1The Coix Genome Provides Insights into Panicoideae Evolution and Papery Hull Domestication显示文摘Coix is a grass crop domesticated as early as the Neolithic era.It is still widely cultivated for both highly nutritional food and medicinal use.However,the genetic study and breeding of this crop are hindered by the lack of a sequenced genome.Here,we report de novo sequencing and assembly of the 1619-Mb genome of Coix,and annotation of 75.39%repeats and 39629 protein-coding genes.Comparative genomics analysis showed that Coix is more closely related to sorghum than maize,but intriguingly only Coix and maize had a recent genome duplication event,which was not detected in sorghum.We further constructed a genetic map and mapped several important traits,especially the strength of hull.Selection of papery hull(thin:easy dehulling)from the stony hull(thick:difficult dehulling)in wild progenitors was a key step in Coix domestication.The papery hull makes seed easier to process and germinate.Anatomic and global transcriptome analysis revealed that the papery hull is a result of inhibition of cell division and wall biogenesis.We also successfully demonstrated that seed hull pressure resistance is controlled by two major quantitative trait loci(QTLs),which are associated with hull thickness and color,respectively.The two QTLs were further fine mapped within intervals of 250 kb and 146 kb,respectively.These resources provide a platform for evolutionary studies and will facilitate molecular breeding of this important crop.Chao Guo Yanan Wang Aiguo Yang Jun He Chaowen Xiao Shanhua Lv Fengming Han Yibing Yuan Yuan Yuan Xiaolong Dong Juan Guo YawenYang-Hailan Liu Ningzhi Zuo Yaxi Hu Kangxu Zhao Zhengbo Jiang Xing Wang Tingting Jiang Yaou Sherf Moju Cao Yuan Wang Zhaobo Long Tingzhao Rong Luqi Huang Shufeng Zhou 2020Molecular Plant2020,13,2:11
2Generation of the regulatory protein rtTA transgenic mice显示文摘AIM: To translate Tet-on system into a conditional mouse model, in which hepatitis B or C virus (HBV or HCV) gene could be spatiotemporally expressed to overcome 'immune tolerance' formed during the embryonic development and 'immune escape' against hepatitis virus antigen(s), an effector mouse, carrying the reverse tetracycline-responsive transcriptional activator (rtTA) gene under the tight control of liver-specific human apoE promoter, is required to be generated. METHODS: To address this end, rtTA fragment amplified by PCR was effectively inserted into the vector of pLiv.7 containing apoE promoter to create the rtTA expressing vector, I.e., pApoE-rtTA. ApoE-rtTA transgenic fragment (-6.9 kb) released from pApoE-rtTA was transferred into mice by pronucleus injection, followed by obtaining one transgene (+) founder animal from microinjection through PCR and Southern blot analysis.RESULTS: rtTA transgene which could be transmitted to subsequent generation (F1) derived from founder was expressed in a liver-specific fashion. 3Temporal Control of Cre Recombinase-mediated in Vitro DNA Recombination by Tet-on Gene Expression System显示文摘Conditional gene expression and gene deletion are important experimental approaches for examining the functions of particular gene products in mouse models. These strategies exploiting Cre-mediated site-specific DNA recombination have been incorporated into transgenic and gene-targeting procedures to allow in vivo manipulation of DNA in embryonic stem cells (ES cells) or living animals. The Cre/lox P system has become widely used in conditional gene targeting, conditional gene repair and activation, inducible chromosome translocation, and chromosome engineering. In this project, we have employed the universal transgenic system and the liver-specific promoter system for tightly temporal and liver-specific control of Cre gene expression in mice that (1) integrates the advantages of the Tet-on gene expression system and Cre/lox P site-mediated gene activation, and (2) simplifies the scheme of animal crosses through a combination of two control elements in a single transgene. A liver-specific apoE promoter was inserted into the promoter cloning site upstream of the rtTA cassette of pCore construct to generate the transgene construct pApoErtTA-tetO-Cre, followed by demonstrating stringent regulation of doxycycline (Dox)-induced Cre-mediated recombination in the lox P-flanked transcription STOP cassette-modified BEL-7402 cells. 4Study on Predicting Coal Spontaneous Combustion in Qingshan Mine显示文摘Based on the measurement experiment of oxidation gas products in the process of temperature-up, with the intention of improving the sensitivity of each sign gas for predicting coal spontaneous combustion and operatingness, this paper has researched carefully the change curves which show oxidation gas products of the coal samples of Qingshan mine changing with coal temperature. According to the actual condition of mine, the initial value k is led into, which is the ratio of the series of sums of carbon monoxide and carbon dioxide in the process of temperature-up to the initial value of carbon dioxide, and it raises the quality and sensitivity of forecast curve of sign gas and auxiliary index, and determines the forecast curve and the corresponding fitting curve of the sign gas for predicting coal spontaneous combustion of Yongzi coal of Qingshan mine. So the experiment data can be provided to forecast and predict the mine fires caused by coal spontaneous combustion. 