2A comparison between chemical and gas hypoxia as models of global ischemia in zebrafish (Danio rerio)显示文摘Background:Zebrafish models for neurovascular diseases offer new methods for elucidation of molecular pathways to tissue damage.External fertilization and high fecundity provide opportunities for transgenics and other forms of genetic manipulation that are more accessible than offered by mammalian models of disease.Furthermore,behavioral analyses of zebrafish allow for connection of molecular pathways to organismal outputs such as locomotion,learning,and memory.Unfortunately,a zebrafish model of hypoxia-ischemia has been slow to catch on,possibly due to hypoxia exposure protocols that are challenging to reproduce and result in high mortality.Methods:In this study,we have introduced a predictable and simple method of hypoxia induction,the addition of sodium sulfite to aquarium water.The effects of this treatment on zebrafish locomotion were compared to those of zebrafish exposed to hypoxia induced by nitrogen gas bubbling,a method used in previous reports.Results:We found that hypoxia induced by sodium sulfite significantly impaired locomotion in the hours following treatment,and its effects did not differ from those caused by nitrogen gas hypoxia.Conclusion:These results indicate that hypoxia by sodium sulfite represents an effective and easily reproducible method for the study of hypoxia-ischemia in zebrafish.Kaitlyn M.Marino Emani R.Silva James A.Windelborn 2020Animal Models and Experimental Medicine2020,3,3:3
3Bile acids and sphingolipids in non-alcoholic fatty liver disease显示文摘Non-alcoholic fatty liver disease (NAFLD) is one of the fastest-growing diseases, and its global prevalence is estimated to increase >50% by 2030. NAFLD is comorbid with metabolic syndrome, obesity, type 2 diabetes, and insulin resistance. Despite extensive research efforts, there are no pharmacologic or biological therapeutics for the treatment of NAFLD. Bile acids and sphingolipids are well-characterized signaling molecules. Over the last few decades, researchers have uncovered potential mechanisms by which bile acids and sphingolipids regulate hepatic lipid metabolism. Dysregulation of bile acid and sphingolipid metabolism has been linked to steatosis, inflammation, and fibrosis in patients with NAFLD. This clinical observation has been recapitulated in animal models, which are well-accepted by experts in the hepatology field. Recent transcriptomic and lipidomic studies also show that sphingolipids are important players in the pathogenesis of NAFLD. Moreover, the identification of bile acids as activators of sphingolipid-mediated signaling pathways established a novel theory for bile acid and sphingolipid biology. In this review, we summarize the recent advances in the understanding of bile acid and sphingolipid-mediated signaling pathways as potential contributors to NAFLD. 14Complications in the spine associated with type 2 diabetes:The role of advanced glycation end-products显示文摘Type 2 diabetes mellitus(T2D)is an increasingly prevalent disease with numerous comorbidities including many in the spine.T2D is strongly linked with vertebral fractures,intervertebral disc(IVD)degeneration,and severe chronic spinal pain.Yet the causative mechanism for these musculoskeletal impairments remains unclear.The chronic hyperglycemic state in T2D promotes the formation of advanced glycation end-products(AGEs)in tissues,and the accumulation of AGEs may play a role in musculoskeletal complications by modifying the extracellular matrix,impairing cellular homeostasis,and perpetuating an inflammatory cascade via its receptor(RAGE).The AGE and RAGE associated alterations in extracellular matrix composition and morphological features of the vertebral bodies and IVDs are likely contributors to the incidence and severity of spinal pathologies in T2D.This review will broadly examine the effects of AGEs on tissues in the spine in the context of T2D,with an emphasis on the changes in the vertebrae and the IVD.Along with the clinical and epidemiological findings,we will provide an overview of preclinical rodent models of T2D that exhibit deficits in the IVD and vertebral bone.Elucidating the role of AGEs and RAGE will be crucial for understanding the disease mechanisms and translation therapies of musculoskeletal pathologies in T2D.Kaitlyn Broz Remy E.Walk Simon Y.Tang 2021Medicine in Novel Technology and Devices2021,,3:1

16Synthesis of new two-dimensional titanium carbonitride Ti_(2)C_(0.5)N_(0.5)T_(x)MXene and its performance as an electrode material for sodium-ion battery显示文摘Two-dimensional(2D)layered transition metal carbides/nitrides,called MXenes,are attractive alternative electrode materials for electrochemical energy storage.Owing to their metallic electrical conductivity and low ion dif-fusion barrier,MXenes are promising anode materials for sodium-ion batteries(SIBs).Herein,we report on a new 2D carbonitride MXene,viz.,Ti_(2)C_(0.5)N_(0.5)T_(x)(T_(x) stands for surface terminations),and the only second carbonitride after Ti_(3) CNT_(x) so far.A new type of in situ HF(HCl/KF)etching condition was employed to synthesize multilayer Ti_(2)C_(0.5)N_(0.5)T_(x) powders from Ti_(2)AlC_(0.5)N_(0.5).Spontaneous intercalation of tetramethylammonium followed by sonication in water allowed for large-scale delamination of this new titanium carbonitride into 2D sheets.Multilayer Ti_(2)C_(0.5)N_(0.5)T_(x) powders showed higher specific capac-ities and larger electroactive surface area than those of Ti_(2)CT_(x) powders.Multi-layer Ti_(2)C_(0.5)N_(0.5)T_(x) powders show a specific capacity of 182 mAh g^(-1) at 20 mA g^(-1),the highest among all reported MXene electrodes as SIBs with excellent cycling stability.Kun Liang Anika Tabassum Ahmad Majed Chaochao Dun Feipeng Yang Jinghua Guo Kaitlyn Prenger Jeffrey JUrban Michael Naguib 2021InfoMat2021,3,12:1

20Effects of personalized exercise prescriptions and social media delivered through mobile health on cancer survivors’physical activity and quality of life显示文摘Purpose This study aimed to examine the effects of a multi-component mobile health intervention(wearable,apps,and social media)on cancer survivors’(CS')physical activity(PA),quality of life,and PA determinants compared to exercise prescription only,social media only,and attention control conditions.Methods A total of 126 CS(age=60.37±7.41 years,mean±SD)were recruited from the United States.The study duration was 6 months and participants were randomly placed into 4 groups.All participants received a Fitbit tracker and were instructed to install its companion app to monitor their daily PA.They(1)received previously established weekly personalized exercise prescriptions via email,(2)received weekly Facebook health education and interacted with one another,(3)received both Conditions 1 and 2,or(4)were part of the control condition,meaning they adopted usual care.CS PA daily steps,quality of life(i.e.,physical health and mental health),and PA determinants(e.g.,self-efficacy,social support)were measured at baseline,3 months,and 6 months.Results The final sample size included 123 CS.The results revealed only the multi-component condition had greater improvements in PA daily steps than the control condition post-intervention(95%confidence interval(95%CI):368–2951;p<0.05).Similarly,those in the multi-component condition had significantly greater increased physical health than the control condition(95%CI:–0.41 to–0.01;p<0.05)over time.In addition,the social media condition had significantly greater increased perceived social support than the control condition(95%CI:0.01–0.93;p<0.05).No other significant differences on outcomes were identified.Conclusion The study findings suggest that the implementation of a multi-component mobile health intervention had positive effects on CS PA steps and physical health.Also,offering social media intervention has the potential to improve CS perceived social support.Zan Gao Suryeon Ryu Wanjiang Zhou Kaitlyn Adams Mohamed Hassan Rui Zhang Anne Blaes Julian Wolfson Ju Sun 2023Journal of Sport and Health Science2023,12,6:0
