein were also predicted correctly(100%accuracy).With the application of the Python programming language,an easy-to-use tool was created to implements our predictor.Conclusions:Robust deep learning model with pre-training vector and attention mechanism mastered the features from the whole genomes of animal-origin coronaviruses and could predict the risk of cross-species infection for early warning of next pandemic. | Zheng Kou Yi‑Fan Huang Ao Shen Saeed Kosari Xiang‑Rong Liu Xiao‑Li Qiang | 2021 | Infectious Diseases of Poverty2021,10,5: | 0 |

| 14 | Activation of the 5-hydroxytryptamine 4 receptor ameliorates tight junction barrier dysfunction in the colon of type 1 diabetic mice显示文摘Hyperglycemia drives dysfunction of the intestinal barrier.5-Hydroxytryptaine 4 receptor(5-HT4R)agonists have been considered therapeutics for constipation in clnic.However,the roles of 5-HT4R activation in mucosa should be fully realized.Here,we investigate the effects of 5-HT4R activation on diabetes-induced disruption of the tight junction(TJ)barrier in the colon.Not surprisingly,the TJ barrier in diabetic mice with or without 5-HT4R is tremendously destroyed,as indicated by increased serum fluorescein isothiocyanate(FITC)-dextran and decreased transepithelial electrical resistance(TER).Simultaneously,decreased expressions of TJ proteins are shown in both wild-type(WT)and 5-HT4R knockout(KO)mice with diabetes.Notably,chronic treatment with intraperitoneal injection of a 5-HT4R agonist in WT mice with diabetes repairs the TJ barrier and promotes TJ protein expressions,including occludin,claudin-1 and ZO-1,in the colon,whereas a 5-HT4R agonist does not improve TJ barrier function or TJ protein expressions in 5-HT4R KO mice with diabetes.Furthermore,stimulation of 5-HT4R inhibits diabetesinduced upregulation of myosin light chain kinase(MLCK),Rho-associated coiled coil protein kinase 1(ROCK1),and phosphorylated myosin light chain(p-MLC),which are key molecules that regulate TJ integrity,in the colonic mucosa of WT mice.However,such action induced by a 5-HT4R agonist is not observed in 5-HT4R KO mice with diabetes.These findings indicate that 5-HT4R activation may restore TJ integrity by inhibiting the expressions of MLCK,ROCK1 and p-MLC,improving epithelial barrier function in diabetes. | Shasha Hu Yueting Kou Xiaochen Liu Weifang Rong Hongxiu Han Guohua Zhang | 2023 | Acta Biochimica et Biophysica Sinica2023,55,12: | 0 |

By converse using the partially blind signature scheme, we build a simple fair e-payment protocol. In the protocol, two participants achieve the goals of exchanging their digital signatures from each other in a simple way. An ad- vantage of this scheme is that this approach does not require the intervention of the third party in any case.