5Temporal and Tight Hepatitis C Virus Gene Activation in Cultured Human Hepatoma Cells Mediated by a Cell-Permeable Cre Recombinase显示文摘Conditional gene expression has greatly facilitated the examination of the functions of particular gene products. Using the Cre/lox P switching expression system, we plan to develop efficient conditional transgeneactivation of hepatitis C virus core protein (HCV-C) cDNA (nucleotide 342–914) in the transgenic mice to overcome “immune tolerance” formed during the embryonic period and “immune escape” against hepatitis virus antigen in our project. To use this system in vivo, the dormant transgenic construct, i.e., pApoE-SCS-EGFP-HCV-C, was generated using techniques of standard molecular biology. The liver- specific human apoE promoter was here used to target expression of genes of interest (EGFP and HCV-C) to murine liver. Prior to generating the transgenic mice, the availability of Cre/lox P system and construct functionality were successfully verified by a cell-free recombination system and via checking the expression of EGFP and HCV-C in the human hepatoma cells at the mRNA and protein levels. 6Hydrocarbons in igneous rock of Brazil:A review显示文摘Igneous rocks are widely developed in many hydrocarbon-bearing sedimentary basins in Brazil,and some igneous rocks play positive roles in the oil and gas accumulation process.But so far,no industrial oil or gas flow is discovered in igneous rocks in Brazilian onshore basins.At present,in some literatures published by some researchers,cases of igneous reservoirs in Brazil are inaccurate.The lithology of reservoirs in the Igarape Cuia oil and gas field,the oil and gas field of Urucu and the Barra Bonita gasfield,were previously thought to be volcanic rocks,but now are clastic rocks according to studies,and the igneous rocks in these three oil and gas field can only be acted as indirect and direct cap rocks.Furthermore,igneous rock reservoirs in the Campos Basin and Santos Basin in the Brazilian offshore are briefly analyzed.The representative Badejo oilfield in the Campos Basin has igneous rock reservoirs dominated by basalts which are intercalated with thin layers of volcaniclastic and sedimentary rocks,the storage space are vesicules,fractures and vugular pores,and the porosity of hydrocarbon bearing interval basalt reservoirs ranges from 10%to 15%.In addition,for the Santos Basin,igneous rocks in this basin are dominated by basic rocks,some alkaline intrusive rocks develop good micro spaces,mainly crystal moldic pores,inter-and intra-crystal dissolution pores and fractures,better reservoir intervals are often concentrated such as at sections near to unconformity surfaces and can have porosity of 6%e9%or higher;local pre-salt igneous rock reservoirs in the Santos Basin have the necessary conditions for hydrocarbon accumulation and it will be a new target layer for hydrocarbon exploration.Kangxu Ren Junfeng Zhao Qian Liu Jian Zhao 2020Petroleum Research2020,5,3:1
7A real-time sensing system based on triboelectric nanogenerator for dynamic response of bridges显示文摘The strain of bridges under traffic loads is time-varying and of small amplitude(~10^(-6)),which is a type of cumulative response and needs long-term continuous monitoring.To precisely capture the time-varying responses,a dynamic strain triboelectric nanogenerator(TENG)sensor with superior response capability,sensitivity,self-powered,and long-term stability is proposed in this paper.An analytical correlation between the structural strain response signal and the detected electrical signal is established for long-term continuous quantitative strain measurements based on the principles of contact electrification and electrostatic induction.A series of experiments are conducted to investigate the output performance of the proposed lateral-sliding mode TENG sensors.The results reveal that,with the loading condition with frequencies lower than 10 Hz,the time-varying strain responses of the steel bridge within the range of 3 to 150 microstrains can also be detected with high precision of 0.1 microstrains.And it achieves long-term stability after 10000 loading cycles compared with commercial sensors.The proposed novel sensing theory and method based on TENG technology can be applied as a new alternative approach for monitoring realtime structural strain information quantitatively with general applicability and feasibility for bridges.ZHANG He HUANG KangXu ZHOU YuHui SUN LiangFeng ZHANG ZhiCheng LUO JiKui 2022Science China(Technological Sciences)2022,65,11:1
